[0002] More specifically, the present invention relates to novel pharmaceutical or nutritional compositions for oral administration comprising multiple vitamins, minerals, coenzymes, amino acids and omega-3 fatty acids; methods for their preparation and their preventive or supportive use as adjuvants to anticonvulsant drugs for the treatment / prevention of various types of epilepsy or seizures.
[0003] More specifically, the present invention relates to novel pharmaceutical compositions or nutritional compositions for oral administration comprising a variety of vitamins, minerals, coenzymes, amino acids and omega-3 fatty acids suitable for all age groups from pediatrics, adolescents, adults to the elderly, a method for preparing the same, and prophylactic or supportive use thereof as an adjuvant to anticonvulsant drugs for the treatment / prevention of various types of epilepsy or epileptic seizures. Background Art
[0004] Epilepsy is a central nervous system (neurological) disorder in which brain activity becomes abnormal, triggering a seizure or a period of unusual behavior, sensations, and sometimes loss of consciousness. Epilepsy affects men and women of all races, ethnic backgrounds, and ages.
[0005] Epilepsy affects approximately 50 million people worldwide. It is characterised by recurring seizures, brief episodes of involuntary movements that may involve one part of the body (localized) or the entire body (generalized), sometimes accompanied by loss of consciousness and loss of control of bowel or bladder function.
[0006] An epileptic seizure is the result of an excessive discharge of electricity from a group of brain cells. Different parts of the brain can be the site of these discharges. An epileptic seizure can range from the briefest inattention or muscle twitch to a severe and prolonged convulsion. The frequency of epileptic seizures also varies, from less than once a year to several times a day.
[0007] The types of epilepsy are as follows:
[0008] 1) Generalized epileptic seizures: “grand mal seizures” or generalized tonic-clonic seizures, absence seizures, myoclonic seizures, clonic seizures, tonic seizures, and atonic seizures;
[0009] and
[0010] 2) Partial (focal) seizures: simple partial (no loss of consciousness), complex partial (impaired consciousness), and partial with secondary generalization
[0011] Anticonvulsants (antiepileptic drugs or antiseizure drugs) are a diverse group of pharmacological agents used to treat epileptic seizures. Different types of drugs are used to treat epilepsy, depending on the category or type of seizure / epilepsy.
[0012] Although the anticonvulsant medications listed above have been found to be extremely helpful, these medications may not be appropriate for everyone and, as with any medication, may carry the risk of side effects.
[0013] There is ongoing research around the world to develop alternative drug or nutritional supplement systems that, when used in combination with existing anticonvulsant drugs, can enhance the anti-epileptic effect with minimal side effects.
[0014] Studies have shown that various vitamins, minerals, and amino acids play important physiological roles in epilepsy, and have established a correlation between the deficiency or excess of these vitamins, minerals, and amino acids and the etiology of certain human disease states (such as epilepsy).
[0015] Therefore, there remains a need to develop improved pharmaceutical or nutritional compositions that overcome the disadvantages associated with the products available on the market.
[0016] The inventors of the present invention have unexpectedly discovered that the use of a synergistic combination of multiple vitamins, minerals, coenzymes, amino acids and omega-3 fatty acids, which provides the right amounts of the right vitamins, minerals, coenzymes, amino acids and omega-3 fatty acids at the right time to ensure adequate intake of the multiple vitamins, minerals, coenzymes, amino acids and omega-3 fatty acids, can be used for preventive or supportive use as an adjuvant to anticonvulsant drugs for the treatment / prevention of various types of epilepsy or epileptic seizures.
[0017] The inventors of the present invention have also discovered that a novel pharmaceutical composition or nutritional composition for oral administration comprising high-quality and high-purity multiple vitamins, minerals, coenzymes, amino acids and omega-3 fatty acids exhibits properties such as, but not limited to, stability, ease of administration, ease of dosage adjustment, minimal side effects, high therapeutic efficiency and better patient compliance, wherein the composition indicates its preventive or supportive use as an adjuvant to anticonvulsant drugs for the treatment / prevention of various types of epilepsy or epileptic seizures.

This vitamin actually includes a group of six chemically related compounds (i.e., isovitaminosis), all of which contain a pyridine ring as their core. These compounds are pyridoxine, pyridoxal, pyridoxamine and their respective phosphorylated derivatives pyridoxine 5'-phosphate, pyridoxal 5'-phosphate and pyridoxamine 5'-phosphate. Pyridoxal 5'-phosphate has the highest biological activity, but other compounds can be converted into this form. Vitamin B6 acts as a cofactor in more than 140 cellular reactions, mainly related to amino acid biosynthesis and catabolism, but also participates in fatty acid biosynthesis and other physiological functions.
[0205] Preferably, vitamin B6 (pyridoxine hydrochloride) is present in an amount of about 0.001 mg to about 5.0 mg, more preferably about 0.001 mg to about 3.5 mg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0206] In one embodiment, vitamin B9 refers to various forms of folate and its related compounds, including tetrahydrofolate (the active form), methyltetrahydrofolate (the major form found in the blood), methenyltetrahydrofolate, folinic acid, folic acid, and pteroylglutamate.
[0207] Preferably, vitamin B9 (folic acid) is present in an amount of about 1.0 mcg to about 400 mcg, more preferably about 25 mcg to about 350 mcg, relative to the total weight of the pharmaceutical or nutritional composition.
[0208] In one embodiment, vitamin B12 or cyanocobalamin is also used to treat pernicious anemia, vitamin B12 deficiency (due to low food intake or inability to absorb due to genetic or other factors), thyrotoxicosis, bleeding, malignancy, liver disease, and kidney disease.
[0209] Vitamin B12 deficiency has a significant effect on the nervous system, causing irreversible lesions, progressive increase in myelinated neurons, demyelination and death of neurons in the spinal cord and cerebral cortex, symptoms such as paresthesias in the hands and feet, decreased vibration and position sense, resulting in instability, confusion, moodiness, and loss of memory and central vision. It may also manifest as delirium, hallucinations, and even frank psychosis.
[0210] Preferably, vitamin B12 (cyanocobalamin) is present in an amount of about 1.0 mcg to 5.0 mcg, more preferably about 1.0 mcg to about 2.2 mcg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0211] In another embodiment, the present invention comprises one or more minerals selected from the group consisting of calcium, phosphorus, potassium, sodium, magnesium, sulfur, iron, chloride, cobalt, copper, zinc, manganese, molybdenum, iodine, selenium, sodium selenite, chromium, etc. or any combination thereof.
[0212] In one embodiment, magnesium is the fourth mineral with the highest content in the body of the human organism, and it is involved in about 325 enzymatic reactions, including the production of cellular energy through ATP-Mg complex formation, cell membrane stabilization and nucleic acid, protein and cytoplasmic organelle synthesis. Therefore, this mineral plays an important role in the electrical stability and integrity of cell membranes, muscle contraction, nerve conduction, vascular tone regulation, etc. It is also associated with anxiety-related disorders. Deficiency (hypomagnesemia) can lead to nervous system irritability, as well as tetany, fatigue, vasodilation, muscle spasms or cramps, convulsions, tremors, depression and psychotic behavior.
[0213] Preferably, magnesium is present in an amount of about 1.0 mg to about 500 mg relative to the total weight of the pharmaceutical or nutritional composition.
[0214] In one embodiment, manganese (Mn), an essential micronutrient for humans, is a trace element necessary for many physiological processes due to its role in the activity of various metalloproteinases. Manganese has 11 known oxidation states, of which Mn2+, Mn3+ and Mn4+ are present in humans and animals.
[0215] It is mainly found in bones, liver, kidneys and pancreas. Manganese helps the body form connective tissue, bones, clotting factors and sex hormones. It also plays a role in fat and carbohydrate metabolism, calcium absorption and blood sugar regulation. Manganese is also necessary for normal brain and nerve function. Low levels of manganese in the body can cause infertility, bone deformities, weakness and seizures.
[0216] Preferably, manganese is present in an amount of about 1.0 mg to 10 mg, more preferably about 1.0 mg to 5.0 mg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0217] In one embodiment, zinc is considered an essential nutrient, meaning your body cannot produce or store it. Zinc is an essential mineral that your body cannot make on its own. It helps with growth, DNA synthesis, immune function, and more. Today, approximately 70 enzymes, many of which are essential to human health, are known to contain zinc, and strong evidence suggests that many, if not all, of these enzymes require zinc to express their activity.
[0218] Preferably, zinc is present in an amount of about 1.0 mg to about 30 mg, more preferably about 1.0 mg to about 20 mg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0219] In one embodiment, selenium is a trace mineral, meaning that the human body only needs a small amount of selenium. It occurs naturally in foods or as a supplement. Selenium is an important component of various enzymes and proteins (called selenoproteins) that help make DNA and protect against cell damage and infection; these proteins are also involved in reproduction and in the metabolism of thyroid hormones. Most of the selenium in the body is stored in muscle tissue, although the thyroid gland holds the highest concentration of selenium because various selenoproteins help thyroid function. Selenium is a component of selenoproteins and enzymes. They have antioxidant properties and help break down peroxides, which can damage tissue and DNA, leading to inflammation and other health problems.
[0220] Preferably, sodium selenite is present in an amount of about 1.0 mcg to 80 mcg, more preferably about 1.0 mcg to 50 mcg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0221] In one embodiment, the present invention comprises one or more amino acids selected from the group consisting of taurine (2-aminoethanesulfonic acid), leucine, arginine, glutamic acid, glycine, and the like.
[0222] In another embodiment, taurine (2-aminoethanesulfonic acid) is a conditionally essential amino acid. It is found free or in simple peptides, but is not used in protein synthesis. In vivo studies have shown that taurine is essential for many aspects of mammalian development. In addition, low levels of taurine are associated with different pathological lesions, including cardiomyopathy, retinal degeneration, and growth retardation. It is well known that taurine plays an important role in many physiological functions, including detoxification, membrane stabilization, osmoregulation, and regulation of cellular calcium levels. Taurine is also used to treat cardiovascular disease, epilepsy and other seizure disorders, macular degeneration, Alzheimer's disease, liver disorders, and cystic fibrosis.
[0223] Preferably, taurine is present in an amount of about 1.0 mg to about 800 mg, more preferably about 1.0 mg to 500 mg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0224] In one embodiment, the present invention comprises one or more omega-3 fatty acids selected from the group consisting of docosahexaenoic acid (DHA), alpha-linolenic acid (ALA), and eicosapentaenoic acid (EPA), or the like, or any combination thereof.
[0225] In another embodiment, ω-3 fatty acids (also referred to as ω-3 oils, ω-3 fatty acids, or n-3 fatty acids) are polyunsaturated fatty acids (PUFAs) characterized by the presence of a double bond, three atoms away from the terminal methyl group in its chemical structure. The three types of ω-3 fatty acids involved in human physiology are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA). ALA is found in plants, while DHA and EPA are found in algae and fish. Marine algae and phytoplankton are the main sources of ω-3 fatty acids. DHA and EPA accumulate in fish that feed on these algae.
[0226] Preferably, the omega-3 fatty acids are present in an amount of about 1.0 mg to about 1600 mg, more preferably about 1.0 mg to 1000 mg, relative to the total weight of the pharmaceutical composition or nutritional composition.
[0227] In one embodiment, the present invention comprises one or more coenzymes, such as but not limited to coenzyme Q10 (ubiquinone) and the like.
[0228] In another embodiment, coenzyme Q, also known as ubiquinone, is a family of coenzymes that are ubiquitous in animals and most bacteria (hence the name ubiquinone). In humans, the most common form is Coenzyme Q10 or ubiquinone-10. It is a 1,4-benzoquinone, where Q refers to the quinone chemical group, and 10 refers to the number of isopyridine chemical subunits at its tail. In natural ubiquinones, this number can be anywhere between 6 and 10. This family is fat-soluble. CoQ has three redox states: fully oxidized (ubiquinone), semiquinone (ubisemiquinone), and fully reduced (ubiquinol). The ability of this molecule to act as a two-electron carrier (moving between the quinone and quinol forms) and a one-electron carrier (moving between semiquinone and one of these other forms) is key to its role in the electron transport chain, as the iron-sulfur cluster can only accept one electron at a time and acts as a free radical scavenging antioxidant.
[0229] Preferably, coenzyme Q10 is present in an amount of about 1.0 mg to about 200 mg, more preferably about 1.0 mg to 150 mg, relative to the total weight of the pharmaceutical composition.
