Description

Technical Field

[0001] The present invention belongs to the field of medical technology, and specifically relates to a class of PROTAC molecules derived from AZD5305 and a preparation method and use thereof. Background Art

[0002] Poly (ADP-ribose) polymerase 1 (PARP1) is a nuclear protein that is mainly responsible for poly ADP-ribosylation modification in organisms. It can recruit many DNA repair factors and promote DNA repair. PARP1 mainly mediates single-strand repair of DNA in the body. If the loss of single-strand DNA is not repaired in time, double-strand DNA damage will occur. Normal cells can repair double-strand DNA damage through HR recombination in the body to ensure the stability of the genome, but some tumor cells lack HR protein. Inhibiting PARP1 protein will induce cells to produce more toxic double-strand DNA damage, thereby killing tumor cells. This mechanism is called synthetic lethality.

[0003] PARP1 inhibitors (PARPi) have made great progress since 2010, but inhibitors still have some defects, such as the clinical toxicity of PARPi, which is caused by the trapping mechanism of PARPi (Yves Pommier et al. Laying a trap to kill cancer cells: PARP inhibitors and their mechanisms of action. Sci. Transl. Med. 8, 362ps17-362ps17 (2016).), which limits the clinical application of PARPi in other diseases.

[0004] A variety of inhibitors have been developed based on PARP1, but due to the potential toxicity of the capture characteristics of PARP1 inhibitors, their clinical use is limited, and drug resistance often occurs in the use of clinical inhibitors. PROTAC molecules have the characteristics of catalytic degradation and can degrade proteins at low doses, which can overcome the drug resistance of PARP1 inhibitors used clinically. In addition, studies have shown that PARP1 degraders can inhibit the skeleton function and catalytic function of PARP1, providing potential applications for the study of the mechanism of PARP1 in diseases.

[0005] AZD5305 is a new generation of PARP inhibitor from AstraZeneca, and its structural formula is as follows:

[0006]

[0007] AZD5305 is a second-generation PARP1 inhibitor. Due to its high selectivity for PARP1, it can avoid the side effects caused by the inhibition of PARP2. Previous PARP1 degraders were based on first-generation inhibitors as targeted warheads. Their poor selectivity for PARP1 / PARP2 may bring potential risks in clinical use. PROTAC designed based on second-generation PARP1 inhibitors may provide a solution. Summary of the invention

[0147] Example 18 PARP1 protein degradation experiment in MDA-MB-231 cells

[0148] MDA-MB-231 cells (ATCC) were cultured in DMEM complete medium at 37°C and 5% CO. 2 DMEM medium was purchased from Gibco and contained 10% FBS (ExCell Bio) and 1% Penicillin-Streptomycin (Gibco). Cells in the logarithmic growth phase were collected and the cell concentration was adjusted to 1.5×105 / ml, add 1ml to each well of a 12-well cell culture plate. After 24h, observe the cell growth status and whether it is evenly distributed under a microscope. The test compound is prepared into a 10mM stock solution using DMSO, and then diluted with DMEM basal culture medium to solutions of different concentrations (0.3nM-1000nM). Discard the original culture medium in the well plate, add different concentrations of drugs respectively, and the volume of solution in each well is 1ml. After 24h of administration, the plate is collected, the original culture medium is discarded, and after washing once with pre-cooled PBS, 1ml PBS is added, and the cells are gently blown off with a gun. This step is completed on ice. The obtained cell suspension was transferred to a 1.5 ml centrifuge tube and centrifuged at 3500 RPM at 4°C for five minutes to obtain a cell pellet. The upper PBS layer was discarded and 60 ul of RIPA lysis buffer containing protease inhibitors (100×) and phosphatase inhibitors (50×) was added (protease inhibitors and phosphatase inhibitors were purchased from Beyotime). After sonication, the tube was lysed on ice for 20 minutes, and then centrifuged at 4°C, 13000 g for 10 minutes. The supernatant was collected to obtain the protein.

[0149] The protein concentration was determined by BCA method, and the protein concentration was normalized and then added to loading buffer (Bio-Rad), vortexed and boiled at 100°C for 8 min to obtain protein samples for subsequent Western blot detection. Western blot experiments used 8% separation gel, PARP1 antibody was purchased from CST (PARP (46D11) Rabbit mAb #9532), and internal reference Anti-alpha Tubulin was purchased from Hangzhou Huaan Biotechnology Co., Ltd. (JM73-24).

[0150] The experimental results are shown in Table 1:
