[0001] The present invention discloses a continuous process for amination of aminopyridines. Particularly, the invention provides a two-step or multi-step process comprising a first continuous nitration step followed by a continuous hydrogenation step to introduce an amino group to an 2-aminopyridine in meta position. Such diaminopyridine compounds are valuable starting materials in the manufacture of active pharmaceutical ingredients.Background of the Invention

[0002] 2-aminopyridine is known as a starting material in the production of active pharmaceutical ingredients (API) like piroxicam, sulfapyridine, tenoxicam, and tripelennamine. Also aminated derivatives of 2-aminopyridine are reported in the art as starting materials for obtaining small molecular APIs via chemical synthesis.

[0008] Linaprazan glurate is a prodrug of Linaprazan and expected to have a longer biological half-life than Linaprazan itself. The gastric acid control ability of Linaprazan Glurate has in studies shown to be superior to current medication. Linaprazan glurate is currently in clinical development.

[0009] There is hence a need for diaminopyridine derivatives as starting materials for the preparation of APIs. The methods disclosed in the prior art for obtaining diaminopyridine derivatives have the drawbacks of limited safety of the nitration reactions, which are highly exothermic reactions.

[0010] The present invention provides a novel continuous process for the preparation of diaminopyridine derivatives. The inventors of present invention have developed a two-step or multi-step process for obtaining diaminopyridines comprising a first continuous nitration step and further a continuous hydrogenation step. The continuous nitration process of present invention is much safer than nitration reactions in batch-mode, as disclosed in the prior art. The hydrogenation process of present invention provides higher efficiency and results in higher purity. The continuous processes of present invention are faster, thereby reducing costs and making it an overall cheaper process.

[0014] The term “CSTR” or “CSTR reactor” refers to a continuous stirred-tank reactor, also known as vatmix reactor, backmix reactor, mixed flow reactor (MFR) , or a continuous-flow stirred-tank reactor (CFSTR) , and is a type of chemical reactor, wherein reactants are continuously fed into a reactor vessel, where they are mixed and allowed to react, producing the desired products. The CSTR consists of a vessel equipped with a stirring mechanism, such as a mechanical stirrer or an agitator. The reactants are introduced into the tank through inlet ports, and the products are continuously removed through outlet ports. The key feature of a CSTR flow reactor is its constant stirring, which ensures that the reactants are evenly distributed throughout the vessel and that the reaction proceeds at a uniform rate. This allows for precise control over the reaction conditions, such as temperature, pressure, and concentration, which can be adjusted by controlling the flow rates of the reactants and products.

/ or wherein R4a and R4b are the same, wherein in the continuous hydrogenation step one equivalent of the nitrated compound of formula (III) is dissolved in a first hydrogenation solvent to obtain a first hydrogenation solution; and / or wherein R4a and R4b are not the same, wherein in the continuous hydrogenation step one equivalent of the nitrated compound of formula (IV) is dissolved in a first hydrogenation solvent to obtain a first hydrogenation solution.18.The process according to any of claims 1 to 18, wherein the continuous hydrogenation step comprises a further purification step, wherein the first hydrogenation solution is precipitated by adjusting the pH of the first hydrogenation solution to basic pH, particularly to pH 8 to 12, using a base, by filtering the obtained precipitate and by redissolving the precipitate in a second hydrogenation solvent to obtain a second hydrogenation solution.19.The process of any of claims 1 to 18, wherein the continuous hydrogenation is performed by mixing the first or second hydrogenation solution with hydrogen and pumping the mixture on a packed bed column comprising catalyst particles.20.The process of any of claims 1 to 19, wherein the continuous hydrogenation is performed by mixing the first or second hydrogenation solution with hydrogen and pumping the mixture on a packed bed column comprising catalyst particles, followed by feeding the obtained product solution into a gas-liquid separator to scavenge unreacted hydrogen.21.A compound of formula (I) obtainable by the process according to any of claims 1 to 20.22.A compound of formula (I) obtainable by the process according to any of claims 1 to 20.23.The invention as described herein before.
