Abstract
Description
Triterpene derivatives or pharmaceutically acceptable salts or solvates thereof and antibacterial uses thereof
[0001] Related applications
[0002] This application claims priority to Chinese patent application No. 202311237384.4, filed on September 22, 2023, entitled “Application of triterpenoid compounds or pharmaceutically acceptable salts thereof in the preparation of antibacterial compositions,” the entire text of which is hereby incorporated by reference. Technical Field
[0003] The present application relates to the field of antibacterial technology, and in particular to triterpene derivatives or pharmaceutically acceptable salts or solvates thereof and their antibacterial uses. Background Art
[0004] With the widespread use of antibiotics, especially their overuse, bacterial resistance to commonly used antibiotics has continued to increase, and the continuous emergence of drug-resistant bacteria has reduced the effectiveness of antibiotics. [0210] The triterpene derivatives of formula (I) can be isolated and purified from the natural plant Cyclocarya paliurus (e.g., compounds C1-C10), or can be obtained by structural optimization of the isolated triterpene derivatives (e.g., compounds C11-C19). The triterpene derivatives of formula (I) are active compounds with a broader antibacterial spectrum, higher efficacy, better drugability, and better safety, and can be used in the preparation of antibacterial drugs and antibacterial agents, and can be applied to the treatment of bacterial infections (including but not limited to drug-resistant bacterial infections). Pharmaceutically acceptable salts of the triterpene derivatives of formula (I) can be prepared using conventional salt-forming methods in the art, such as, but not limited to, cooling crystallization, antisolvent methods, and the like.
[0211] Traditionally, Cyclocarya paliurus is believed to have the effects of clearing heat and relieving summer heat, lowering blood sugar, and lowering blood pressure. Recent research has focused on the hypoglycemic pharmacology and the underlying substance of its efficacy. In this application, a specific separation and purification method is used to obtain triterpene derivatives from Cyclocarya paliurus leaf extract, and further derivatize new triterpene derivatives. The separation and purification method used in this application has a higher extraction efficiency.
[0212] After extensive experimental exploration, the applicant of this application unexpectedly discovered that triterpene derivatives of formula (I) have strong killing effects on a variety of common pathogenic bacteria, such as Staphylococcus aureus, Staphylococcus epidermidis, Staphylococcus saprophyticus, Streptococcus pneumoniae, alpha-hemolytic Streptococcus, beta-hemolytic Streptococcus, Escherichia coli, Enterococcus faecalis, vancomycin-resistant Enterococci, Pseudomonas aeruginosa, Mycobacterium abscessus, Klebsiella pneumoniae, and Helicobacter pylori. Triterpene derivatives of formula (I) also have high bactericidal activity against some antibiotic-resistant strains, such as methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Enterococci, and linezolid-resistant Enterococci.
[0213] The minimum inhibitory concentration (MIC) of the triterpene derivative of formula (I) for one or more of the above-mentioned bacteria can reach less than 150 μg / mL, further can reach less than or equal to 75 μg / mL, further can reach less than or equal to 37.5 μg / mL, and further can reach less than or equal to 9.375 μg / mL.
[0214] As a non-limiting example, for example, the MIC of compound C8 against alpha-hemolytic streptococci, beta-hemolytic streptococci, and Streptococcus pneumoniae may meet ≤9.375 μg / mL, the MIC against Staphylococcus aureus, MRSA, Staphylococcus epidermidis, and Escherichia coli may meet ≤18.75 μg / mL, the MIC against Staphylococcus saprophyticus may meet ≤37.5 μg / mL, the MIC against Enterococcus faecalis, vancomycin-resistant enterococci, and linezolid-resistant enterococci may meet ≤75 μg / mL, and the MIC against Pseudomonas aeruginosa, Mycobacterium abscessus, Klebsiella pneumoniae, and Helicobacter pylori may meet ≤150 μg / mL.
[0215] In some embodiments, Compound C8 and Compound C11 can be used to target Streptococci (such as group A / B hemolytic Streptococci and Streptococcus pneumoniae) and Staphylococci (such as methicillin-resistant / sensitive Staphylococcus aureus and Staphylococcus epidermidis). They can be used to treat, but are not limited to, skin and soft tissue infections, respiratory tract infections, heart valve infections, and bone and joint infections. In some embodiments, Compound C11 has a slightly stronger killing effect on Staphylococci than Compound C8.
[0216] In some embodiments, compound C14 has a killing effect on streptococci (such as alpha / beta hemolytic streptococci and streptococci pneumoniae) and staphylococci (such as methicillin-resistant / sensitive Staphylococcus aureus and Staphylococcus epidermidis), and can also have a killing effect on Bacillus (Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Mycobacterium abscessus, Helicobacter pylori, etc.). Compound C14 has a stronger killing effect on E. coli than compounds C8 and C11. In addition, compound C14 also has a certain killing effect on Enterococcus faecalis, vancomycin-resistant enterococci, and linezolid-resistant enterococci. Compound C14 can also be used for skin and soft tissue infections, respiratory tract infections, heart valve infections, and bone and joint infections. In addition, it may also be suitable for digestive tract infections, bacteremia, etc.
[0217] In some embodiments, compound C15 has a good effect on Streptococcus, Staphylococcus aureus, Bacillus, and Enterococcus. Based on the above application examples of compound C14, compound C15 can also be used for urinary tract infections.
[0218] In some embodiments, relative to Lactobacillus acidophilus, the triterpene derivatives of formula (I) exhibit certain selectivity in their antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant enterococci and linezolid-resistant enterococci, Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Staphylococcus saprophyticus, Streptococcus pneumoniae, alpha-hemolytic Streptococcus, beta-hemolytic Streptococcus, Enterococcus faecalis, Pseudomonas aeruginosa, Klebsiella pneumoniae, Helicobacter pylori and Mycobacterium abscessus.
[0219] In this application, MIC (Minimal Inhibitory Concentration) can be used to characterize antibacterial activity. The lower the MIC value, the lower the effective antibacterial concentration and the stronger the antibacterial efficacy. The triterpene derivatives of formula (I) in this application can achieve antibacterial effects by targeting FtsZ.
[0220] The bacterial division Z ring, a ring-shaped structure formed by the filamentous temperature-sensitive mutant Z (FtsZ) protein at the site of cell division, is essential for bacterial survival and proliferation. Because it is present in most bacterial species, highly conserved, and crucial for cell division, cell wall formation, cell morphology, and structural stability, it is a potential target for new antibiotics. Although a few compounds targeting FtsZ have been successfully synthesized, such as PC190723, no antibacterial drugs targeting FtsZ have been marketed globally due to poor drugability and high toxicity.
[0221] The triterpene derivatives of formula (I) provided in the present application target FtsZ, occupy the binding domain of FtsZ and its active substrate guanosine triphosphate (GTP), hinder the binding of FtsZ and GTP, lead to GTP hydrolysis disorder, block the formation of Z ring, and make the cells after nucleic acid replication unable to complete division, and eventually the bacteria rupture and die. Therefore, the triterpene derivatives of formula (I) that can bind to the GTP site have antibacterial effects, and the antibacterial efficacy is positively correlated with its binding force. As a drug target that has not yet been clinically applied, bacteria have no resistance to drugs targeting FtsZ. The triterpene derivatives of formula (I) have the effect of treating drug-resistant bacterial infections.
[0222] Mammalian cells do not secrete FtsZ. Therefore, it is believed that the triterpene derivatives of formula (I) have no known toxic side effects in humans. In addition, the triterpene derivatives of formula (I) are derived from natural plants that have both edible and medicinal properties, and have the advantage of good drugability.
[0223] The top and bottom structures of the FtsZ protein have significant geometric complementarity. In the multimeric state, a wide hydrophobic interaction region is formed between the bottom of the FtsZ protein at the top (C-terminal domain) and the top of the FtsZ protein at the bottom (N-terminal domain). The GTP binding domain in the FtsZ structure is composed of 85 carbon atoms, 24 nitrogen atoms and 27 oxygen atoms, forming a pocket-like structure. The pocket volume is Has a surface area of Depth up to Theoretically, FtsZ has 17 hydrogen bond donor units and 34 acceptor units, with 20 hydrophobic binding sites. The hydrophilic / hydrophobic ratio is 0.28. A schematic diagram of the FtsZ binding pocket can be found in Figure 3, where the light-colored portion represents the binding pocket region.
[0224] Molecular docking revealed that the binding site of the triterpene derivative of formula (I) may be in the GTP binding domain between the N-terminus and the H7 helix of FtsZ. Its binding mechanism may be similar to the reported crystal structure of the FtsZ-GTP complex (PDB ID: 5MN5).
[0225] Taking compounds C8 and C11 as examples, Figures 1A-1B and 2A-2B illustrate the binding modes of compounds C8 and C11 with FtsZ, respectively. Figure 1A is a visualization of the docking of compound C8 with FtsZ, and Figure 1B is a surface diagram of the interaction between compound C8 and FtsZ. Figure 2A is a visualization of the docking of compound C11 with FtsZ, and Figure 2B is a surface diagram of the interaction between compound C11 and FtsZ.
[0226] See Figures 1A-1B and Table 4. Molecular docking revealed that compound C8 deeply inserts into the GTP-binding pocket of FtsZ, forming hydrogen bonds with amino acid residues Gly108, Thy109, Glu139, and Asn166. It also binds to amino acid residues Asp46, Pro135, and Phe183 via hydrophobic interactions, and to amino acid residue Arg143 via ionic bonds. Analysis revealed a docking index of -5.593 for compound C8 with FtsZ, comparable to the docking index of -5.612 for the natural ligand GTP. It is speculated that compound C8 competitively binds to the GTP-binding site of FtsZ, resulting in the loss of GTPase activity and the inability of FtsZ to catalyze Z-ring formation, ultimately leading to bacterial death.
[0227] PC190723 is the most studied compound targeting FtsZ. Its binding site is located in the gap formed by the H7 helix and the C-terminal domain, rather than directly interacting with the GTP-binding domain of FtsZ. However, upon binding to FtsZ, compounds C8 and PC19023 inhibited FtsZ's GTPase activity. The triterpene derivatives of formula (Ib) and formula (Ic) differ from the triterpene derivative of formula (Ia) in that the triterpene derivatives of formula (Ib) and formula (Ic) are ring-opened tetracyclic triterpene derivatives, while the triterpene derivative of formula (Ia) is a tetracyclic triterpene derivative with a complete structure. Among the compounds contained in the plant Cyclocarya paliurus, ring-opened tetracyclic triterpene derivatives typically have carboxyl groups, which contain active hydrogens and are prone to hydrogen bonding, potentially helping to stabilize this binding and achieve a better antibacterial effect.
[0228] It has been verified that the triterpene derivatives of formula (I) provided in the present application have good FtsZ binding ability, which can be seen in the following Examples.
[0229] In yet another aspect of the present application, a method for preparing a triterpene derivative of formula (I) is provided.

[0234] Method 2: Based on the triterpene derivative obtained in Method 1, triterpene derivatives with other structures can be obtained through conventional chemical reactions such as esterification, addition, acylation, and alkylation. If necessary, a suitable hydroxyl protecting agent can be used to protect the hydroxyl group during the reaction.
[0235] The above-mentioned reagents for group modification of the extract compounds can be selected from one or more of the following: acetic anhydride, benzoyl chloride, trifluoroacetyl ester, benzyl ether, methanol, ethanol, propanol, isopropanol, n-butanol, tert-butanol, sec-butanol, isobutanol, cyclopropanol, cyclobutanol, cyclopentanol, cyclohexanol, acetyl chloride, dimethyl sulfate and benzyl chloride.
[0236] The triterpene derivatives of formula (I) can also be prepared by reacting other triterpene compounds or triterpene derivatives.
[0237] The above-mentioned compounds provided in this application can be derived from natural plants. They not only have high antibacterial efficacy and a broad antibacterial spectrum, but also have obvious inhibitory and killing activities against a variety of antibiotic-resistant strains. Elute with dichloromethane-methanol (90%-0%) gradient, that is, gradient elution from dichloromethane: methanol volume ratio of 90%:10% to 0%:100%, the elution volume is 3 column volumes, and 6 fractions (Fr.1-6) are obtained. Fr.1 (55 g) was applied to an 800 g silica gel column and eluted with a gradient of petroleum ether-ethyl acetate (100%-20%, i.e., from a petroleum ether:ethyl acetate volume ratio of 100%:0% to 20%:80%) over 3 column volumes to obtain five fractions (Fr.1a-e). Fr.1b (2 g) was repeatedly subjected to C18 preparative chromatography with a gradient of methanol-water (20%-100%, i.e., from a methanol:water volume ratio of 20%:80% to 100%:0%) over 4 column volumes to obtain compounds C5 (208 mg) and C6 (112 mg). Fr.2 (62 g) was applied to an 800 g silica gel column using a gradient elution of petroleum ether-ethyl acetate (80%-20%, i.e., from a petroleum ether:ethyl acetate volume ratio of 80%:20% to 20%:80%) over 3 column volumes to yield five fractions (Fr.2a-e). Fr.2b (3 g) was repeatedly subjected to C18 preparative chromatography using a gradient elution of methanol-water (20%-100%, i.e., from a methanol:water volume ratio of 20%:80% to 100%:0%) over 4 column volumes to yield compounds C7 (230 mg) and C8 (183 mg). Fr.3 (80 g) was loaded onto an 800 g silica gel column and gradient eluted with petroleum ether-ethyl acetate (80%-20%, i.e., from a petroleum ether:ethyl acetate volume ratio of 80%:20% to 20%:80%) over a 3 column volume to give four fractions (Fr.3a-d). Fr.2c (2 g) was repeatedly subjected to C18 preparative chromatography using methanol-water (20%-100%, i.e., from a methanol:water volume ratio of 20%:80% to 100%:0%) with a gradient elution volume of 4 column volumes to give compound C3 (106 mg), compound C4 (115 mg), compound C9 (95 mg) and compound C10 (150 mg).Fr.4 (30 g) was placed on a 300 g silica gel column and gradient eluted with petroleum ether-ethyl acetate (70%-10%, i.e., from a petroleum ether:ethyl acetate volume ratio of 70%:30% to 10%:90%) with an elution volume of 3 column volumes to obtain 4 fractions (Fr.4a~d). The fractions were repeatedly subjected to preparative chromatography with methanol-water (20%-100%, i.e., from a methanol:water volume ratio of 20%:80% to 100%:0%) with a gradient elution volume of 4 column volumes to obtain compound C1 (103 mg) and compound C2 (120 mg).

[0346] (VIII) Synthesis and Preparation of Compound C17
[0347] Trifluoroacetic anhydride (1.09 mL of a 0.5 M solution in dichloromethane, 0.545 mmol) was added dropwise to a solution of C2 (0.519 mmol) and pyridine (0.419 mL, 5.18 mmol) in anhydrous dichloromethane (3.46 mL) at -78°C. The bath was slowly warmed to 0°C over 90 minutes, and the reaction was stirred at 0°C for an additional 20 minutes. The reaction was then quenched with 1N aqueous HCl. The reaction was diluted with n-hexane, stirred vigorously, and warmed to room temperature. The aqueous phase was extracted three times with n-hexane, and the organic layer was washed with saturated brine, dried over Na2SO4, and concentrated under reduced pressure. The product was purified by flash column chromatography on silica gel (n-hexane / ethyl acetate mixed solvent, 100:1 to 10:1 by volume) to obtain 25 mg of the product (compound C17) in a 58.1% yield.

Lactobacillus acidophilus ATCC 4356 was cultured in MRS medium. Enterococci faecalis, vancomycin-resistant Enterococci, and linezolid-resistant Enterococci were cultured in trypticase soy broth supplemented with 0.6% yeast extract. Culture conditions: 37°C, shaking at 150 rpm. Helicobacter pylori was cultured in H. pylori medium at 37°C in an incubator with 5% O2 and 5% CO2. Mycobacterium abscessus was cultured in Roche's basal medium at 30°C.
[0372] (III) Bactericidal effect test of triterpenoid derivatives
[0373] Preparation of test bacteria: Inoculate each bacterium into 5 mL of the corresponding culture medium, culture overnight at 37°C in a shaking incubator, adjust the turbidity of the bacterial solution to 0.5 McFarland units, and then dilute the bacterial solution 100 times for later use. The final bacterial concentration is 5×10 5 cfu / mL.
[0374] Preparation of triterpene derivative mother solution: Accurately weigh 7.5 mg of triterpene derivative powder and dissolve it in 1 mL of DMSO to obtain a 7.5 mg / mL triterpene derivative mother solution.
[0375] Preparation of a triterpene derivative drug concentration gradient: Dilute the drug stock solution with the corresponding culture medium to an initial triterpene derivative concentration of 300 μg / mL. Add 100 μL of this drug solution to wells 1 and 2 of a 96-well plate. From well 2 onward, add 100 μL of culture medium to perform serial dilutions up to well 9. Set well 10 as the culture medium control. From wells 1 to 9, the concentrations of the triterpene derivative are 150 μg / mL, 75 μg / mL, 37.5 μg / mL, 18.75 μg / mL, 9.375 μg / mL, 4.6875 μg / mL, 2.34375 μg / mL, 1.171875 μg / mL, and 0.5859375 μg / mL, respectively.
[0376] To test for antibacterial activity, add 100 μL of the bacterial solution prepared above to the wells containing each triterpene derivative concentration and the culture medium control wells. After thorough mixing, incubate overnight at 37°C in a humidified chamber. The lowest drug concentration resulting in no bacterial growth is determined as the minimum inhibitory concentration (MIC).
[0377] If “no colony growth” could not be achieved at any of the above concentrations, the MIC was recorded as “>150 μg / mL”.
[0378] MIC can be used to characterize antibacterial activity. The lower the MIC value, the lower the effective antibacterial concentration and the stronger the antibacterial efficacy.
[0379] The test results can be found in Table 2-3.
[0380] (4) Test results
[0381] As can be seen from Tables 2 to 4, these triterpene derivatives not only have a killing effect on the standard strain of common Staphylococcus aureus (ATCC25923), but also have a significant killing effect on various MRSA strains. In addition, triterpene derivatives also have a good killing effect on Staphylococcus epidermidis, Staphylococcus saprophyticus, Streptococcus pneumoniae, alpha-hemolytic Streptococcus, and beta-hemolytic Streptococcus. Some triterpene derivatives also have a good killing effect on Escherichia coli, Enterococcus faecalis, vancomycin-resistant Enterococcus, linezolid-resistant Enterococcus, Pseudomonas aeruginosa, Klebsiella pneumoniae, Helicobacter pylori, and Mycobacterium abscessus. At the same time, each triterpene derivative shows certain selectivity to different strains, for example, the MIC for Lactobacillus acidophilus is higher. When the drug containing these triterpene derivatives is delivered in an oral manner, the impact on the normal flora of the gastrointestinal tract is limited.
[0382] Furthermore, Tables 2 through 4 show that while C0 and C4-C15 share similar core structures, different R1, R4, and R8 components enable some compounds to achieve enhanced killing effects against key bacterial strains. For example, at the highest concentration tested, C0 had no inhibitory effect against vancomycin-resistant Enterococci, linezolid-resistant Enterococci, and Klebsiella pneumoniae. However, among C4-C15 compounds, C5 exhibited significantly enhanced inhibitory effects against these bacteria compared to C0.

[0387] Analysis of the antibacterial mechanism of diterpenoid and triterpenoid derivatives
[0388] Analysis method: The two-dimensional molecular formula of the triterpene derivative of formula (I) was drawn in ChemDraw 18.0 software and exported as a .sdf format structure as a ligand. The FtsZ / GTP complex structure was used as a template, and the X-ray crystal structure of the complex was obtained from the PDB database (PDB ID: 5MN5). Molecular docking was performed using Schrödinger The Glide software package was run in its SP mode. Ligand compounds were pretreated using LigPrep with default parameters, and protein complexes were pretreated using Protein Preparation. After docking, the docking results were analyzed using PyMol software.
[0389] The "docking score" of each compound on FtsZ, a representative of Staphylococcus aureus, is used to characterize the binding strength of the compound to FtsZ. A more negative docking score indicates a stronger binding affinity between the compound and FtsZ.
[0390] The test results can be found in Table 4.
[0391] Compound C15 had the most negative docking score, indicating the strongest binding to FtsZ and relatively stronger antibacterial activity against various bacteria, consistent with the test results in Tables 2 and 3. This is likely due to the presence of a carboxyl group in compound C15 and the introduction of an electron-withdrawing ketone carbonyl group into R8, which allows compound C15 to form hydrogen bonds with Gly110 and Asn44 in the GTP-binding domain of FtsZ, resulting in stronger binding between compound C15 and FtsZ.
[0392] For compound C1, the binding force between compound C1 and FtsZ may be weaker due to the larger molecular radius of compound C1, which increases the steric hindrance with the GTP binding domain of FtsZ and weakens the binding strength between the molecule and the binding domain.
[0393] The binding force between compounds C12 and C13 and FtsZ is relatively weak, which may be because R4 is methyl or ethyl, the active hydrogen disappears, and hydrogen bonds cannot be formed.
[0394] Compound C0 showed no inhibitory activity against vancomycin-resistant Enterococci, linezolid-resistant Enterococci, and Klebsiella pneumoniae at the highest concentration tested. This may be due to the fact that compound C0's glycoside is an arabinopyranoside, while compound C8's glycoside is a rhamnopyranoside. Rhamnopyranose, compared to arabinopyranose, is more easily able to cross the bacterial cell membrane and reach the cytoplasm to bind to FtsZ. Furthermore, compound C0's docking score with FtsZ was -4.955, while C8's was -5.593, indicating that compound C0's binding to FtsZ was weaker than C8's. Compounds C11, C14, and C15, derived from C8 through chemical synthesis, increased their binding to FtsZ and enhanced their antibacterial activity. These differences in glycoside binding lead to differences in binding affinity, suggesting differences in antibacterial efficacy.
[0395] Table 4. Molecular docking results (binding strength)
[0396] Furthermore, the amino acid sequence and protein structure of FtsZ differ between different bacteria, potentially leading to varying antibacterial activity against different bacteria. For example, the sequence similarity between Staphylococcus aureus FtsZ and Pseudomonas aeruginosa FtsZ is only 53%. Compound C0 inhibits the growth of S. aureus but has no effect on P. aeruginosa. To address this phenomenon, molecular docking simulations were conducted to investigate the docking scores of compound C0 with P. aeruginosa FtsZ. Data analysis revealed that compound C0 had a docking score of -1.005, indicating poor binding to P. aeruginosa FtsZ. Compound C5 exhibited significant differences in R8 from C0, enabling C5 to inhibit not only the growth of S. aureus but also that of P. aeruginosa. Molecular docking analysis revealed that C5 had a docking score of -2.115 with P. aeruginosa FtsZ, indicating enhanced binding compared to compound C0. Therefore, C5 also exhibited growth inhibition against P. aeruginosa.
