Description

BACILLUS STRAINS FOR ANIMAL FEED

[0001] Reference to a deposit of biological material

[0002] This application contains a reference to a deposit of biological material, which deposit is incorporated herein by reference. See the Examples section for further information.Field of the Invention

[0003] The present invention relates to animal feed or feed additive that is useful in the prevention or reduction of bacterial infection in livestock animals, in particular a monogastric animal such as poultry or swine, where the animal feed or feed additive comprises one or more selected Bacillus strains. Background of the Invention

[0004] Swine and poultry meat account for a large portion of world-wide meat consumption. However, swine and poultry farms are often accompanied by diseases. Once a disease breaks out, it will not only slow down the growth of swine and poultry and reduce the supply of meat, but can also cause large economic losses for farmers.

[0005] Antibiotics have been widely used for many years to combat these diseases, but the long-term and extensive use of antibiotics has led to an increase in antibiotic resistance, and there is therefore increasing pressure to ban the use of antibiotics in animal feed. As an alternative to prophylactic use of antibiotics in animal feed, probiotics can have a significant effect in promoting animal health when ingested in sufficient quantities.

[0006] Bacillus-based probiotic feed additives are known for their positive effects on health in animal feed. These products are relevant for the feed industry because spores are heat stable and can survive the pelletizing process at temperatures up to 90-95℃. The Bacillus species B. subtilis, B. licheniformis and B. pumilus have been given the status Generally Regarded as Safe (GRAS) by the U. S. Food and Drug Administration (FDA) and are acceptable for inclusion in an animal diet.

[0007] US2020359653 discloses a composition comprising three or four Bacillus species selected from Bacillus amyloliquefaciens, Bacillus subtilis, Bacillus licheniformis and Bacillus coagulans.

[0008] US2020222472 provides a Bacillus subtilis strain selected from the group consisting of a) the strain deposited as DSM32324, b) the strain deposited as DSM32325, and c) a mutant strain of (a) or (b) which has sensitivity for ampicillin, vancomycin, gentamicin, kanamycin, streptomycin, erythromycin, clindamycin, tetracycline, and chloramphenicol, and has inhibitory activity against E. coli and Clostridium perfringens, as well as compositions comprising at least one such Bacillus subtilis strain.

[0009] US2022088094 discloses probiotic compositions that include one or more isolated novel Lactobacillus reuteri strains which colonize the gastrointestinal tract to increase the health of an animal.

[0010] WO2019141815 discloses a Bacillus subtilis strain deposited as DSM 32685 and mutant strains thereof which are susceptible to relevant antibiotics, have inhibitory activity against Staphylococcus, Streptococcus, E. coli and Salmonella enterica, and have the ability to facilitate degradation of non-starch polysaccharides and thereby increase the amount of available oligosaccharides (sugar) in animal feed.

[0011] US20170246222 discloses Bacillus strains having antimicrobial activity against Clostridium perfringens.

[0012] In spite of the existence of Bacillus strains and feed compositions such as those outlined above, there is still a need for animal feed products and additives comprising novel combinations of probiotic bacteria strains to address unmet needs, for example to prevent or reduce pathogenic bacterial infections in animals such as poultry and swine.Summary of the Invention

[0024] Definitions

[0025] In accordance with this detailed description, the following definitions apply. Note that the singular forms "a, " "an, " and "the" include plural references unless the context clearly dictates otherwise.

[0026] Unless defined otherwise or clearly indicated by context, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this invention belongs.

[0027] Animal: The term "animal" as used herein refers to domesticated animals raised as livestock, in particular monogastric animals, and includes, but is not limited to, equine (e.g., horses) and porcine animals (e.g. swine, such as piglets, weaners, growers, finishers, hocks, polts, gilts, sows and gestation sows) and rabbits; as well as avians including, but not limited to, poultry such as broilers, breeders, layers, ostriches, quails, turkeys, ducks and geese. Preferred animals in the context of the present invention are poultry and swine.

[0028] Animal feed: The term “animal feed” refers to any compound, preparation or mixture suitable for, or intended for intake by an animal. Animal feed for a production animal comprises concentrates as well as for example vitamins, minerals, enzymes, amino acids and / or other feed ingredients (such as in a premix) . The animal feed may further comprise forage.

[0029] Average Daily Gain: The Average Daily Gain (ADG) of an animal is the increase in body weight of the animal per day expressed in g / day. Typically the weight of the animal is measured at a few time points during the experiment and the difference in weight is then divided by the number of days in the measured period to get the Average Daily Gain.

[0030] Blend: the term “blend” means more than one of the bacterial strains described herein.

[0031] Body Weight Gain: The Body Weight Gain of an animal is the increase of body weight of the animal over a specified time period. The body weight gain can readily be determined by weighing the animal at two timepoints and calculating the gain in weight by subtracting the weight at the first timepoint from the weight at the second timepoint.

[0032] Composition: The term “composition” refers to a composition comprising a carrier and at least one bacterial strain as described herein. The composition may be a liquid or solid composition, for example a solid composition in the form of a powder, a granulate or pellets. The compositions described herein may be mixed with an animal feed to obtain a “mash feed” , extruded or pressed feed pellets, or liquid feed.

[0034] Direct Fed Microbial (DFM) : The term “direct fed microbial” or “DFM” means live micro-organisms including spores which, when administered in adequate amounts, confer a benefit, such as improved digestion or health, on the host.

[0035] Effective amount / concentration / dosage: The terms “effective amount” , “effective concentration” , or “effective dosage” are defined as the amount, concentration, or dosage of the bacterial strain (s) sufficient to improve the digestion or yield of an animal. The actual effective dosage in absolute numbers depends on factors including: the state of health of the animal in question, other ingredients, additives, or drugs present. The “effective amount” , “effective concentration” , or “effective dosage” of the bacterial strains may be determined by routine assays known to those skilled in the art.

[0036] FCR (Feed Conversion Ratio) : FCR is a measure of an animal's efficiency in converting feed mass into increases of the desired output, where the output is the mass gained by the animal. The term is used for animals that are raised for meat, such as e.g. swine, poultry and fish. Specifically, FCR is the mass of the food eaten divided by the output, all over a specified period. Improvement in FCR means reduction of the FCR value. A FCR improvement of 2%means that the FCR was reduced by 2%.

[0037] Feeding an animal: The terms “feeding an animal” or “fed to an animal” means that the composition of the present invention is administered orally to the animal one or more times in an effective amount. The oral administration is typically repeated, e.g., one or more times daily over a specified time period such as several days, one week, several weeks, one months or several months. Accordingly, the terms “feeding” or “fed” mean any type of oral administration such as administration via an animal feed or via drinking water.

[0038] Isolated: The term “isolated” means that the one or more bacterial strains described herein are in a form or environment which does not occur in nature, that is, the one or more bacterial strains are at least partially removed from one or more or all of the naturally occurring constituents with which it is associated in nature.

[0039] Monogastric: The term “monogastric” refers to animals having a stomach with only a single compartment. Examples of monogastric animals are poultry, pigs or swine and horses. In the context of the present invention, the monogastric animals are domesticated animals raised as livestock.

[0040] Mutant: A "mutant bacterium" or a "mutant strain" refers to a natural (spontaneous, naturally occurring) mutant bacterium or an induced mutant bacterium comprising one or more mutations in its genome (DNA) which are absent in the parent strain DNA. An "induced mutant" is a bacterium where the mutation was induced by human treatment, such as treatment with any conventionally used mutagenization treatment including treatment with chemical mutagens, such as a chemical mutagen selected from (i) a mutagen that associates with or become incorporated into DNA such as a base analogue, e.g. 2-aminopurine or an interchelating agent such as ICR-191, (ii) a mutagen that reacts with the DNA including alkylating agents such as nitrosoguanidine or hydroxylamine, or ethane methyl sulphonate (EMS) or N-methyl-N'-nitro-N-nitroguanidine (NTG) , UV-or gamma radiation etc. In contrast, a "spontaneous mutant" or "naturally occurring mutant" has not been mutagenized by man.

[0041] A mutant may have been subjected to several mutagenization treatments (asingle treatment should be understood one mutagenization step followed by a screening / selection step) , but it is preferred that no more than 20, or no more than 10, or no more than 5, treatments (or screening / selection steps) are carried out. In preferred mutants less than 1%, less than 0.1%, less than 0.01%, less than 0.001%or even less than 0.0001%of the nucleotides in the bacterial genome have been replaced with another nucleotide, or deleted, compared to the mother strain.

[0042] Mutant bacteria as described above are non-GMO, i.e. not modified by recombinant DNA technology. As an alternative to the above preferred method of providing the mutant by random mutagenesis, it is also possible to provide such a mutant by site-directed mutagenesis, e.g. by using appropriately designed cloning techniques.

[0043] When the mutant is provided as a spontaneously occurring mutant the strain is subjected to the selection step without any preceding mutagenization treatment.

[0121] The term "CFU / kg" refers to the weight of the composition including carriers and other components present in the composition.

[0122] In one embodiment, the composition may be administered to an animal such as a swine to provide a bacterial count of each Bacillus strain of between 1x103 and 1x1013 CFU / animal / day, preferably between 1x105 and 1x1011 CFU / animal / day, more preferably between 1x106 and 1x1010 CFU / animal / day and most preferably between 1x107 and 1x109 CFU / animal / day.

[0123] The compositions of the invention include at least one Bacillus strain of the invention and at least one carrier, excipient and / or diluent and / or other component that make the composition suitable for feeding to an animal or as an additive for drinking water. There are a myriad of such agents available which may be added. Without intending to be limiting, examples include standardizing agents, extenders, wetting agents and lubricating agents, preservative agents, lipids, stabilizers, solubilizers, free flowing agents, and emulsifiers. Examples that may be particularly useful in administration to an animal include ground corn cobs, salt, ground limestone, calcium carbonate, sodium bentonite, zeolites, ground soy hulls, citrus pulp, dairy by-products, animal protein products, grain products, plant protein products, processed grain products and by-products, roughage products, molasses products fermentation by-products such as dried distillers grains and / or solubles, citric acid and glutamic acid fermentation by-products and the like.

[0124] Bacillus bacteria exist as spores and vegetative cells which can divide to produce more vegetative cells. When reference is made herein to Bacillus, this relates to both spores and vegetative cells unless the context indicates otherwise.

[0125] In the compositions of the present invention, the Bacillus strain or strains are preferably provided as spores. The primary function of sporulation is generally to ensure the survival of a bacterium through periods of environmental stress. They are therefore resistant to ultraviolet and gamma radiation, desiccation, lysozyme, temperature, starvation, and chemical disinfectants. The spore coat is impermeable to many toxic molecules and may also contain enzymes that are involved in germination. The core has normal cell structures, such as DNA and ribosomes, but the spore is metabolically inactive.

[0126] The vegetative form of the bacteria produces effectors which may reduce the numbers of bacterial pathogens or have other beneficial effects in the gastrointestinal tract of an animal. Thus, reactivation and germination of the spores after administration to the animal is important.

[0127] Probiotic bacteria such as the Bacillus strains of the invention should be able to survive and proliferate in the gut of animals by being able to tolerate a low pH and be resistant to bile salt in order to be useful as probiotic compositions for animal feed. The ability to be able to form endospores makes Bacillus particularly suitable as probiotics. The one or more bacterial strains are therefore preferably present in the composition in the form of a spore such as a stable spore, for example a stable spore that will germinate in the intestine and / or stomach of the monogastric animal.

[0128] In one embodiment, the one or more bacterial strains are stable when subjected to pressures applied / achieved during an extrusion process for pelleting. In a particular embodiment, the one or more bacterial strains are stable at pressures ranging from 1 bar to 40 bar, particularly 10 bar to 40 bar, more particularly 15 bar to 40 bar, even more particularly 20 bar to 40 bar, for example from 35 bar to 37 bar.

[0129] In a particular embodiment, the one or more bacterial strains are stable at high temperatures. In particular, the bacterial strains are stable when they are subjected to temperatures achieved during an extrusion process for pelleting. In an even more particular embodiment, the one or more bacterial strains are stable at temperatures ranging from 60℃ to 120℃, particularly temperatures ranging from 90℃ to 120℃, even more particularly temperatures ranging from 95℃ to 120℃.

[0130] In one embodiment the composition further comprises a carrier, which may comprise one or more of the following compounds: water, glycerol, ethylene glycol, 1, 2-propylene glycol or 1, 3-propylene glycol, sodium chloride, sodium benzoate, potassium sorbate, sodium sulfate, potassium sulfate, magnesium sulfate, sodium thiosulfate, calcium carbonate, sodium citrate, dextrin, maltodextrin, glucose, sucrose, sorbitol, lactose, wheat flour, wheat bran, corn gluten meal, starch, cellulose farigel, cassava cores, sodium aluminium silicate, colloidal amorphous silica,  50S, polyethylene glycol 200, polyethylene glycol 400, polyethylene glycol 600, polyethylene glycol 1000, polyethylene glycol 1500, polyethylene glycol 4000 and / or

[0131] The compositions of the invention can be manufactured by methods known in the art, for example as a mash composition (non-pelleted) or as pelleted composition. The one or more Bacillus strains and optionally other components can be added as solid or liquid formulations. For example, for mash composition a solid or liquid culture formulation may be added before or during the ingredient mixing step. Typically, a liquid culture preparation comprises the one or more Bacillus strains of the invention optionally together with a polyol, such as glycerol, ethylene glycol or propylene glycol, and is added to pellets after the pelleting step, such as by spraying the liquid formulation onto the pellets.

[0132] A solid formulation may for example be in the form of a granule, a powder such as a spray dried powder, pellets or an agglomerate (e.g. as disclosed in WO 2000 / 70034) . The formulating agent may comprise a salt (organic or inorganic zinc, sodium, potassium or calcium salts such as calcium acetate, calcium benzoate, calcium carbonate, calcium chloride, calcium citrate, calcium sorbate, calcium sulfate, potassium acetate, potassium benzoate, potassium carbonate, potassium chloride, potassium citrate, potassium sorbate, potassium sulfate, sodium acetate, sodium benzoate, sodium carbonate, sodium chloride, sodium citrate, sodium sulfate, zinc acetate, zinc benzoate, zinc carbonate, zinc chloride, zinc citrate, zinc sorbate or zinc sulfate) , starch or a sugar or sugar derivative (such as sucrose, dextrin, glucose, lactose or sorbitol) .

[0133] In one embodiment, the composition may be a solid composition in the form of a powder, for example mixed with a salt such as calcium carbonate and a suitable amount of a vegetable oil. Examples of vegetable oils include soybean oil, rapeseed oil, sunflower seed oil and flax seed oil. Alternative solid forms include, for example, pelletized compositions, granulate compositions and incorporation in feed pellets.

[0205] A total of 64 mixed-sex piglets weaned at 21d of age (day 0) with initial body weight (BW) of 6.76±0.45kg were assigned to one of two treatments in a randomized block design, with eight replicates per treatment and four piglets per pen for 42 days (day 0 to day 42) . During the suckling period, the piglets did not receive any dietary supplement besides sows’ milk. After weaning, piglets were fed with corn-soybean meal-based diets, formulated to meet the nutritional requirements during the two phases of the nursery period (day 0 to day 14 and day 15 to day 42) , as recommended by NRC (Nutritional Research Council) 2021. The treatments consisted of a control group and a 0.05%Bacillus subtilis SB3295 treatment group. The probiotic product (Bacillus subtilis SB3295) provides a spore count of 2x109 CFU / g.

[0206] Piglets had free access to feed and water throughout the experimental period. During the trial, feed was weighed before feeding and feed wastage and leftovers were collected and weighed daily to determine average daily feed intake. Piglets were individually weighed on days 0 and 42 of the experimental period to determine BW, average daily gain (ADG) and feed conversion ratio (FCR) . On day 42 , one piglet with BW closest to the pen average from each treatment was selected for blood collection via venipuncture in orbital sinus into 5 ml uncoated vacuum tubes, and the blood was centrifuged to separate serum. Serum was stored at -20℃ until analysis by total antioxidant capacity (T-AOC) and catalase (CAT) .

[0207] The growth performance results are summarized in Table 4 below.

[0208] Table 4. Growth performance of piglets

[0209] Data analyzed by ANOVA compared to the control:

[0210] **means differs (P<0.01) from the control

[0211] The key findings of this study are the following:

[0212] · The Bacillus subtilis SB3295 treatment resulted in 13.5%higher body weight gain and 5.9%lower FCR compared to the control group.

[0213] · The mortality rate decreased by 50%in the Bacillus subtilis SB3295 treatment group compared to the control group.

[0214] The Bacillus treatment also resulted in a reduction in diarrhea as summarized in Table 5 below:

[0215] Table 5. Diarrhea reduction in piglets

[0216] The key findings in relation to diarrhea as as follows:

[0217] · The diarrhea incidence (percentage of days with diarrhea) decreased by 33.3%in the Bacillus subtilis SB3295 treatment group compared to the control group.

[0218] · The Bacillus subtilis SB3295 treatment resulted in 11.7%higher T-AOC and 202%higher CAT compared to the control group.

[0219] The present study indicates that Bacillus subtilis SB3295 can significantly enhance the health and production efficiency of piglets. The experimental data demonstrate that Bacillus subtilis SB3295 achieves the following beneficial effects: (1) improved growth performance, evidenced by increased body weight gain and optimized feed conversion ratios; (2) reduced incidence of diarrhea, which is believed to be obtained by stabilizing intestinal microbiota and inhibiting pathogenic colonization; and (3) mitigated systemic oxidative stress by modulating antioxidant enzyme activity and reducing lipid peroxidation markers. This innovation offers a sustainable, antibiotic-free solution to swine industry challenges, with statistically validated efficacy in large scale trials.

[0221] Feces of late nursey feed piglets were collected from a pig farm and transported to a lab at a temperature below 4℃. The feces were mixed once they arrived at the lab. 50 g of feces were dissolved in 156 ml 0.1 M PBS (32%w / v) to prepare a fecal slurry. 16 ml of medium (beef extract powder 2.4 g / L, peptone 20 g / L, L-cysteine-HCl 0.6 g / L, glucose 2.5 g / L, yeast extract powder 5 g / L, sodium chloride 5 g / L, soluble starch (from corn) 1 g / L, and molasses 5 g / L; cysteine is sensitive to temperature, so it is added to the medium after the medium has been autoclaved) and 4 ml fecal slurry (20%v / v) were added to a 100 ml serum bottle.

[0222] Single colonies of Bacillus subtilis SB3295 were inoculated to LB broth and incubated at 37℃ for one day at 180 rpm. The fermentation broth was tested at OD600, and the inoculated fermentation broth was calculated based on CFU numbers. The final CFU number inoculated to the bottle was 1.28×107 CFU / bottle. The procedure was repeated in triplicate. Bottles were sealed with an aluminum lid and a syringe to create a vacuum (removal of 40 ml of air) and placed at 37℃ with oscillation at 30 rpm for a 24h fermentation period.

[0223] Once incubation had finished, the bottles were submerged in ice for 15 min, and aliquots were taken as needed at the end of the 15 min. 10 ml of fermentation broth was sent directly to Azenta Life Sciences for 16S rRNA sequencing. The sequencing found effects of the Bacillus strain on five key bacterial genera, namely three beneficial genera (Prevotella, the Prevotellaceae_NK3B31 group, Succinivibrio) and two harmful genera (Streptococcus, Escherichia-Shigella) . The results for the beneficial bacteria are shown in Figure 1, and the results for the harmful bacteria are shown in Figure 2.

[0224] The significance of these genera is briefly discussed below.

[0225] Prevotella: Prevotella is one of the most predominant genera across the large intestine of pigs. The Prevotella-driven enterotype has shown positive associations with animal traits including feed intake, feed efficiency, weight gain and incidence of diarrhea, suggesting that Prevotella is important in mediating growth performance and disease resilience in pigs.

[0226] Prevotellaceae_NK3B31 group: The Prevotellaceae_NK3B31 group can effectively alleviate intestinal inflammatory responses, promote nutrient absorption in the gut, and reduce immune rejection in autoimmune diseases.

[0227] Succinivibrio: In swine, the genus Succinivibrio has been considered as a member of the core microbiome of the proximal colon, or cecum, and can metabolize various carbohydrate sources, resulting in fermentation products such as acetate and succinate, forming propionate in the further decarboxylation process.

[0228] Streptococcus: Among the bacterial group of gram-positive cocci comprising the genera Streptococcus, Enterococcus and Peptostreptococcus, streptococci constitute the most consequential pathogens in swine.

[0229] Escherichia-Shigella: Escherichia-Shigella encompasses species of swine enteric pathogens of the genera Escherichia and Shigella. These are believed to play an important role in the development of diarrhea in piglets.

[0230] The key findings of the 16s rRNA sequencing were as follows:

[0231] Bacillus subtilis SB3295 supplementation was found to regulate the intestinal microbiota of piglets, increasing the content of beneficial bacteria related to pig growth and health (Prevotellaceae_NK3B31_group, Prevotella, Succinivibrio) while reducing the content of harmful bacteria that may induce diseases (Escherichia-Shigella, Streptococcus) .

[0232] The Bacillus subtilis 3295 strain was thus able to provide a beneficial effect on the intestinal microbiota of piglets, increasing the content of beneficial bacteria and reducing the content of harmful bacteria.

[0233] Example 6: Animal trial to measure performance of Bacillus strains

[0234] For this trial, 360 1-day old Ross 308 broiler chicks with similar body weight were purchased from Xiangjia hatchery (Hunan, China) . Chicks were weighed and divided into 2 treatment groups. Each group consisted of 9 replicates with 20 birds (10 males and 10 females) per replicate with a total of 27 floor pens (area of 2 x 4m) covered with fresh wood shavings. Birds were given ad libitum access to water and food throughout the experiment of 35 days. Chicks were fed with diets in pellet form as follows: a control group, and a treatment group receiving Bacillus subtilis SB3295. All diets were corn and soybean meal based diets, formulated to meet the nutritional requirements during the two phases of the nursery period (day 0 to day 14, and day 15 to day 35) , as recommended by NRC. The probiotic product (Bacillus subtilis SB3295) provided a spore count of 2x109 CFU / g.

[0235] During the trial, feed was weighed before feeding, and feed wastage and leftovers were collected and weighed daily to determine average daily feed intake. Feed intake and body weight of broilers was measured per replicate as a unit (i.e. per replicate of 20 chicks, calculating the average body weight) on day 1 and day 35. Mortality was monitored daily. Dead birds were recorded and weighed to adjust the estimates of gain, intake and feed conversion ratios as appropriate. The average body weight gain, feed intake (FI) and feed: gain ratio (FCR) were calculated.

[0236] The growth performance results are summarized in Table 6 below:

[0237] Table 6. Growth performance of broilers

[0238] Data analyzed by ANOVA:

[0239] **means differ (P<0.01) from each other

[0240] During the total trial from day 1 to day 35, there was a significant effect on body weight gain and FCR of the treatment group compared with control group (P<0.01) . There was a lower mortality rate in the Bacillus subtilis SB3295 treatment group in this trial, although it was not statistically significant. In conclusion, the results showed that a Bacillus subtilis SB3295 diet supplementation can support broiler performance by increasing body weight gain and reducing FCR.
