[0082] Experimental Example

[0083] The powder for treating Hashimoto's thyroiditis prepared using the method of Example 2 is referred to as HT-FF.

[0084] 1. Animal modeling, grouping, and intervention methods

[0085] 1) Twenty healthy SPF-grade female SD rats were selected and acclimatized for one week. Four rats were randomly selected as the normal control group and fed with normal feed and water. The remaining 16 rats were used to establish an EAT rat model using an immune-mediated high-iodine method.

[0086] 2) After successful preparation of the EAT model, the models were randomly assigned to the following 4 groups: model group, model + low dose HT-FF (HT-FF-L) group, model + high dose HT-FF (HT-FF-H) group, and model + selenium yeast tablet group.

[0087] 3) HT-FF was respectively prepared with 0.5% sodium carboxymethyl cellulose solution in an amount of 0.5-1 mL / 100 g / d according to the corresponding dose (HT-FF-L: 1 g / kg·d; HT-FF-H: 3 g / kg·d), and then the model + HT-FF-L group and the model + HT-FF-H group of rats were given intragastric administration. The administration time and interval: once a day at 09:00-11:00. The model + selenium yeast tablet group was given intragastric administration according to the dose of 20 μg / kg·d. The normal control group and the simple model group of rats were given intragastric administration with the same amount of 0.5% sodium carboxymethyl cellulose solution. All rats were given normal diet and water, and the intragastric administration was continued for 8 weeks. After the last administration, the rats in each group were fasted for 12 h, and then 1.5 mL / kg of 1% sodium pentobarbital solution was injected intraperitoneally. After the rats were anesthetized, the abdominal aorta was opened, 5 mL of blood was taken, and then the blood was naturally coagulated at room temperature. After centrifugation at 3000 r / min for 10 min, the supernatant was taken, 0.5 mL / tube was divided into EP tubes, and then the tubes were labeled and stored at -80°C for detection. After the abdominal aorta was taken, the thyroid gland of the rat was removed, washed with PBS, and then the excess tissue was cut off. The filter paper was used to dry, and then the weight was measured and the scale was photographed. Then the left and right lobes of the thyroid gland were separated, and then the left side was fixed with 4% paraformaldehyde, dehydrated with gradient concentration alcohol, treated with xylene, and then paraffin-embedded, sliced, HE-stained, and mounted. Three non-consecutive sections were randomly selected from each group, and then the pathological changes of the thyroid tissue were observed under a microscope.

[0088] 4) During the experiment, the mental state, activity, and hair color of the rats were observed every day. The food and water intake of the rats in each group was recorded by weighing the water at a fixed time every day. The body weight of the rats was measured once a week.

[0089] 2. Evaluation index

[0090] The thyroid organ index (thyroid weight / rat body weight), the serum thyroid function and thyroid antibody of the rats were detected by ELISA, and the pathological changes of the thyroid tissue of the rats in each group were observed by HE staining, so as to judge the effect of the traditional Chinese medicine composition on Hashimoto's thyroiditis.

[0091] 3. Experimental results

[0092] After 8 weeks of continuous administration, the body weight and the wet weight of the thyroid gland of the rats in each group were shown in Tables 1 and 2. The thyroid organ index of the model group of rats was significantly higher than that of the control group (0.14±0.02 vs. 0.09±0.01, P<0.01); the thyroid organ index of the model + HT-FF-H group was significantly lower than that of the model group (0.10±0.01 vs. 0.14±0.02, P<0.05); and the thyroid organ index of the model + selenium yeast tablet group was also significantly lower than that of the model group (0.11±0.01 vs. 0.14±0.02, P<0.05). The results were shown in Figure 1 .

[0096]

[0097] After 8 weeks of administration, the TSH level of the model group was higher than that of the control group (5.30 ± 0.36 vs. 1.13 ± 0.17, P < 0.01), and the TPOAb and TGAb levels were significantly higher than those of the control group (436.53 ± 19.63 vs. 109.78 ± 5.13, P < 0.01; 139.13 ± 11.48 vs. 39.55 ± 2.50, P < 0.05), and the TT4, TT3, FT4, and FT3 levels were significantly lower than those of the control group, as shown in Table 3. The TSH level of the model + HT-FF-L group was significantly lower than that of the model group (3.98 ± 0.22 vs. 5.30 ± 0.36, P < 0.05), but there was no significant difference in the TPOAb, TGAb, TT4, TT3, FT4, and FT3 levels between the two groups. The TSH level of the model + HT-FF-H group was more significantly decreased than that of the model group (3.38 ± 0.28 vs. 5.30 ± 0.36, P < 0.01), and the TPOAb level was significantly decreased (337.45 ± 13.34 vs. 436.53 ± 19.63, P < 0.05), and the TT4 and TT3 levels were significantly increased (106.83 ± 4.42 vs. 82.53 ± 3.70, P < 0.01; 4.0 ± 0.18 vs. 2.67 ± 0.25, P < 0.05) compared with the model group. The TSH level of the model + selenium yeast tablet group was also significantly decreased compared with the model group, and the TPOAb, TT4, and TT3 levels were also significantly improved, as shown in Table 3.

[0101] Note: * indicates a significant difference compared with the control group, P < 0.05; ** indicates a very significant difference compared with the control group, P < 0.01; # indicates a significant difference compared with the model group, P < 0.05; ## indicates a very significant difference compared with the model group, P < 0.01.

[0102] After 8 weeks of continuous administration, the thyroid tissue of rats in each group was observed under a microscope after HE staining. The results showed that the thyroid follicle structure of rats in the normal group was complete, and there was no inflammatory lymphocyte infiltration. The thyroid follicle of the model group was diffusely damaged, the follicular cell integrity was destroyed, and the cell nucleus was dispersed in the thyroid parenchyma. The model + low-dose traditional Chinese medicine group and the model group had no great difference, and also appeared diffuse follicle damage and inflammatory cell infiltration. Compared with the model group, the model + high-dose traditional Chinese medicine group was improved, the follicle was relatively complete, only a small amount of inflammatory cell infiltration, and the thyroid parenchyma was full. Compared with the model group, the thyroid follicle of the model + selenium yeast tablet group was relatively complete, and there was a small amount of inflammatory infiltration, as shown in Table 2. Figure 2

[0103] The traditional Chinese medicine composition can significantly improve the thyroid organ index of EAT rats, improve the damage of thyroid follicle and inflammatory cell infiltration of EAT rats, and also has a significant improvement and inhibition on the abnormality of EAT rat thyroid function and thyroid related antibodies. The results show that the traditional Chinese medicine composition has a certain therapeutic effect on Hashimoto's thyroiditis.
