Description

TECHNICAL FIELD

[0001] The present application relates to the technical field of biotechnology, in particular to a preparation method of D-psicose and sodium gluconate. BACKGROUND

[0002] D-psicose is a very important rare ketohexose, with a sweetness of 70% of sucrose, but its energy is almost 0, and its solvent properties and taste are similar to sucrose. In the food and medicine industry, D-psicose is an ideal substitute for sucrose. In addition, D-psicose has the functions of inhibiting obesity, reducing blood sugar, reducing blood lipids, etc., and is very suitable for consumption by diabetic patients and obese people, so D-psicose has significant efficacy in medical and health care. At present, the production of D-psicose is mainly based on the isomerization reaction of D-fructose catalyzed by isomerase. This reaction is a reversible reaction, and there is a problem of low conversion rate, with a conversion rate of about 30%.

[0003] Sodium gluconate, also known as sodium pentahydroxyhexanoate, is a white or light yellow crystalline powder, easily soluble in water and slightly soluble in alcohol, insoluble in ether. As an organic acid sodium salt with wide application, it is mainly used in construction, chemical industry, food industry, pharmaceutical industry and other industries. At present, the production method of sodium gluconate is mainly biological fermentation by using strains such as Aspergillus niger or double enzyme oxidation by using glucose oxidase and catalase. The biological fermentation method has the characteristics of stable and mild production, easy scale industrial production, etc., but also has some defects, such as difficult control of product color, high degree of sterilization requirement, etc. The double enzyme oxidation method has the characteristics of fast speed, simple process, high product purity, low energy consumption, etc., but also has some shortcomings: the current enzyme preparation price is high, which increases the production cost. Therefore, in view of the above problems, a preparation method for co-production of D-psicose and sodium gluconate is established. SUMMARY

[0004] The technical problem to be solved by the present application is to provide a preparation method for co-production of D-psicose and sodium gluconate, which simultaneously produces D-psicose and sodium gluconate by using the preparation method, thereby reducing the production cost.

[0005] To solve the above technical problems, the technical scheme of the present application is as follows:

[0006] A preparation method for co-production of D-psicose and sodium gluconate, the preparation method comprising the following steps:

[0007] (1) taking D-fructose and D-glucose as substrates, adding a solvent and a whole-cell catalyst Ⅰ co-expressing three enzymes, adjusting the pH of the reaction system to be weakly acidic, controlling the temperature of the reaction system, and centrifuging the supernatant collected after the reaction to obtain a mixed solution of D-allulose and sodium gluconate;

[0008] (2) taking the supernatant in step (1), adjusting the pH to be neutral, adding a whole-cell catalyst Ⅱ co-expressing two enzymes under ventilation and stirring, and obtaining a mixed solution of D-allulose and sodium gluconate after the reaction;

[0009] (3) taking the mixed solution in step (2), passing through a cation resin column first, collecting the effluent, then passing through an anion resin column, evaporating, concentrating and crystallizing the collected effluent to obtain an allulose product, and using sodium hydroxide solution to flush the anion resin column, evaporating, concentrating and crystallizing the collected effluent to obtain a sodium gluconate product.

[0012] As an improved technical solution, the preparation of the whole-cell catalyst Ⅰ includes the following operations: linking an allulose epimerase gene (DPE), a trehalose dehydrogenase gene (RDH) and a glucose dehydrogenase gene (GDH) to a pYB1s plasmid, transforming the obtained recombinant plasmid pYB1s-DPE-RDH-GDH into Escherichia coli BW25113 to obtain the whole-cell catalyst Ⅰ;

[0013] The preparation of the whole cell catalyst II comprises the following steps: connecting a gene of a mannitol dehydrogenase (RDH) and a gene of an NADH oxidase (NOX) to a pYB1s plasmid, transforming the obtained recombinant plasmid pYB1s-RDH-NOX into Escherichia coli BW25113 to obtain the whole cell catalyst II.

[0018] The reaction equations in steps (1) and (2) in the preparation method of the present application are as follows:

[0019] The reaction of step (1) is as follows:

[0020]

[0021] The reaction of step (2) is as follows:

[0022]

[0023] The application has the following beneficial effects:

[0024] The application uses D-fructose and D-glucose as substrates, adjusts the pH of the reaction system to be weakly acidic, uses whole-cell catalyst I which simultaneously expresses allulose epimerase (DPE) gene, tetritol dehydrogenase (RDH) gene and glucose dehydrogenase (GDH) gene to obtain an NADH regeneration system, and catalyzes D-fructose and D-glucose into D-tetritol and sodium gluconate respectively; then, whole-cell catalyst II which simultaneously expresses tetritol dehydrogenase (RDH) gene and NADH oxidase (NOX) gene is used to oxidize D-tetritol into D-allulose, and sodium gluconate remains in the reaction system. In this synthesis route, two NADH regeneration systems are completed through reduction driven by GDH and oxidation driven by NOX respectively. By using this two-step multi-enzyme cascade biotransformation system, D-fructose and D-glucose are catalyzed to co-produce D-allulose and sodium gluconate, and the production cost is reduced. [0072]

[0073] From the data in Table 1, it can be found that the preparation method of the present application can be suitable for industrial production. D-psicose and sodium gluconate are co-produced by a multi-enzyme system catalyzing D-fructose and D-glucose, which reduces the production cost. In Example 4, the supernatant after step (1) reaction was determined by high performance liquid chromatography (Aminex HPX-87C column (7.8*300 mm), detector (RID), mobile phase: double distilled water, column temperature: 80 °C, flow rate: 0.6 ml / min) to determine the concentrations of dulcitol and sodium gluconate. The specific detection is shown in the following table. Figure 1 .

[0074] The mixture after the reaction of step (2) in Example 4 was subjected to high performance liquid chromatography (Aminex HPX-87H column (7.8*300 mm), detector (DAD, 210 nm), mobile phase: 5 mM sulfuric acid, column temperature 50°C, flow rate: 0.5 ml / min) to determine the concentrations of sodium gluconate and D-allulose. The detailed detection is shown in the following Table 1. Figure 2 .

[0075] The allulose epimerase protein sequence, the mannosonate dehydrogenase protein sequence, the glucose dehydrogenase protein sequence, the mannosonate dehydrogenase protein sequence, and the NADH oxidase protein sequence are shown in the sequence listing.

[0076] The above merely provides the preferred embodiments of the present application, and is not intended to limit the present application. Any modification, equivalent replacement, and improvement made within the spirit and principle of the present application shall fall within the protection scope of the present application.
