Description

[Technology Field]

[0001] The present invention relates to a novel compound, a method for preparing the same, and its use; more specifically, to a novel 3-aryl-2-propen-1-one derivative, a method for preparing the same, a pharmaceutical composition containing the same, and the use of these for promoting myocardial regeneration. [Background technology]

[0002] Myocardial infarction is a common acute heart disease in clinical practice, caused by a sudden interruption of blood circulation in a portion of the heart muscle. Consequently, the cardiomyocytes are damaged by oxygen deprivation. Generally, once a myocardial infarction occurs, the damaged and dead cardiomyocytes cannot regenerate. Only fibrous scars are formed, covering and repairing the area containing the damaged and dead cardiomyocytes. While fibrous scars help maintain the integrity of the heart, hard fibrous scars cannot contract, further hindering cardiac reconstruction and causing dysfunction. These can lead to heart diseases such as heart failure, arrhythmias, and cardiac shock.

[0003] Currently, treatment for myocardial infarction includes interventional therapies and drug administration. For example, interventional therapies include balloon angioplasty, stenting, and coronary artery bypass surgery, while drug administration includes thrombolytic agents, calcium channel blockers, nitrates, angiotensin-converting enzyme inhibitors, beta-antagonists, and narcotic analgesics.

[0004] However, the above treatment methods developed in clinical practice and basic medical research have not been able to improve the regeneration of myocardial tissue and may cause side effects. Currently, there is no effective treatment other than waiting for the damaged myocardial tissue to self-repair.

[0005] Therefore, in both clinical practice and basic medical research, it is urgent to develop related technologies to promote myocardial regeneration and to provide effective pharmaceuticals for treating myocardial infarction. [Overview of the Initiative]

[0113] Test Example 1: Activity Evaluation of Compounds 1 to 46 for Myocardial Regeneration Myosin is a functionally and structurally important component of the cardiomyocyte and is composed of myosin heavy chains. Among these, α-cardiac myosin heavy chain (α-MHC) is encoded by the MYH6 gene, which is specifically expressed in cardiomyocytes during the early stages of cardiac development. The MYH6 gene is mainly expressed in the atria and plays a crucial role in the contractile action of cardiomyocytes. Furthermore, this gene serves as a clue to recognizing the heart in transcriptional regulation involved in the induction and maintenance of cardiomyocyte lineages. α-MHC is an important protein in the early stages of cardiac development, and the expression level of α-MHC in the H9C2 cell line (cardiomyoblasts) is an appropriate indicator for evaluating the effect of studies that promote stem cell differentiation and maintain the prognosis of myocardial infarction.

[0114] 1.1: Cell culture of the H9C2 cell line The H9C2 cell line is a rat-derived cardiomyocyte, purchased from the Food Industry Development Research Institute (Taiwan). The culture medium is 90% (v / v) Dulbecco modified Eagle medium containing 4 mM L-glutamine (Gibco®, Cat. 12800017) and 10% (v / v) fetal bovine serum (Gibco®, Cat. 10437028), adjusted to contain 4.5 g / L glucose and 1.5 g / L sodium bicarbonate (Sigma, Cat. S5761). Cells were cultured at 37°C in 5% (v / v) CO2 under an atmospheric environment.

[0115] 1.2: MTT test for cell viability testing To find a suitable compound concentration that does not cause cytotoxicity, cells were cultured at a 90% culture density and placed in 24-well plates at a concentration of 1.5 × 10⁶ 4 Cells were seeded at the specified cell / well concentration. After 24 hours, compounds 1 to 46 and DMSO (Sigma, Cat. D4540) were added at concentrations of 0.2 to 30 μg / ml and treated for 48 hours (each group was treated three times). After washing with PBS buffer (phosphate-buffered saline), 0.5 mg / ml MTT (thiazolyl blue tetrazolium bromide; Sigma, Cat. M2128) was added, and the 24-well plates were incubated at 37°C for 4 hours. Subsequently, blue-violet crystals were observed under a microscope. The supernatant was then removed, and 200 μL of DMSO was added to dissolve the crystals. Each sample was mixed again using a pipette, and the absorbance at 570 nm was measured (the value is the average of three measurements). The value for the DMSO group was 100%, meaning that the cell viability was 100%. In the following tests, the concentration of the compounds was selected so that the cell viability was within the range of 80% to 120%, and it was confirmed that the growth of cardiomyocytes was not affected, as shown in Figure 1 below.

[0116] 1.3: Cell destruction and harvesting H9C2 cells were cultured at approximately 90% culture density and placed in 24-well plates in a 1.5 × 10⁶ container. 4Cells were seeded at the specified cell / well concentration. After 24 hours, compounds 1 to 46 and DMSO were added for 48 hours (each group was treated three times). The concentrations of compounds 1 to 46 are listed in Table 5 below. After washing with PBS buffer, 0.05% (w / v) 200 μL / well of trypsin was added for 5 minutes. The cell culture medium was then added to stop the trypsin reaction. The cell suspension was centrifuged at 4°C and 1000 rpm for 3 minutes, and the cells were washed three times with PBS buffer. The cells were then resuspended in PBS and frozen at -80°C. The cells were gently mixed to thaw. The freeze-thaw cycle was repeated three times to disrupt the cells. Finally, the solution was centrifuged at 4°C and 3000 rpm for 15 minutes, the supernatant was collected, and then stored at -20°C.

[0117] [Table 5]

[0118] 1.4: Determination of α-MHC expression levels α-MHC expression levels were determined using a rat myosin heavy chain 6, cardiac alpha (MYH6) ELISA kit (MyBioSource, Cat.MBS753946) and an ELISA reader. The absorbance values at 450 nm read by the ELISA reader were then recorded (the values are the average of three readings). The DMSO group's value was set to 100%, and the relative expression levels of the α-MHC groups treated with compounds 1 to 46 were calculated.

[0119] As shown in Figure 1, after treating H9C2 cells with compounds 1 to 46, the cells nevertheless grew normally (cell viability in the range of 80% to 120%), and α-MHC expression levels increased in all treatment groups compared to the DMSO-treated group. In other words, compounds 1 to 46 actually promote α-MHC expression levels in H9C2 cells. This indicates that compounds 1 to 46 can promote and repair myocardial regeneration.

[0120] Test Example 2: Evaluation of the effects of compounds 1 to 46 on myocardial regeneration in zebrafish. Frostbite-induced cardiac damage (up to 30%) in zebrafish repairs itself within 21 days. Therefore, adult zebrafish are a suitable animal model for studying cardiac injury. After cardiac injury in zebrafish, the regeneration process involves three stages: the first is the proliferation of cardiomyocytes, the second is the regeneration of pericardial cells, and the final is the regeneration of vascular endothelial cells.

[0121] To evaluate the effects of compounds for myocardial regeneration and repair, transformed zebrafish (Tg-fli1-eGFP) were anesthetized and cardiac frostbite was performed. Therefore, the vascular endothelial cells of transformed zebrafish can express green fluorescent protein. After frostbite damage was inflicted on the zebrafish heart, the damaged cells could not express green fluorescent protein, allowing for the evaluation of the damaged area of the heart.

[0122] 2.1: Performing cardiac injury in adult zebrafish Cardiac cryogenic injury was performed in zebrafish using a 6 mm long and 0.8 mm wide cryoprobe. To perform cardiac injury, anesthetized adult zebrafish were placed ventrally upward in a sponge slit. The skin and muscle between the two pectoral fins were incised, and then the underlying silvery subcutaneous tissue was gently opened with forceps to access the heart. The cryoprobe was then gently inserted into the chest to make contact with the heart for cryogenic injury.

[0123] To evaluate the regeneration and repair of zebrafish hearts, cardiac aspiration was performed. During cardiac aspiration, the branchial arches in the thoracic region were incised to access the ventricles and visualize the arterial ball and ventricles. The arterial ball was then removed using arterial forceps, and the heart was excised to complete the cardiac aspiration.

[0124] In the present invention, all experimental steps relating to cryogenic injury or cardiac collection are the same as those described above.

[0125] 2.2: Evaluation of the effects of compounds 1 to 46 on myocardial regeneration in zebrafish After inducing cryogenic injury in transformed zebrafish, they were given a 12.5 μg / g test dose daily for 7 days. On day 8, the zebrafish were sacrificed, the area of cardiac injury was calculated, and cardiac collection was performed to further evaluate the effect of promoting myocardial regeneration and repair in the zebrafish.

[0126] Since the vascular endothelial cells of transformed zebrafish can express green fluorescent protein, the fluorescent regions can indicate undamaged areas. In other words, areas without fluorescence are damaged areas, and by calculating the proportion of these areas relative to the heart, the percentage of cryogenically damaged areas can be obtained.

[0127] The group given a test sample containing only the solvent is shown as the solvent group. To confirm the percentage of damage area in the solvent group, zebrafish with cardiac injuries were given a test sample containing only the solvent for 7 days. The percentage of the low-temperature damage area was 27%. Next, zebrafish with cardiac injuries were given test samples containing compounds 1 to 46 for 7 days, and the percentage of the low-temperature damage area for each group was calculated and is shown in Figure 2. As shown in Figure 2, the percentage of the low-temperature damage area in all groups given test samples containing compounds 1 to 46 for 7 days was clearly reduced compared to the result with the solvent group. In other words, compounds 1 to 46 can actually promote myocardial regeneration and repair in zebrafish.
