Abstract
Description
Technical Field
[0001] The invention belongs to the field of organic synthesis, and in particular relates to a method for preparing a trifluorovinyl alkyl ether compound and an intermediate thereof. Background Art
[0002] There are two main categories of trifluorovinyl ethers, one is trifluorovinyl aryl ether and the other is trifluorovinyl alkyl ether.
[0003] Trifluorovinyl aryl ether is an important monomer for polymer synthesis, mainly used for synthesizing polymers containing hexafluorocyclobutane structure. The most widely used method for synthesizing the monomer of trifluorovinyl aryl ether is to use phenol as raw material, directly react with the easily available BrCF2CF2Br under alkaline conditions to obtain 2-bromo-1,1,2,2-tetrafluoroaryl ether (ArOCF2CF2Br), and then obtain trifluorovinyl aryl ether by zinc powder dehalogenation. The synthesis of trifluorovinyl aryl ether is relatively simple because it is relatively easy to synthesize 1,2-dihalotetrafluoroethyl aryl ether compounds directly from phenol, and the reagents used are safe and easy to operate. However, there is a significant disadvantage in the direct zinc powder dehalogenation step: a considerable amount of by-product 1,1,2,2-tetrafluoroaryl ether (ArOCF2CF2H) is generated, and the boiling point and polarity of this by-product are very close to those of the main product, which brings great difficulties to separation and purification (CN106631716A, 2017).
[0004] The research on the corresponding trifluorovinyl alkyl ethers mainly focuses on perfluoro(alkyl vinyl ether) (R f OCF=CF2, PAVE), such as perfluoro(methyl vinyl ether) (CF3OCF=CF2, PMVE), perfluoro(ethyl vinyl ether) (CF3CF2OCF=CF2, PEVE) and perfluoro(propyl vinyl ether) (CF3CF2CF2OCF=CF2, PPVE). This class of compounds is widely used in the synthesis of fluororesins.
[0005] Compared with the above two widely studied and applied trifluorovinyl ethers, another type of trifluorovinyl alkyl ether (oxygen atom is directly connected to non-perfluoroalkyl carbon atom) is rarely studied. This is mainly because the reported synthesis methods for this type of trifluorovinyl alkyl ether are few and not efficient. There are three main synthesis methods: one is Pellerite's report on the use of hexafluoropropylene oxide to obtain trifluorovinyl ether by thermal decomposition of potassium salt of 2-alkoxytetrafluoropropionic acid (J. Fluor. Chem. 1990, 49, 43). Even after continuous improvement, the yield is not ideal. Only the yield of trifluorovinyl ether substituted with trifluoroethoxy (CF3CH2O-) can reach a moderate yield (67%), while the yield of trifluorovinyl ethyl ether (EtOCF=CF2) substituted with potassium salt of tetrafluoropropionic acid substituted with ethoxy (EtO-) can only obtain about 10% of the yield.
[0006] The second is the direct reaction of sodium alcoholate and tetrafluoroethylene to prepare trifluorovinyl alkyl ether reported by Dixon and Kojima et al. (US Patent 2,917,548,1959; Chem.Soc.Jpn.1962,35,532). This is the method commonly used to synthesize trifluorovinyl alkyl ether. However, this method requires the use of tetrafluoroethylene gas at a relatively high pressure and heating, and has strict requirements on experimental operation. If the operation is improper, there will be a risk of explosion. In addition, it is difficult to obtain high-purity sodium alcoholate, and when the sodium alcoholate reacting with tetrafluoroethylene is mixed with raw alcohol, under this reaction condition, the alcohol can react with tetrafluoroethylene to generate a by-product tetrafluoroethyl alkyl ether (ROCF2CF2H). This method is mainly used to prepare some simple trifluorovinyl alkyl ethers, such as CF2=CFOCH3 and CF2=CFOCH2CH3, and the yield is generally low, especially for some complex trifluorovinyl alkyl ethers (for example, the examples given in the US patents show that the calculated yield of nBu ether is 40%, and that of nPr ether is 10%; the yields of Me ether, Et ether and iPr ether in Japanese literature are 28%, 53% and 26% respectively).
[0007] Third, Liu Bo et al. recently disclosed that tetrafluoropropyl trifluorovinyl ether was prepared from trifluoroethylene gas through the reaction of 1,2-dibromo-1,1,2-trifluoroethane intermediate and 2,2,3,3-tetrafluoropropanol (CN111018679A, 2020), but it was limited to one report of tetrafluoropropyl trifluorovinyl ether, and 1,2-dibromo-1,1,2-trifluoroethane was prepared from trifluoroethylene; however, trifluoroethylene gas is a by-product produced in the production process of tetrafluoroethylene and is not easy to obtain.
[0008] The synthesis of trifluorovinyl alkyl ether (ROCF=CF2) is generally prepared by the second method mentioned above, that is, the alcohol substrate is reacted with sodium alcoholate and tetrafluoroethylene gas (there is a risk of explosion, and tetrafluoroethylene gas is not easy to be transported outside the place of production), instead of using phenolic substrates to react with safe and easily available XCF2CF2X (X is chlorine or bromine) and then dehalogenating with zinc powder to obtain trifluorovinyl alkyl ether as in the synthesis of trifluorovinyl aryl ether. The main reason is that there is currently a lack of universal methods for the direct reaction of alcohol substrates with XCF2CF2X to efficiently generate 2-X-1,1,2,2-tetrafluoroethyl alkyl ether. There are only a few sporadic cases at present:
[0009] The first example is the direct reaction of ethanol with BrCF2CF2Br under the action of potassium hydroxide, which can only obtain the target product CH3CH2OCF2CF2Br with a yield of 31% (J. Fluorine Chem. 1979, 13, 123);
[0010]
[0011] The second example is that an alcohol containing an oxygen atom at the β-position directly reacts with BrCF2CF2Br in DMF under the action of NaH to obtain the target product 2-bromo-1,1,2,2-tetrafluoroethyl ethyl ether with a yield of 88%. However, the structure of the reaction substrate is special, and the β-position of the hydroxyl group must contain an oxygen atom substituent (EP3670501A1, 2006);
[0012]
[0013] The third example is the direct reaction of fluoroalkyl-substituted ethanol with BrCF2CF2Br in DMF under the action of NaH to generate 2-bromo-1,1,2,2-tetrafluoroethyl ether with good to excellent yields. Similarly, the reaction substrate has a special structure and contains fluorine atoms at the β-position of the hydroxyl group (JP2007332060A, 2007);
[0014]
[0015] However, the above conditions are not universal. For example, benzyl alcohol can directly react with BrCF2CF2Br under the action of potassium to obtain 2-bromo-1,1,2,2-tetrafluoroethylated product PhCH2OCF2CF2Br with a yield of only 2.6% (J. Fluorine Chem. 2015, 171, 139).
[0016]
[0017] In addition, we used phenylbutanol as the template substrate and directly reacted it with BrCF2CF2Br in DMF with sodium hydride as the base to give Ph(CH2)4OCF2CF2Br with a yield of only 27%.
[0018]
[0019] It can be seen that, unlike aryl trifluorovinyl ethers, there is currently no universal and practical methodology for ordinary laboratories to synthesize trifluorovinyl alkyl ethers from alcohols. Therefore, it is very important to successfully react alcohols with BrCF2CF2Br to generate 2-bromo-1,1,2,2-tetrafluoroethyl alkyl ethers. This is also the biggest challenge and research focus in the synthesis of trifluorovinyl alkyl ethers from alcohols and BrCF2CF2Br.
[0020] Therefore, the art is in urgent need of providing a safe, efficient and widely applicable method for synthesizing trifluorovinyl ether compounds and 2-halogen-tetrafluoroethyl alkyl ether compounds. Summary of the invention

[0079] In the fourth aspect of the present invention, there is also provided an application of the 2-halogen-1,1,2,2-tetrafluoroethyl ether compound as described above in the preparation of trifluorovinyl ether compounds; wherein the 2-halogen-1,1,2,2-tetrafluoroethyl ether compound is represented by the formula R'-OCF2CF2X as described above.
[0080] In one embodiment of the application, the steps include:

[0082]
[0083] The operation and reaction conditions of the dehalogenation reaction may be as shown in the operation and reaction conditions of the preparation method of trifluorovinyl alkyl ether described above.
[0084] Unless otherwise indicated, the following definitions used herein shall apply. For purposes of the present invention, chemical elements are consistent with the Periodic Table of the Elements, CAS version, and Handbook of Chemistry and Physics, 75th edition, 1994. In addition, general principles of organic chemistry can be found in "Organic Chemistry", Thomas Sorrell, University Science Books, Sausalito: 1999, and "March's Advanced Organic Chemistry" by Michael B. Smith and Jerry March, John Wiley & Sons, New York: 2007, the entire contents of which are incorporated herein by reference.
[0085] In this specification, groups and substituents thereof can be selected by those skilled in the art to provide stable structural moieties and compounds. When substituents are described by conventional chemical formulas written from left to right, the substituents also include chemically equivalent substituents obtained when the structural formula is written from right to left.
[0086] Certain chemical groups defined herein are preceded by simplified notations to indicate the total number of carbon atoms present in the group. For example, a C1-C6 alkyl group refers to an alkyl group as defined below having a total of 1, 2, 3, 4, 5, or 6 carbon atoms. The total number of carbon atoms in the simplified notation does not include carbons that may be present in substituents of the group.
[0087] As used herein, numerical ranges defined in substituents such as 0 to 4, 1-4, 1 to 3, etc. indicate integers within the range, such as 1-6 is 1, 2, 3, 4, 5, 6.
[0088] In addition to the foregoing, when used in the specification and claims of the present application, the following terms have the meanings indicated below unless otherwise specifically stated.
[0089] The term "comprising" is an open expression, that is, including the contents specified in the present invention but not excluding other contents.
[0090] The term "substituted" means that any one or more hydrogen atoms on a particular atom are replaced by a substituent as long as the valence state of the particular atom is normal and the compound after the substitution is stable.
[0091] In general, the term "substituted" means that one or more hydrogen atoms in a given structure are replaced by a specific substituent. Further, when the group is substituted by more than one substituent, the substituents are independent of each other, that is, the more than one substituent may be different from each other or the same. Unless otherwise indicated, a substituent group may be substituted at each substitutable position of the substituted group. When more than one position in the given structural formula can be substituted by one or more substituents selected from a specific group, the substituents may be substituted at each position in the same or different manner. When a substituent group and a substituted group are both cyclic structures, the substitution may be that the substituent group is connected to the substituted group by a single bond, or that the substituent group is connected to the substituted group by a ring.
[0092] In various parts of this specification, substituents of compounds disclosed in the present invention are disclosed according to group types or ranges. It is particularly pointed out that the present invention includes each independent subcombination of the individual members of these group types and ranges. The term "C x -C y "Alkyl" refers to a straight or branched chain saturated hydrocarbon containing x to y carbon atoms. For example, the term "C1-C6 alkyl" or "C 1-6 "Alkyl" specifically refers to methyl, ethyl, C3 alkyl, C4 alkyl, C5 alkyl and C6 alkyl disclosed independently; "C 1-4 The term "alkyl" specifically refers to the independently disclosed methyl, ethyl, C3 alkyl (i.e. propyl, including n-propyl and isopropyl), C4 alkyl (i.e. butyl, including n-butyl, isobutyl, sec-butyl and tert-butyl).
[0093] As used herein, the terms "part", "moiety", "chemical moiety", "group", "chemical group" refer to a specific fragment or functional group in a molecule. A chemical moiety is generally considered to be a chemical entity embedded in or attached to a molecule.
[0094] When a substituent is listed without indicating the atom through which it is attached to a chemical formula (including but not specifically mentioned compounds), such a substituent may be bonded via any atom thereof. Combinations of substituents and / or their variants are permitted only if such combinations result in stable compounds.
[0095] When any variable (such as R z ) appears multiple times in the definition of a compound, the definition of the variable at each position is independent of the definition of the other positions, and their meanings are independent of each other and do not affect each other. z group-substituted, that is, the group may be replaced by up to 3 R z Replace, the position R z Definition and other positions Rz In addition, combinations of substituents and / or variables are permissible only if such combinations result in stable compounds.
[0096] When there is no explicit indication that a group has a substituent, such group refers only to unsubstituted groups. For example, when "C1-C4 alkyl" is not preceded by "substituted or unsubstituted", it only refers to "C1-C4 alkyl" itself or "unsubstituted C1-C4 alkyl".
[0097] In various parts of the present invention, linking substituents are described. When the structure clearly requires a linking group, the Markush variable listed for that group should be understood as a linking group. For example, if the structure requires a linking group and the Markush group definition for that variable lists "alkyl", it should be understood that the "alkyl" represents a linked alkylene group.
[0098] In some specific structures, when an alkyl group is clearly indicated as a linking group, the alkyl group represents a linked alkylene group, for example, the C1-C6 alkyl in the group "halo-C1-C6 alkyl" should be understood as a C1-C6 alkylene.
[0099] The term "halogen" refers to fluorine, chlorine, bromine or iodine, in particular to F or Cl.
[0100] The term "ether" refers to a product in which the hydrogen of the hydroxyl group of an alcohol or phenol is replaced by a hydrocarbon group (including an alkyl group, an alkenyl group, an alkynyl group, a cycloalkyl group, a heterocycloalkyl group) or an aromatic hydrocarbon group (including an aryl group or a heteroaryl group).
[0101] In the present application, as a group or part of other groups (for example, in haloalkyl, deuterated alkyl and the like), the term "alkyl" refers to a saturated aliphatic hydrocarbon group including branched and straight chains with a specified number of carbon atoms, consisting only of carbon atoms and hydrogen atoms, and connected to the rest of the molecule by a single bond. For example, it has 1 to 30 (preferably 1 to 20, more preferably 1 to 10, more preferably 1 to 6) carbon atoms; wherein propyl is C3 alkyl (including isomers, such as n-propyl or isopropyl); butyl is C4 alkyl (including isomers, such as n-butyl, sec-butyl, isobutyl or tert-butyl); pentyl is C5 alkyl (including isomers, such as n-pentyl, 1-methyl-butyl, 1-ethyl-propyl, 2-methyl-1-butyl The hexyl group is a C6 alkyl group (including isomers, such as n-hexyl, 1-ethyl-2-methylpropyl, 1,1,2-trimethylpropyl, 1,1-dimethylbutyl, 1,2-dimethylbutyl, 2,2-dimethylbutyl, 1,3-dimethylbutyl, 2-ethylbutyl, 2-methylpentyl, 3-methylpentyl, 4-methylpentyl, 2,3-dimethylbutyl). For example, methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl, sec-butyl, tert-butyl, n-pentyl, 2-methylbutyl, 2,2-dimethylpropyl, n-hexyl, n-heptyl, 2-methylhexyl, 3-methylhexyl, n-octyl, nonyl, decyl and the like.
[0102] In the present application, as a group or part of other groups, the term "alkylene" means a saturated divalent hydrocarbon group obtained by removing two hydrogen atoms from a saturated straight or branched hydrocarbon group; that is, one hydrogen in an alkyl group is replaced, and the definition of alkyl group is as described above. Examples of alkylene groups include methylene (-CH2-), ethylene {including-CH2CH2- or-CH(CH3)-}, isopropylene {including-CH(CH3)CH2- or-C(CH3)2-}, and the like.

The present invention provides a new method for synthesizing 2-halogen-1,1,2,2-tetrafluoroethyl ether compounds and trifluorovinyl alkyl ether compounds with high efficiency and wide versatility. Various alcohols can be conveniently converted into 2-halogen-1,1,2,2-tetrafluoroethyl ether compounds and further converted into trifluorovinyl alkyl ether compounds by the method of the present invention. The method of the present invention has good compatibility of substrate functional groups and little effect on other functional groups of the substrate, which can further expand the substrate range, improve the yield and reduce the cost.

It is very suitable for industrial production. The obtained product has great application potential in biomedicine, pesticides and materials science. DETAILED DESCRIPTION
[0122] The present invention is further described below by way of examples, but the present invention is not limited to the scope of the examples. The experimental methods in the following examples without specifying specific conditions are carried out according to conventional methods and conditions, or selected according to the product specifications.
[0123] In the present invention, room temperature refers to ambient temperature, which is 10°C-35°C.
[0124] Before conducting the experimental research, we first explored why phenolic substrates can react well with BrCF2CF2Br under the action of base to generate 2-bromo-1,1,2,2-tetrafluoroethyl aryl ether, but it is more difficult for alcohols to react with BrCF2CF2Br.
[0125] In 1982, Wakselman and Rico reported the first case that thiophenol and potassium salt of phenol can react with BrCF2CF2Br to generate the corresponding 2-bromo-1,1,2,2-tetrafluoroethyl aryl (thio) ether. It was specifically mentioned that potassium salt of phenol cannot react spontaneously with BrCF2CF2Br and needs a small amount of thiol to initiate the reaction (J. Fluorine Chem. 1982, 20, 759).
[0126] ArXK+BrCF2CF2Br→ArXCF2CF2Br
[0127] X=O,S
[0128] The authors proposed the following ionic chain reaction mechanism: As to why potassium phenolate requires an initiator to react, the authors believe that this is because the phenol anion is too "hard" compared to the thiophenol anion, and cannot attack BrCF2CF2Br smoothly during the initiation stage of the reaction, causing the reaction to fail to proceed spontaneously.
[0129] Initiation:RS - +BrCF2CF2Br→RSBr+-CF2CF2Br
[0130] Propagation: - CF2CF2Br→CF2=CF2+Br -
[0131] ArX - +CF2=CF2→ArXCF2CF 2-
[0132] ArXCF2CF2 - +BrCF2CF2Br→ArXCF2CF2Br+ - CF2CF2Br
[0133] termination: ArXCF2CF2 - +″H″[solvent]→ArXCF2CF2H
[0134] Subsequently, in 1984, Jiang Xikui et al. corrected the conclusion of Wakselman and Rico and found that potassium phenolate can spontaneously react with BrCF2CF2Br to generate 2-bromo-1,1,2,2-tetrafluoroethyl aryl ether, and the reaction can also be extended to more dibromoperfluoroalkyl groups (Tetrahedron Lett. 1984, 25, 4937). The author reported that this reaction can be carried out in a variety of solvents, as long as the potassium salt and BrCF2CF2Br are carefully purified and dried, a higher yield than before can be obtained without an initiator. They also proposed an anionic chain reaction mechanism, believing that the phenolate anion can initiate the reaction by nucleophilic attack on the bromine atom in BrCF2CF2Br, as shown below:
[0135] Initiation: Aro - +BrCF2CF2Br→ArOBr+ - CF2CF2Br
[0136] Propagation: - CF2CF2Br→CF2=CF2+Br -
[0137] AO - +CF2=CF2→ArOCF2CF2 -
[0138]
[0139]
[0140] In 2007, Li et al. further revealed the experimental details of the reaction of phenol with BrCF2CF2Br (Tetrahedron Lett. 2007, 48, 7516). Based on the previous work, the reaction conditions were further optimized, and cesium carbonate was used as a base to achieve the efficient preparation of 2-bromo-1,1,2,2-tetrafluoroethyl aryl ether by the reaction of phenol with BrCF2CF2Br. Previously, the raw materials needed to be carefully purified, especially potassium phenolate, which needed to be dried, otherwise the reaction yield would drop significantly. Since potassium phenolate easily absorbs water, the authors adopted the method of preparing potassium phenolate in situ, and found that cesium carbonate could significantly shorten the reaction time and increase the yield by screening the base. Finally, the authors also proposed the anion chain reaction mechanism, and also believed that the SRN1 mechanism involving single electron transfer could not be completely ruled out.
[0141]
[0142] In summary, it can be found that the reason why phenol can react well with BrCF2CF2Br to generate 2-bromo-1,1,2,2-tetrafluoroethyl aryl ether under the action of alkali is that the phenol oxygen anion can successfully initiate the reaction. For alcohol, the alcohol oxygen anion is difficult to attack the bromine atom in BrCF2CF2Br with nucleophilicity and cannot successfully initiate the reaction, making the reaction of alcohol with BrCF2CF2Br particularly difficult. In order to solve the difficulty of the reaction between alcohol and BrCF2CF2Br, we explored and tried to add various initiators to the system to promote the reaction.
[0143] Example 1
[0144] First, a catalytic amount of tetrafluoroethylene (12.5 mol%) was introduced to promote the reaction, and Ph(CH2)4OCF2CF2Br was obtained in a yield of 34%. Compared with the reaction without the additive, the yield of Ph(CH2)4OCF2CF2Br was indeed improved.
[0145]
[0146] Under nitrogen protection, weigh (2mmol, 1.0equiv) was placed in a 25-mL sealed tube, anhydrous DMF (5mL) was added, and then NaH (101mg, 95% purity, 4.0mmol, 2.0equiv) was slowly added. After stirring at room temperature for 6 hours, 1,2-dibromotetrafluoroethane (1.04g, 4.0mmol, 2.0equiv) was added. After stirring for 5 to 10 minutes, tetrafluoroethylene (0.25mmol) was introduced at -78°C. After sealing, the temperature was raised to 60°C for reaction for 12 hours, and ethyl acetate was added for dilution. Trifluorotoluene was used as the internal standard. The yield was 34% as determined by fluorine spectrum.
[0147] Example 2-37 Typical operation of synthesizing 2-bromo-1,1,2,2-tetrafluoroethyl ether from alcohol
[0148]
[0149] Under nitrogen protection, alcohol (0.5mmol, 1.0equiv) and zinc powder (3.3mg, 0.05mmol, 0.1equiv) were weighed in a 25mL sealed tube, anhydrous DMF (5mL) was added, and NaH (25.2mg, 95% purity, 1.0mmol, 2.0equiv) was slowly added. After stirring at room temperature for 4 hours, 1,2-dibromotetrafluoroethane (389.7mg, 1.5mmol, 3.0equiv) was added. After stirring for 5 to 10 minutes, the mixture was stirred at -5 to 0°C for 6 hours. After returning to room temperature, the reaction was continued at room temperature for 12 hours, ethyl acetate was added for dilution, saturated ammonium chloride aqueous solution was added for quenching, water was added, and ethyl acetate was extracted three times. The organic phases were combined, washed with saturated sodium chloride solution, dried over anhydrous sodium sulfate, concentrated, mixed with silica gel, and separated by rapid column chromatography with petroleum ether as eluent to finally obtain the target product.
[0150]
[0151] a Reaction conditions: ROH (0.5 mmol, 1.0 eq.) BrCF2CF2Br (1.5 mmol, 3.0 eq.), NaH (1.0 mmol, 2.0 eq.) and Zn (0.05 mmol, 0.1 eq.) in DMF (5 mL). Isolated yield. b The yields in brackets are given by 19 F NMR was measured with PhCF3 as the internal standard.
[0152] It can be seen that whether it is primary alcohol, secondary alcohol, or tertiary alcohol, it can react with BrCF2CF2Br to generate the corresponding bromotetrafluoroethyl alkyl ether with moderate to good yields. For primary alcohols, the reaction yield of alcohols with longer chains is relatively high, among which the yield of benzyl alcohol is relatively low, but the yield of electron-rich benzyl alcohol is slightly higher. For secondary and tertiary alcohols, their overall yields are higher than those of primary alcohols. Whether they are linear or cyclic secondary and tertiary alcohols, they can react well and give good yields.

[0277] Investigation of material ratio and solvent
[0278]
[0279] Under nitrogen protection, weigh (0.2mmol, 1.0equiv) and zinc powder (x mmol) were placed in a 25-mL sealed tube. After adding dry solvent (2mL), NaH (10.8mg, 95% purity, 0.4mmol, 2.0equiv) was slowly added. After stirring at room temperature for 4 hours, 1,2-dibromotetrafluoroethane (ymmol) was added. The temperature was raised to 60°C and the reaction was continued for t hours. Trifluorotoluene was used as the internal standard and the fluorescence spectrum was used for detection. The yield.
[0280] Using phenylbutanol (1) as a template substrate, 2 equivalents of sodium hydride as a base, and zinc powder as an initiator, its reaction with BrCF2CF2Br in DMF solvent was investigated (Table 1). First, 0.2 equivalents of zinc powder were added as an initiator, and the reaction was heated at 60°C for 9 hours to obtain bromotetrafluoroethylphenylbutyl ether 2 with a 25% fluorine spectrum yield.
[0281] Since extending the reaction time to 16 hours did not significantly improve the reaction yield, the equivalent of BrCF2CF2Br was optimized. The amount of BrCF2CF2Br was increased from the initial 2.5 equivalents to 3 equivalents, 3.5 equivalents, and 4 equivalents, and it was found that when the amount of BrCF2CF2Br added was 3 equivalents, it was better and the yield could be increased to 58%.
[0282] Then we tried to reduce the equivalent of zinc powder to 0.1 equivalent, and found that the yield did not drop too much, and a yield of 54% could be obtained when the reaction time was 12 hours. When zinc powder was not added, the reaction could also occur, but the yield dropped to 27%.
[0283] Then the solvent DMF was replaced with DMSO, and it was found that the yield decreased significantly.

[0287] Examples 54-61
[0288] Study on reaction temperature
[0289]
[0290] Under nitrogen protection, weigh (0.2mmol, 1.0equiv) and zinc powder (0.02mmol, 0.1equiv) were placed in a 25-mL sealed tube, and dry DMF (2mL) was added. Then, NaH (10.8mg, 95% purity, 0.4mmol, 2.0equiv) was slowly added. After stirring at room temperature for 4 hours, 1,2-dibromotetrafluoroethane (0.6mmol) was added. The reaction was carried out at different temperatures T for t hours. Trifluorotoluene was used as the internal standard and the fluorescence spectrum was used for detection. The yield.
[0291] When the reaction temperature was raised to 80°C, the reaction yield dropped to 40%. Attempting to lower the temperature yielded better results, with the yield increasing to 67% at 50°C. Further lowering the temperature to 40°C increased the reaction yield to 71%, and reacting at room temperature for 16 hours also yielded 69%, which shows that lowering the reaction temperature is beneficial to improving the reaction yield.
[0292] Then we tried to react at a lower temperature. We first placed the reaction system at -5 to 0°C for 6 hours, and then returned it to room temperature for 12 hours. We could get a yield of 83%. We placed the reaction system at -10°C and -40°C for 6 hours, and the yields decreased to 75%. Finally, we screened the reaction time at -5 to 0°C and found that extending the reaction time did not increase the yield.
[0293] Thus, the optimal conditions for the reaction of alcohol with BrCF2CF2Br to generate 2-bromo-1,1,2,2-tetrafluoroethyl alkyl ether were obtained. In addition, it can be seen from the process of condition optimization that low temperature is more conducive to the reaction, but it cannot inhibit the formation of protonated byproduct tetrafluoroethyl alkyl ether.

[0297] The debromination reaction of the above-prepared bromotetrafluoroethyl alkyl ether (ROCF2CF2Br) was carried out to investigate the universality of the substrate.
[0298]
[0299] In the glove box, zinc powder (29.4 mg, 0.45 mmol, 1.5 equiv) and ZnCl2 (12.3 mg, 0.09 mmol, 0.3 equiv) were weighed into a dried 25-mL sealed tube, and 2-bromo-1,1,2,2-tetrafluoroethyl alkyl ether (0.30 mmol, 1.0 equiv) and anhydrous CH3CN (6 mL) were added under N2 protection after removal from the glove box. The mixture was heated at 95°C for 12 hours. After the reaction was completed, the mixture was concentrated by rotary evaporation, mixed with silica gel, and separated by column chromatography using petroleum ether as the eluent to obtain the target product.
[0300]
[0301] a Reaction conditions: ROCF2CF2Br (0.3mmol, 1.0equiv), Zn (0.45mmol, 1.5equiv), ZnCl2 (0.09mmol, 0.3equiv), MeCN 6.0mL. Isolated yield. b EtOAc 0.5 mL was added as a co-solvent. PhOCF3 was used as an internal standard. 19 The yield was determined by FNMR.
[0302] 2-Bromo-1,1,2,2-tetrafluoroethyl ethers of various primary, secondary, and tertiary alcohols can be converted to trifluorovinyl ethers in moderate to excellent yields, with little hydrogenation byproduct (ROCF2CF2H) being observed. Due to the poor solubility of some substrates in acetonitrile, it is difficult for the raw materials to react completely. Adding ethyl acetate to aid solubilization can promote the reaction.
