[0006] Human interleukin-2 (IL-2) muteins or variants thereof are provided herein. In particular, provided are IL-2 muteins that have a decreased binding capacity for IL-2Rβγreceptor. Such IL-2 muteins find use, for example, as IL-2 partial agonists and antagonists in applications where reduction or inhibition of one or more IL-2 and / or IL-15 functions is useful (e.g., cancer) . Also provided are nucleic acids encoding such IL-2 muteins, methods of making such IL-2 muteins, pharmaceutical compositions that include such IL-2 muteins and methods of treatment using such IL-2 muteins. Also provided are fusion proteins that have decreased toxicity and have pharmacological activity.

[0281] The affinities of the generated IL-2-Fc fusions to human IL-2Rβγ (ACRO Biosystems, Cat#: ILG-H5283) and mouse IL-2Rβγ (ACRO Biosystems, Cat#: ILG-M5283) were measured. Briefly, a streptavidin (SA) reagent kit (Sartorius, Cat#: 18-5019) was used to capture the fusion proteins at a final concentration of 5 μg / mL to the surface of the SA biosensor. Thereafter, the sensor was placed in IL-2Rβγ solutions with serially diluted concentrations (the highest concentration being 200 nM, 2-fold gradient dilution, 7 concentrations in total) for 4 minutes. After the binding phase, the sensors were transferred to 0.02%PBST (PBS supplemented with 0.02% 20) for dissociation for 10 minutes. Kinetic association rates (ka) and dissociation rates (kdis) were obtained simultaneously by fitting the data globally to a 1: 1 Langmuir binding model (Karlsson, R. Roos, H. Fagerstam, L. Petersson, B., 1994. Methods Enzymology 6.99-110) using Data Analysis HT 12 software. Affinities were deduced from the equation

[0289] Example 3: Capability of monovalent N-terminal IL-2 fusion and bivalent C-terminal IL-2 fusion in inducing pSTAT5 phosphorylation

[0290] The capability of the IL-2 Fc fusions to induce STAT5 phosphorylation was evaluated. Cryopreserved human peripheral blood mononuclear cells (PBMCs) were thawed and resuspended in PBS at a density of 5 × 106 cells / mL. Cells were stained with Fixable Viability Stain 510 (FVS510; BD Biosciences, 1: 1000 dilution) for 15 min at room temperature (RT) , washed once with complete RPMI-1640 medium (Gibco, supplemented with 10%FBS and 1%penicillin-streptomycin) , and adjusted to 3 × 106 cells / mL.

[0291] PBMCs (1.5 × 105 cells / well in 50 μL) were seeded into 96-well V-bottom plates (Corning #3897) and treated with 50 μL of serially diluted test wild type and mutant IL-2 molecules (10-point dilution series, starting at 100 nM) . After 25 min incubation at 37℃ / 5%CO2, reactions were terminated by adding 100 μL pre-warmed fixation buffer (BD Biosciences #554655; 37℃, 12 min) . Cells were centrifuged (900 × g, 7 min) and permeabilized with 100 μL 1× Perm III buffer (BD Biosciences #558050) for 30 min on ice.

[0318] High dose IL-2 therapy is associated with severe toxic side effects. Similar effects were also observed in mice. Administration of IL-2 causes death of mice at doses intolerable to the animals, usually shortly after the second weekly dosing. This observation provides an easy and convenient means to evaluate the toxicity of IL-2 constructs.

[0319] H16W and V91Y muteins of IL-2 were found to reduce its binding to IL-2Rβγ and its capability in inducing STAT5 phosphorylation. Monovalent, N terminal and bivalent C-terminal fusions of IL-2-Fc, as well as their H16W and V91Y versions were tested for causing death of C57 mice. In some cases R38L / F42A mutations (designated laIL-2) with reduced binding to IL-2Rα were also incorporated.

[0320] The 10 constructs listed in FIG. 9C were i. v. injected into C57 mice at indicated doses, two mice at each dose for each construct, on Day 0 and D7, and animal deaths were recorded on Day 8. For each construct, the highest doses that did not cause animal death were recorded as highest non-lethal dose (NLD) (FIGS. 9A-9C) .

[0324] Interestingly, the R38L / F42A mutations, which slightly increased the toxicity of IL-2-Fc fusions in monovalent N-terminal format, reduced the toxicity in the bispecific, C-terminal fusion format. When comparing the wildtype IL-2 versions in regard to IL-2Rβγ binding, the low IL-2Rα binding (la-IL-2) version with R38L / F42A mutant increased the highest NLD by above 33 fold. When comparing the H16W IL-2 versions in regard to IL-2Rβγ binding, the la-IL-2 version with R38L / F42A mutations increased the highest NLD by above 9 fold.

[0325] These results suggest that IL-2 toxicity can be reduced by inducing low affinity IL-2Rβγ binding and low affinity IL-2Rα binding mutations. Furthermore, toxicity reduction by both low affinity IL-2Rβγ and IL-2Rα binding mutations are format dependent. Toxicity reduction is more evident in bispecific, C-terminal IL-2-Fc fusions.
