Description

[0001] Combinations of polypeptide having DNase activity and soil release polymer for use in laundry detergent compositions

[0002] Field of the invention

[0003] The invention relates to the use of a polypeptide having DNase activity and a soil release polymer in a laundry process for reducing redeposition of malodor on a textile, for removing malodor from a textile and / or for maintaining or improving whiteness of a textile, and to laundry detergent compositions comprising one or more polypeptides having DNase activity and one or more terephthalic acid-based soil release polymers.

[0004] Background of the invention

[0005] Laundry detergent compositions that lead, upon application in a laundry process, to a reduction of malodor redeposition on a textile, to a removal of malodor from a textile or to the maintenance or improvement of whiteness of a textile are already known. However, there is still a need for laundry detergent compositions which demonstrate all of the above-mentioned properties and / or show an improvement in one or more of the respective properties.

[0006] Therefore, it was an object of the present invention to provide ingredients or combinations of ingredients for laundry detergent compositions which lead to an advantageous reduction of malodor redeposition on a textile, to a beneficial removal of malodor from a textile and / or to an advantageous maintenance or improvement of whiteness of a textile when applied in a laundry process.

[0007] Surprisingly, it has been found that this problem is solved by the combined use of a polypeptide having DNase activity and a soil release polymer in a laundry process.

[0008] Therefore, a subject matter of the present invention is the use of a polypeptide having DNase activity and a soil release polymer in a laundry process for reducing redeposition of malodor on a textile, for removing malodor from a textile and / or for maintaining or improving whiteness of a textile.

[0009] The combined use of a polypeptide having DNase activity and a soil release polymer in a laundry process leads to an advantageous reduction of malodor redeposition on textiles, to a beneficial removal of malodor from textiles and / or to an advantageous maintenance or improvement of whiteness of textiles. More particularly, the combination of a polypeptide having DNase activity and a soil release polymer has been found to provide a synergistic effect on malodor and whiteness compared to use of either a polypeptide having DNase activity or a soil release polymer alone, which is illustrated in the examples.

[0034] Definitions

[0035] The term “removing malodor from textiles” in the context of the invention consists of two different terms “malodor removal from primary wash” and “malodor repellent”.

[0036] “Malodor removal from primary wash” refers to the fact that malodor on fabrics is removed by washing malodorous fabrics with a laundry detergent composition.

[0037] “Malodor repellent” refers to the fact that a malodor repellent film is formed on fabrics whilst the fabrics are washed with a laundry detergent composition. This malodor repellent film enables malodor on fabric to be washed off easily in a subsequent wash process with a laundry detergent composition.

[0038] The term “reducing redeposition of malodor on a textile” in the context of the invention refers to the fact that malodor, which is washed off from malodorous fabrics in a wash process using a laundry detergent composition, cannot redeposit from the wash liquor onto the fabric in the same wash process, or that there is a reduced level of malodor redeposition. "Soil release” polymer as used herein means a polymer that enhances soil removal during laundering by modifying the surface of the fabric that is laundered, preferably by increasing surface polarity.

[0039] The term “DNase” means a polypeptide having DNase (deoxyribonuclease) activity that catalyzes the hydrolytic cleavage of phosphodiester linkages in DNA, thus degrading DNA. DNases belong to the esterases (EC number 3.1), a subgroup of the hydrolases. The DNases may be classified in several subclasses, in particular EC 3.1.21 and its subclasses. The term “DNase” as used herein is also intended to include nucleases that cleave both RNA and DNA, for example the endoribonucleases in EC 3.1.30 that are active on both RNA and DNA. For purposes of the present invention, DNase activity may be determined according to the procedure described in Assay I. The terms “DNase” and “a polypeptide with DNase activity” may be used interchangeably throughout the application.

[0040] The term “variant” means a polypeptide having DNase activity comprising at least one alteration, i.e. a substitution, an insertion (including extension), and / or a deletion (e.g., truncation), at one or more positions. A substitution means replacement of the amino acid occupying a position with a different amino acid; a deletion means removal of the amino acid occupying a position; and an insertion means adding 1-5 amino acids (e.g., 1-3 amino acids, in particular one amino acid) adjacent to and immediately following the amino acid occupying a position. The alterations in a DNase variant are typically substitutions.

[0041] Sequence identity: The relatedness between two amino acid sequences is described by the parameter “sequence identity”.

[0042] For purposes of the present invention, the sequence identity between two amino acid sequences is determined as the output of “longest identity” using the Needleman-Wunsch algorithm (Needleman and Wunsch, 1970, J. Mol. Biol. 48: 443-453) as implemented in the Needle program of the EMBOSS package (EMBOSS: The European Molecular Biology Open Software Suite, Rice et al., 2000, Trends Genet. 16: 276-277), preferably version 6.6.0 or later. The parameters used are a gap open penalty of 10, a gap extension penalty of 0.5, and the EBLOSUM62 (EMBOSS version of BLOSUM62) substitution matrix. In order for the Needle program to report the longest identity, the -nobrief option must be specified in the command line. The output of Needle labeled “longest identity” is calculated as follows:

[0043] (Identical Residues x 100) / (Length of Alignment - Total Number of Gaps in Alignment) It is known that a host cell may produce a mixture of two of more different mature polypeptides, and that different host cells process polypeptides differently. For example, in the case of the mature polypeptide shown in SEQ ID NO: 1 , it has been found that this may be expressed with different N-terminal truncations, for example as the polypeptide of SEQ ID NO: 2 (206 amino acid residues, corresponding to amino acids 16 to 221 of SEQ ID NO: 1) or SEQ ID NO: 3 (204 amino acid residues, corresponding to amino acids 18 to 221 of SEQ ID NO: 1). Since SEQ ID NOs: 1 , 2 and 3 are identical other than truncation at the N-terminal, any reference herein to a polypeptide having a given percent sequence identity to SEQ ID NO: 1 should be understood as also including polypeptides having that percent sequence identity to SEQ ID NO: 2 or to SEQ ID NO: 3.

[0044] The term “wash liquor” refers to an aqueous solution comprising a polypeptide having DNase activity and a soil release polymer. A wash liquor is a solution, e.g. found in a washing machine, containing water and a detergent composition comprising the polypeptide having DNase activity and the soil release polymer. The detergent composition, prior to being mixed with water to form a wash liquor, may be in any form as described elsewhere herein, for example a liquid or powder.

[0140] The laundry detergent compositions of the invention comprise the one or more terephthalic acid-based soil release polymers of component Z2) preferably in an amount of at least 0.1 wt.- %, more preferably in an amount from 0.1 to 10 wt.-%, even more preferably in an amount from 0.2 to 5 wt.-% and particularly preferably in an amount from 0.2 to 3 wt.-%, in each case based on the total weight of the laundry detergent composition.

[0141] The soil release polymers used according to the invention and the terephthalic acid-based soil release polymers contained in the laundry detergent compositions of the invention may be prepared by methods known to the person skilled in the art.

[0142] It is to be understood that the soil release polymer, and in particular the polyester, used according to the invention or the terephthalic acid-based soil release polymers, and in particular the terephthalic acid-based polyesters, used according to the invention or contained in the laundry detergent composition of the invention are typically prepared by polycondensation processes. This leads to statistically determined mixtures of polymers or polyesters in which a mixture of molecular species with a distribution around a molar average is obtained.

[0143] For the preparation of the polyesters used according to the invention, and in particular for the preparation of the terephthalic acid-based polyesters used according to the invention or contained in the laundry detergent compositions of the invention, e.g. a two-stage process of transesterification of diesters and diols, followed by a polycondensation reaction under reduced pressure, can be used. For example, the process for the preparation of the terephthalic acid-based polyesters used according to the invention or contained in the laundry detergent compositions of the invention comprises the steps of: heating one or more substances of the formula Q1-OOC-CeH4-COO-Q2, wherein Q1 and Q2, independently of one another, are e.g. selected from (Ci-C4)-alkyl and preferably are CH3, and one or more glycols H-O(CnH2n)O-H or mixtures thereof, wherein “n” has the meaning given above, and one or more substances of the formula R1-[O-R2]a-OH or mixtures thereof, wherein R1, R2and “a” have the meaning given above, with the addition of a catalyst, to temperatures of from 160 to 220°C, preferably beginning at atmospheric pressure, and then continuing the reaction under reduced pressure at temperatures of from 160 to 240°C.

[0144] Reduced pressure preferably means a pressure of from 0.1 to 900 mbar and more preferably a pressure of from 0.5 to 500 mbar.

[0145] Typical transesterification and condensation catalysts known in the art can be used for the process for the preparation of the polyesters used according to the invention or contained in the laundry detergent compositions of the invention, such as antimony, germanium and titanium- based catalysts. Preferably, tetraisopropyl orthotitanate (I PT) and sodium acetate (NaOAc) are used as the catalyst system in the process for the preparation of the polyesters used according to the invention or contained in the laundry detergent compositions of the invention.

[0146] In order to obtain polyesters comprising one or more polyalkyleneglycol-derived structural units of the formula -O-[R3-O]f- (IV-a) or mixtures thereof, the respective one or more polyalkyleneglycols of the formula HO-[R3-O]f-H or mixtures thereof, may be employed in the process for the preparation of the polyesters used according to the invention or contained in the laundry detergent compositions of the invention.

[0147] The preparation of soil release polyesters is also described in e.g. WO 2019 / 224030 A1 or WO 2021 / 233987 A1. Soil release polymers are also commercially available.

[0148] The laundry detergent compositions of the invention preferably comprise Z3) one or more surfactants.

[0149] Surfactants assist in removing soil from textile materials and also assist in maintaining removed soil in solution or suspension in the wash liquor. [0239] EXAMPLES

[0240] The examples below are intended to illustrate the invention in detail without, however, limiting it thereto. Unless explicitly stated otherwise, all percentages given are percentages by weight (weight%, % by wt. or wt.-%).

[0241] Assay I: Testing of DNase activity

[0242] DNase activity may be determined on DNase Test Agar with Methyl Green (BD, Franklin Lakes, NJ, USA), which is prepared according to the supplier’s manual. Briefly, 21 g of agar is dissolved in 500 ml water and then autoclaved for 15 min at 121 °C. Autoclaved agar is adjusted to 48°C in a water bath, and 20 ml of agar is poured into petri dishes and allowed to solidify by incubation overnight at room temperature. On solidified agar plates, 5 pl of enzyme solutions are added and DNase activity is observed as colorless zones around the spotted enzyme solutions.

[0243] Example 1 - Preparation of liquid laundry detergent compositions

[0244] Liquid detergent compositions according to Table 1 were prepared, where amounts of the components are given in weight% by total weight of the composition, except for the DNase, where the amount is given in ppm active enzyme product based on the total weight of the composition.

[0247] Example 2 - Preparation of sebum / DNA mixture

[0248] 1. Preparation of DNA solution:

[0249] Salmon DNA (1 g, CAS no. 438545-06-3, purchased from Sigma) was weighted in a beaker. Demineralized water (99 g) was added to prepare a final solution, which contains 1 weight% of DNA. The mixture was stirred on a magnet stirrer for 1 hour at 400 rpm (rpm: resolution per minute), at 70°C, until completely dissolved. The beaker was covered closely with aluminum foil to avoid evaporation while stirring.

[0250] 2. Preparation of sebum / DNA mixture:

[0251] Tween 80 (Polysorbate 80, 200 pL) was added to the DNA solution (100 g) just before adding 0.3 g Sebum Bey (purchased from CFT), so that the final DNA solution contains 0.2 weight%

[0252] Tween 80. Sebum Bey (0.3 g) was then added to the DNA solution. The mixture was stirred at 400 rpm at 70°C for 5-10 minutes and then ULTRA-TURRAX at a high mix speed of 16000 rpm was applied to the mixture for 1 minute. The mixture was afterwards cooled to room temperature and was used within 3 days.

[0253] Example 3 - Preparation of DNA / sebum soiled swatches for malodor removal from primary wash and reducing redeposition of malodor on a textile

[0254] DNA / sebum mixture (200 pL) from Example 2 was added on each swatch (knitted polyester, WFK 33A, 6 cm x 6 cm). The strains were left to dry overnight in the dark at room temperature before being used in the following experiments.

[0255] Example 4 - Preparation of DNA / sebum soiled swatches for malodor repellent

[0260] Artificial malodor composition (50 pL) was dosed on top of the sebum / DNA stain on each dried sebum / DNA soiled swatch from Example 3 (prepared without pre-wash) and Example 4 (prepared with pre-wash). The swatches were then left to dry for 30 minutes in a fume hood at room temperature.

[0261] Artificial malodor composition: The artificial malodor composition was prepared by mixing 50 mg of 2,3-butanedione, 25 mg of octanal, 15 mg of isovaleraldehyde, 1500 mg of isovaleric acid (IVA) and 250 mg of hexanal with 500 g of water and 500 g of isopropanol. The artificial malodor composition was prepared and used on the same day.

[0262] Example 6 - Fabric washing for malodor removal from primary wash

[0263] Washing of fabrics:

[0264] Malodor soiled swatches from Example 5 (prepared without pre-wash) were then washed with liquid laundry detergent compositions A, B, C and D under the conditions shown in Table 3.

[0265] Table 3. Washing conditions for malodor removal from primary wash

[0266] Test swatches were rinsed under tap water 3 times and then under demineralized water 3 times after wash. Excess water was then wringed out.

[0267] Analytical procedure:

[0268] The washed swatches were then packed into Headspace GC vials without further drying steps and the vials were then closed tightly and analyzed.

[0269] Analytical evaluation of malodor in the vials was conducted through a headspace measurement. A Gas Chromatograph Agilent 9000 Intuvo equipped with an autosampler Agilent 7697A equipped with a headspace extracting syringe, a Mass Detector Agilent MSD 5977B was used. The headspace vials were equilibrated at 60°C for 30 minutes at the autosampler before the measurement. A gas phase sample of 1000 pL was extracted from the vial and injected into the GC column.

[0270] Carrier gas: Helium 22.7 psi, split 5:1

[0271] Column: DB-624 III (length 60 m, diameter 250 pm, size of film 1.4 pm)

[0272] Oven: Initial 40°C - Hold time 2 minutes - Rate 20 °C / minute - Temperature 260°C - Hold time 5 minutes Mass detector parameter: run in select ion mode. The peak area of an odorant, Hexanal as marker, was then integrated and recorded.

[0273] Results are shown in Table 4.

[0274] Table 4. Relative peak areas* for the marker odorant (Hexanal) measured in sample headspace from fabric washed for malodor removal from primary wash

[0275] * The peak areas of the individual odorants in the example containing fabrics washed with liquid laundry detergent composition A were normalized as 100.

[0276] From Table 4, it can be clearly seen that the malodor intensity from the fabrics washed with the liquid laundry detergent composition D (inventive) was significantly reduced.

[0277] Example 7 - Fabric washing for reducing redeposition of malodor on a textile

[0278] Washing of fabrics:

[0279] Malodor soiled swatches from Example 5 (prepared without pre-wash) and fresh unsoiled swatches were then washed with liquid laundry detergent compositions A, B, C and D under the conditions shown in Table 5.

[0280] Table 5. Washing conditions for reducing redeposition of malodor on a textile Washed fresh unsoiled swatches were rinsed under tap water 3 times and then under demineralized water 3 times after wash. Excess water was then wringed out.

[0281] Analytical procedure:

[0282] Same as described in Example 6.

[0283] Results are shown in Table 6.

[0284] Table 6. Relative peak areas* for the marker odorant (Hexanal) measured in sample headspace from fabric washed for reducing redeposition of malodor on a textile

[0285] * The peak areas of the individual odorants in the example containing fabrics washed with liquid laundry detergent composition A were normalized as 100.

[0286] From Table 6, it can be clearly seen that the malodor intensity from the fabrics washed with the liquid laundry detergent composition D (inventive) is not detectable anymore, which means no malodor was redeposited onto the fresh unsoiled swatches anymore.

[0287] Example 8 - Fabric washing for malodor repellent

[0288] Washing of fabrics:

[0289] Malodor soiled swatches from Example 5 (prepared with pre-wash) were then washed with liquid laundry detergent compositions A, B, C and D under the conditions shown in Table 7.

[0290] Table 7. Washing conditions for malodor repellent Washed swatches were rinsed under tap water 3 times and then under demineralized water 3 times after wash. Excess water was then wringed out.

[0291] Analytical procedure:

[0292] Same as described in Example 6.

[0293] Results are shown in Table 8.

[0294] Table 8. Relative peak areas* for marker odorant (Hexanal) measured in sample headspace from fabric washed for malodor repellent

[0295] * The peak areas of the individual odorants in the example containing fabrics washed with liquid laundry detergent composition A were normalized as 100.

[0296] From Table 8, it can be clearly seen that the malodor intensity from the fabrics washed with the liquid laundry detergent composition D (inventive) is not detectable anymore, which means all malodor was removed by washing and the swatches pre-washed with liquid laundry detergent D had good malodor repellent effect.

[0297] Example 9: Whiteness benefit of soil release polymer (SRP) and DNase

[0298] Fabric

[0299] Woven polyester / cotton (65% polyester 135% cotton), WFK 20A (Center For Testmaterials BV, the Netherlands), was prewashed as described below and cut into small round swatches with a diameter of 2 cm.

[0300] Prewash without SRP

[0301] Prewash of textiles was done primarily to remove starch, carboxymethyl cellulose (CMC) and other additives from the textile. Amylase (Stainzyme® 12L, Novozymes A / S, Denmark), cellulase (Celluclean® ST, Novozymes) and protease (Savinase® 16L, Novozymes) were added to the prewashes to remove these additives. The textiles were washed three times in 86.1 g / wash detergent W-ECE-2 (wfk Testgewebe GmbH, Germany) using water with 15°dH water hardness (3.00 mL of 0.713 mol / L CaCh, 1.50 mL of 0.357 mol / L MgCh and 0.3371 g of NaHCOs in 1 L deionized water) and containing the following enzymes:

[0302] Prewash with SRP

[0303] The above pre-washed textile was washed three times in 5 g / L of liquid detergent (composition A in Table 1 , Example 1) containing 2% of a soil release polymer, TexCare® SRN 260 (Clariant, Germany), using water with 15°dH water hardness as described above, at 40°C. After wash, the textile was line-dried and cut into small round swatches with diameter of 2 cm.

[0304] Preparation of microbial extracellular DNA (eDNA) stain

[0305] Pseudomonas fluorescens was pre-grown on Tryptone Soya Agar (TSA) (pH 7.3) (CM0131 , Oxoid Lid. Basurgstukc, UK) for 3 days at 30°C. From a single colony, a loop-full was transferred to each of the 3 tubes containing 10 mL of Tryptone Soya Broth (TSB) and incubated for 1 day at 30°C with shaking (200 rpm). The optical density (OD) at 600 nm (ODeoo) was measured using a Spectrophotometer (Clariostar, BMG Labtech, Germany). The bacteria culture was diluted to an ODeoo of 0.03 using half-strength TSB, and 200 ml of the diluted culture was transferred into one 500 mL sterile flask (4 flasks in total). The flasks with Pseudomonas fluorescens were incubated at 30°C for 3 days with shaking at 120 rpm on a sharker (MaxQ™ 4000).

[0306] After propagation, Pseudomonas fluorescens was pelleted by centrifugation (Sorvall LYNX 6000) (10,000 rpm for 10 min) and resuspended in 2 mL of phosphate buffered saline (PBS) (pH 6.9). All the resuspended pellets were transferred into one tube. This tube with resuspended bacteria culture was stored in a cold room and used for all four washes in the multi-cycle wash described below. Before each wash, the ODeoo value of the culture was measured on the spectrophotometer (Clariostar), and was diluted to reach ODeoo = 2 using PBS. For each wash, every swatch was spotted with 50 pl bacteria culture (ODeoo = 2) and was then dried in a clean bench for one hour.

[0307] Detergent

[0308] The detergent was the liquid detergent of composition A described above in Table 1 of Example 1.

[0309] Wash assay - Multi-cycle Mini Launder-O-Meter (miniLOM)

[0310] The miniLOM assay is a small-scale version of the Launder-O-Meter (LOM). A MiniLOM basically consists of closed test tubes being rotated in a heating cabinet for a given time and temperature. Each test tube represents one small washing machine. It is mainly used in testing of detergents and enzymes at European wash conditions.

[0311] In this experiment, three microbial eDNA stains prewashed without SRP were placed in a 50-ml plastic tube and 10 ml of wash solution (15° dH water with 4.3 g / L liquid detergent and 0.7 g / L pigment soil (I LG 09V (wfk Tesgewebe GMB, Germany)) was added to each tube. The following four wash conditions were tested: 1) detergent only (no SRP, no DNase); 2) only SRP, no DNase; 3) no SRP, only DNase; and 4) both SRP and DNase. The SRP concentration was 1% by weight of the detergent, and the DNase concentration in the wash solution was 0.5 ppm.

[0312] The DNase used in this example is a variant of SEQ ID NO: 1 with the substitutions D32E, V35I, K65E, K67A, K105N, A111 P, G181 N and S182Y, where position numbers are based on SEQ ID NO: 1 . The SRP was TexCare® SRN 260.

[0313] The tubes were then mounted in a Mini-Laundr-O-Meter (a Stuart Tube Rotator SB3) and washed at 30°C for 60 minutes at 20 rpm. After wash, the rotator was placed at room temperature while swatches from one tube at a time were rinsed with 15°dH water and placed back into the rotator. Each plastic tube was rinsed two times in 20 mL 15°dH water. After wash, the swatches were dried and evaluated as described below. The whole process (adding culture to swatches, washing, evaluation) was repeated four times in total.

[0314] In parallel, the multi-cycle MiniLOM washes were repeated with the microbial eDNA stains prewashed with SRP under the same wash conditions and wash cycles as described above.

[0315] Evaluation of the whiteness benefits

[0316] After drying, all swatches were measured on a Datacolor 800V spectrophotometer (Datacolor, Lawrenceville, NJ, USA) using the CIE Standard llluminant D65 and the CIE 1964 10° Standard Observer. The measurements were made without UV in the incident light, and remission values at 460 nm were recorded for the stains. The whiteness benefits were visually evident when there was a remission difference between stains of two units or more. Results

[0317] The results are shown in Tables 9 and 10 below, where Table 9 provides the results for swatches that were pre-washed without SRP, and Table 10 provides the results for swatches that were pre-washed with SRP. The “SRP benefit”, “DNase benefit” and “SRP+DNase benefit” were calculated by subtracting the remission value of condition 1 (detergent only) from the remission values of condition 2, 3 and 4, respectively. The “Presumed additive benefit of SRP+DNase” was calculated by adding the values of “SRP benefit” and “DNase benefit”. The “Difference between experimental and presumed additive benefit of SRP+DNase” was calculated by subtracting the value of “Presumed additive benefit of SRP+DNase” from the value of “SRP+DNase benefit”, and represents the synergistic benefit of the combination of the soil release polymer and the DNase.

[0318] Table 9: Remission values (460 nm) measured for swatches pre-washed without SRP after 1, 2,

[0319] 3 or 4 wash cycles with SRP and / or DNase Table 10: Remission values (460 nm) measured for swatches pre-washed with SRP after 1 , 2, 3 or 4 wash cycles with SRP and / or DNase

[0320] The data in Tables 9 and 10 show that the combination of SRP and DNase provided a synergistic effect on whiteness in the multi-cycle wash for both types of stains, i.e. prewashed with or without SRP, starting from the 2ndwash cycle. It is also seen that the synergistic effect of the combination increases with each additional wash using SRP and DNase from the 2ndto the 4thwash cycle. This synergistic effect starting from the 2ndwash cycle was observed with both swatches pre-washed without SRP and those pre-washed with SRP, although the benefit was greater for swatches pre-washed with SRP.
