[0136] The following examples further illustrate various aspects of the present disclosure. However, they are in no way limiting to the teachings or disclosure as described herein.
[0137] Example
[0138] Example 1—Preparation of Compound 6: 4-(4-chloro-3-hydroxy-5,5,8,8-tetramethyl-5,6,7,8-tetrahydronaphthalene-2-carboxamido)-2,6-difluorobenzoic acid
[0139] The preparation method of compound 6 is as follows Figure 1 As shown, starting from commercially available compound 1 (2,5-dichloro-2,5-dimethylhexane), it can also be generally prepared as described in steps 2-5 below, starting from commercially available compound 2 (5,5,8,8-tetramethyl-5,6,7,8-tetrahydronaphthalene-2-ol).
[0140] Step 2—Synthesis of Compound 3: 3-Hydroxy-5,5,8,8-tetramethyl-5,6,7,8-tetrahydronaphthalene-2-carboxylic acid. In a pressure reactor, 100 g of 5,5,8,8-tetramethyl-5,6,7,8-tetrahydronaphthalene-2-acid sodium (Na salt of Compound 2) and 40 g of 5,6,7,8-tetrahydro-5,5,8,8-tetramethyl-β-naphthol were injected in a high boiling point solvent at a temperature of 170°C and a carbon dioxide pressure of 10 kg / cm 2 (G) and stirred for 2 hours. The reaction mixture was cooled and injected with 500 ml of water, and then separated into a reaction medium layer and an aqueous layer at 90°C. The aqueous layer was extracted with 50 ml of xylene. The aqueous layer was made acidic with dilute sulfuric acid, and compound 3 was isolated.
[0141] Step 3 - Synthesis of Compound 4: 4-Chloro-3-hydroxy-5,5,8,8-tetramethyl-5,6,7,8-tetrahydronaphthalene-2-carboxylic acid. Sulfuryl chloride (12.7 kg, 1 equivalent) was added in portions to a solution of Compound 3 (23.0 kg, 1 equivalent) in tetrahydrofuran, and the mixture was heated and stirred until the reaction was complete. The mixture was cooled, and ethyl acetate was added. The mixture was washed with brine, and the organics were then concentrated and evaporated with heptane. Heptane was added and the slurry was cooled, and the product was collected by filtration and rinsed with heptane. The product Compound 4 was dried (17.7-23.8 kg, yield 67-91%).
[0142] Step 4 - Synthesis of Compound 5. Phosphorus trichloride (4.2 kg, 1.44 equivalents) was added to a suspension of Compound 4 (18 kg, 1.0 equivalents) in toluene, and the mixture was heated and stirred until a solution was formed. A solution of 3,5-difluoroaniline (9 kg, 1.09 equivalents) in toluene was then added, and heating was continued until the reaction was complete. The mixture was cooled slightly, then partially concentrated and co-evaporated with isopropanol. Isopropanol was added, and the slurry was then heated to dissolve the product and partially concentrated again. The resulting slurry was cooled and the product was collected by filtration. The filtrate was partially concentrated and then cooled to precipitate a second fraction, which was collected by filtration. It was then dried in vacuo at a temperature less than 60°C to obtain Compound 5 (18.1-20.1 kg, 72-80% yield).
[0143] Step 5 - Synthesis of Compound 6. A mixture of Compound 5 (15 kg, 1 eq.) and 2,2,6,6-tetramethylpiperidine (10.7 kg, 2 eq.) in tetrahydrofuran was cooled. While maintaining the temperature, n-hexyllithium (43.1 kg, 2.5 M, in hexane, 4 eq.) was slowly added to the mixture, and the mixture was stirred for a certain time. The mixture was quenched on dry ice (about 186 kg), and then heated and stirred until all solid dry ice disappeared. Water was added, and the layers were separated, and the organic matter was extracted a second time with water. The aqueous layers were combined, acidified with hydrochloric acid (15.01 kg, 4 eq.), and then extracted twice with ethyl acetate. The combined organic matter was concentrated and co-evaporated with toluene. Toluene was added, and the mixture was cooled. The resulting slurry was filtered and rinsed with cold toluene. The product compound 6 was dried (40-50°C) and then refrigerated in the dark (11.7-15.9 kg, yield 70-95%). 1 HNMR(CD3OD): d1.34(s,6H), 1.54(s,6H), 1.68(m,2H), 1.75(m,2H), 7.53(d,J HF =10.2Hz,2H).
