Abstract
Description
Technical Field
[0001] The present invention relates to the technical field of traditional Chinese medicine, and particularly relates to a traditional Chinese medicine composition for treating chronic atrophic gastritis and its application. Background Art
[0002] Chronic atrophic gastritis is one of the most common chronic digestive system diseases clinically and is generally considered a precancerous lesion state of gastric cancer. The clinical symptoms of chronic atrophic gastritis include stomachache, loss of appetite, weight loss, etc. The pathological features are chronic inflammation of the gastric mucosa and reduction or loss of cells in the gastric glandular part, often accompanied by intestinal metaplasia, which will ultimately lead to the occurrence of gastric cancer. Reversing precancerous lesions and improving the state of chronic atrophic gastritis should become an important means to avoid the occurrence of gastric cancer. The pathogenesis of chronic atrophic gastritis is currently unclear, but it is generally considered to be caused by repeated damage to the gastric mucosa by physical and chemical factors, Helicobacter pylori infection and other factors.
[0003] Currently, Western medicine has no specific therapy for chronic atrophic gastritis. The treatment methods include eradicating Helicobacter pylori, protecting the gastric mucosa, promoting gastric motility (such as domperidone), supplementing mucosal nutrition (such as coenzyme complex), etc. The curative effect is poor, the recurrence rate is high, and it is difficult to cure. Therefore, there is an urgent need to develop a new traditional Chinese medicine composition for treating chronic atrophic gastritis. Traditional Chinese medicine has a long history and unique advantages in the treatment of gastrointestinal diseases. Summary of the Invention
[0004] In view of this, the purpose of the present invention is to provide a traditional Chinese medicine composition for treating chronic atrophic gastritis, which has a good therapeutic effect on chronic atrophic gastritis.
[0005] The purpose of the present invention is achieved through the following technical solutions:
[0006] A traditional Chinese medicine composition for treating chronic atrophic gastritis is made from the following raw materials in parts by weight:
[0007] Codonopsis pilosula 8 - 15 parts, Atractylodes macrocephala 8 - 15 parts, Poria cocos 8 - 15 parts, Sparganium stoloniferum 8 - 12 parts, Ficus hirta 25 - 35 parts, Scutellaria barbata 10 - 20 parts, Dioscorea opposita 25 - 35 parts, Dendrobium officinale 8 - 15 parts, Pinellia ternata 6 - 10 parts, Bupleurum chinense 8 - 15 parts, Curcuma aromatica 8 - 15 parts, Glycyrrhiza uralensis 4 - 10 parts.
[0008] As a further preferred scheme, the traditional Chinese medicine composition for treating chronic atrophic gastritis is made from the following raw materials in parts by weight:
[0009] Codonopsis pilosula 10 parts, Atractylodes macrocephala 10 parts, Poria cocos 10 parts, Sparganium stoloniferum 10 parts, Ficus hirta 30 parts, Scutellaria barbata 15 parts, Dioscorea opposita 30 parts, Dendrobium officinale 10 parts, Pinellia ternata 9 parts, Bupleurum chinense 10 parts, Curcuma aromatica 10 parts, Glycyrrhiza uralensis 6 parts.
[0010] The Chinese medicine composition of the present invention comes from the clinical experience prescription, with Codonopsis pilosula, Atractylodes macrocephala and Poria cocos as the main ingredients. These three medicines are sweet in taste, all enter the spleen meridian, belong to qi-tonifying medicines, and are used in combination. The main effects are to invigorate qi and strengthen the spleen, warm but not dry, tonic but not harsh, and slowly, which conforms to the characteristics of spleen and stomach liking sweetness and mild medication. The spleen likes dryness and hates dampness, and spleen deficiency is prone to dampness. In addition to strengthening the spleen, Atractylodes macrocephala and Poria cocos can also dry dampness and promote diuresis, and are more suitable for patients with spleen deficiency and dampness. Chronic gastritis is seen in poor appetite, fatigue, hunger and loss of appetite, and other symptoms of spleen deficiency and yin deficiency of stomach qi. The treatment should invigorate the spleen and invigorate qi, but it should not be harsh, and the emphasis should be on tonifying the middle qi. Five-fingered ginseng, Chinese yam, Tripterygium wilfordii, and Curcuma zedoaria are the assistants. Five-fingered ginseng, also known as Nanqi, is a Lingnan local medicinal material. It has the effect of nourishing qi and dispelling dampness, and assists the main medicine to nourish without stagnation; Chinese yam nourishes the spleen and produces body fluid, and assists the main medicine to nourish qi, and takes into account the chronic atrophic gastritis patients with long-term illness that damages yin and insufficient stomach yin; chronic atrophic gastritis has a long course of disease, and the disease becomes blood stasis after a long time, and it is common to have a dark tongue, stabbing pain in the stomach, and the pain is fixed. Tripterygium wilfordii and Curcuma zedoaria can promote blood circulation and relieve pain, and Curcuma zedoaria can promote qi and relieve depression, and take into account the adjustment of the emotions of patients with long-term illness. Scutellaria barbata, Dendrobium officinale, Pinellia ternata, and Bupleurum chinense are the assistants. Scutellaria barbata is pungent, bitter, and cold. It has the effects of clearing away heat and detoxifying, promoting blood circulation and removing blood stasis, and relieving swelling and pain. It is the counter-adjuvant of this prescription, making this prescription warm and nourishing without being warm and dry. It slightly adds the power of attack and dispersion to the tonic medicine, expelling evil without hurting the body. It can still expel evil by detoxifying and reducing swelling. It is suitable for chronic gastritis accompanied by mucosal atrophy, intestinal metaplasia, and dysplasia. Dendrobium is sweet and belongs to the stomach meridian. It nourishes yin and benefits the stomach. It assists yam in nourishing stomach fluid. Spleen deficiency fails to transport and transform, and the fine food accumulates, forming phlegm and dampness in the middle. Pinellia and Bupleurum are used to promote qi, eliminate stagnation, and dry dampness. Bupleurum enters the liver meridian and is a good medicine for promoting qi and soothing the liver. It assists the whole prescription in replenishing the middle and helps Curcuma regulate emotions. Licorice is used to replenish qi and harmonize the middle, and harmonize all the medicines. It is a guiding medicine. The whole prescription is based on the methods of invigorating Qi and strengthening the spleen, promoting blood circulation and dispersing blood stasis, taking into account the methods of regulating Qi, dispersing nodules and nourishing Yin. It is suitable for patients with spleen deficiency and blood stasis syndrome in chronic atrophic gastritis, and has a definite therapeutic effect on chronic atrophic gastritis.
[0011] The present invention also provides a method for preparing the above-mentioned Chinese medicine composition for treating chronic atrophic gastritis, comprising the following steps:

[0049] After 7 days of adaptive feeding with standard diet and water (24 - 25 °C, humidity 70 - 75%, 12-hour light / dark cycle), the rats were randomly divided into 2 groups, including a blank group and a model group. The blank group drank water freely, and the model group was continuously orally administered a 200 μg / ml MNNG solution combined with irregular feeding (fasting for one day and feeding for one day) for 8 weeks. After 8 weeks, the rats in the model group were randomly divided into 4 groups, namely the model group, the positive drug coenzyme Q10 group (0.6 g / kg), the low-dose group of Shenzhu Yangwei Fang (7.191 g / kg), and the high-dose group of Shenzhu Yangwei Fang (14.39 g / kg). The low-dose and high-dose groups of Shenzhu Yangwei Fang were intragastrically administered the corresponding doses of the drug at 1 ml / 100 g per day. The model group and the positive drug group were respectively given an equivalent amount of normal saline and coenzyme Q10, and the drug administration continued for 4 weeks. At the end of the drug administration period, the animals were anesthetized and blood was collected from the abdominal aorta, and then the animals were sacrificed and the rat stomachs were taken. Calculate the organ index: Organ index = organ mass / rat body weight.

[0052] 3. Results and discussion
[0053] 3.1 Effect of Shenzhu Yangwei Fang on the body weight of rats with chronic atrophic gastritis
[0054] As shown in Table 1 and Figure 1 as shown, after the 12-week modeling and drug administration period ended, the weight gain rate of the model group was slower than that of the blank group. There was a significant difference in weight between the model group and the blank group at the 12th week (P < 0.001). The low-dose and high-dose groups of the Shenzhu Yangwei formula could increase the weight of rats with chronic atrophic gastritis (P < 0.01).

[0056]
[0057]
[0058] Note: #p < 0.05, ##p < 0.01, p < 0.001 vs blank control group, the difference was statistically significant. *p < 0.05, **p < 0.01, ***p < 0.001 vs model group, the difference was statistically significant.
[0059] 3.2 Effects of the Shenzhu Yangwei formula on the gastric organ index of rats with chronic atrophic gastritis
[0060] As shown in Table 2 and Figure 2 as shown, the gastric organ index of rats in the model group was significantly increased compared with that of the blank group (P < 0.001), while the low-dose and high-dose groups of the Shenzhu Yangwei formula could significantly reduce the gastric organ index of rats with chronic atrophic gastritis (P < 0.01), alleviating the degree of gastric atrophy.

[0062]
[0063] Note: #p < 0.05, ##p < 0.01, p < 0.001 vs blank control group, the difference was statistically significant. *p < 0.05, **p < 0.01, ***p < 0.001 vs model group, the difference was statistically significant.
[0064] 3.3 Effects of the Shenzhu Yangwei formula on the gastric appearance of rats with chronic atrophic gastritis
[0065] As Figure 3 shown, the gastric mucosa color of rats in the blank group was dark red, the gastric folds were deeper, and the mucosal layer was rich; the color of the gastric tissue of rats in the model group was lighter than that of the blank group, and the gastric folds were shallower and fewer. The gastric tissue of rats in the low-dose and high-dose groups of the Shenzhu Yangwei formula was darker in color and had more gastric folds compared with the model group.
[0066] 3.4 Effects of the Shenzhu Yangwei formula on the serum biochemical indexes of rats with chronic atrophic gastritis
[0067] As shown in Table 3 and Figure 4 as follows, compared with the blank group, the contents of PGI and GAS in the model group decreased, while the content of PGII increased (P<0.01). The low-dose and high-dose groups of the Shenzhu Yangwei formula could effectively increase the contents of PGI and GAS (P<0.001), and decrease the content of PGII (P<0.01).

[0071] Note: #p<0.05, ##p<0.01, p<0.001 vs the blank control group, the difference was statistically significant. *p<0.05, **p<0.01, ***p<0.001 vs the model group, the difference was statistically significant.
[0072] As shown in Table 4 and Figure 5 as follows, compared with the blank group, the content of IL-6 in the model group increased. The low-dose and high-dose groups of the Shenzhu Yangwei formula could significantly decrease the level of IL-6 and reduce the inflammation level in rats with chronic atrophic gastritis.
[0073] Table 4. Effects of the Shenzhu Yangwei formula on serum inflammatory factor levels in rats with chronic atrophic gastritis (n=6)
[0074]
[0075] Note: #p<0.05, ##p<0.01, p<0.001 vs the blank control group, the difference was statistically significant. *p<0.05, **p<0.01, ***p<0.001 vs the model group, the difference was statistically significant.
[0076] 4. Conclusion
[0077] The traditional Chinese medicine composition (Shenzhu Yangwei formula) of the present invention can effectively improve the weight loss and gastric atrophy degree in rats with chronic atrophic gastritis, improve the reduction of gastric folds in rats with atrophic gastritis, increase the contents of PGI and GAS in rats with chronic atrophic gastritis, decrease the contents of PGII and IL-6 levels, indicating that it can improve the gastric hormone level and inflammation level in rats with chronic atrophic gastritis. In summary, the Shenzhu Yangwei formula can effectively reverse MNNG-induced chronic atrophic gastritis.
