Abstract

[0001] A PERSONAL CARE COMPOSITION

[0002] Field of the invention

[0003] The present invention relates to a personal care composition. Particularly, the present invention relates to a personal care composition that provides protection from sun-induced damages.

[0004] Background of the invention

[0005] Solar radiation includes about 5% ultraviolet (UV) radiation, wavelength of which is between 200 nm and 400 nm. It is further divided into three regions: from 320 to 400 nm (UV-A), 280 to 320 nm (UV-B) and from 200 to 280 nm (UV-C). Substantial portion of UV-C radiation is absorbed by ozone. Scientific studies have indicated that exposure to UV-A and UV-B radiation for short period causes pigmentation, reddening of the skin and localized irritation, whereas continued or prolonged exposure can lead to sunburn, melanoma, formation of wrinkles and age spots. UV radiation is also thought to cause significant damage to hair. Studies have also indicated that exposure to UV also leads to generation of reactive oxygen species (ROS) and thereby exert oxidative stress. Therefore, it is desirable to protect the skin and other keratinous substrates of the human body from such harmful effects of both UV-A and UV-B radiation.

[0006] On the other hand, solar radiations also include visible spectrum, wavelength of which is between 400 to 700 nm which includes blue light (about 450 to 495 nm). Blue light amongst the visible spectrum is also reported to induce undesired effects e.g. ROS generation.

This is achieved through a composition comprising a combination of an oil-soluble sunscreen, a water-soluble sunscreen, a non-ionic surfactant having an HLB value in the range of 9 to 20, fatty acid, humectant; and a polymer selected from one of more of dimethicone / vinyldimethicone crosspolymer and hydrophobically modified silica.

[0011] However, a need still exists to provide compositions that protect the skin from sun-induced damages, particularly from oxidative stress / reactive oxygen species generation. Such ROS generation may be due to exposure to UV light or blue light.

[0012] It is thus an object of the present invention to provide for a personal care composition that provides protection from sun-induced damages, particularly from oxidative stress I generation of reactive oxygen species.

[0013] It is another object of the present invention to provide such personal care composition that, in addition, delivers desired sensorial attributes when applied on the skin e.g. moisturization, matte feel, easy spreadability and gentle on skin. It has now been found that when a personal care composition comprising a skin care benefit agent selected from acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, or a salt, or mixtures thereof, were combined with an oil-soluble sunscreen, a -water soluble sunscreen and non-ionic surfactant having select HLB value and select polymers, protection from sun-induced damages, particularly from oxidative stress / ROS generation, was obtained. In addition, the desired sensorial attributes mentioned above were also obtained.

[0015] Accordingly, in the first aspect, the present invention relates to a personal care composition comprising:

[0016] (i) 0.0001 to 10 wt% of a skin care benefit agent selected from acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, or a salt, or mixtures thereof,

For the avoidance of doubt, any feature of one aspect of the present invention may be utilized in any other aspect of the invention. The word "comprising" is intended to mean "including" but not necessarily "consisting of" or "composed of." In other words, the listed steps or options need not be exhaustive. It is noted that the examples given in the description below are intended to clarify the invention and are not intended to limit the invention to those examples per se. Unless specified otherwise, amounts as used herein are expressed in percentage by weight based on total weight of the composition and may be abbreviated as “wt%”. [0025] The composition of the invention is meant to be used for personal care or for cosmetic use and could also be referred to as a cosmetic composition. By “personal care composition” as used herein, is meant to include a composition for topical application to sun-exposed areas of the skin and / or hair of humans. Such a composition may be classified as leave-on or rinse off (also known as wash-off), and includes any product applied to a human body for improving appearance, cleansing, odor control or general aesthetics. The composition of the present invention can be in the form of a liquid, a lotion, a cream, foam, a stick or a gel. Non-limiting examples of such leave-on personal care compositions include skin lotions, creams, antiperspirants, deodorants, foundations, mascara, sunscreen lotions; and that of wash-off personal care compositions include face wash, hand I body wash compositions, shampoos, conditioners and shower gels. The composition of the present invention is preferably a leave- on composition. "Skin" as used herein is meant to include skin on the face and body (e.g., neck, chest, back, arms, underarms, hands, legs and scalp) and especially to the sun exposed parts thereof. Compositions of the invention are also of relevance to applications on any other keratinous substrates of the human body other than skin e.g. hair, where products may be formulated with the aim of improving photoprotection.

[0033] A composition as per the present invention (the composition) comprises a skin care benefit agent selected from acetyl glutamic acid (e.g., N-acetyl glutamic acid), acetyl glutamine (e.g., N-acetyl glutamine), acetyl methionine (e.g., N-acetyl methionine), acetyl tyrosine (e.g., N- acetyl tyrosine), and combinations thereof, including as a free acid, amide, ester, lactone, isomer, and any derivatives or salts thereof. Preferably, the skin care benefit agent is selected from acetyl methionine, or a derivative or a salt or mixture thereof. More preferably, acetyl methionine is selected as the skin care benefit agent. The term “derivative(s)” as used with reference to the skin care benefit agent as used herein refers to an amide, ester, ether, carboxyl, amino, acetyl, and / or alcohol forms of the particular ingredient. “Salts,” as used herein, includes but is not limited to ammonium, calcium, magnesium, manganese, potassium, sodium, tetraalkylammonium, trialkylammonium, and a combination thereof.

By ‘oil-soluble’, it is meant those organic sunscreens which have solubility in water less than 10 g / l, preferably less than 5 g / l, more preferably less than 2 g / l, at 25°C. Oil-soluble sunscreens could be of IIV-B type or of IIV-A type.

Some of the well known brands under which the above sunscreens are sold are Octisalate®, Homosalate®, Neo Heliopan®, Neo Heliopan® AV, Neo Heliopan® OS, Octocrylene® and Parsol® MCX. The oil-soluble UV-B sunscreen has Amax from 280 to 320 nm.

When included, it is selected from one or more of a dibenzoylmethane compound, diethylamino hydroxybenzoyl hexyl benzoate and menthyl anthranilate. Preferably, a water-soluble sunscreen is selected from a UV-A water-soluble sunscreen and / or a UV-B water-soluble sunscreen described below. By ‘water soluble’, it is meant those organic sunscreens which have solubility in water higher than 10 g / l, preferably higher than 50 g / l, at 25°C.

[0046] The preferred water-soluble UVB sunscreen for use in the present invention is phenyl benzimidazole sulphonic acid (PBSA), benzophenone-4 or benzylidene camphor sulfonic acid, preferably PBSA. [0051] The composition of the invention comprises a non-ionic surfactant having an HLB value in the range of 9 to 20. More preferably the nonionic surfactant is selected from those having HLB value in the range 12 to 20, further more preferably 12 to 18.

Preferred are emollients that can be used, especially for products intended to be applied to the face, to improve sensory properties and are chosen from the group of oils that do not form stiff gels with 12HSA; these include polypropylene glycol-14 butyl ether otherwise known as Tegosoft® PBE, or PPG15 stearyl ether such as Tegosoft® E, other oils such as esters, specifically, isopropyl myristate, isopropyl palmitate, other oils could include castor oils and derivatives thereof.

The CTFA Cosmetic Ingredient Handbook, Second Edition, 1992, which is incorporated by reference herein in its entirety, describes a wide variety of non-limiting cosmetic and pharmaceutical ingredients commonly used in the skin care industry, which are suitable for use in the compositions of the present invention. Examples include antioxidants, binders, biological additives, buffering agents, colorants, astringents, fragrance, opacifying agents, conditioners, exfoliating agents, pH adjusters, essential oils, skin sensates, skin soothing agents and skin healing agents.

[0113] Preparation of compositions

[0114] The compositions of examples A to C (control) and a composition of example 1 (as per the present invention), as shown in table 1 below, were prepared as follows:

[0115] The polymers Ultrez 10 and xanthan gum were pre-dispersed in water in a separate beaker. The oil phase ingredients such as fatty acid mix, non-ionic surfactant, emollients, and oilsoluble sunscreens were added to a glass beaker and melted at 75°C. If needed, the sensory agents DC 9701 and Ganzpearl 820 were pre-dispersed in the molten oil phase before adding to emulsification process. The water-soluble sunscreens were pre-neutralized in a separate beaker using alkali (NaOH or KOH) in water. In the main container, glycerine and water were homogenized at 75°C. The pre-neutralized water-soluble sunscreens were added and were homogenized. After homogenization, the molten oil phase was added to the main container and homogenization was continued. After complete mixing, the pre-dispersed polymers were added and neutralized as required. After complete mixing, other ingredients e.g. Acemet, that was pre-neutralized with 0.1 N KOH, was added at 45°C.

[0116] Cell culture and exposure to UV or blue light

[0117] Keratinocytes (HaCaT cells) were seeded in 48-well plates at a density of 3.5 x 104cells / well and were maintained in the DMEM medium reconstituted as described above, for 24 to 48 h. The cells were exposed to UV light using a Solar simulator comprising a Xenon arc lamp and to blue light using LED source, as required. Cells that were not exposed either to UV or to blue light, were used as control. Prior to exposure, compositions (as shown in table 1 below) were applied evenly on a PMMA plate (2 mg / cm2). For the UV / Blue light exposure, the media was carefully removed, and the cells were gently washed with sterile 1X Phosphate buffered saline (PBS). Exposure was carried out with a PMMA plate on top of the culture plate with a lid and minimal levels of 1X PBS coating the cell layer. After exposure, PBS was removed and replaced with the media in all wells. To mimic the penetration of either Acemet or N-acetyl glutamic acid or N-acetyl tyrosine into the skin, cells were treated with media containing either Acemet or N-acetyl glutamic acid or N-acetyl tyrosine, as shown in tables below. Plates were incubated in 5% CC>2 / 37OC incubator for 6 h and post incubation, cells were harvested and measured ROS level by DCFDA assay.

[0119] Post 6 h, cells were washed with 1X PBS and treated with media containing DCFDA reagent and incubated at 37°C in the CO2 incubator for 30 minutes. Then cells were washed with 1x PBS once followed by the addition of fresh PBS. The plates were then wrapped with aluminum foil and taken reading at an excitation wavelength of 485 nm and emission wavelength of 535 nm. The amount of ROS generated was calculated as percentage control after background subtraction.

[0121] The invitro SPF and UVAPF values for the samples shown in table 4 below were measured using the following procedure:

[0122] Thin film transmittance measurements were done using Labsphere’s Ultraviolet transmittance analyzer. In this study, 70 x 70 mm PMMA plate with ~6 pm roughness from Schonberg GmbH & Co were used. The percent transmittance of the various compositions was measured using a procedure as outlined below. 2 mg / cm2of sample was applied on PMMA plate, distributed uniformly using a syringe, and spread uniformly. The drying time for the PMMA plates was 30 minutes. After the drying time, sample plates were exposed to UV light and transmittance scan was recorded. This scan gives the transmittance as a function of wavelength (290 - 400 nm) for a given sample. For a single plate six to nine different spots were scanned. The same was repeated for 2 or 3 plates. The data reported is thus an average over 12 -18 readings. The reference transmittance scan was obtained using a blank plate, with no sample on the PMMA plates with glycerin spread on it. The transmittance values were used to arrive at the SPF values using the UV -2000s application provided with the instrument.

Claims

Claims1. A personal care composition comprising:(i) 0.0001 to 10 wt% of a skin care benefit agent selected from acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, or a salt, or mixtures thereof,(ii) 0.1 to 25 wt% of an oil-soluble sunscreen,(iii) 0.1 to 5 wt% of a water-soluble sunscreen,(iv) 0.1 to 5 wt% of a non-ionic surfactant having an HLB value in the range of 9 to 20, wherein the composition further comprises 0.25 to 5 wt% of a polymer selected from one or more of (a) dimethicone / vinyldimethicone crosspolymer and b) hydrophobically modified silica, wherein solubility of the oil-soluble sunscreen is less than 10 g / l, in water, at 25°C; and wherein solubility of the water-soluble sunscreen is higher than 10 g / l, in water, at 25°C.

2. The composition as claimed in claim 1 wherein the skin care benefit agent is selected from acetyl methionine, or a derivative or a salt or mixtures thereof.

3. The composition as claimed in claims 1 or 2 wherein the skin care benefit agent is acetyl methionine.

4. The composition as claimed in any one of claims 1 to 3 wherein the composition further comprises 3 to 25 wt% of a C10-C22 fatty acid.

5. A composition as claimed in claim 4 wherein the fatty acid is selected from one or more of stearic acid and palmitic acid, preferably a mixture of stearic acid and palmitic acid.

6. The composition as claimed in any one of claims 1 to 5 wherein the composition further comprises 0.1 to 10 wt% humectant.

7. A composition as claimed in any one of claims 1 to 6 wherein the oil-soluble sunscreen is chosen either from a IIV-B sunscreen selected from one or more of octyl salicylate, 3,3,5-trimethylclohexyl 2- hydroxy benzoate, ethylhexyl salicylate, 2-ethylhexyl 2-cyano- 3,3-diphenyl-2-propenoate, 2-ethylhexyl-4-methoxycinnamate andoctylmethoxycinnamate; and / or from a IIV-A sunscreen selected from one or more of a dibenzoylmethane compound, diethylamino hydroxy benzoyl hexyl benzoate and menthyl anthranilate.

A composition as claimed in claim 7 wherein the dibenzoylmethane compound is selected from one or more of t-butylmethoxydibenzoylmethane, 2- methyldibenzoylmethane, 4-methyl-dibenzoylmethane, 4-isopropyldibenzoyl-methane, 4-tert-butyldibenzoylmethane, 2,4-dimethyldibenzoylmethane, 2,5- dimethyldibenzoylmethane, 4,4'-diisopropyl-dibenzoylmethane, 2-methyl-5-isopropyl- 4'-methoxydibenzoylmethane, 2-methyl-5-tert-butyl-4'-methoxy-dibenzoyl methane, 2,4-dimethyl-4'-methoxy dibenzoylmethane and 2,6-dimethyl-4-tert-butyl-4'-methoxy- dibenzoylmethane.

9. A composition as claimed in any one of claims 1 to 8 wherein the water-soluble sunscreen is either chosen from a IIV-B sunscreen selected from one or more of 2- phenylbenzimidazole-5-sulfonic acid, benzophenone-4 and benzylidene camphor sulfonic acid; and / or from a water-soluble IIV-A sunscreen selected from one or more of di sodium phenyl dibenzimidazole tetra sulfonate and terephthalylidene dicamphor sulfonic acid.

10. A composition as claimed in any one of claims 1 to 9 wherein the non-ionic surfactant has an HLB value in the range of 12 to 20.

11. A composition as claimed in any one of claims 1 to 10 wherein the non-ionic surfactant is selected from one or more of a fatty alcohol ethoxylate, fatty acid ethoxylate, alkyl phenol ethoxylate, and polyoxyethylene sorbitan alkyl esters; preferably a fatty alcohol ethoxylate or a fatty acid ethoxylate.

12. A method of protecting the skin from sun-induced damages: a. applying the composition as claimed in any one of claims 1 to 11 on to the skin; and b. optionally, rinsing the skin with water.

13. Use of the composition as claimed in any one of claims 1 to 11 for providing protection from sun-induced damages.

14. Use of a skin care benefit agent selected from acetyl glutamic acid, acetyl glutamine, acetyl methionine, acetyl tyrosine, or a derivative, or a salt, or mixtures thereof, in a personal care composition comprising an oil soluble sunscreen, a water-soluble sunscreen and a nonionic surfactant having HLB value in the range from 9 to 20, wherein the composition further comprises 0.25 to 5 wt% of a polymer selected from one or more of (a) dimethicone / vinyldimethicone crosspolymer and b) hydrophobically modified silica, for protecting skin from sun-induced damages.
