[0510] In brief, mice were immunized by using the recombinant protein CD38-His (ACRO Biosystems, Cat: CD8-H5224) , which contains a human CD38 extracellular domain with a His tag, as an antigen, and positive mouse plasma cells (binding CD38 and expressing IgG) were separated and screened by the beacon single-cell photoconductive system. Furthermore, the single-cell sequencing was used to obtain a sequence encoding a variable region of an anti-CD38 antibody in the positive mouse plasma cells. Appropriate candidate antibodies were selected by analyzing the positive binding of the antibody secreted by each cell to CD38 and the gene encoding the antibody sequence for the antibody thereof.
