The method comprises using a Schiff base and a cyclic anhydride as monomers and performing alternating copolymerization to prepare the spontaneously degradable cyclic poly(ester-peptide); the Schiff base has a general structural formula as shown in the following formula (I), wherein R1 and R2 are independently selected from branched or linear alkyl groups with 1 to 10 carbon atoms, branched or linear alkenyl groups with 2 to 5 carbon atoms, branched or linear alkynyl groups with 2 to 5 carbon atoms, branched or linear alkynyl groups with 3 to 10 carbon atoms, aromatic groups with 5 to 20 carbon atoms, and the above-mentioned groups containing one or more heteroatoms selected from O, S, N, Si, P, F, Cl, Br, and I; the present invention successfully prepares a cyclic poly(ester-peptide) with a clear chain structure and a fully alternating structure; the cyclic poly(ester-peptide) has spontaneous degradability and can be completely depolymerized and converted into small molecules by simple pyrolysis or exposure to air for a period of time, thereby achieving complete degradation.
