01.07.2024]The genus "Lactobacillus" has, for a long time, been a collecting basin in bacterial taxonomy. All Gram-positive, lactic acid-producing, rod-shaped bacteria incapable of producing spores were almost automatically assigned to this genus. As a consequence, the genus Lactobacillus contains numerous bacteria which are genetically very different. This prompted scientists from Belgium, Canada, China, Germany, and Ireland, Italy, amongst which were scientists belonging to the Subcommittee on the Taxonomy of Lactobacillus and related Organisms, to bring order to the chaos in the taxonomy of species belonging to the genus Lactobacillus. Using a polyphasic approach, the genus Lactobacillus could be reclassified into 23 novel genera. Particularly, Lactobacillus casei became Lacticaseibacillus casei and Lactobacillus rhamnosus became Lacticaseibacillus rhamnosus. The results of the study were published in the peer-reviewed journal International Journal of Systematic and Evolutionary Microbiology (Zheng J, Wittouck S, Salvetti E, Franz CMAP, Harris HMB, Mattarelli P, O'Toole PW, Pot B, Vandamme P, Walter J, Watanabe K, Wuyts S, Felis GE, MG, Lebeer S. A taxonomic note on the genus Lactobacillus: Description of 23 novel genera, emended description of the genus Lactobacillus Beijerinck 1901, and union of Lactobacillaceae and Leuconostocaceae. Int J Syst Evol Microbiol. 2020; 70 (4) : 2782-2858) .Probiotic is defined as live microorganisms that, when administered in adequate amounts, confer a health benefit on the host. In general, the effects of probiotics are associated with normalization of perturbed microbiota and the production of useful metabolites or enzymes. However, some strain-specific effects at the intestinal or extraintestinal level can be found in certain probiotics. Letrozole is a non-steroidal aromatase inhibitor. By blocking the conversion of androgen to estrogen, letrozole induced high androgen levels and symptoms similar to women with PCOS, such as acyclic, polycystic ovary, increased body weight and testosterone level, and insulin resistance (Osuka S, Nakanishi N, Murase T, Nakamura T, Goto M, Iwase A, et al. Animal models of polycystic ovary syndrome: A review of hormone-induced rodent models focused on hypothalamus-pituitary-ovary axis and neuropeptides. Reprod Med Biol. 2018; 18 (2) : 151-60) . However, there is no effective drug specifically for treatment of the indication of PCOS.The present disclosure unexpectedly found the beneficial effects of Lacticaseibacillus paracasei on PCOS symptoms. The compositions described herein comprise a bacterial strain of the genus Lacticaseibacillus, which is useful for relieving, treating and / or preventing PCOS.Examples of Lacticaseibacillus strains for use in the present disclosure include Lacticaseibacillus paracasei. Lacticaseibacillus paracasei (commonly abbreviated as L. paracasei) is a gram-positive, homofermentative species of lactic acid bacteria that are commonly used in dairy product fermentation and as probiotic cultures. L. paracasei is a type of probiotic commonly found in the human intestinal tract and mouth. It is also found in fermented foods and dairy. Strains of L. paracasei have been isolated from a variety of environments including dairy products, plants or plant fermentations, and from the human and animal gastrointestinal tracts. Lacticaseibacillus paracasei's genome contains circular DNA and varies slightly among the different strains isolated. A particular embodiment of the Lacticaseibacillus paracasei is L. paracasei K83 (K83) , which was isolated from healthy human feces and was deposited at Leibniz Institute DSMZ -German Collection of Microorganisms and Cell Cultures GmbH at Inhoffenstraβe 7B 38124 Braunschweig Science Campus Braunschweig-Süd GERMANY on 27 June 2013 under DSMZ Accession No. DSM 27449. The 16S rRNA gene sequence of L. paracasei K83 is listed as follows. The original name of Lacticaseibacillus paracasei was Lactobacillus paracasei when the deposit of biological material was made. However, the name of Lactobacillus was changed for the reason described in paragraph

[0095] .The L. paracasei K83 deposited under accession number DSM 27449 was tested in the Examples. A 16S rRNA sequence for K83 that was tested is provided in SEQ ID NO: 1. The genome of K83 comprises a chromosome and plasmids.K83 16S rRNA sequenceBacterial strains closely related to the strain tested in the examples are also expected to be effective for relieving, treating and / or preventing PCOS. In certain embodiments, the bacterial strain for use in the present disclosure has a 16S rRNA sequence that is at least 95%, 96%, 97%, 98%, 99%, 99.5%or 99.9%identical to the 16S rRNA sequence of a bacterial strain of L. paracasei. Preferably, the bacterial strain for use in the invention has a 16S rRNA sequence that is at least 95%, 96%, 97%, 98%, 99%, 99.5%or 99.9%identical to SEQ ID NO: 1. Preferably, the bacterial strain for use in the present disclosure has the 16S rRNA sequence represented by SEQ ID NO: 1. In certain embodiments, the bacterial strain for use in the invention has a 16S rRNA sequence that is at least 95%, 96%, 97%, 98%, 99%, 99.5%or 99.9%identical to the 16S rRNA sequence of a bacterial strain of L. paracasei K83.Other L. paracasei strains that are useful in the compositions and methods of the present disclosure may be identified using any appropriate method or strategy, including the assays described in the examples. For instance, strains for use in the present disclosure may be identified by culturing in a culture medium, cultured at 37 ℃ for 18 hours and then assessed by genetic typing and / ugation, and then embedded and lyophilized with protective agents and excipients to form a heat-treated K83 powder. Prior to oral administration to animals, the live K83 or heat-treated K83 concentration was adjusted to 1 × 109 colony-forming units (CFU)  / mL or 1 ×109 cells / mL in phosphate-buffered saline (PBS) , respectively.Animals and treatmentsEight-week-old female Sprague Dawley rats (BioLASCO Taiwan Co., Ltd., Taipei City, Taiwan) were purchased and housed under standard laboratory conditions with ad libitum access to food and water. All animal procedures listed herein were approved by the Institutional Animal Care and Use Committee of the National Yang Ming Chiao Tung University (protocol number 1100802) . The rats were randomly divided into 5 groups: control, letrozole, Diane-35, K83, and HT-K83. Letrozole is a non-steroidal aromatase inhibitor and was used to induce PCOS symptoms in this study (Kafali H, Iriadam M, Ozardali I, Demir N. Letrozole-induced polycystic ovaries in the rat: a new model for cystic ovarian disease. Arch Med Res. 2004; 35 (2) : 103-8) . The rat's PCOS symptoms were induced on days 15–35 of experiment by received oral administration of 1 mg / kg body weight of letrozole dissolved in 1%carboxymethyl cellulose (CMC) , while the control rats received 0.5 ml of 1%CMC only. Diane-35 (one tablet containing 2 mg cyproterone acetate and 35 μg ethinylestradiol, dissolved in 25 mL of 1%CMC and oral administered at 4.5 mL / kg body weight) was used as treatment control. L. paracasei K83 (1 × 109 CFU) or HT-K83 (1 × 109 cells) was given to the rats prior to PCOS induction and throughout the study (day 1 to day 42) . Estrous cycle was monitored from day 10 to day 42. At the end of the experiment, all rats were sacrificed for subsequent analysis.Determination of estrous cycle phasesThe phases of estrous cycle were determined by the predominant cell type in vaginal smears (Marcondes FK, Bianchi FJ, Tanno AP. Determination of the estrous cycle phases of rats: some helpful considerations. Braz J Biol. 2002; 62 (4a) : 609-14) . Briefly, approximately 150–200 μL of PBS was flushed into the rat's vagina and back out 2 or 3 times to collect the vaginal cells for microscopic examination (Cora MC, Kooistra L, Travlos G. Vaginal Cytology of the Laboratory Rat and Mouse: Review and Criteria for the Staging of the Estrous Cycle Using Stained Vaginal Smears. Toxicol Pathol. 2015; 43 (6) : 776-93) . The estrous cycle is classified into the four phases of proestrus, estrus, metestrus, and diestrus, and the phases were identified based on the relative proportion of nucleated epithelial cells, anucleated keratinized epithelial cells, and leukocytes.Histological and immunohistochemical (IHC) analysisAfter the rats were sacrificed, ovaries were immediately harvested and fixed with 10%formalin fixative (JT Baker, Phillipsburg, NJ, USA) . Ovaries were embedded in paraffin and sectioned into 5-μm-thick slices. The sections were stained with hematoxylin and eosin (H&E) to observe the numbers of cystic follicles and corpus lutea.To evaluate the expression of androgen receptor (AR) , and apoptosis related proteins (Bax) in ovarian tissues, IHC staining was conducted. Ovary sections were subjected to antigen retrieval and incubated with primary antibodies against AR (Cell Marque Corporation, CA, USA) or Bax (Santa Cruz Biotechnology, Inc., CA, USA) . Then, sections were washed, incubated in specific secondary antibody, and counterstained with hematoxylin. The sections were scanned, quantified the positive staining by ImageJ software.Hormone and miRNA expression profileBlood was collected in tubes with or without anticoagulant (EDTA) to obtain plasma or serum. Collected serum or plasma was aliquoted and stored at -80 ℃ for further analysis.The levels of serum testosterone were measured using Elecsys assay on Cobas immunoassay analyzers (Roche Diagnostics, Meylan, France) .For the RNA isolation from plasma, RNA fragments of less than 1000 nucleotides were isolated from 100 μL plasma samples using the LiqmiR microRNA Extraction Kit (Topgen Biotech, Kaohsiung City, Taiwan) according to the manufacturer’s instructions. For the RNA isolation from ovary, total RNAs were extracted from 20–30 μg of tissues using the RNeasy Mini Kit (Qiagen, Hilden, Germany) following manufacturer’s protocol. RNA isolated from ovaries and plasma were stored at -80 ℃ for further use. cDNA synthesis was performed using the miRscript RT Kit (Topgen Biotech) with specific microRNA stem-loop reverse transcribed primer listed in table 1. Real time polymerase chain reaction (PCR) was performed on StepOne Plus Real-Time PCR system (Applied Biosystems, CA, USA) . The amplification data were normalized by U6 snRNA or cel-miR-39-3p expression of ovary samples and plasma samples, respectively. The relative expression was quantified by the 2-ΔΔCt relative quantification method (Livak KJ, Schmittgen TD. Analysis of relative gene expression data using real-time quantitative PCR and the 2 (-Delta C (T) ) Method. Methods. 2001; 25 (4) : 402-8) .Table 1. The primer and probe sequences used in cDNA synthesis and miRNA quantification.Statistical analysisThe software GraphPad Prism 9 was used to process the experimental data. All data are shown as mean ± standard error of the mean. Between-group differences were analyzed using one-way analysis of variance (ANOVA) by Tukey’s post hoc test. P < 0.05 was considered as statistically significant.Example 1 Genetic typing of Lacticaseibacillus paracasei K83The following table lists those organisms whose DNA sequences of 16S rRNA and Tuf gene show the highest similarity values compared to the 16S rRNA and Tuf gene sequence of K83.Comparison of 16S rRNA and Tuf gene sequence indicates that K83 belongs to Lacticaseibacillus paracasei subsp. paracasei.Example 2 API typingSugar utilization of K83 was investigated using API 50 CHL kit (bioMerieux, France) . The fermentation test indicates that K83 harbors a biochemical property similar to Lacticaseibacillus paracasei subsp. paracasei.+, positive; -, negative.Example 3 K83 alleviates ovarian dysfunction in letrozole-induced PCOS-like ratsReproductive health and ovarian function of rats were evaluated through monitoring of estrous cycle and examination of ovarian morphology. The control rats exhibited regular estrous cycles of 4-5 days, whereas a majority of PCOS-like rats were completely acyclic after letrozole treatment. However, the disrupted estrous cycles were improved after K83 treatment. A total of 25%of PCOS-like rats in K83 group and 40%in HT-K83 group showed restored estrous cycles by the end of the experiment, which were similar or better effects than those of the treatment control Diane-35 (Fig. 1A) . Histological examination of ovarian tissues showed multiple growing follicles in different development stages and a higher number of corpora lutea in control rats. However, PCOS-like rats exhibited an increased number of cystic follicles along with fewer corpora lutea (Fig. 1B) . Remarkably, K83 and HT-K83 administration significantly decreased the number of cystic follicles and slightly recovered the number of corpora lutea compared with those in the letrozole group (Fig. 1B, 1C) . Overall, these data indicated that K83 and HT-K83 improved the function and pathological changes of ovaries significantly.Example 4 K83 reduced the level of serum testosterone and expression of AR in the ovarian tissuesElevated level of testosterone is one of the typical characteristics of PCOS. The serum testosterone level of letrozole group was significantly higher compared with those in the control group (Fig. 2) . Administration of K83 significantly reduced the testosterone level compared with those in the letrozole group (Fig. 2) and returned to a level similar to those of control rats. As the effects of testosterone are mediated through the interaction with its receptor, AR, we further examined the expression of AR in the ovarian tissues of control and PCOS-like rats. An increased immunoreactive signal was observed in PCOS-like rats, but the AR-positive area in K83 group was significantly lower compared with that in the letrozole group (Fig. 3) .Thus, our results suggest that K83 could significantly improve ovarian dysfunction, probably by restoring abnormal testosterone level and reducing the AR-positive area in the ovarian tissue in PCOS-like rats. Although HT-K83 did not reduce the serum testosterone level significantly, HT-K83 might improve the ovarian function through another mechanism.Example 5 K83 reduced the expression of specific miRNAs and apoptosis protein in PCOS-like ratsMiRNAs dysregulation was often found in PCOS patients and PCOS animal models. According to previous studies, abnormal expression of miR-30 has been found to be related to apoptosis of ovarian cells (He H, Li D, Tian Y, Wei Q, Amevor FK, Sun C, Yu C, Yang C, Du H, Jiang X, Ma M, Cui C, Zhang Z, Tian K, Zhang Y, Zhu Q, Yin H. miRNA sequencing analysis of healthy and atretic follicles of chickens revealed that miR-30a-5p inhibits granulosa cell death via targeting Beclin1. J Anim Sci Biotechnol. 2022 Apr 12; 13 (1) : 55) . To further clarify the role of miRNAs in PCOS, we evaluated the expression of miR-30a-5p which was reported to be differentially expressed in the plasma and ovarian tissues of PCOS rats. We discovered that the expression of miR-30a-5p in plasma and ovarian tissues was significantly increased in the letrozole group compared with those in the control group (Fig. 4) . K83 administration significantly reduced the expression of miR-30a-5p compared with those in the letrozole group both in plasma and ovarian tissues (Fig. 4) .Coincidentally, the dysregulation of miRNAs and excess androgen production have been found to be involved in the stimulation and promotion of apoptosis in ovarian cells. Thus, we examined the pro-apoptotic protein Bax in ovarian tissues by performing the IHC staining. As with the results presented in Fig. 5, the expression of Bax was significantly increased in the letrozole group compared with the control group, whereas the administration of K83 significantly reduced the expression of Bax in the ovarian tissues compared with that in the letrozole group (Fig. 5) .
