METHODS AND DEVICES FOR DETECTION OF ANTI-LIPOIDAL ANTIBODIESTechnical Field

[0001] This disclosure generally relates to methods and immunoassay devices useful, for example, for detection of anti-lipoidal antibodies and / or diagnosis of a disease such as syphilis.Background

[0002] Syphilis is a sexually transmitted disease (STD) caused by the spirochete bacterium Treponema pallidum (T. pallidum) . The immune response in syphilis includes production of treponemal antibodies, and (ii) anti-lipoidal antibodies.

[0003] Tests for anti-lipoidal antibodies, sometimes called "non-treponemal tests" , are typically based on naturally occurring cardiolipin, lecithin and cholesterol. Treponemal tests are based on antigens derived from T. pallidum and include T. pallidum particle agglutination (TP-PA) , the fluorescent treponemal antibody-absorbed test (FTA-ABS) and immunoassays. Non-treponemal tests are considered a better choice for monitoring treatment or testing for reinfection, because anti-lipoidal antibody titers decrease with treatment of syphilis, eventually disappearing in most patients, while treponemal antibodies titers remain high for years or even a lifetime. A combination of a non-treponemal test and a treponemal test is useful for syphilis screening and diagnostic purposes.

[0004] There is still a need of a rapid, point-of-care test for anti-lipoidal antibodies, and also a long-felt need of an easy-to-use, rapid, point-of-care test capable of concurrently detecting both non-treponemal and treponemal antibodies.SUMMARY

[0005] The inventors of the present disclosure found that anti-lipoidal antigens made up of at least cardiolipin and phosphatidylcholine (also referred to as "lecithin" ) can be reliably attached to a solid substrate (such as, a microporous membrane) without utilizing a medium such as a proteinaceous carrier, while maintaining the antigenicity and specificity of the anti-lipoidal antigens for anti-lipoidal antibodies. The inventors of the present disclosure further found that a higher amount of lipoidal antigens attached to the solid substrate is useful for maintaining the antigenicity and specificity of the anti-lipoidal antigens for anti-lipoidal antibodies. The lateral flow assay device as disclosed herein is also capable of detecting the anti-lipoidal antibodies in finger stick blood from a syphilis-infected subject, and meet the WHO target profile for Syphilis POCT detection.
