[0001] Eukaryotic chromatin is composed of macromolecular complexes called nucleosomes. A nucleosome has 147 base pairs of DNA wrapped around a protein octamer having two subunits of each of histone protein H2A, H2B, H3, and H4. Histone proteins are subject to post-translational modifications which in turn affect chromatin structure and gene expression. One type of post-translational modification found on histones is methylation of lysine and arginine residues. Histone methylation plays a critical role in the regulation of gene expression in eukaryotes. Methylation affects chromatin structure and has been linked to both activation and repression of transcription (Zhang and Reinberg, Genes Dev. 15: 2343-2360, 2001) . Enzymes that catalyze attachment and removal of methyl groups from histones are implicated in gene silencing, embryonic development, cell proliferation, and other processes.

[0002] One class of histone methylases is characterized by the presence of a Suppressor of Variegation Enhancer of Zeste Trithorax (SET) domain, comprising about 130 amino acids. Enhancer of Zeste Homolog 2 (EZH2) is an example of a human SET-domain containing methylase. EZH2 associates with EED (Embryonic Ectoderm Development) and SUZ12 (suppressor of zeste 12 homolog) to form a complex known as PRC2 (Polycomb GroupRepressive Complex 2) having the ability to tri-methylate histone H3 at lysine 27 (Cao and Zhang, Mol. Cell 15: 57-67, 2004) . PRC2 complexes can also include RBAP46 and RBAP48 subunits. Another example is the related methylase EZH1.

[0003] The oncogenic activities of EZH2 have been shown by a number of studies. In cell line experiments, over-expression of EZH2 induces cell invasion, growth in soft agar, and motility while knockdown of EZH2 inhibits cell proliferation and cell invasion (Kleer et al., 2003, Proc. Nat. Acad. Sci. USA 100: 11606-11611; Varambally et al., (2002) , “The polycomb group protein EZH2 is involved in progression of prostate cancer, ” Nature 419, 624-629) . It has been shown that EZH2 represses the expression of several tumor suppressors, including E-cadherin, DAB2IP and RUNX3 among others. In xenograft models, EZH2 knockdown inhibits tumor growth and metastasis. Recently, it has been shown that down modulation of EZH2 in murine models blocks prostate cancer metastasis (Min et al., “An oncogene-tumor suppressor cascade drives metastatic prostate cancer by coordinately activating Ras and nuclear factor-kappaB, ” Nat Med. 2010 Mar; 16 (3) : 286-94) . EZH2 overexpression is associated with aggressiveness of certain cancers such as breast cancer (Kleer et al., Proc. Nat. Acad. Sci. USA 100: 11606-11611, 2003) . Recent studies also suggest that prostate cancer specific oncogenic fusion gene TMPRSS2-ERG induces repressive epigenetic programs via direct activation of EZH2 (Yu et al., “An Integrated Network of Androgen Receptor, Polycomb, and TMPRSS2-ERG Gene Fusions in Prostate Cancer Progression, ” Cancer Cell. 2010 May 18; 17 (5) : 443-454) .

[0145] Temperature cycling experiments were performed by preparing slurries by weighing 200 mg of amorphous Compound 1 and adding 1 of 48 solvents  / solvent mixtures. See Table 1. Solvents were selected to provide a variety of polarities and functional groups with which to explore the polymorphic landscape. Solvent mixtures were selected due to the high solubility observed in the solubility assessment so that a slurry could be maintained during thermal cycling. After solvent addition, the vials were sealed and temperature cycled (with agitation) between ambient (approximately 20 ℃) and 40 ℃, in 4-hour cycles. After 3 days, observations were made, and any solids were isolated via centrifuge filtration and were analyzed by XRPD. Solids were dried under vacuum at 40 ℃ for 24 hours and analyzed by XRPD. The saturated solutions from the isolated solids were split into equal portions for further polymorph screening experiments. Novel patterns recovered at any stage were dried under vacuum at 40 ℃ and re-analyzed by XRPD. [0161] The primary salt screen of Compound 1 was carried out using 36 counter ions in six solvent systems (2-propanol, acetone, acetonitrile, ethanol, ethyl acetate, and tetrahydrofuran) . The counter ions are presented in Table 6. Stock solutions for each solvent system were prepared by dissolving 180 mg of amorphous Compound 1 was dissolved in 1.2 mL of each solvent at ambient temperature (concentration of 150 mg / mL, ca. 20 ℃) .

[0162] For liquid counter ions, 0.2 mL of stock solution (30 mg of Compound 1) was added to a vial with a magnetic stirrer bar. Then, 1.05 mol. equivalents of concentrated acid were added to the stock solutions with magnetic stirring throughout.

[0169] Analysis by PLM showed irregular shaped particles that were birefringent under crosspolarized light. TGA / DSC showed 0.3 eq. water or 0.1 eq. ethyl formate lost between 70 and 110 ℃. (FIG. 2) A larger loss of 3.7 % (0.29 equivalents of ethyl formate) occurred at a temperature concurrent with solvent loss in the simultaneous DSC trace. The simultaneous DSC trace showed a small endothermic event at ca. 121 ℃ consistent with loss of solvent. DSC showed in the first heating step a sharp endothermic event (melt) with onset of 123 ℃, followed by a shallow endothermic event with an onset of 159 ℃. DSC (cooling step) showed a vitrification event with a midpoint at 117 ℃, confirming that on solvent loss the material becomes amorphous. The solid sample was confirmed to be crystalline Form II by XRPD (FIG. 1) . XRPD peaks are summarized in Table 8. [0187] The solid sample was confirmed to be crystalline 2-Naphthalene sulfonic acid salt form by XRPD (FIG. 11) . XRPD peaks are summarized in Table 14. There was no change in crystalline form on drying. PLM analysis of the 2-naphthalenesulfonic salt indicated that the material consisted of small birefringent particles with no clearly defined morphology. TG analysis indicated that there was a loss of 1.8 wt. %from the onset of heating to 200 ℃related to surface moisture. Simultaneous DSC analysis indicated that there was an endothermic event with an onset of 179.5 ℃, and an endothermic event with an onset of 225 ℃ (FIG. 12) . DSC analysis was carried out with the material sealed non-hermetically with a pierced lid. The first heat of the DSC analysis indicated that there was an endothermic event relating to the evaporation of acetone with an onset of 43.67 ℃ (peak at 64.14 ℃) , then a melting of the material with an onset of 179.45 ℃ (peak at 188.02 ℃) and a probable subsequent melting with an onset of 222.42 ℃ (peak at 233.58 ℃) . VT-XRPD analysis indicated that crystallinity was lost on heating. Poorly crystalline 2-naphthalenesulfonic acid salt form was observed at 195 ℃, and all crystallinity was lost on heating to 235 ℃. No recrystallization was observed on further heating and on cooling. A clear glass was observed at the end of the experiment. KF analysis indicated that there was a moisture content of 0.544 and 0.372 %w / w (0.24 and 0.16 mol eq. water) based on an average of two titrations. The result was consistent with the TG analysis. DVS analysis indicated that the crystalline 2-naphthalenesulfonic acid salt form was slightly hygroscopic, with a moisture uptake of 2.00 wt. %at 80 %RH. XRPD analysis indicated that there was no change in form of crystalline 2-naphthalenesulfonic acid salt form after the DVS experiment was completed. HPLC analysis of the solid indicated that the crystalline 2-naphthalenesulfonic acid salt form had a purity of 99.66 %area. HPLC organic acid concentration analysis resulted in a 2-naphthalenesulfonic acid content of 21.6 %w / w which was consistent with a mono 2-naphthalenesulfonic acid salt. 1H NMR analysis of crystalline 2-naphthalenesulfonic acid salt form indicated that there was 0.017 mol eq. of acetone. The 7-day stability assessment of crystalline 2-naphthelenesulfonic acid salt form showed that there was no change in form at ambient light 80 ℃, and 40℃ / 75 %RH, and that the purity was maintained at both conditions. The solubility of crystalline 2-naphthalenesulfonic acid salt form was > 400 mg / mL in the solutions with Aw of 0.8 and 0.5. Crystalline 2-naphthalenesulfonic acid salt form did not change form on stirring in the ethanol: water solutions. [0188] The solid sample was confirmed to be crystalline p-toluene sulfonic acid salt by XRPD (FIG. 13) . XRPD peaks are summarized in Table 15. There was no change in form on drying. PLM analysis of the crystalline p-toluene sulfonic salt indicated that the material consisted of small birefringent particles with no clearly defined morphology. TG analysis indicated that there was a loss of 1.0 wt. %from the onset of heating to 200 ℃ related to surface moisture. Simultaneous DSC analysis indicated that there was an endothermic event with an onset of 198 ℃ (FIG 14) . DSC analysis was carded out with the material sealed non-hermetically with a pierced lid. The first heat of the DSC analysis indicated that there was an endothermic event relating to the evaporation of acetone with an onset of 35.57 ℃ (peak at 65.51 ℃) , then an endothermic event with an onset of 121.99 ℃ (peak at 136.16 ℃) and a melting event with an onset of 194.61 ℃ (peak at 201.98 ℃) . KF analysis indicated that there was a moisture content of 0.827 and 0.874 %w / w (0.36 and 0.38 mol eq. water) based on two titrations. The result was consistent with the TG analysis. DVS analysis indicated that the crystalline p-toluene sulfonic acid salt was slightly hygroscopic, with a moisture uptake of 3.10 wt. %at 80 %RH. XRPD analysis indicated that there was no change in form of the crystalline p-toluene sulfonic acid salt after the DVS experiment was completed. HPLC analysis of the solid indicated that the crystalline p-toluene sulfonic acid salt had a purity of 99.77%area. HPLC organic acid concentration analysis resulted in a p-toluene sulfonic acid content of 25.4 %w / w which was consistent with a mono p-toluene sulfonic acid salt. 1H NMR analysis of crystalline p-toluene sulfonic acid form indicated that there was 0.034 mol eq. of acetone. The 7-day stability assessment of the crystalline p-toluene sulfonic acid salt showed that there was no change in form at ambient light 80 ℃, and 40℃ / 75 %RH, and that the purity was maintained at both conditions. The solubility of crystalline p-toluene sulfonic acid salt was > 400 mg / mL in the solutions with Aw of 0.8, 0.5 and 0.2.
