[001] The present invention relates to processes and intermediates for the preparation of a dual GIP / GLP1 agonist peptide, tirzepatide, or a pharmaceutically acceptable salt thereof.
[002] Diabetes mellitus is a chronic disorder characterized by hyperglycemia resulting from defects in insulin secretion, insulin action, or both. In type 2 diabetes mellitus (T2D), the combined effects of impaired insulin secretion and insulin resistance are associated with elevated blood glucose levels. The dual GIP / GLP1 agonist, tirzepatide, is described and claimed in U.S. Patent 9,474,780 (Patent 780). Tirzepatide may be useful in the treatment of T2D.
[003] US9474780 documents generally describe peptides and a method for preparing a dual GIP / GLP1 agonist.
[004] There is a need for processes and intermediates to enable an improved technology for the production of tirzepatide with a combination of advantages, including commercially desirable purity. Similarly, there is a need for efficient and environmentally green processes, including stable intermediates, to provide tirzepatide with fewer purification steps. An improved technology is also needed to provide tirzepatide manufacturing processes that produce minimal waste streams for enhanced operator and environmental safety. The large-scale preparation of pharmaceutically acceptable tirzepatide presents a number of technical challenges that can affect overall yield and purity.There is a need for processes to avoid the use of transition metals and / or adverse reaction conditions that are incompatible with peptide synthesis. [005] The present invention seeks to meet these needs by providing new intermediates and processes useful in the manufacture of tirzepatide (SEQ ID NO: 1) or a pharmaceutically acceptable salt thereof. The improved tirzepatide manufacturing processes of the present invention provide intermediates and process reactions that incorporate a combination of advancements, including an efficient pathway with fewer steps while at the same time maintaining high quality and purity. Importantly, the improved processes and intermediates decrease resource intensity and minimize waste streams.

1 / 2 CLAIMS 1. Process for desulfurizing a cysteine analog of tirzepatide to form tirzepatide SEQ ID NO: 1 or a pharmaceutically acceptable salt thereof, characterized in that it comprises contacting the cysteine analog of tirzepatide with a radical initiator.
2. Process according to claim 1, characterized in that the radical initiator is a water-soluble radical initiator.
3. Process according to claim 1, characterized in that the radical initiator is an azo initiator.
4. Process according to claim 1, characterized in that the radical initiator is selected from the group consisting of 2,2'-azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride and 2,2'-azobis(2-methylpropionamidine) dihydrochloride.
5. Process according to claim 1, characterized in that the thiol comprises 2,2,2-trifluoroethanethiol or mercaptophenylacetic acid.

Process according to claim 7, characterized in that the process comprises: coupling the thioester peptide intermediate SEQ ID NO: 34 with the tirzepatide intermediate SEQ ID NO: 33 to form the tirzepatide cysteine analog SEQ ID NO: 35; and bringing the tirzepatide cysteine analog SEQ ID NO: 35 into contact with a radical initiator to form tirzepatide (SEQ ID NO: 1).

Process according to claim 8, characterized in that the process comprises: (a) synthesizing a tirzepatide intermediate SEQ ID NO: 32 by solid-phase peptide synthesis; (b) contacting the tirzepatide intermediate of step (a) with a thiol to form the thioester peptide intermediate SEQ ID NO: 34; (c) coupling the thioester peptide intermediate with the tirzepatide intermediate SEQ ID NO: 33 to form the cysteine analog of tirzepatide SEQ ID NO: 35; and (d) contacting the cysteine analog of tirzepatide (SEQ ID NO: 35) with a radical initiator to form tirzepatide (SEQ ID NO: 1).
