Abstract

AQUEOUS ANTIMICROBIAL COATING COMPOSITIONFIELD

[0001] The present invention relates to an aqueous antimicrobial coating composition, particularly, affording antiviral properties, and a method of preparing the same.

[0002] INTRODUCTION

[0003] Silver ions and silver element are widely used in coating compositions to provide antimicrobial properties. The higher the silver content is in the compositions, the better the antimicrobial properties. However, coatings (dry coating films) made from coating compositions containing inorganic microbiocides on which metal ions are supported often undergo conspicuous changes in coloration upon exposure to sun light. Accordingly, the use of these microbiocides is effectively limited to systems for which such changes in coloration can be tolerated. Discoloration of coatings can be improved by incorporating counter ions into coating compositions to form more stable silver salts, such as silver chloride (AgCl) in wet state, however, the antimicrobial efficacy of the coatings is usually compromised. Incorporation of silver complexed with polymers bearing imidazole pendant groups as antimicrobial additives into polymer binders may improve discoloration of the polymer binders in wet state when the polymer binders have good compatibility with the antimicrobial additives. These coatings may demonstrate antibacterial properties, but their antiviral properties need further improvement. Viruses can be divided into two types, enveloped viruses and non-enveloped viruses. According to U.S. Environmental Protection Agency’s Microbiological Disinfection Hierarchy, non-enveloped viruses such as enteroviruses, rhinoviruses, Coxsackie viruses, MS2 phage, EV71, and norovirus are more resistant to common disinfectants than fungi, bacteria, and enveloped viruses. Thus, more powerful and costly disinfectants are needed to kill non-enveloped viruses. Simply increasing the silver concentration in coating compositions (e.g., up to 600 ppm of silver relative to coating composition weight) is not effective in improving coatings’ antiviral activity against non-enveloped viruses and may also lead to discoloration of coatings. It is therefore desirable to provide an aqueous antimicrobial coating composition that affords coatings made therefrom with improved antiviral activity.SUMMARY

[0004] The present invention solves the problem of discovering an aqueous antimicrobial coating composition without improved antiviral properties. The aqueous antimicrobial coating composition (hereinafter also referred to as “coating composition” ) of the present invention comprises a novel combination of a specific emulsion polymer bearing pendant groups selected from specific heterocyclic groups (such as imidazole groups) , acetoacetyl functional groups, or combinations thereof; (B) a silver salt, and (C) a protease. The coating composition affords coating films (i.e., coatings) made therefrom antiviral properties, as indicated by antiviral activity ≥ 0.82 against MS2  phage after exposure for 24 hours, as measured according to modified Chinese T / CNCIA 03002-2020 for MS2 phage.

[0005] In a first aspect, the present invention is an aqueous antimicrobial coating composition comprising:

[0006] (A) an emulsion polymer comprising, by weight based on the weight of the emulsion polymer,

[0007] (a1) from 0 to 10%of structural units of an acetoacetyl functional monomer; (a2) from 0 to 8%of structural units of a monomer containing at least one heterocyclic group selected from the group consisting of imidazole, benzotriazole, and benzimidazole; and (a3) structural units of an additional monoethylenically unsaturated nonionic monomer;

[0008] provided that the total concentration of (a1) structural units of the acetoacetyl functional monomer and (a2) structural units of the monomer containing at least one heterocyclic group is at least 0.1%;

[0032] The emulsion polymer (A) may comprise (a1) structural units of an acetoacetyl functional monomer, (a2) structural units of the heterocyclic monomer, or combinations thereof; provided that the total concentration of (a1) structural units of an acetoacetyl functional monomer and (a2) structural units of the heterocyclic monomer is at least 0.1%, by weight based on the weight of the emulsion polymer (A) .

[0033] The emulsion polymer (A) may comprise (a3) structural units of an additional monoethylenically unsaturated nonionic monomer that is other than monomers (a1) and (a2) , hereinafter referred to as “monomer (a3) ” . The additional monoethylenically unsaturated nonionic monomer may be selected from an alkyl (meth) acrylate, an itaconate ester, a vinyl aromatic monomer, a vinyl ester monomer, acrylonitrile, methacrylonitrile, or mixtures thereof. Desirably, the additional monoethylenically unsaturated nonionic monomer is selected from one or more alkyl (meth) acrylates.  “Alkyl (meth) alkylates” refers to alkyl esters of (meth) acrylic acid containing a linear, branched, or cyclic alkyl group. The alkyl (meth) acrylate can be a C1-C24-alkyl (meth) acrylate (i.e., an alkyl (meth) acrylate having an alkyl with from 1 to 24 carbon atoms) , desirably a C1-C10-alkyl (meth) acrylate, more desirably a C1-C4-alkyl (meth) acrylate. Examples of suitable alkyl (meth) acrylate include methyl acrylate (MA) , methyl methacrylate (MMA) , butyl methacrylate (BMA) , ethyl methacrylate (EMA) , butyl acrylate (BA) , ethyl acrylate (EA) , 2-ethylhexyl acrylate, 2-ethylhexyl methacrylate, a cycloalkyl (meth) acrylate such as cyclohexyl (meth) acrylate, or mixtures thereof. Desirably, the alkyl (meth) acrylate is a C1-C4-alkyl (meth) acrylate such as BA, EA, or mixtures thereof. Examples of suitable itaconate esters include dibutyl itaconate, dihexyl itaconate, dioctyl itaconate, didecanyl itaconate, or mixtures thereof. Examples of suitable vinyl aromatic monomers include styrene and substituted styrene, styrene, and substituted styrene such as . alpha. -methyl styrene, α-ethylstyrene, p-methyl styrene, t-butyl styrene, trans-beta-methylstyrene, 2, 4-dimethylstyrene, ethylstyrene, o-, m-, and p-methoxystyrene; p-trifluoromethylstyrene, vinyl xylene, or mixtures thereof. The concentration of structural units of the monoethylenically unsaturated nonionic monomer (a3) in the emulsion polymer (A) may be in a range of from 75%to 99.99%, and can be 80%to 99.9%, 85%to 99.5%, 86%to 99.3%, or 87%to 99%, desirably, 85%to 99%, by weight based on the weight of the emulsion polymer (A) .

[0038] When the emulsion polymer (A) comprises (a2) structural units of the heterocyclic monomer (e.g., 1-vinyl imidazole) , the emulsion polymer (A) can be a multistage polymer. The multistage polymer may comprise, or can consist of, 78%to 97%of the first-stage polymer and 3%to 22%of the second-stage polymer, by weight based on the multistage polymer weight. Desirably, the concentration of the first-stage polymer can be from 80%to 97%, 85%to 96%, or 85%to 95%; and the concentration of the second stage polymer can be 3%to 20%, 4%to 15%, or 5%to 15%, by weight based on the multistage polymer weight (i.e., the weight of the emulsion polymer (A) ) . The multistage polymer may comprise structural units of the heterocyclic monomer such as 1-vinyl imidazole, desirably, in the second-stage polymer, and optionally, in the first-stage polymer. The total concentration of structural units of the heterocyclic monomer in the multistage polymer can be in a range of from 0.1%to 1%, by weight based on the multistage polymer weight. The heterocyclic monomer for the first-stage polymer and second-stage polymer can be the same or different. Desirably, from 60%to 100%of structural units of the heterocyclic monomer in the multistage polymer are present in the second-stage polymer, and can be 65%or more, 70%or more, 75%or more, 80%or more, 85%or more, 90%or more, 95%or more, or even 100%of structural units of the heterocyclic monomer are present in the second-stage polymer. For example, the concentration of structural units of the heterocyclic monomer in the second-stage polymer may be in a range of from 0.4%to 22%, and can be 0.4%or more, 0.5%or more, 0.6%or more, 0.8%or more, 1.0%or more, 1.2%or more, 1.4%or more, 1.6%or more, 1.8%or more, 2%or more, 2.5%or more, 3.0%or more, 3.5%or more, even 3.75%or more while at the same time is generally 22%or less, 20%or less, and can be 18%or less, 16%or less, 15%or less, 14%or less, 13%or less, or even 12%or less, desirably, 3.0%to 14%, more desirably, 3.5%to 13%, most desirably, from 3.75%to 12%, by weight based on the weight of the second-stage polymer. The concentration of structural units of the heterocyclic monomer in the first-stage polymer may be, by weight based on the weight of the first-stage polymer, in a range of from zero to 0.9%, and can be less than 0.9%, less than 0.8%, less than 0.6%, less than 0.4%, less than 0.2%, less than 0.1%, or even zero.

[0082] Firstly, a monomer emulsion 1# (ME1) was prepared by mixing deionized (DI) water (329.16 grams (g) ) , AM (21.19 g, 40%) , MAA (25.82 g) , BA (1022.12 g) , MMA (475.55 g) , AAEM (50.85 g) , and RS-610 S25 surfactant (98.41 g, 25%) . A monomer mix 2# (MM2) was prepared by mixing BA (110.73 g) and VI (8.56 g) .

[0083] Secondly, in a one-gallon vessel equipped with a reflux condenser and a stirrer, DI water (940 g) was added at an agitation rate of 130 revolutions per minute (rpm) . Meanwhile, the temperature of the reaction vessel was raised to 91℃. Then FES 32 surfactant (5.40 g, 31%) and a buffer solution of sodium carbonate (Na2CO3) (3.41 g in 31.37 g DI water) were introduced into the reaction vessel.

[0084] Thirdly, the ME1 (93.13 g) and an initiator solution of APS (3.40 g in 28 g DI water) were  injected into the reaction vessel. The reaction mixture was held at a temperature between 80 and 95℃ for 5 minutes (min) . Thereafter, the remainder of the ME1 was added into the reaction vessel over the span of 100 min. After completing the feed of the ME1, the MM2 was added into the reaction vessel over the span of 10 min. During the addition of the ME1 and MM2, another shot of an initiator solution consisting of APS (1.55 g) and DI water (167.33 g) was co-fed into the reaction vessel over the span of 120 min. The reaction temperature was being held at somewhere between 84 to 86 ℃. After the above mixing steps were completed, the reaction vessel was cooled down. While cooling the contents of the reaction vessel to room temperature, an initial reductant solution consisting of ferrous sulfate (0.0091 g) , EDTA sodium salt (0.0549 g) , and DI water (10 g) , a secondary reductant solution consisting of FF6 (1.05 g) and DI water (50 g) , and an initiator solution consisting of 70%aqueous solution of t-BHP (1.03 g) , 35%aqueous solution of H2O2 (0.45 g) , and DI water (50 g) , were injected into the reaction vessel when the temperature had dropped to 70 ℃. Then a solution of H2O2 and t-BHP (6.55 g 35%aqueous solution of H2O2, 6.45 g 70%aqueous solution of t-BHP in 40 g H2O) was added into the reaction vessel when the temperature was over 50℃. Then a neutralizer solution comprising 15 g of MEA, 17.5 g of TERGITOLTM 15-S-40 surfactant (70%) , and 32.50 g of H2O were added into the reaction vessel. Thus, PD-1 was obtained.

[0113] Antiviral activity properties against non-enveloped virus MS2 phage, according to modified T / CNCIA 03002-2020 (Test method for determining antiviral activity of coating) , which includes sample preparation and antiviral activity test procedure. Details are provided below:

[0114] A coating composition sample ( “test sample” ) was applied onto plastic charts using a 100 micrometers (μm) film applicator and dried at room temperature for 7 days. A blank plastic chart was used as an untreated control sample ( “control sample” ) . The non-enveloped virus MS2 phage were inoculated on both test samples and control samples, then covered with a thin, sterile film to ensure  the virus suspension close contact with the sample surfaces and incubated for 24 hours at 25 ℃ and 90%relative humidity (RH) . The virus titers (Plaque Forming Unit (PFU)  / ml) on the surfaces of the control sample and the test sample were determined at “time zero” , denoted as “V0-control” and “V0-test” , respectively. After the virus suspension contacted with the sample surfaces, the covered control sample and test sample were allowed to incubate undisturbed at 25 ℃ and 90%RH for 24 hours. After incubation, residual virus titers on the test sample ( “VTest” ) and control sample ( “VControl” ) were determined. The antiviral activity is calculated as logarithm reduction of virus titer in the test sample comparing with the control sample as below:

[0118] A coating composition was evaluated for antiviral activity against non-enveloped virus EV71, according to T / CNCIA 03002-2020 (Test method for determining antiviral activity of coating) , which includes sample preparation and antiviral activity test procedure. Details are provided below:

[0119] A test coating composition was applied onto Aluminum carriers using a 50 μm film applicator, after drying at room temperature for 2 hours, then applied again using a 50 μm film applicator, followed by drying at room temperature for 7 days, thereby forming a test sample. The standard Control Sample without any biocidal components supplied by Guangdong Detection Center of Microbiology was used as an untreated control sample ( “Control Sample” ) .

[0122] Characterization results of antiviral activity properties of the coatings are given in Table 3.

[0123] As shown in Table 3, IEs 1-15 coating composition samples comprising the specified emulsion polymer (A) with structural units of AAEM and / or VI, (B) silver salt, and (C) protease all passed the antiviral activity test with antiviral activity ≥ 0.82 for non-enveloped virus MS2 phage after exposure for 24 hours. Comparing with CEs C and D samples, IE-1 comprising the novel combination of silver salt and protease with the specified emulsion polymer (A) has demonstrated synergistic effects on improving antiviral activity properties against non-enveloped virus MS2 phage. After heating the coating composition of IE-5 (wet state) at 50 ℃ for 2 weeks (thus forming IE-5-A sample) or after storing the coating film (dry state) made from IE-5 composition at room temperature for 2 weeks (thus forming IE-5-B sample) , the obtained samples of IE-5-A and IE-5-B, respectively, both passed the antiviral activity test with antiviral activity ≥ 0.82 against MS2 phage after exposure for 24 hours.

[0124] In contrast, CEs samples all failed the antiviral activity test against MS2 phage. Comparing with IE-1 and IE-2, CEs A and C samples that do not contain protease (even at a higher concentration of silver for CE-C) , CE-D sample that does not contain any silver salt, and CE-B sample that contains insufficient amount of silver salt (less than 15 ppm) all failed to meet the antiviral activity requirement. Comparing with IE-8, CE-E sample that does not contain a protease even with a higher silver concentration and CE-F sample does not contain silver even with a higher protease concentration still failed the antiviral activity tests. CE-G sample that contains PD-5, which does not contain structural units of the acetoacetyl functional monomer nor the heterocyclic monomer, failed the antiviral activity test as indicated by antiviral activity against MS2 phage less than 0.82. CE-H and CE-I samples that contain the specified components (A) , (B) , and (C) but with insufficient amounts of protease, even in combination with other types of enzymes (e.g., amylase, lipase, and cellulase) (CE-H) , both failed the antiviral activity tests.

1.An aqueous antimicrobial coating composition comprising:(A) an emulsion polymer comprising, by weight based on the weight of the emulsion polymer,(a1) from 0 to 10%of structural units of an acetoacetyl functional monomer;(a2) from 0 to 8%of structural units of a monomer containing at least one heterocyclic group selected from the group consisting of imidazole, benzotriazole, and benzimidazole; and(a3) structural units of an additional monoethylenically unsaturated nonionic monomer;provided that the total concentration of (a1) structural units of the acetoacetyl functional monomer and (a2) structural units of the monomer containing at least one heterocyclic group is at least 0.1%;(B) a silver salt present in an amount to provide 15 to 1000 ppm of silver, by weight based on the weight of the aqueous antimicrobial coating composition; and(C) from 0.3%to 5%of a protease, by weight based on the weight of the aqueous antimicrobial coating composition.2.The aqueous antimicrobial coating composition of claim 1, wherein the monomer containing at least one heterocyclic group is 1-vinyl imidazole.3.The aqueous antimicrobial coating composition of claim 1 or 2, wherein the emulsion polymer (A) comprises, by weight based on the weight of the emulsion polymer, (a1) from 0.5%to 8%of structural units of the acetoacetyl functional monomer.4.The aqueous antimicrobial coating composition of any one of claims 1-3, wherein the acetoacetyl functional monomer is selected from the group consisting of acetoacetoxyethyl methacrylate, acetoacetoxyethyl acrylate, acetoacetoxypropyl methacrylate, acetoacetoxybutyl methacrylate, 2, 3-di (acetoacetoxy) propyl methacrylate, allyl acetoacetate, and mixtures thereof.5.The aqueous antimicrobial coating composition of any one of claims 1-4, wherein the weight ratio of silver to protease is in a range of from 1: 20 to 1: 1000.6.The aqueous antimicrobial coating composition of any one of claims 1-5, wherein the emulsion polymer (A) is a multistage polymer comprising a first-stage polymer and a second-stage polymer, wherein the second-stage polymer comprises structural units of the monomer containing at least one heterocyclic group, and the total concentration of structural units of the monomer containing at least one heterocyclic group in the multistage polymer is in a range of from 0.1%to 1%, by weight based on the weight of the multistage polymer.7.The aqueous antimicrobial coating composition of claim 6, wherein the multistage  polymer comprises 78%to 97%of the first-stage polymer and 3%to 22%of the second-stage polymer, by weight based on the multistage polymer weight;wherein the first-stage polymer comprises, by weight based on the first-stage polymer weight, from 0 to 0.9%of structural units of the monomer containing at least one heterocyclic group; from 0.5%to 8%of structural units of the acetoacetyl functional monomer; from 85%to 99%of structural units of a C1-C4-alkyl (meth) acrylate; and from 0.1%to 14.5%of structural units of a monoethylenically unsaturated functional monomer carrying at least one functional group selected from a carboxyl, carboxylic anhydride, sulfonic acid, amide, sulfonate, phosphoric acid, phosphonate, phosphate, alkoxysilane, or hydroxyl group; a salt thereof; or combinations thereof; andwherein the second-stage polymer comprises, by weight based on the second-stage polymer weight, from 0.4%to 22%of structural units of 1-vinyl imidazole and from 75%to 99%of structural units of a C1-C4-alkyl acrylate.8.The aqueous antimicrobial coating composition of any one of claims 1-5, wherein the aqueous antimicrobial coating composition meets one of the following conditions:(1) when the emulsion polymer comprises (a2) structural units of the monomer containing at least one heterocyclic group, the silver salt is present in an amount to provide 15 to 100 ppm of silver, by weight based on the weight of the aqueous antimicrobial coating composition; and(2) when the emulsion polymer (A) is free of structural units of the monomer containing at least one heterocyclic group, the silver salt is present in an amount to provide 120 to 600 ppm of silver, by weight based on the weight of the aqueous antimicrobial coating composition.9.The aqueous antimicrobial coating composition of any one of claims 1-8, wherein the silver salt (B) is selected from silver nitrate, silver chloride, or mixtures thereof.10.The aqueous antimicrobial coating composition of any one of claims 1-9, wherein the protease (C) is selected from an alkaline protease, a neutral protease, or mixtures thereof.11.A process for preparing the aqueous antimicrobial coating composition of any one of claims 1-10, comprising: admixing the emulsion polymer (A) , the silver salt (B) , and the protease (C) .12.A method of providing an antimicrobial coating, comprising the steps of:a) applying the aqueous antimicrobial coating composition of any one of claims 1-10 to a substrate;and b) drying, or allowing to dry, the applied antimicrobial coating composition.
