Abstract
Description
[0001] Specification Technical Field
[0002] The present invention relates to cosmetics. More particularly, the present invention relates to a single-agent cosmetic comprising a solid portion of the parenchyma of an aloe plant and a coating having a waxy nature that completely encapsulates the solid portion.
[0003] The present invention also relates to a method for manufacturing the cosmetic. Background Art
[0004] Aloe vera belongs to the Asphodelaceae family of plants. It is a fleshy, cold-intolerant plant with a high water content (about 99 - 99.5%). The solid content is 0.5 - 1% and consists of a variety of active components, such as fats and water-soluble minerals, vitamins, simple / complex polysaccharides, organic acids, enzymes, and phenolic compounds.
[0005] The fleshy leaves arranged in a rosette consist of three layers. The outer layer consists of the stem bark or exocarp, which accounts for 20% to 30% of the weight of the whole plant and is green or blue-green in color; the middle layer is called the parenchyma, also known as the fillet, pulp, or gel, which is transparent and has a gelatinous and fibrous matrix and accounts for 50% to 60% of the total weight of the plant; and between the exocarp and the parenchyma, occupying the entire inner surface of the leaf, are the aloin ducts, which are a group of longitudinal ducts through which acybar (also known as latex) circulates.
[0006] The therapeutic potential of aloe is well known. A variety of beneficial effects of aloe have been reported, including immunomodulation, wound and burn healing, hypoglycemic, anti-cancer, gastroprotective, antifungal, and anti-inflammatory properties. These beneficial therapeutic properties of aloe have been used in several commercial applications.
[0007] As a cosmetic ingredient, aloe is widely used in a variety of mainstream skin care products and cosmetics. The plant is very skin-friendly and has a low risk of causing allergies, sensitivities, or skin reactions, and it is also multi-purpose. It is common in after-sun products as well as facial, hair, and body care products.
[0008] Cosmetics or compositions containing a high percentage of aloe are always accompanied by other compounds or active ingredients, such as preservatives or antimicrobials, as well as other chemical products that help to stabilize and preserve the cosmetics or compositions to prevent deterioration of the aloe properties.
[0009] For example, WO2009112558A1 describes a composition for use as a cosmetic base, which comprises 50% to 99% by weight of aloe vera gel and an emulsifier comprising acryloyldodecyl methacrylate copolymer, a nonionic surfactant consisting of esters of polyethoxylated sorbitol, and an oil of natural origin.
[0010] JP2003252719A relates to a cosmetic mainly composed of aloe vera gel juice. The product contains the juice of aloe vera gel (75 - 83% by weight), 1,3 - butanediol, glycerol, and sodium hyaluronate. The active ingredients moisturize the skin, prevent skin spots and freckles, and promote the regeneration of collagen and superoxide - degrading enzymes in the skin.
[0011] However, the use of some compounds in these compositions can have an adverse effect on the health of consumers. There is also a growing interest in 100% natural cosmetics that do not incorporate these compounds or other additives.
[0012] However, it is difficult to maintain the properties of the compounds in cosmetics without using such compounds. In the case of aloe vera, all the benefits of aloe vera are most abundant when the aloe vera product is obtained directly from the plant, but over time the active ingredients of aloe vera deteriorate and it is complex to maintain all the properties.
[0013] One of the methods used to prevent the deterioration of the properties of pure aloe vera and increase the shelf - life is to subject the gel of aloe vera to a freezing treatment, as described in EP3067060A1.
[0014] EP3067060A1 relates to a method that enables the use of a pure aloe vera gel containing all the natural properties of the plant, based on the introduction of a stabilization process achieved by freezing, whereby cold cream, lotion, pomade, or ointment can be locally applied, achieving all the typical uses of aloe vera, but mainly used to relieve various burns. The gel is stored in a separate PET container or a similar plastic container designed to withstand temperatures as low as - 19°C without any damage. The resulting product is a natural aloe vera cold gel without a stabilizer.
[0015] The disadvantages of EP3067060A1 are as follows: The steps of the method used to obtain the product, such as squeezing and filtering aloe vera, change its natural structure and cause the loss of functional molecules of aloe vera such as polysaccharides, vitamins, and minerals. In addition, this processing results in the loss of endophytic microorganisms living in the aloe vera plant tissue.
[0016] In addition, the aloe vera gel has undergone a freezing process in which the gel is stabilized. However, the freezing process inactivates certain microorganisms and is not completely effective unless combined with other processes (heating, irradiation, acidification, etc.).
[0017] In addition, due to temperature differences, exposure to light, and other storage factors that directly affect the shelf life, storing in such plastic containers does not prevent the degradation of aloe vera gel. The last aspect is also affected by the fact that the product must be kept at low temperatures, and this poses difficulties during transportation, storage, and handling because special conditions are required.
[0018] Therefore, there is a need to discover new aloe-based cosmetics that incorporate all the properties of pure aloe and are stable over time without the need to incorporate compounds such as stabilizers or preservatives and / or use complex manufacturing processes. Summary of the Invention
[0019] In view of the problems of the prior art, the main object of the present invention is to provide a single-agent cosmetic that comprises a solid part of the parenchyma of the aloe vera plant, a coating with a waxy nature that completely encapsulates the solid part, and at least one tear strip located between the coating and the solid part.
[0020] The cosmetic of the present invention solves the technical problem of maintaining the properties and stability of a 100% aloe vera product without the use of additives.
[0021] The main technical difference compared to conventional cosmetics lies in the fact that this new cosmetic is like directly applying 100% pure / natural plant to the skin, without other ingredients that weaken its effectiveness, without preservatives, without colorants, without artificial fragrances, and without undesirable substances that reduce the quality of the cosmetic.
[0022] Another unexpected technical difference is that the cosmetic of the present invention can keep various endophytic microorganisms that naturally live in the plant alive, such as species Pantoea agglomerans, Yersinia kristensenii, Erwinia billingiae, Rahnella victoriana, Pantoea allii, Serratia liquefaciens, Yersinia ruckeri, Rahnella woolbedingensis, Leclercia adecarboxylata, Serratia grimesii, Rahnella inusitata, and Pantoea ananatis. These endophytic microorganisms are natural probiotics present in different plant tissues (parenchyma) and have beneficial effects on the skin immune system and the treatment of skin diseases.
[0023] Conventional cosmetics, whether in the form of gels, creams or ointments, cannot contain these endophytic microorganisms because the production processes they undergo always damage the aloe vera plant tissues in which these endophytic microorganisms live.
[0024] On the other hand, as mentioned above, conventional cosmetics are commercially available in the form of gels, creams or ointments. The application of these cosmetics by the user is sometimes insufficient because the product does not allow for the correct dosage or the texture of the product is difficult to handle.
[0025] The cosmetic of the present invention is in the form of a natural block of a plant covered with a coating having a waxy nature, which allows for better handling of aloe vera and also preserves its properties. In addition, the cosmetic of the present invention contains pieces of aloe vera parenchyma, which are an amount suitable for a single dose.
[0026] In addition, the coating having a waxy nature facilitates the manipulation of the pieces of aloe vera parenchyma because it is a smooth product and, therefore, its application can be complex for the user.
[0027] The coating having a waxy nature has several functions. The coating protects the pieces of aloe vera parenchyma from deterioration due to dehydration. In addition, the coating forms a physical barrier against microorganisms, preventing them from attaching to the surface of the pieces of aloe vera parenchyma and reducing the growth of microorganisms.
[0028] In addition, there is at least one tear strip to assist in the use of the cosmetic. Specifically, the tear strip helps to remove a portion of the coating, leaving a portion of the solid part uncoated and another portion still coated. The uncoated solid part can be applied to any area of the user's body.
[0029] In a first aspect, the present invention relates to a single-dose cosmetic comprising a solid part of the parenchyma of an aloe vera plant, a coating and at least one tear strip, wherein the coating completely encapsulates the solid part and is made of a material having a waxy nature selected from hydrocarbon waxes and natural waxes or mixtures thereof, and wherein the tear strip is made of a non-woven material and is located between the coating and the solid part.
[0030] Examples of natural waxes include carnauba wax, candelilla wax, beeswax, Japanese wax, rice bran wax and sunflower seed wax. Among the above waxes, beeswax is more preferred.
[0031] Examples of hydrocarbon waxes include polyethylene wax, paraffin wax, ozokerite and microcrystalline wax.
[0032] Optionally, the coating of the cosmetic of the present invention further contains natamycin to prevent fungal contamination of the product. A dosage of 3 to 20 ppm can be used.
[0033] Optionally, the coating of the cosmetic of the present invention further contains at least one synthetic dye or natural dye. Preferably, the dye is copper chlorophyllin.
[0034] In the present invention, the aloe parenchyma can be extracted from any aloe plant, such as Aloe barbadensis Miller, Aloe Arborescens, Aloe Ferox, Aloevera (L.) Burn, Aloe officinalis, Aloe Vulgaris Lamarck, etc. Preferably, the aloe plant is Aloe barbadensis Miller.
[0035] The tear strip is made of 100% non-woven material. This fabric material is a flat porous sheet directly manufactured from fibrils. It has properties that make it the best choice for achieving its functions, especially its tear strength and its ability to adhere to the coating.
[0036] In another embodiment, the single-agent cosmetic of the present invention further comprises at least one cosmetic active ingredient inserted into the parenchyma, and the at least one cosmetic active ingredient is selected from vitamins, minerals, essential oils, and mixtures thereof.
[0037] Inserting at least one cosmetic active ingredient into the parenchyma can be accomplished by simple diffusion through a molecular concentration gradient or by forced diffusion into the parenchyma.
[0038] The active ingredients in the cosmetic composition for the single-agent cosmetic of the present invention can have various purposes and functions. Generally, such components will provide or at least be expected to provide some benefits to the user, preferably the user's skin or hair, such as anti-aging, cell regeneration, antioxidant, disinfection, moisturization, healing effects, etc.
[0039] The amount of one or more active ingredients for the present invention can vary within a wide range depending on the specific application, and all such amounts can be determined by a person skilled in the art. For example, the amount of one or more active ingredients can vary depending on the ability of the ingredients to penetrate through the skin, onto the skin, or into the pores of the skin. Other factors that can affect the relative amount of the active ingredient include the specific active ingredient involved, the specific benefit required, the sensitivity of the user to the active ingredient, the health condition, age, and skin condition of the user, and so on. In short, one or more active ingredients are preferably used in a "safe and effective amount", which is an amount high enough to bring the desired skin benefits.
[0040] In the second aspect of the present invention, there is a method for manufacturing the single-agent cosmetic of the first aspect of the present invention. The method comprises the following steps:
[0041] (i) Cleaning and disinfecting the surface of the aloe leaf;
[0042] (ii) Remove the cuticle and the outer pericarp of the aloe vera leaf to obtain aloe vera parenchyma;
[0043] (iii) Cut the aloe vera parenchyma into solid parts and macerate the solid parts in a solution of a processing aid;
[0044] (iv) Remove the solution of the processing aid from the parts by filtration;
[0045] (v) Attach at least one tear strip made of a non-woven material to each solid part;
[0046] (vi) Cover the parts with the tear strip of step (v) with a coating made of a material having waxy properties selected from hydrocarbon waxes and natural waxes or mixtures thereof; and
[0047] (vii) Apply a high-pressure process to the product obtained in step (vi) to obtain a cosmetic, wherein the pressure is from 400 MPa to 600 MPa.
[0048] In the present invention, a "processing aid" is an auxiliary technical substance that is added indirectly for manufacturing purposes. According to Good Manufacturing Practice for Pharmaceuticals, these substances that are added to a cosmetic during the manufacturing process to keep the raw materials stable for the duration of the manufacturing process are removed from the cosmetic, after which the cosmetic is packaged in its final form. Any person skilled in the art will know how to select the said processing aids for this purpose.
[0049] In another embodiment, when the single-dose cosmetic of the first aspect of the present invention further comprises at least one cosmetic active ingredient, there is another step of inserting at least one cosmetic active ingredient selected from vitamins, minerals, essential oils and mixtures thereof into the parenchyma before step (iv).
[0050] Inserting at least one cosmetic active ingredient into the parenchyma can be accomplished by simple diffusion through a molecular concentration gradient or by forced diffusion into the parenchyma.
[0051] In the method of the present invention, preferably the time of the high-pressure process of step (vii) is from 3 to 6 minutes. More preferably, the time of the high-pressure process of step (vii) is 3 minutes.
[0052] Optionally, the method includes step (viii), wherein the product obtained in step (vii) is packaged in a stand-up pouch or a sealed bag. Description of the Drawings
[0053] The following drawings are described below. These illustrate exemplary embodiments and do not limit their scope.
[0054] Figure 1 Shows the unopened cosmetic.
[0055] Figure 2 Shows the cosmetics during the opening process.
[0056] Figure 3 Shows the opened cosmetics, where part of the coating has been removed and part of the solid part of the parenchyma is exposed for coating.
[0057] Figure 4 Shows, for each aloe vera form analyzed (the cosmetic of the present invention, aloe vera gel, and aloe vera cream), the percentage loss of each active compound (Acemannan (aloverose), glucomannan, mannose, and vitamins A and C) relative to its initial concentration after ten months of storage.
[0058] Description of Embodiments
[0059] Example 1. Method for Manufacturing the Cosmetic of the Present Invention
[0060] First, a person skilled in the art is aware of the instruments and equipment suitable for carrying out the method according to the present invention and will select a suitable system based on their professional experience, so no more detailed details need to be given here.
[0061] Initially, the rough mud on the surface of the aloe vera leaves is removed by immersion in an immersion tank. Subsequently, the aloe vera skin is disinfected by soaking the aloe vera leaves in a 1:3 ratio of water and sodium hypochlorite for 5 minutes. Then they are rinsed by soaking the aloe vera leaves in water in the tank for 3 minutes.
[0062] Next, the outer skin of the aloe vera leaves is mechanically brushed in an industrial washing machine and then rinsed with a pressurized water jet. The upper and lower parts of the aloe vera leaves are trimmed with a cutting blade and then rinsed with a pressurized water jet.
[0063] Subsequently, the aloe vera leaves are skinned, the cutin layer and the exocarp of the leaves are removed to obtain aloe vera parenchyma blocks, which are cut into several pieces (about 5×2.5×1.5 cm) using a shear blade or a high-pressure water jet.
[0064] The cut parts of the aloe vera parenchyma are subjected to industrial blanching with boiling water (100 °C) for 30 - 60 seconds. Then the parts are softened in a solution of processing aids, which allows the cut parts to be stored for another 18 - 20 hours. This softening helps the aloe vera parenchyma blocks not to lose physical properties such as color and texture and also prevents contamination during the storage period.
[0065] In this embodiment, the processing aid used is a composition comprising sorbate (0.2 - 0.4% w / w), benzoate (0.05 - 0.1% w / w), ascorbic acid (0.1 - 0.2% w / w), L-cysteine (0.3 - 0.6% w / w), water (99.35 - 98.3% w / w) and citric acid (to pH 3.4).
[0066] Subsequently, the solution of the technical aid described in the previous step is completely removed by gravity filtration through a perforated mesh for 10 minutes. Once the solution of the technical aid has been removed, the blocks are separated from each other.
[0067] On each block, a tear strip made of non-woven material is attached at the central part of the block and along the length and width of the block. The tear strip is not fixed in any way but is simply placed by hand on the surface of the block.
[0068] Subsequently, the part with the tear strip is covered with a coating having a waxy nature. In this embodiment, the coating composition is 70% - 80% ester, 10% - 15% free acid and 10% - 20% paraffin wax.
[0069] The coating is heated at a temperature of 75 - 100 °C for 2 - 4 hours until it is completely melted to obtain a liquid state. The liquid coating is applied to the said part and then a cooling phase is carried out using a cold tunnel system (<14 °C).
[0070] Finally, the product obtained in the previous step is processed using a high-pressure process to obtain the cosmetic of the present invention. The pressure of the high-pressure process is 600 MPa for 3 minutes.
[0071] The product obtained is shown in Figure 1 . For use, the user pulls the tear strip, partially removes the coating and exposes the solid part of the parenchyma ([[]] Figure 2 and 3 ). Then the user can apply the solid part of the parenchyma to the desired area.
[0072] Example 2. Comparative Test of the Properties of a Product Containing Aloe Vera and the Cosmetic of the Present Invention
[0073] The beneficial effects of aloe vera are directly related to the active compounds it contains. The methods used to preserve and stabilize the products obtained from aloe vera can have a great influence on the concentration of these compounds and thus on the intensity of the beneficial effects attributed to them.
[0074] For this reason, 4 samples of different forms of aloe vera were analyzed by high performance liquid chromatography (HPLC). Table 1 shows the composition of each sample.
[0075] The concentrations of the main compounds of aloe vera, acetylated mannan (aloe polysaccharide), glucomannan, mannose, and vitamins A and C, were analyzed in each sample. Table 2 shows the results obtained.
[0076] Table 1. Composition of the samples analyzed
[0077]
[0078] Table 2. Concentrations of acetylated mannan (aloe polysaccharide), glucomannan, mannose, and vitamins A and C in the samples analyzed
[0079]
[0080] Table 3 shows the percentage of each compound present in each type of sample relative to the concentration of pure aloe vera (Sample 1).
[0081] Table 3. Percentage of each compound relative to the concentration of pure aloe vera
[0082]
[0083] The cosmetics of the present invention exhibit concentrations of each analyzed compound more similar to those of pure aloe vera (Sample 1). Specifically, the concentrations of the polysaccharides (acetylated mannan and glucomannan) and the monosaccharide mannose are similar to those of the natural product, i.e., these compounds remain intact and have not been affected by the processing. Vitamin C has been degraded to a lesser extent during processing but retains 92.7% of its concentration compared to pure aloe vera (Sample 1). On the other hand, the slightly higher concentration of vitamin A (<100%) presented by Sample 2 may be due to the non-uniformity between the samples, but in any case, it indicates that it does not degrade during the process.
[0084] Aloe vera gel (Sample 3) reflects a reduction of approximately 10% in sugars, 23.5% in vitamin C, and 16% in vitamin A. In the cream form (Sample 4), none of the compounds exceed a concentration of 15% compared to pure aloe vera (Sample 1).
[0085] In addition, compared to commercial products, the stability of the cosmetics of the present invention was evaluated to estimate its shelf life and degradation over time.
[0086] Samples 2, 3, and 4 were stored in a temperature control room (20 °C) and monitored over time. The samples were analyzed at the start of the study and then regularly every 2 months until 10 months. The main compounds of aloe vera mentioned above were used for shelf life monitoring. The analytical method was high performance liquid chromatography (HPLC). The results obtained are presented in Tables 4, 5, and 6 and Figure 4 in.
[0087] Table 4. Percentage of loss of each compound in the cosmetics of the present invention
[0088]
[0089] Table 5. Percentage loss of each compound in aloe vera gel
[0090]
[0091] Table 6. Percentage loss of each compound in aloe vera cream
[0092]
[0093] As shown in the above table, the initial concentration of the compounds analyzed in the cosmetic of the present invention is higher compared to the other two products (gel and cream), and is approximately 10 times higher in the cosmetic of the present invention mainly compared to the aloe vera cream.
[0094] Furthermore, taking into account that the difference between the initial and final concentrations of the compounds analyzed is lower than the analytical uncertainty (the percentage loss is not significant), after ten months of storage, the product of the present invention does not undergo any significant loss of the compounds analyzed (see Figure 4 ).
[0095] In contrast, the gel underwent significant losses of vitamin A (11.7%), vitamin C (10.7%) and aloin (11.35%), and similarly, the cream underwent losses of vitamin C (18.9%), vitamin A (7.88%) and mannose (6.5%).
[0096] As a conclusion of the tests performed, the concentration of any of the compounds analyzed (active ingredients, to which the beneficial health properties of aloe vera are attributed) in the cosmetic of the present invention is higher than in the gel, and significantly higher than in the aloe vera cream (approximately ten times higher).
[0097] Furthermore, the cosmetic of the present invention retains the same beneficial properties as pure aloe vera. In addition, the cosmetic of the present invention exists in a single-dose form and has been treated with high-pressure treatment (a physical treatment that preserves the original characteristics of the product), and thus, unlike other forms, it does not require the use of preservatives.
[0098] Another advantage of the product of the present invention is that it is less allergenic than other forms (gel and cream) because it does not contain any preservatives, which can cause allergic reactions by ingestion or contact.
[0099] Example 3. Metagenomic Analysis of the Cosmetic of the Present Invention
[0100] DNA was extracted from samples of the product of the present invention using two different commercial DNA extraction kits: (1) DNeasy UltraClean Microbial Kit (Qiagen) and (2) DNeasy PowerLyzer PowerSoil Kit (Qiagen).
[0101] The resulting DNA was quantified using Qubit (Thermofisher).
[0102] The 16S rRNA gene in the samples was amplified using the maximum amount of DNA. This gene is conserved in all bacterial species, allowing the identification and establishment of the phylogeny between different bacterial species based on the differences in the genetic sequences of the gene.
[0103] The results obtained showed that the samples of the cosmetic product of the present invention contained bacteria of the following 5 genera: Pantoea, Yersinia, Rahnella, Erwinia, and Serratia. More specifically, the main species present were Pantoea agglomerans, Yersinia kristensenii, Erwinia billingiae, Rahnella victoriana, Pantoea allii, Serratia liquefaciens, Yersinia ruckeri, Rahnella woolbedingensis, Leclercia adecarboxylata, Serratia grimesii, Rahnella inusitata, and Pantoea ananatis.
[0104] These endophytic microorganisms are natural probiotics present within plant tissues (parenchyma). By keeping the parenchyma intact, the product of the present invention keeps these microorganisms alive, thereby producing the consequent health effects on the skin.
[0105] In conventional cosmetic products, whether in the form of gels, creams, or ointments, it is impossible to keep these microorganisms alive due to their production processes, as the tissues of the aloe vera plants in which the microorganisms live must be "destroyed" or "wrecked" in order to produce these products.
