A method for producing immuno-tolerant hair follicle stem cells (HFSCs) for enhanced hair implantation therapy, comprising: a. isolating and culturing HFSCs characterized by CD105 expression and CD45 absence; b. establishing a biosensor system in the HFSCs by CRISPR / Cas9 knock-in to link CD105 to GFP (green fluorescence for undifferentiated HFSCs) and CD45 to RFP (red fluorescence for differentiated cells); c. selecting undifferentiated HFSCs based on green fluorescence emission using fluorescence-activated cell sorting (FACS) to remove differentiated red fluorescent cells; d. expanding the selected green fluorescent undifferentiated HFSCs while maintaining the biosensor system for real-time sternness monitoring and quality control; e. genetically modifying the expanded HFSCs using CRISPR / Cas9 to create HLA-A / B / C triple knockout HFSCs by targeting HLA-A, HLA-B, and HLA-C genes; f. confirming the HLA-A / B / C triple knockout by genomic sequencing; g. expanding single-clone HLA-A / B / C triple knockout HFSCs with the integrated biosensor system; and h. evaluating hair follicle growth potential of the modified HFSCs in vitro (differentiation into osteocytes and adipocytes) and in vivo (mouse model implantation).

2. The method of claim 1 , wherein undifferentiated HFSCs are selectively expanded based solely on green fluorescence emission during FACS, ensuring maintenance of sternness purity and preventing differentiation in large-scale production.

3. The method of claim 1 , wherein the biosensor system enables real-time monitoring of cell populations, with only green fluorescent cells expanded to preserve regenerative capabilities and immuno-tolerant properties of HFSCs.

4. The method of claim 1 , wherein the genetic modification in step (e) uses designed and synthesized guide RNAs (gRNAs) targeting exon regions of HLA-A, HLA-B, and HLA-C genes for precise triple knockout.

5. The method of claim 4, wherein the gRNAs comprise sequences targeting: o(SEQ ID NO: 1) or equivalent; o(SEQ ID NO: 2) or equivalent;HLA-C or shared:(SEQ ID NO: 3) or equivalent, achieving at least 85-95% knockout efficiency as confirmed by sequencing and reduced surface HLA expression.

6. The method of claim 1 , wherein the HLA-A / B / C triple knockout reduces immune rejection risk, enabling universal donor HFSCs without donor dependency or large scars in hair implantation therapy.

7. The method of claim 1 , further comprising optimizing protocols for large-scale production of HLA-A / B / C triple knockout HFSCs with integrated biosensor system.

A system for producing immuno-tolerant hair follicle stem cells (HFSCs) for hair implantation therapy, comprising: a. a culture medium optimized for HFSC isolation, culture, and expansion; b. a CRISPR / Cas9 gene-editing toolkit for: (i) knock-in of a biosensor system linking CD105 to GFP and CD45 to RFP, and (ii) triple knockout of HLA-A / B / C genes; c. a biosensor system configured to select and monitor undifferentiated HFSCs via green fluorescence emission during FACS and expansion; d. a fluorescence-activated cell sorting (FACS) setup for selecting green fluorescent undifferentiated HFSCs; e. a genomic sequencing setup to confirm HLA- A / B / C triple knockout and validate single-clone expansion stability; and f. in vitro and in vivo assay setups to evaluate hair follicle growth potential of the modified HFSCs.

9. The system of claim 8, wherein the biosensor system includes CD105 linked GFP for green fluorescence in undifferentiated HFSCs and CD45 linked RFP for red fluorescence in differentiated cells.

10. The system of claim 8, wherein the biosensor system facilitates high-throughput FACS based on green fluorescence emission to maintain sternness purity during expansion.
