Description

2-PHENYL-1,2,4-TRIAZINE-3,5(2H,4H)-DIONE DERIVATIVES AS THR-BETA AGONIST OR MODULATORS FOR THE TREATMENT OF OBESITYCross-reference to related applicationsThe present disclosure claims the benefits of the PCT application No. PCT / CN2024 / 122793 entitled “ATHR-β AGONIST OR MODULATOR AND METHODS OF USING THEREOF” filed September 30, 2024, PCT application No. PCT / CN2024 / 129959 entitled “ATHR-β AGONIST OR MODULATOR AND METHODS OF USING THEREOF” filed November 5, 2024, PCT application No. PCT / CN2025 / 072969 entitled “ATHR-β AGONIST OR MODULATOR AND METHODS OF USING THEREOF” filed January 17, 2025, which are incorporated herein by reference in their entireties.FIELDThe present disclosure generally relates to a THR-β agonist or modulator and using the same.BACKGROUNDThyroid hormones are critical for normal growth and development and for maintaining metabolic homeostasis (Paul M. Yen Physiological Review, Vol. 81 (3) : pp. 1097-1126 (2001) ) . Circulating levels of thyroid hormones are tightly regulated by feedback mechanisms in the hypothalamus / pituitary / thyroid (HPT) axis. Thyroid dysfunction leading to hypothyroidism or hyperthyroidism clearly demonstrates that thyroid hormones exert profound effects on cardiac function, body weight, metabolism, metabolic rate, body temperature, cholesterol, bone, muscle and behavior. The thyroid hormone receptors (THR) are derived from two separate genes, α and β. These distinct gene products produce multiple forms of their respective receptors through differential RNA processing. The major thyroid hormone receptor isoforms are α1, α2, β1 and β2. Thyroid hormone receptors α1, β1 and β2 bind thyroid hormone. It has been shown that the thyroid hormone receptor (THR) subtypes can differ in their contribution to particular biological responses. Recent studies suggest that THR-β1 plays an important role in regulating TRH (thyrotropin releasing hormone) and on regulating thyroid hormone actions in the liver. THR-β2 plays an important role in the regulation of TSH (thyroid stimulating hormone) . THR-β1 plays an important role in regulating heart rate.SUMMARYOne aspect of the present disclosure provides a compound of Formula (I) ,or an isomer, a pharmaceutically acceptable salt, a deuterated compound, a prodrug thereof, or a metabolite thereof,wherein,each of X1 and X2 is independently selected from the group consisting of methyl, ethyl, -Cl, -Br, and -I;G is selected from the group consisting of -CH2-and -O-;or G and X1 together with carbon atoms to which they are attached form C3-6 cycloalkyl, wherein C3-6 cycloalkyl is optionally substituted with one or more substituents independently selected from the group consisting of H, halogen and C1-6 alkyl;each of R1 and R2 is independently selected from the group consisting of H, C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl, C0-6 alkyl-aryl, C0-6 alkyl-cycloalkyl, C0-6 alkyl-heterocycloalkyl and C3-8 cycloalkyl; wherein each of R1 and R2 is optionally substituted with one or more substituents independently selected from the group consisting of H, halogen, C1-6 alkyl, C1-6 haloalkyl, C2-C6 alkenyl, C3-6 cycloalkyl, 5-to 10-membered heterocycle, aryl and heteroaryl;or R1 and R2 together with carbon atoms to which they are attached form C3-15 carbocyclic ring, wherein C3-15 carbocyclic ring is optionally substituted with one or more substituents independently selected from the group consisting of H, halogen, C1-6 alkyl, C1-6 haloalkyl, C2-6 alkenyl, C3-6 cycloalkyl, 5-to 10-membered heterocycle, aryl and heteroaryl;R3 is selected from the group consisting of H and -COR5; wherein R5 is selected from the group consisting of C1-30 alkyl, C2-30 alkenyl and C2-30 alkynyl; andR4 is selected from the group consisting of -NH2 and-C≡CH.Another aspect of the present disclosure provides a pharmaceutica composition comprising the compound, the isomer, the pharmaceutically acceptable salt, the deuterated compound, the prodrug, or the metabolite thereof of any one of the present disclosure, and a pharmaceutically acceptable excipient.Another aspect of the present disclosure provides a method for treating or preventing obesity, hyperlipidemia, hypercholesterolemia, diabetes mellitus, nonalcoholic steatohepatitis (NASH) , hepatic steatosis, arteriosclerosis, cardiovascular disease, hypothyroidism, overweight, fibrosis and thyroid cancer, comprising administering to a subject in need thereof an effective amount of the compound, the isomer, the pharmaceutically acceptable salt, the deuterated compound, the prodrug, or the metabolite thereof of any one of the present disclosure, or the pharmaceutical composition of the present disclosure.Another aspect of the present disclosure provides a method for chronic weight management or weight management, comprising administering to a subject in need thereof an effective amount of the compound, the isomer, the pharmaceutically acceptable salt, the deuterated compound, the prodrug, or the metabolite thereof of any one of the present disclosure, or the pharmaceutical composition of the present disclosure.Another aspect of the present disclosure provides a method for treating or preventing alopecia, comprising administering to a subject in need thereof an effective amount of the compound, the pharmaceutically acceptable salt, the deuterium substitute, the isomer, the prodrug, or the metabolite thereof of any one of the present disclosure, or the pharmaceutical composition of the present disclosure.BRIEF DESCRIPTION OF THE DRAWINGSFig. 1 shows in vivo body weight reduction in high-fat diet induced obese (DIO) mice model.Fig. 2 shows the result of photography record in the C57BL / 6J alopecia mouse model4.1 Mice and modelingThe male mice (C57BL / 6J) were allowed a 7-day acclimation period. One day before model induction, each mouse was depilated on the dorsal midline to expose a 2.5 cm × 2.5 cm area; only animals whose skin turned uniformly pink, without bluish discoloration or residual hair, were selected for modeling. Alopecia was induced by a single subcutaneous injection of 100 μL testosterone propionate solution (80 mg / kg) . Seven days post-induction, mice still free of bluish pigmentation and hair regrowth were randomized into five groups of six animals each and treatment was initiated for 18 consecutive days. Photographs were taken on the day of depilation, the day of randomization, and twice weekly during the dosing period.4.2 Study design(1) Formuation:The compound was dissolved in PEG300 : ethanol : water (v
