[0006] Cytokines belonging to the common γ chain (γc) family are central regulators of the development, proliferation, survival and differentiation of multiple cell lineages of the innate and adaptive immune system, and as such are of high interest for anticancer therapeutic applications.

[0007] Among them, IL-2 is the most comprehensively one. It became the first immunotherapy approved by the United States Food and Drug Administration (FDA) for cancer treatment nearly 30 years ago. However, IL-2 has the effect of promoting the proliferation of regulatory T cells and CD4+ helper T cells. It can also eliminate self-relative T cells via activation-induced cell death (AICD) to prevent autoimmunity. Thus, IL-15 has gradually emerged as an alternative to IL-2 in cancer treatment because of its functional similarity to IL-2 with several added benefits. For example, the physiological / pathological roles and mechanisms of action of IL-15 in immunity have been more thoroughly investigated. IL-15 is essential in the growth, mobilization and activation of immune cells such as natural killer (NK) cells, CD8+ T cells, γδ T cells and natural killer T (NKT) cells. IL-15 can also enhance the anti-tumor activity of immune cells.

[0008] However, wildtype IL-15 is relatively unstable. Thus, there is a need to develop cancer therapies targeting the IL-15 pathway with improved stability.SUMMARY

[0009] This disclosure relates to engineered IL-15 variants, protein constructs (e.g., fusion proteins or protein complexes) , and methods of use thereof. In some embodiments, the engineered IL-15 variants and protein constructs include a non-native disulfide bond formed by mutating two or more residues (e.g., a pair of residues) of wildtype IL-15 to cysteines, without interfering the overall structure or residues critical for IL-15 receptor interaction. As a result, the engineered IL-15 variants or protein constructs thereof can be more stable than a wildtype IL-15 or protein construct thereof. In some embodiments, the protein constructs showed enhanced thermostability (e.g., increased melting temperatures) , a medium to high T cell response, and / or a comparable CD122 / CD132-binding affinity than a wildtype IL-15 or protein construct thereof. These engineered IL-15 variants and protein constructs thereof described herein can be used for cancer therapies.

[0225] PBMCs (STEMCELL TECHNOLOGIESTM, Cat#: 70025) were seeded at a density of 1 × 104 cells / well in 50 μL of Complete RPMI (10%FBS (GibcoTM, Cat#: A52567-01) +90%RPMI 1640 medium, HEPES (GibcoTM, Cat#: 22400-089) ) with equal volume of titrated doses of H8-sushi-IL15_wt and its variants in 96-well F-bottom microplates (Greiner Bio-one, Cat#: 655098) and cultured for 4 days in a 37℃ incubator. Each concentration was tested in triplicate. The proliferation was assessed by the CellTiter- Luminescent Cell Viability Assay (Promega, Cat#: G7573) . In brief, the plates were removed from the incubator and conditioned at room temperature for 15 minutes, and a volume of pre-mixed Reagent equal to the volume of cell culture medium present in each well was added. The contents were mixed by gently tapping on each side of the walls, followed by incubating at room temperature in the dark for 10 minutes. The luminescent signals were then measured with a microplate reader (VarioskanTM LUK, Thermo Scientific, type3020) . The results were further analyzed using GraphPad Prism software.

[0230] The protein stability of H8-sushi-IL15_wt and its variants was determined by differential scanning fluorimetry (DSF) . Assays were performed with a Bio-Rad CFX ConnectTM Real-Time PCR System (Hercules, CA, USA) into MultiplateTM 96-Well PCR Plates, low profile, unskirted, clear (Bio-Rad; Hercules, CA, USA) . Wells were filled to 25 μL, each composed of 2.5 μL of Orange (Sigma-Aldrich; Cat#: S5692) 5,000×, for a final concentration of 5×; 10 μL of each tested sample at a final protein concentration of 0.4 mg / mL; and the remaining 12.5 μL with the appropriate buffer. The protocol was set with a temperature analysis range from 20℃ to 95℃, with steps of 1℃ increasing every 10 seconds and fluorescence recorded at the end of each step. Raw fluorescence data were analyzed to determine the melting temperature (Tm) , from non-linear fitting of thermal denaturation data.
