Description

TECHNICAL FIELD

[0001] The application belongs to the technical field of agricultural planting, and relates to a multicolor peanut, in particular to a method for planting a multicolor peanut with improved sucrose content. BACKGROUND

[0002] Peanut is one of the world's three major oil crops, and is a major source of edible oil and an important source of protein. China is the world's leading peanut producer. China's average yield, total production and total consumption of peanuts are the highest in the world, and peanuts are one of the target crops for expanding acreage and increasing yield. Edible peanuts are an important part of Chinese peanuts, accounting for about 45% of total peanut consumption.

[0003] Edible peanuts, especially fresh peanuts, require high sucrose content (6-8%), low oil content (about 45%) and high protein content (25-30%) for nutritional health, and have outstanding "sweet, fragrant, crisp and glutinous" characteristics, i.e. a sweet, soft, crisp and glutinous taste when eaten. Traditional peanuts generally have an oil content of about 50%, a sucrose content of less than 3% and a protein content of about 20%. Reducing oil content, increasing sucrose content and protein content are the directions for improving the quality of edible, especially fresh, peanut varieties.

[0004] Peanut kernel sucrose content is a heritable quantitative trait. Through variety improvement, domestic peanut breeding units have bred a batch of high-sucrose-content varieties with sucrose content exceeding 5%, with the highest sucrose content reaching more than 10%. However, as the sucrose content of the variety increases, the peanut kernel becomes hard and no longer has a soft, crisp and glutinous taste, which reduces the eating quality of peanuts.

[0005] Multicolor peanuts are a Yunnan specialty edible peanut brand and a representative of Chinese specialty edible peanuts, and were selected as one of the most influential peanut brands in China in 2023. The promotion of multicolor peanuts has transformed Yunnan peanuts from a traditional peasant industry that meets the consumption needs of farmers themselves to a rich people industry that meets the consumption needs of domestic large and medium-sized cities and a new pillar industry for rural revitalization in the production area. Currently, the planting area of multicolor peanuts has reached more than 300,000 mu, accounting for about 40% of the peanut production area in Yunnan Province.

[0006] Yunhua 24 is a representative seven-colored peanut variety bred and registered by the Institute of Economic Crops, Yunnan Academy of Agricultural Sciences. This variety is for consumption and has a growth period of 127 days. The main stem is 59.96 cm tall, lateral branches are 65.76 cm long, with a total of 4.1 branches and 3.7 fruiting branches. Each plant produces 17.1 fully-filled pods. The weight of 100 pods is 241.83 g, with a full pod rate of 78.62%. The weight of 100 kernels is 59.99 g, with a kernel yield of 63.86%. The seed coat is multicolored (dark purple and light brown); the inner seed coat is multicolored (purple and dark yellow). The kernel contains 50.20% oil, 23.20% protein, 44.40% oleic acid, 34.90% linoleic acid, and 6.40% sucrose.

[0007] Colorful peanuts, a specialty peanut product of Yunnan Province, show a significant decline in their edible quality (sucrose content, taste, etc.) when grown outside Yunnan, especially in major peanut-producing areas of northern my country. This indicates that the variety of colorful peanuts is closely related to the growing environment. In the cross-regional identification of over 400 core peanut germplasm resources, we found that simultaneous planting in Kunming and Wuhan increased the average sucrose content of peanuts from approximately 1.9% in Wuhan to 5.8%, an increase of about three percentage points. Similarly, in different ecological regions of Yunnan, the sucrose content and taste of Yunhua 24 peanuts grown in different areas showed significant differences. It is evident that peanut edible qualities, such as sucrose content, are not only regulated by genes but also interact with environmental factors. Therefore, this study conducts in-depth research on the correlation between planting environment and climatic conditions and sucrose content, providing a method for cultivating colorful peanuts to improve sucrose content. Summary of the Invention

[0031] Unless otherwise specified, the experimental methods used in the following examples are conventional methods.

[0032] Unless otherwise specified, all materials and reagents used in the following examples are commercially available.

[0033] To make the objectives, technical solutions, and advantages of this invention clearer, the invention will be further described in detail below with reference to specific embodiments. It should be understood that the specific embodiments described herein are merely illustrative and not intended to limit the invention.

[0034] Example

[0035] The sucrose content in peanuts was detected by high performance liquid chromatography (HPLC) according to GB / T 30390-2013 in order to screen the planting conditions for colorful peanuts.

[0036] 1. Screening of planting methods

[0037] 1.1 Relationship between sucrose content and temperature

[0038] Six different production environments were selected in Yunnan Province, and the same cultivation and management measures were applied to plant the colorful peanut variety Yunhua 24 and the control variety Yunhua 3. Environmental temperature was recorded during the peanut growth period. Mature peanut pods were harvested, and the sugar content of the peanut kernels was measured after shelling. By analyzing the relationship between the sugar content of colorful peanuts and environmental temperature, planting environments conducive to sucrose accumulation were selected.

[0039] 1.2 Sowing at the appropriate time

[0040] Five temperature gradients (8℃, 10℃, 12℃, 14℃, and 16℃) were set up in a constant-temperature incubator to investigate peanut germination potential (the proportion of germinated peanut kernels on day 7 of the experiment) and germination rate (the proportion of germinated peanut kernels on day 10 of the experiment), thus studying the lower limit of the temperature range for seedling emergence of colorful peanuts. A method for early sowing was proposed, which involves artificially supplementing soil moisture using sprinkler or drip irrigation to maintain a suitable soil moisture content of 60-70%.

[0041] 1.3 Ridge Cultivation

[0042] To address the problems of poor drainage and irrigation in traditional flat cultivation, which leads to waterlogging in the field during peanut growth due to rainfall and other factors, increasing disease incidence and reducing yield and variety, three planting methods were set up: flat cultivation, ridge cultivation, and raised ridge cultivation (Table 1). Under the same management conditions, a disease survey and peanut yield and quality were conducted at harvest time for comparison.

[0043] Table 1 Treatment methods for different planting methods

[0044]

[0045] 1.4 Planting Method

[0046] Using the same field planting and management methods, a field comparison experiment was conducted on Yunhua 24 peanuts under plastic film mulching and open field cultivation to analyze the effects of plastic film mulching on peanut yield and quality, and to explore the feasibility of applying plastic film mulching and open field cultivation in the cultivation of colorful peanuts.

[0047] 1.5 Comprehensive Nutrient Supply and Plant Protection Program

[0048] Based on the screening of a large number of chemical fertilizers and pesticides introduced in the early stage, a complete nutrient supply and plant protection program was established. Then, a field comparison was carried out between the complete nutrient supply and plant protection program and the traditional nutrient supply and plant protection program (Table 2). Under the same management conditions, a disease survey at harvest time and a comparison of peanut yield and quality were conducted.

[0049] Table 2 Treatment methods for different nutrient supply and plant protection programs

[0050]

[0051]

[0052] Note: All percentages are volume fractions.

[0053] 1.6 Harvest at the appropriate time

[0054] Referring to the peanut pod classification criteria of Boote et al. (1982), the development and size of peanut kernels were divided into six stages: the young pod stage (R1), the pod shaping stage (R2), the kernel filling stage (R3), the pod full stage (R4), the maturity stage (R5), and the harvest stage (R6). Samples were taken from each of these six stages, and the sucrose content of the sucrose standard solution and peanut kernels collected at different stages was detected by high performance liquid chromatography-refractive index (HPLC-RID) according to Li Weitao (2021). The accumulation law of sucrose content in Yunhua 24 kernels was studied, and the harvest time was selected.

[0055] 2. Experimental Results

[0056] 2.1 Relationship between sucrose content and temperature

[0057] In the early stages of this invention, 400 peanut core germplasm accessions were simultaneously planted and identified in Wuhan, Hubei Province and Kunming, Yunnan Province. The results showed that the average sucrose content of the tested germplasm in Kunming and Wuhan was 5.823% and 1.908%, respectively, a difference of 3 times, indicating that different planting environments have a significant impact on sucrose content.

[0058] To further analyze the relationship between sucrose content and the planting environment, and to screen out the varieties most suitable for increasing the sucrose content of colorful peanuts, colorful peanut cultivation was carried out in six different production environments in Yunnan.

[0059] Peanut growth stages were divided into seedling stage, flowering and pegging stage, pod-setting stage, pod-filling stage, and maturity stage based on the plant's growth status. Analysis showed that peanut kernel sucrose content was closely related to temperature during the growth period, and significantly negatively correlated with the average maximum temperature during pod-setting, the average minimum temperature from flowering to maturity, and the average air temperature. This indicates that higher temperatures are less conducive to sucrose accumulation in the kernels, while cooler habitats are more favorable for cultivating edible peanuts with high sucrose content (Table 3).

[0060] Yunhua 24 in six different planting environments in Yunnan (Table 3, Figure 1 Under the conditions of [unclear], six experimental sites were set up in Dali, Dehong, Yanshan, Xundian, Yanjin, and Lincang. The sucrose content of the harvested peanut kernels ranged from 4.21% to 6.79%, a difference of 1.6 times. Among them, the average maximum temperature of pilot sites 4 and 5 was 26.00–26.48℃, the average minimum temperature was 15.74–17.66℃, and the average temperature was 20.87–22.07℃, with the sucrose content of the harvested peanut kernels reaching 6.79%–6.87%.

[0061] Based on the analysis of the above experimental results, the suitable planting environment for producing high-sucrose-content colorful peanuts is determined to be: a minimum average temperature of 15-18℃, a maximum average temperature of 26-27℃, and an average temperature of 20-22℃ during the peanut growth period.

[0062] Table 3. Main Overview of the Test Sites

[0063]

[0064] 2.2 Sowing at the appropriate time

[0065] The main factors affecting peanut sowing and emergence are soil temperature and soil moisture. Traditional peanut cultivation often waits for natural rainfall until the soil moisture content stabilizes at 60-70% before sowing, leading to a concentrated market supply, lower prices, and reduced profitability. For multi-colored peanuts, timely early sowing is crucial, ensuring the temperature and soil moisture meet the germination requirements. Early sowing and early market access maximize the profitability of multi-colored peanut cultivation.

[0066] Table 4 shows the germination statistics of colorful peanut seeds under different temperatures. The results indicate that during the period of 8–12℃, the germination potential and germination rate of Yunhua 24 seeds slowly increased with the increase of temperature; at 14–16℃, the germination potential and germination rate reached more than 80% and 95% of the normal levels, respectively. This shows that the minimum germination temperature for Yunhua 24 colorful peanut seeds is 14℃, and colorful peanuts can germinate normally as long as the soil temperature is stable between 14℃ and 16℃.

[0067] Based on the above results, the following method is proposed to improve the production efficiency of colorful peanuts: when the soil temperature is stable above 14℃ in early spring, artificial water supplementation methods such as sprinkler irrigation and drip irrigation should be used to ensure suitable soil moisture and sow as early as possible.

[0068] Table 4. Statistics on the emergence of colorful peanut seedlings at different temperatures.

[0069]

[0070] 2.3 Ridge Cultivation

[0071] Table 5 shows the yield and quality of colorful peanuts under different planting methods. Compared with flat planting, ridge cultivation significantly reduced the incidence of peanut root rot, stem rot, and pod rot, and increased the yield of fresh peanut pods, marketability, and sucrose content of peanut kernels. In particular, ridge cultivation ensures smooth irrigation and drainage in the field, preventing significant waterlogging even during heavy rainstorms. The disease incidence rate was reduced to 6.5%, having virtually no impact on peanut production and the final yield and quality. The yield of fresh peanut pods reached 836 kg per mu, an increase of 55.4% and 30.2% compared to flat planting and ridge cultivation, respectively; the marketability of peanut pods reached 75%, an increase of 10 and 7 percentage points, respectively; and the sucrose content of peanut kernels reached 6.43%, an increase of 59.4% and 22.7%, respectively.

[0072] Based on the above experimental results, the proposed planting method for colorful peanuts is as follows: ridge cultivation, with ridges made up to a height of 40-45cm, a width of 50cm, and a furrow width of 30cm, followed by spot sowing.

[0073] Table 5. Peanut Yield and Quality Analysis under Different Planting Methods

[0074]

[0075] 2.4 Cultivation Methods

[0076] Mulching is currently recognized as an effective way to increase peanut yield. However, in a comparative study of Yunhua 24 peanuts grown in the same field (Table 6), although the yield of fresh peanut pods per mu (667 square meters) decreased by 10.0% compared to mulching cultivation, the quality of peanut pods and kernels was significantly improved. The marketable rate of peanut pods increased by 5 percentage points, and the sucrose content of peanut kernels increased from 3.56% to 4.87±0.63%, an increase of 36.8%. This resulted in high-quality, multicolored peanuts. Based on this, open-field cultivation is recommended in suitable areas.

[0077] Table 6 Comparison of peanut yield and quality under different cultivation methods

[0078]

[0079] 2.5 Effects of the Application of the Comprehensive Nutrient Supply and Plant Protection Program

[0080] The most notable characteristics of the colorful peanut variety are its tall stature and vigorous vegetative growth; the peanut pods are large, but often empty or shriveled, reducing the marketable yield. Scientific field management focuses on increasing calcium fertilizer application, controlling plant growth and pest and disease occurrence, and improving the marketable yield of harvested peanut pods.

[0081] The comprehensive nutrient supply and plant protection program reduced the incidence of root, stem, and fruit rot to 5.4%, a decrease of 40 percentage points, by increasing the application of calcium, magnesium, and phosphate fertilizers, timely supplementing micronutrients, and promptly preventing diseases, pests, and weeds. This essentially eliminated any impact on peanut yield and quality. The commercial pod rate of colorful peanuts increased from 60.8% to 82.5%, an increase of more than 20 percentage points. At the same time, the sucrose content of peanut kernels also increased slightly (Table 7).

[0082] Table 7 Analysis of Peanut Yield and Quality under Different Planting Methods

[0083]

[0084] 2.6 Harvest at the appropriate time

[0085] The accumulation pattern of sucrose content in kernels is shown in the results. Figure 2The sucrose content of Yunhua 24 peanut kernels exhibits a trend of first decreasing, then increasing, and then decreasing again as the kernels develop. The sucrose content is highest during the young pod stage (chicken beak stage), gradually decreasing as the plant grows, reaching its lowest point during the pod-filling stage, increasing again at maturity, and then slightly decreasing at harvest. This indicates that multi-colored peanuts should be harvested promptly at maturity, as the sucrose content decreases after maturity.

[0086] Based on this, a suitable harvesting method for high-sucrose-content colorful peanuts is proposed: harvest the peanuts promptly when they are mature to avoid a decrease in sucrose content after maturity.

[0087] Application Examples

[0088] The above embodiments establish a method for cultivating colorful peanuts with increased sucrose content. Based on this method, a standardized, high-value, and efficient cultivation technique for colorful peanuts has been innovated, and significant results have been achieved in production demonstrations.

[0089] Example 1

[0090] In 2022, Yanjin County, Yunnan Province, introduced the colorful peanut variety Yunhua 24 and applied this technology to establish a 30-mu demonstration area for the new specialty peanut variety, achieving significant results in increasing production and income.

[0091] 1. Planting Plan

[0092] (1) Planting environment: Peanuts were sown on May 20, 2022. The average minimum temperature during the peanut growing season was 16℃, the average maximum temperature was 27℃, and the average temperature was 21.5℃, which met the planting environment conditions of this technology.

[0093] (2) Sowing method: Implemented according to this technical method. Raised ridges for open field cultivation, with ridge height of 40cm, ridge width of 50cm, and furrow width of 30cm; 2 rows per ridge, with 2 sowing furrows on each ridge, and single seeds sown in the furrows with a seed spacing of 6cm.

[0094] (3) Sowing time: When sowing was carried out on May 20, 2022, the local area had entered the rainy season and the soil moisture content was about 65%, so there was no need to supplement the water supply for seedling emergence through artificial irrigation.

[0095] (4) Full-process nutrient supply and plant protection plan.

[0096] The demonstration area implements the full-process nutrient supply and plant protection plan in strict accordance with the unified procurement of materials.

[0097] Before sowing: Coat the seeds with 50 times diluted 15% high-efficiency chlorpyrifos·thiamethoxam·fludioxonil microcapsule suspension;

[0098] At sowing time: use 30 kg of compound fertilizer with N:P2O5:K2O = 15:15:15 and 50 kg of calcium magnesium phosphate fertilizer per mu as seed fertilizer, and use 33% succinate·propyne fluoride microcapsule suspension for pre-emergence weed control.

[0099] Post-seedling stage: Apply 25ml of 20% imidacloprid microemulsion, 2g of 0.136% 28-epichlorohydrin·red·indole seed treatment dispersible powder and 50ml of amino acid liquid fertilizer per mu, plus 200ml of BT + 100g of acetamiprid to supplement nutrition and prevent diseases and pests;

[0100] After flowering and pegging stage: Apply 50g of 325g / L pyraclostrobin SC, 2g of 0.136% 28-epi-glucosinolate seed treatment dispersible powder, 40g of boron-molybdenum fertilizer, 40g of calcium-magnesium fertilizer and 50ml of amino acid-containing liquid fertilizer per mu to supplement micronutrients and prevent diseases and pests.

[0101] Early stage of fruit fullness: Apply 50g of 325g / L pyraclostrobin SC, 25g of 50% benzoyl propiconazole EC, 2g of 0.136% 28-epichlorohydrin seed treatment dispersible powder, 50ml of amino acid liquid fertilizer, and 40g of high phosphorus and potassium foliar fertilizer per mu to prevent pests and diseases and prevent premature aging.

[0102] (5) Harvest at the appropriate time. Harvest peanuts in a timely manner when they are ripe to avoid a decrease in sucrose content after ripening.

[0103] 2. Implementation Results

[0104] Following a field survey and yield measurement organized by the Yunnan Provincial Crop Science Society on September 24, 2022, the incidence of root, stem, and fruit rot in the demonstration area was low (8.7%), which had virtually no impact on yield. The yield of the colorful peanut variety Yunhua 24 was 720.5 kg of fresh pods per mu (approximately 0.067 hectares), equivalent to 360.25 kg of dried pods. This represents an increase of 11.9% and 7.9% respectively compared to the non-demonstration area (which used the local main variety Yunhua 4 and was cultivated using traditional techniques), which yielded 667.0 kg of fresh pods per mu (approximately 0.067 hectares) and 333.5 kg of dried pods per mu (approximately 0.067 hectares). The marketable rate of harvested colorful peanut pods reached 72.4%, an increase of 4.1 percentage points compared to 68.3% in the non-demonstration area. In particular, the sucrose content of the peanut kernels reached 6.54%, nearly double the 3.32% in the non-demonstration area (97.0%) (Table 8).

[0105] Table 8. Achievements of the New Variety Demonstration Zone Construction

[0106]

[0107]

[0108] Example 2

[0109] In 2023, Menglian County, Yunnan Province, planted the new colorful peanut variety Yunhua 24 and applied this technology to build a high-quality peanut production base. This included a core demonstration area of 3,000 mu and a production demonstration base of 30,000 mu.

[0110] 1. Planting Plan

[0111] (1) Planting environment: Peanuts were sown on March 10, 2023. The average minimum temperature during the peanut growth period was 16.5℃, the average maximum temperature was 26.3℃, and the average temperature was 21.8℃, which met the planting environment conditions of this technology.

[0112] (2) Sowing method: Implemented according to this technical method. Raised ridges for open field cultivation, with a ridge height of 45cm, a ridge width of 50cm, and a furrow width of 30cm; 2 rows per ridge, with 2 sowing furrows on each ridge, and single seeds sown in the furrows with a seed spacing of 8cm.

[0113] (3) Sowing time: When sowing was carried out on May 20, 2022, the local area was in the early spring dry season. The demonstration area used drip irrigation pipes to artificially supplement water and keep the soil moisture content at about 65%, thus achieving the goal of early sowing.

[0114] (4) Full-process nutrient supply and plant protection plan.

[0115] The demonstration zone supplies materials through unified procurement and strictly implements the full-process nutrient supply and plant protection plan in accordance with this method.

[0116] Before sowing: Coat the seeds with 50 times diluted 15% high-efficiency chlorpyrifos·thiamethoxam·fludioxonil microcapsule suspension;

[0117] At sowing time: use 30 kg of compound fertilizer with N:P2O5:K2O = 15:15:15 and 50 kg of calcium magnesium phosphate fertilizer per mu as seed fertilizer, and use 33% succinate·propyne fluoride microcapsule suspension for pre-emergence weed control.
