[0003] Retinal diseases and conditions, such as glaucoma, macular degeneration, and retinitis pigmentosa, are a major cause of vision loss worldwide. Retinal diseases and conditions are often associated with retinal degeneration, impaired vision, light (daytime) blindness, dark (nighttime) blindness, light sensitivity, loss of peripheral vision, reduced macular function, and reduced visual acuity. Left untreated, retinal diseases and conditions can progress to total vision loss.

[0005] Sirtuin 6 (SIRT6) plays various roles in the retina such as protecting against oxidative stress, regulating glucose metabolism, and activating autophagy in the retina. An increased expression of SIRT6 has also shown favorable effects in the processes of inflammation and DNA repair. Among others, oxidative stress, inflammation, and defects in autophagy are common features in retinal diseases and conditions.

[0006] Accordingly, the present invention is based on the determination that SIRT6 activators provide a significant therapeutic effect for retinal diseases and conditions.

[0007] Thus, one aspect of the invention relates to a method of treating, preventing, ameliorating, or slowing the progression of a retinal disease or condition in a human subject in need thereof, comprising administering to the subject a therapeutically effective amount of a SIRT6 activator, thereby treating, preventing, ameliorating, or slowing the progression of theretinal disease or condition. In some embodiments, the method further comprises administering to the subject a therapeutically effective amount of an additional therapeutic agent for treating a retinal disease or condition.

In some embodiments, the additional therapeutic includes, but is not limited to, an anti-angiogenesis agent (e.g., angiostatin, anecortave acetate, thrombospondin, ranibizumab and bevacizumab, pegaptanib, sunitinib, and sorafenib), a known ophthalmic drug (e.g., acetbutolol, atenolol, bisoprolol, carvedilol, asmolol, labetalol, nadolol), a carbonic anhydrase inhibitor agent (e.g., acetozolamide, brinzolamide, dorzolamide and methazolamide, ethoxzolamide, diamox, and dichlorphenamide), a mydriatic-cycloplegic agent (e.g., atropine, cyclopentolate, succinylcholine, homatropine, phenylephrine, scopolamine and tropicamide), a prostaglandin (e.g., prostaglandin F2 alpha, bimatoprost, latanoprost, travoprost and unoprostone), an immunomodulator (e.g., glatiramer acetate, teriflunomide, dimethyl fumarate), or any combination thereof.

28. The method of claim 26, wherein the compound is (2S,5’R)-7-chloro-3’,4-dimethoxy- 5’-methyl-6-(5-methyl-l,3,4-oxadiazol-2-yl) spiro [benzofuran-2,4’-cyclohex-2-ene]-l’,3- dione or a pharmacologically acceptable salt thereof.

29. The method of claim 26, wherein the compound is (2S,5’R)-7-chloro-6-(5-ethyl- l,3,4-oxadiazol-2-yl)-3’,4-dimethoxy-5’-methyl-spiro [benzofuran-2,4’-cyclohex-2-ene]- 1’, 3-dione or a pharmacologically acceptable salt thereof.

30. The method of claim 26, wherein the compound is (2S,5’R)-7-chloro-6-[3-(l- hydroxy- 1 -methyl -ethyl)- 1 ,2,4-oxadiazol-5-yl]-3 ’ ,4-dimethoxy-5 ’-methyl-spiro [benzofuran- 2, 4’-cyclohex-2-ene]-l’, 3-dione or a pharmacologically acceptable salt thereof.

31. The method of claim 26, wherein the compound is (2S,5’R)-7-chloro-4-ethoxy-3’- methoxy-5’-methyl-6-(5-methyl-l,3,4-oxadiazol-2-yl) spiro [benzofuran-2,4’-cyclohex-2- ene]-l’, 3-dione or a pharmacologically acceptable salt thereof.
