Description

Skin soothing cosmetic composition containing seven natural complexes

[0001] The present invention relates to a cosmetic composition for skin soothing containing seven kinds of natural complexes, and more particularly, to a natural complex extract having the effect of alleviating skin inflammation by suppressing the expression of mRNA of inflammatory factors (IL-1α, IL-6) that cause inflammation by ultraviolet rays (UVB), and a method for producing the same.

[0002] The skin, the tissue that covers the body's surface, protects against physical stimuli like shock, heat, and sunlight. It also protects against chemical stimuli and bacterial invasion, and helps the body adapt to environmental changes by maintaining a constant body temperature. However, because it is so closely exposed to the external environment, it is also susceptible to sensitization from exposure to UV rays, environmental pollution, and chemicals.

[0003] Cytokines, such as tumor necrosis factor (TNF-α) and interleukins (IL-1α, IL-6), are key components of the inflammatory response and are collectively referred to as inflammatory factors. Inflammation mediated by these inflammatory factors is one of the body's defense mechanisms to minimize the response when cells or tissues are damaged by any cause and to restore the damaged area to its original state. Inflammation is also a primary immune response to identify and eliminate various foreign substances, such as pathogens, that have invaded the body to maintain homeostasis. However, this inflammation causes damage to cells and connective tissues through proteolytic enzymes, and damage to connective tissues reduces skin elasticity, leading to wrinkles and rapid skin aging.

[0004] Meanwhile, reactive oxygen species (ROS) refer to highly reactive radicals containing oxygen, and nitric oxide (NO) is also called reactive nitrogen species (RNS) containing nitrogen atoms (N), but is generally considered a subgroup of reactive oxygen species. Nitric oxide or reactive oxygen species can easily be generated due to environmental pollution, ultraviolet rays, stress, smoking, etc., and if produced in excess, it destroys collagen and elastin, which are fibrous structures in the skin, causing a loss of elasticity, which can lead to aging symptoms such as "wrinkles" and "sagging." In addition, reactive oxygen species are considered to be substances that acidify the skin, cause waste products to accumulate under the skin, and block blood circulation, accelerating skin aging.

[0005] Therefore, suppressing inflammation and reactive oxygen species and soothing irritated skin are crucial elements in preventing skin aging. Consequently, the development of functional materials with anti-inflammatory and antioxidant effects is becoming increasingly important to prevent various skin inflammations and aging.

[0006]

[0007] The purpose of the present invention is to provide a skin soothing cosmetic composition containing seven kinds of natural complexes and a method for producing the same.

[0008] Another aspect of the present invention is to provide a cosmetic composition that effectively suppresses the expression of mRNA of IL-1α and IL-6, which are inflammatory factors that cause inflammation in the skin, and the generation of active oxygen directly related to skin aging.

[0009] Specifically, the purpose of the cosmetic composition of the present invention is to confirm the excellent skin anti-inflammatory and antioxidant effects of an extract obtained by mixing seven kinds of materials, including Phellodendron amurense, Oryzae, Fennel, Prunus mume, Lithospermum officinalis, Schisandra chinensis, and Tangerine, in a specific ratio, and to provide a cosmetic composition comprising the same.

[0010] The technical tasks to be achieved in the embodiments are not limited to those mentioned above, and other technical tasks not mentioned can be considered by a person having ordinary skill in the art from the various embodiments described below.

[0011]

[0012] To achieve these purposes, the present invention provides a cosmetic composition for skin soothing and anti-inflammation, comprising extracts of at least one selected from the group consisting of Phellodendron amurense, Oryza sativa, Fennel, Prunus mume, Lithospermum officinalis, Schisandra chinensis, and Tangerine.

[0013] Specifically, the cosmetic composition may be formed by including a mixed extract obtained by mixing and extracting all of the above Phellodendron amurense, Alder tree, Fennel, Yellow plum, Lithospermum chinensis, Schisandra chinensis, and Tangerine chinensis, and more specifically, may be formed by using a mixed extract obtained by mixing in a ratio of 10 to 20 wt% of Phellodendron amurense, 21 to 28 wt% of Alder tree, 2 to 7 wt% of Fennel, 2 to 7 wt% of Yellow plum, 10 to 14 wt% of Lithospermum chinensis, 29 to 35 wt% of Schisandra chinensis, and 5 to 12 wt% of Tangerine chinensis. In one embodiment of the present invention, the cosmetic composition was manufactured by including 15 wt% of Phellodendron amurense, 24 wt% of Alder tree, 4 wt% of Fennel, 4 wt% of Yellow plum, 12 wt% of Lithospermum chinensis, 32 wt% of Schisandra chinensis, and 9 wt% of Tangerine chinensis, but is not limited thereto.

[0014] In the present invention, the phellodendron amurense is the dried bark of Phellodendron amurense, a member of the Zanthoxylum family. Phellodendron amurense is a deciduous broad-leaved tree that grows deep in the mountains, reaching a height of approximately 10 m. Its bark is covered with cork and its inner bark is yellow. The leaves are lanceolate and obovate, and the leaf margins have fine teeth and soft hairs. The berberine component of Phellodendron amurense exhibits hypotensive effects, antibacterial effects against pathogenic microorganisms, and anti-inflammatory effects.

[0015] The alder tree described herein can reach a height of 20 m, has hairy young branches, and its leaves grow alternately and are oblong. Alder trees grow in low-lying wetlands and are cultivated throughout Korea, as well as in Japan, Manchuria, and Siberia. In oriental medicine, the bark or buds are used as a medicinal herb for fever reduction, hemostasis, and digestive inflammation.

[0016] The fennel plant in the present invention is an annual plant of the Apiaceae family, native to the Mediterranean coast. It grows 1.5 to 2 meters tall, has a straight, hollow stem, and its leaves are long, sheath-shaped, pointed, and finely divided into three to four feather-like lobes. It is used worldwide as a food or spice, and its diverse pharmacological properties have led to its use in Oriental medicine. The phenolic compounds in fennel are known to exhibit excellent analgesic and anti-inflammatory effects.

[0017] The plum in the present invention is the fruit of the plum tree, native to China. It grows to a height of 5-10 m, has alternate, ovate leaves, sharply serrated edges, and hairs on both sides. Its light red flowers bloom before the leaves, a characteristic feature. Plums are rich in organic acids, promoting carbohydrate metabolism and relieving fatigue. They also have excellent detoxification properties, helping to treat conditions like stomach upset and food poisoning.

[0018] The lily of the valley plant of the present invention is a perennial herb belonging to the family Boraginaceae. Its stem is straight, and its roots branch out in a ginseng-like pattern. The purple pigment in the root forms around the outer skin, resulting in a strong purple color. The presence of purple pigment in the root is a distinguishing characteristic. The root has been used as a dye and medicinal ingredient since ancient times, and is known to be particularly effective in treating skin inflammation, including dermatitis, freckles, and rashes.

[0019] The five flavors mentioned in the present invention are named after the five flavors they possess, a harmonious balance of sour, sweet, spicy, bitter, and salty. They grow in Korea, Japan, China, and other countries. Omija contains ingredients such as schizandrin and gomisin, which are known to have health benefits such as strengthening the heart, lowering blood pressure, and boosting immunity.

[0020] In the present invention, the tangerine is the fruit of the tangerine tree. The tangerine tree is a deciduous broadleaf shrub belonging to the Rutaceae family, growing approximately 3 m tall and bearing white flowers in May. The tangerine fruit is round, ripens to a yellow color, and has a pleasant fragrance, but is inedible. The tangerine contains essential oils such as limonene, naringin, and ponsyrin. In oriental medicine, it has been used to treat bronchial diseases.

[0048]

[0049] Example 1. Preparation of natural product extracts

[0050] The cosmetic composition of the present invention is manufactured using seven kinds of natural products, and extracts of each of these products were manufactured.

[0051] Specifically, seven kinds of natural products, Schisandra chinensis, Oryzae, Phellodendron amurense, Lithospermum Parkii, Tangerine, Fennel, and Yellow Plum, were each prepared in a dried state, and 20 g of each of these natural products were separately soaked in 200 g of 70% ethanol, extracted at 60℃ for 4 hours, filtered through a filter paper (Whatman No. 2), and concentrated. The obtained concentrate was dissolved in a solvent (butylene glycol 30%, purified water 70%) to 1.0% (w / w), and then filtered through a filter paper (Durapore 0.22 μm, PVDF) to obtain natural product extracts for each of the seven kinds of natural products of the present invention.

[0052]

[0053] Experimental Example 1. Confirmation of the skin soothing effect through inhibition of IL-1α activity of individual materials.

[0054] It was evaluated whether each of the above natural product extracts exhibited an effect of suppressing skin inflammation by inhibiting the expression of interleukin-1α (IL-1α).

[0055] Specifically, to confirm the true efficacy, an IL-1α gene expression experiment was performed. HaCaT (immortalized keratinocyte) cells were seeded in 60 mm dishes and cultured for 24 hours. The medium was replaced with serum-free medium, and 50 mJ / cm2 of Ultraviolet B (UVB), which causes inflammatory reactions such as sunburn, erythema, and edema in HaCaT, was irradiated. The samples were treated at various concentrations and cultured for 4 hours in a 37°C, CO25% incubator. After culture, the cells were harvested with QIAzol™ Lysis Reagent (Qiagen) and RNA was isolated according to the manufacturer's method. The extracted total RNA was quantified, and cDNA was synthesized from 1 μg of RNA using a cDNA synthesis kit on a ProFlex PCR system (Applied Biosystems). To confirm gene expression, a 7500 fast real-time PCR system (Applied Biosystems) was used, and the results are shown in Table 1 below.

[0056] As a result of the cytotoxicity evaluation, it was confirmed that there was no toxicity when treated at a concentration of 1% for Schisandra chinensis, Oryzae, Phellodendron amurense, and Zizyma chinensis, and at a concentration of 0.5% for Tangerine, Fennel, and Yellow Plum, and the experiment was conducted accordingly.

[0058] As a result of the IL-1α expression inhibition evaluation, as shown in Table 1, Schisandra chinensis showed the highest IL-1α expression inhibition activity at 82.8%. All seven materials were confirmed to be effective in inhibiting IL-1α expression, and in particular, Schisandra chinensis, Oryzae, Phellodendron amurense, Lithospermum chinensis, and Tangerine chinensis showed a superior inhibition rate than the positive control, quercetin.

[0059] Experimental Example 2. Confirmation of the skin soothing effect through the inhibition of IL-6 activity of individual materials.

[0060] We evaluated whether each of the above natural product extracts exhibited an effect of suppressing skin inflammation by inhibiting the expression of interleukin-6 (IL-6).

[0061] Specifically, an IL-6 gene expression experiment was performed to confirm the true efficacy. HaCaT (immortalized keratinocyte) cells were seeded on 60 mm dishes and cultured for 24 hours, then replaced with serum-free medium, and irradiated with Ultraviolet B (UVB) at 50 mJ / cm2, which causes inflammatory reactions such as sunburn, erythema, and edema in HaCaT, and the samples were treated at various concentrations and cultured for 4 hours in a 37℃, CO25% incubator. After culture, the cells were collected with QIAzol™ Lysis Reagent (Qiagen) and RNA was isolated according to the manufacturer's method. The extracted total RNA was quantified, and then cDNA was synthesized from 1 μg of RNA using a cDNA synthesis kit using a ProFlex PCR system (Applied Biosystems). To confirm gene expression, a 7500 fast real-time PCR system (Applied Biosystems) was used, and the results are shown in Table 2 below.

[0062] As a result of the cytotoxicity evaluation, it was confirmed that there was no toxicity when treated at a concentration of 1% for Schisandra chinensis, Oryzae, Phellodendron amurense, and Lithospermum chinensis, and at a concentration of 0.5% for Tangerine, Fennel, and Yellow Plum, and the experiment was conducted accordingly.

[0064] As a result of the IL-6 expression inhibition evaluation, as shown in Table 2, Omija showed the best activity at 82.0%. All seven materials were confirmed to be effective in inhibiting IL-6 expression, and in particular, Omija, Oryzae, and Phellodendron amurense showed a superior interleukin-6 expression inhibition rate than the positive control, quercetin.

[0065] Examples 2 to 5. Preparation of seven types of composite extracts

[0066] The composite extracts of Examples 2 to 5 of the present invention were prepared by mixing in the weight ratios shown in Table 3 below.

[0067] The composite extract of Example 2 was prepared by mixing seven materials in equal proportions, and for Examples 3 to 5, the extracts were mixed and extracted in the weight ratios shown in Table 3 below, taking into account the results of Experimental Examples 1 and 2, so as to exhibit the best anti-inflammatory effect.

[0068] 50 g of 7 kinds of materials mixed in the ratios of Examples 2 to 5 were immersed in 500 g of 70% ethanol, extracted at 60°C for 4 hours, filtered through filter paper (Whatman No. 2), and concentrated. The obtained concentrate was dissolved in a solvent (butylene glycol 30%, purified water 70%) to a concentration of 1.0% (w / w), filtered through filter paper (Durapore 0.22 μm, PVDF), and then used.

[0070] Experimental Example 3. Confirmation of the skin soothing effect of the complex extract through inhibition of IL-1α activity.

[0071] In order to evaluate whether the composite extracts of Examples 2 to 5 inhibit the expression of IL-1α and are effective in suppressing skin inflammation, an experiment was conducted using the same method as Experimental Example 1, and the results are shown in Table 4 below.

[0072] Sample UVB Concentration (%) IL-1α Inhibition Rate (%) Control--93.6 UVB 50mJ-0.0 Quercetin (μM) 50mJ 2534.2 Example 250mJ 3.0 11.6 Example 350mJ 3.0 38.2 Example 450mJ 3.0 24.7 Example 550mJ 3.0 27.3

[0073] As shown in Table 4, Example 3 showed an inhibition rate of 38.2%, which was close to the positive control. On the other hand, Example 2 showed a low inhibition rate of 11.6%. In addition, it was confirmed that the effect was significantly superior in the ratio of Example 3 compared to the results of Example 5, which contained the largest amount of Schisandra chinensis, which was the most effective when used alone, and Example 4, which contained a larger amount of Oryzae.

[0074] Experimental Example 4. Confirmation of the skin soothing effect of the complex extract through inhibition of IL-6 activity.

[0075] In order to evaluate whether the complex extracts of Examples 2 to 5 are effective in suppressing skin inflammation by inhibiting the expression of IL-6, an experiment was conducted using the same method as Experimental Example 2, and the results are shown in Table 5 below.

[0076] Sample UVB Concentration (%) IL-6 Inhibition Rate Control--95.8 UVB 50mJ-0.0 Quercetin (μM) 50mJ 2546.5 Example 250mJ 3.0- Example 350mJ 3.0 22.9 Example 450mJ 3.0 10.8 Example 550mJ 3.0 12.1

[0077] As shown in Table 5, in the case of Example 3, an inhibition rate of 22.9% was confirmed, whereas in the case of Example 2, the expression rate was 113.2%, showing that the expression of IL-6 was rather promoted. In addition, the results of Examples 4 and 5 showed lower effects than Example 3, and considering that this is the same as Experimental Example 3, the contents of Schisandra chinensis and Oryzae, which showed the best effect as a single raw material, increased, it can be seen that this is the result of the synergistic effect according to the mixing ratio of Example 3.

[0078] Experimental Example 5. Confirmation of antioxidant effect through inhibition of ROS production.

[0079] To evaluate whether the composite extracts of Examples 2 to 5 exhibit antioxidant effects by inhibiting the production of reactive oxygen species (ROS), experiments were conducted using the following method.

[0080] To confirm the antioxidant efficacy, a Reactive Oxygen Species (ROS) assay was performed. HaCaT (immortalized keratinocyte) cells were seeded in 96-well plates and cultured for 24 hours. After replacing with serum-free medium, samples were treated at various concentrations and cultured for 24 hours in a 37°C, CO25% incubator. After culture, the cells were washed with HBSS, treated with H2DcFDA solution, and incubated in an incubator for 30 minutes in a light-shielded environment. After washing with HBSS, 100 mM H2O2 solution was treated, incubated in an incubator for 10 minutes, and the fluorescence value was measured to determine the amount of ROS produced. The results are shown in Table 6 below.

[0081] Sample Concentration (%) ROS Production Inhibition Rate (%) Control-0.0 EGCG (ppm) 1.0 2 6.48 Example 20.75 4.56 Example 30.75 19.23 Example 40.75 7.50 Example 50.75 9.87

[0082] As shown in Table 6, Example 3 showed an inhibition rate of 19.23%, whereas Example 2 showed a low inhibition rate of 4.56%. In addition, Example 4 showed 7.50%, and Example 5 showed 9.87%, which were higher than Example 2, but only about half the effect compared to Example 3. When these experimental results are combined, it can be confirmed that the seven types of complex extracts of the present invention and the cosmetic composition containing the same can effectively suppress the expression of mRNA of inflammatory factors IL-1α and IL-6 and the generation of active oxygen directly related to skin aging.
