NOVEL ANTI-CD47 ANTIBODIES AND USES THEREOFCROSS REFERENCE TO RELATED APPLICATIONS

[0003] The present application relates to anti-CD47 antibodies, methods of producing such antibodies, and use of such antibodies in the treatment of diseases and disorders associated with CD47. BACKGROUND OF THE APPLICATION

[0004] Extensive clinical and experimental evidence suggests that macrophages play a crucial role in cancer immunotherapy. Cluster of differentiation (CD) 47, which is found on both healthy and malignant cells, regulates macrophage-mediated phagocytosis by sending a "don't eat me" signal to the signal regulatory protein alpha (SIRPα) receptor. Increasing evidence demonstrates that blocking CD47 interaction with SIRPα can enhance cancer cell clearance by macrophages. Additionally, inhibition of CD47 / SIRPα interaction can increase antigen cross-presentation, leading to T-cell priming and an activated adaptive antitumor immune response. Therefore, inhibiting CD47 / SIRPα axis has a significant impact on tumor immunotherapy.

[0005] Studies on CD47 monoclonal antibodies are at the forefront of research, and impressive results have been obtained. Nevertheless, hematotoxicity, especially anemia, has become the most common adverse effect of the CD47 monoclonal antibody. See e.g., Biomark Res 11, 15 (2023) .

[0241] CD47 delivers an antiphagocytic signal on macrophages through its interaction with SIRPα. To evaluate the phagocytosis activity of Anti-CD47 antibodies, ADCP (antibody dependent cell phagocytosis) assay on OCI-LY19 and RKO tumor cell lines was performed. Briefly, Macrophage  was generated via 7 days’ induction by 50ng / mL M-CSF on Monocyte which was isolated from Human PBMC with CD14+ isolation beads. On the day of ADCP assay,  was labelled with cell trace Far red, and RKO was labeled with Violet at 37℃ with 5%CO2 for 15 minutes. After labelling,  and tumor cells were washed with DPBS buffer with 0.1%BSA, resuspended with MACS buffer [PBS, pH 7.2, 0.5%bovine serum albumin (BSA) , and 2 mM EDTA] to the density of 6E5 / mL and tumor cell to 1.8E6 / mL respectively. Anti-CD47 antibodies, benchmark antibody 5F9 and isotype control antibody were serially diluted with MACS buffer, incubated with 50 μL  cell and 50 μL tumor cell at 37℃ for 2 hours. After incubation, cells were washed once with DPBS buffer, resuspended with 100 μL FACS buffer and analyzed with Beckman CytoFlex. Phagocytosis was measured by gating on Far red+ cells and then assessed the percent of Violet+ cells.
