Description

[0001] This published application claims priority over pending Iranian invention patent application No. 139850140003008438, registered on December 15, 2019, entitled "Herbal Combination for the Treatment of Female Infections, Vulvovaginal and Pelvic Organ Inflammations", which is fully incorporated herein by reference. Technical Field

[0002] The present application relates to herbal compounds and methods of use thereof for treating vulvovaginitis, restoring normal vulvovaginal flora, treating vaginal-related diseases, and inhibiting infection or inflammation of pelvic organs. Background Art

[0003] Vulvovaginitis is an inflammation of the lower genitals that affects the vagina and vulva. These inflammatory symptoms include abnormal vaginal discharge, odor, itching and / or discomfort. Vulvovaginitis is mainly caused by bacteria or fungi. Infections caused by protozoa (mainly Trichomonas vaginitis) only account for 10% of these infections. Bacterial infections account for about 50% to 60% of all vulvovaginitis. The most common microorganisms observed in this type of infection (inflammation) include Gardnerella vaginalis, Gardnerella mobiluncus, Chlamydia trachomatis, Mycoplasma hominis, Peptostreptococcus or Ureaplasma urealyticum.

[0004] Approximately 70% of women will have a vulvovaginal fungal infection during their lifetime. In addition, approximately 40% of women with a vulvovaginal bacterial infection will develop a yeast infection. Bacterial and protozoal infections are mainly treated with antibiotics and antiparasitic drugs from the nitroimidazole family such as metronidazole. Fungal infections are usually treated with antifungals from the imidazole-derived drug family such as econazole, miconazole and fluconazole. Despite all the above-mentioned drug treatment modalities, statistics show that half of women treated for a vulvovaginal bacterial infection will be infected again within the next 12 months. It is important to mention that antibiotics used to treat infections in women often lead to a disruption of the normal flora that protects the vaginal area, which can lead to recurrence of the infection. Therefore, the management of Candida infections strongly requires the introduction of new drugs with antifungal properties.

[0005] Therefore, according to the above points, the existing chemotherapy for vulvovaginitis and other diseases of the female pelvic organs is mostly unsatisfactory and causes various side effects including, recurrence, development of drug resistance, disruption of the balance of normal flora, and discomfort of other organs such as stomach, intestines and liver due to systemic absorption of drugs. Therefore, the research on active ingredients, including natural or herbal ones, can be a good solution to this problem.

[0006] Some natural or herbal remedies can exhibit different and sometimes undesirable symptoms at high doses or when used directly for treatment. To address the above problems, a combination (drug) and prescription approach for vulvovaginitis and other diseases of the female pelvic organs is needed. Summary of the invention

Example

[0073] Example 1: Preparation of herbal disinfectant composition

[0074] Plant species, including citron, myrrh, myrtle, and rose, were first obtained and dried.

[0075] The gum of myrrh, the fruit of watermelon, the leaves of myrtle and the buds of rose are ground using industrial grinders.

[0076] 30% by weight of watermelon fruit powder, 20% by weight of myrrh gum, 30% by weight of myrtle leaves, and 20% by weight of rose buds were mixed.

[0077] A fiber bag made of multi-layer (3-6 layers) cotton fiber with circular vertical weave was prepared, which was 5-10 cm in length, 2-5 cm in area, and cone-shaped. One end of the bag had a rope 8-10 cm long, woven together with the cylinder.

[0078] Place 2-7 grams of the herbal disinfectant composition as a powder into each fiber bag.

[0079] Example 2: Preparation of herbal disinfectant composition

[0080] The powder obtained by mixing 30% by weight of the powder of the fruit of the medicinal watermelon, 20% by weight of the gum of myrrh, 30% by weight of the leaves of myrtle and 20% by weight of the rose buds will be made into granules of 1-5 cm in size. Here, after the granules are prepared, 2-7 grams of the prepared granules are placed in each fiber bag.

[0081] The prepared granules contained 8.5% total ash, 4% acid insoluble ash and 6-6.6% moisture and pH = 3.83. The moisture content of the granules was studied by both direct and azeotropic methods.

[0082] Example 3: Investigation of drug release

[0083] The effects of three patterns on the amount of drug released were investigated. These patterns all relate to the effect of the weaving of the fiber packages on the released content, and include four types of simple weaves, vertical weaves, low-pilled weaves, and high-pilled weaves. To conduct this test, the fiber packages containing the drug were placed in a buffer solution at a temperature of 37°C in a stability test chamber instrument, and the pH of the buffer solution was close to the pH of the vagina (pH≈4). Samples were taken from the buffer solution within 5-24 hours, and the content of the active agent of the drug watermelon called colocynthis was measured using an HPLC method ( Figure 3 ). The results showed that the packaging woven using the vertical weaving method resulted in the highest drug release. Using this weaving method, all components of the drug were released into the environment after 20 hours.

[0084] Example 4: Study of inflammatory effects

[0085] To ensure that the topical administration of the drug prepared by the present invention is safe and harmless, two groups of rabbits (n = 3) including a target group (a fiber pack treated with a drug in the intravaginal method) and a control group (a fiber pack treated with no drug) were studied by histological study. Therefore, a fiber pack containing a drug and a fiber pack without a drug were placed in the vagina of the target group and the control group every day and left for 24 hours for 14 days. After 14 days, tissue samples were taken from the vaginal area of these animals and slides were prepared. The prepared slices of the incision were studied by optical microscopy. Figure 4 The histological results of samples prepared from vaginal tissues are shown. The results showed that the herbal disinfectant composition, target group (Fig.-4, d, e and f) did not induce any significant allergic inflammatory response in these animals as compared to the target group (Fig.-4, a, b and c).

[0086] Example 5: Patient treatment methods and clinical trials

[0087] Each complete drug pack consists of 14 fiber packs, and the patient places one in the vagina for up to 24 hours a day (15-24 hours), then removes it and uses another pack. To completely treat vaginitis, the patient should use the fiber packs for 14 days as described above. For chronic infections or pelvic inflammatory disease, treatment can continue for several courses (6-8 months) until the desired effect is achieved. According to field studies, several courses of use of this drug cause no or minimal side effects to volunteers and patients.

[0088] This fiber pack was tested on 30 patients after obtaining their acquiescence regarding drug allergy and effectiveness testing. The patients did not experience any side effects. In addition, recovery of patients affected by vaginal infections was confirmed after one or several courses of use.
