AI Technical Summary

Technical Problem

The existing MsPA bonding method uses nitrogen methylsulfur sulfonylamide (MsN) that is explosive, hydrolyzed and decomposed, and generates toxic and explosive ammonia nitrogen gases, which are at high risk.

Method used

Methionamide (MsNH) is used as a replacement reagent to form MsPA bonding by reacting with derivatives such as dimethyl adenylate (NVY), thereby avoiding the use of explosive substances.

Benefits of technology

A method of safely synthesizing MsPA bonds is realized, avoiding explosions and toxic gas generation, and improving the safety and reliability of the process.

Description

Compound or salt thereof, and method for producing same

[0001] The present invention relates to a compound or a salt thereof that can be used to produce, for example, an oligonucleotide having an MsPA structure or an amidite block having an MsPA structure, or as a precursor thereof, and a method for producing the same.

[0002] Mesyl phosphoramidate (MsPA) conjugates have recently attracted attention as an alternative to PS for RNase H1-mediated knockdown of microRNA targets and modulation of mRNA splicing, and as a next-generation chemical platform for antisense oligonucleotide (ASO) drugs (see, for example, Non-Patent Documents 1-3).

[0003] To date, the synthesis of MsPA conjugates has been performed using methanesulfonyl azide (MsN 3 ) has been reported (see, for example, Non-Patent Documents 4 and 5). 3 MsN has the property of exploding when exposed to heat or impact. It also decomposes easily in water, producing hydrogen azide, a toxic and explosive gas. 3 Therefore, it is desirable to develop a novel method for synthesizing MsPA conjugates without using .

[0005] In view of the above circumstances, an object of the present invention is to provide a novel method for producing a compound having an MsPA bond, which is non-explosive and does not generate toxic gases.

[0006] Another object of the present invention is to provide a compound or a salt thereof that can be used to produce, for example, an oligonucleotide having an MsPA structure or an amidite block (a phosphoramidite oligomer having a length of several bases) having an MsPA structure, or as a precursor thereof.

[0007] As a result of extensive research to solve the above problems, the present inventors have discovered methanesulfonamide (MsNH), which is non-explosive and does not generate toxic gases. 2 The present invention has been completed by successfully developing a synthetic method using a ) derivative.
