Description

Technical Field

[0001] The present invention relates to co-crystals of methylxanthines with stilbenes as coformers, their polymorphs and methods for their preparation. The debittering of methylxanthines is carried out by co-crystals using stilbenes as coformers. The present invention also relates to the preparation of polymorphs of the debittered co-crystals and their use in nutraceuticals and pharmaceutical compositions.

[0002] Background and Prior Art

[0003] Methylxanthines such as caffeine, theophylline, theobromine, methylcaproline, theophylline, paraxanthine are purine alkaloids that are said to have similar neuroenergetic effects. These are found in the seeds and leaves of various coffee (Coffea) species. Initially, caffeine accumulates in the young leaves of coffee trees, but as it grows into adult leaves, it is gradually replaced by theophylline, theobromine and methylcaproline.

[0004] Pterostilbene, chemically known as trans-3,5-dimethoxy-4'-hydroxystilbene, is a naturally occurring stilbene found in nature, such as many tree barks and various berries (including grapes), as well as plants commonly used in traditional folk medicine and structurally related to resveratrol. It is considered a nutraceutical with potential antioxidant, anticancer, anti-inflammatory, pro-apoptotic, anti-tumor, cytoprotective properties, and has also been reported to reduce plasma glucose levels and also reduce the LDL / HDL cholesterol ratio. Pterostilbene is reported to exert its antioxidant activity by scavenging reactive oxygen species (ROS) after administration, thereby preventing oxidative stress and ROS-induced cell damage.

[0005] US2009 / 0069444 describes the use of pterostilbene for improving oxidative stress and improving working memory and compositions comprising pterostilbene. As pterostilbene is believed to have health benefits; the consumption of foods containing this compound, such as berries and grapes, has increased.

[0006] WO 2011097372A3 discloses co-crystals of pterostilbene, including pterostilbene:caffeine co-crystals, pterostilbene:carbamazepine co-crystals, pterostilbene:glutaric acid co-crystals and pterostilbene:piperazine co-crystals.

[0007] US9468645B2 discloses a stable aqueous composition of at least one biologically active form of a purine alkaloid compound structurally related to caffeine for oral administration, the composition comprising a) 1,3,7,9-tetramethyluric acid (Theacrine), (O(2),2-methoxy-1,9-dimethyl-7H-purine-6,8-dione (Liberine) and (O(2),1,7,9-trimethyluric acid (Methylliberine), collectively referred to as theophyllines, wherein the alkaline uric acid structure has been partially or fully methylated at the 2, 3 or 4 positions; and b) at least one aqueous buffer solution having a pH range of about 1.5-9.0.

[0008] EP3068240A1 discloses a dietary supplement comprising theophylline and optionally other active ingredients that modulate the effects of theophylline.

[0009] US20210100266 discloses an energy supplement composition comprising caffeine and methylcafrine.

[0010] Although methylxanthines are known to have very good health benefits, their use is limited due to their bitter taste and there is a need to mask the bitterness or to make these nutraceuticals edible by debittering them to improve palatability. Unacceptable palatability is a major problem in the development of nutraceuticals as it often hinders patient compliance. Therefore, masking the bitterness of methylxanthines is a major goal in the development of nutraceuticals and is widely considered a key aspect of modern drug development.

[0011] Co-crystallization of two or more pure nutraceutical compounds by crystal engineering to create a novel functional material with improved taste and bioavailability has great industrial significance for both the pharmaceutical and food industries. Therefore, there is still a need in the art to provide a composition comprising methylxanthines that has a good taste, is not bitter, and has improved bioavailability.

[0012] Purpose of the Invention

[0013] Therefore, an object of the present invention is to provide co-crystals of methylxanthines and stilbenes which have a good taste and are free from bitterness.

[0014] Another object of the present invention is to provide co-crystals of methylxanthines using stilbenes, more particularly pterostilbene and resveratrol as crystal formers for taste masking of methylxanthines.

[0015] A further object of the present invention is to provide a method for preparing co-crystals of methylxanthines using stilbenes, more particularly pterostilbene and resveratrol as crystal formers and compositions comprising the same.

[0016] A further object of the present invention is to provide a method for preparing polymorphic forms of co-crystals of methylxanthines with pterostilbene and resveratrol. Summary of the invention

[0071] The foregoing descriptions of specific embodiments of the present invention have been presented for purposes of illustration and description. They are not intended to be exhaustive or to limit the invention to the precise forms disclosed, and obviously many modifications and variations are possible in light of the above teachings.

[0072] Pterostilbene, resveratrol, theophylline, theobromine, methylcroftan, paraxanthine, caffeine and theophylline used in the present invention are commercially available synthetic compounds.

[0073] The terms "coformer," "cocrystal," "nutraceutical," and "drug" are used in this application with the same meanings as used in the scientific literature.

[0074] Taste assessment is an increasingly important aspect in the development of any oral dosage form. Bitterness is the most problematic due to its intuitive association with toxicity. Various taste masking techniques exist to suppress bitterness, such as polymer coating, co-crystal formation or complexation with cyclodextrins.

[0075] The present inventors surprisingly found that when methylxanthines such as theophylline, theobromine, methylcaffeine, caffeine and paraxanthine are prepared into co-crystals using pterostilbene, or when theophylline is prepared into co-crystals using resveratrol, the bitterness of theophylline, theobromine, methylcaffeine, paraxanthine, caffeine and theophylline is completely reduced or eliminated.

[0076] In an embodiment, the present invention relates to co-crystals of methylxanthines with stilbenes as coformers, wherein the co-crystals exhibit a pleasant taste and lack the bitter taste of methylxanthines.

[0077] The methylxanthines are selected from the group consisting of theophylline, theobromine, methylcrocaffeine, paraxanthine, caffeine and theophylline.

[0079] The co-crystal exhibits a pleasant taste and does not contain the bitter taste of methylxanthines.

[0080] The stilbenes are selected from hydroxy derivatives of styrene, preferably pterostilbene and resveratrol.

[0142] These polymorphs are reliably characterized by the peak positions in the X-ray diffraction pattern, which produces a fingerprint of the crystalline form and enables it to be distinguished from all other crystalline and amorphous forms of theophylline or theophylline or theobromine or caffeine or paraxanthine and pterostilbene. The 2θ values are measured with an accuracy within ±0.2 degrees. All powder diffraction patterns were measured on a PANalytical X'Pert3 X-ray powder diffractometer equipped with a copper-K-alpha radiation source.

[0143] In an embodiment, the present invention discloses a composition comprising an effective amount of a co-crystal of a coformer of a methylxanthine selected from theophylline, theobromine, methylcaproline, caffeine, paraxanthine and theophylline and a stilbene selected from theophylline or a polymorph thereof and one or more excipients.

[0144] The excipient is selected from one or more of the following: binders, fillers, lubricants, emulsifiers, suspending agents, sweeteners, preservatives, buffers, wetting agents, disintegrants, diluents, flavoring agents, colorants, suitable polymer effervescent agents, additives and mixtures thereof.

[0145] The additive is selected from the group consisting of microcrystalline cellulose, lactose, sucrose, fructose, glucose, dextrose, calcium hydrogen phosphate, calcium sulfate, cellulose, methyl cellulose, cellulose derivatives, kaolin, mannitol, lactitol, maltitol, xylitol, sorbitol, sugar alcohols, dry starch, dextrin, maltodextrin, polysaccharides and mixtures thereof.

[0146] In yet another embodiment, the present invention relates to a nutraceutical or pharmaceutical dosage form comprising a co-crystal of a coformer of a methylxanthine selected from theophylline, theobromine, methylcrocarpine, caffeine, paraxanthine and theophylline and a stilbene selected from theophylline or a polymorph thereof.

[0147] In another embodiment, the present invention relates to a pharmaceutical composition for dermatological use, comprising an effective amount of a co-crystal of a coformer of a methylxanthine selected from theophylline, theobromine, methylcaproline, caffeine, paraxanthine and theophylline and a stilbene selected from theophylline or a polymorph thereof and one or more excipients.

[0148] The nutraceutical or pharmaceutical dosage form, wherein the dosage form is an oral dosage form selected from the group consisting of tablets, capsules, powders, suspensions and lozenges.

[0149] The coformer selected from pterostilbene or resveratrol used in the present invention masks the bitter taste of methylxanthines by blocking the receptors responsible for taste perception.

[0150] Taste is the sensation achieved when a nutraceutical or a drug is placed in the mouth. Preference tests provide information on whether the tester likes or dislikes the product. Taste evaluation tests are designed by using statistical tools to minimize or reduce the bias in responses within and between human volunteers. For sensory analysis, one of the important parts is testing the taste buds of healthy volunteers.

[0151] Smell, taste and texture are the three main components that affect the palatability of a product. Palatability refers to the voluntary acceptance and consumption of a pharmaceutical composition, which can be measured by acceptance, preference and consumption tests, and when a substance has a voluntary acceptance rate of 90% or more among the test subjects; the product is considered palatable.

[0152] Therefore, round table evaluations were performed every six days by a group of eight panelists. The panelists were asked to take water and unsalted crackers to cleanse their palates before the test.

[0153] On the first day, all panelists took theophylline and equal concentrations (quantities) of theophylline: pterostilbene cocrystals for sensory analysis.

[0154] On the second day, all panelists took theobromine and an equal concentration (amount) of theobromine:pterostilbene co-crystal for sensory analysis.

[0155] On the third day, all panelists took caffeine and an equal concentration (amount) of caffeine:pterostilbene cocrystal for sensory analysis.

[0156] On the fourth day, all panelists were given caffeine and an equal concentration (amount) of caffeine:pterostilbene co-crystal for sensory analysis.

[0157] On the fifth day, all panelists were given paraxanthine and an equal concentration (amount) of paraxanthine:pterostilbene co-crystal for sensory analysis.

[0158] On the sixth day, all panelists took theophylline and equal concentrations (amounts) of theophylline:resveratrol co-crystals for sensory analysis.

[0159] The results were recorded by each panelist in their own form, indicating their preference (like) and dislike (dislike) for taking. The results of the sensory test showed that all eight panelists had a clear preference for the co-crystals of the present invention compared to pure theophylline, theobromine, methyl-caffeine, caffeine, paraxanthine and theophylline, because the bitter taste of theophylline, theobromine, methyl-caffeine, caffeine, paraxanthine and theophylline was completely masked in the co-crystal form. The results are provided in Table 1 below.

[0160] As is evident from the table below, the panelists showed a clear preference for the co-crystallized methylxanthines compared to the pure methylxanthines, which ultimately suggests that pterostilbene and resveratrol de-bitter the methylxanthines by masking their bitter taste.

[0161] In other embodiments, the co-crystal compositions of the present invention produce various health benefits based on the dosage form, such as improved memory, alertness, concentration, relaxation, enhanced energy and endurance.

[0162] Furthermore, the crystal formers, namely pterostilbene and resveratrol, are safe and tested nutraceuticals and thus synergistically improve the nutraceutical effects of theophylline, theobromine, methylcrocarpine, caffeine, paraxanthine and theophylline.

[0163] In an embodiment, dissolution studies of co-crystals of methylxanthines with the coformers pterostilbene and resveratrol were evaluated. It was observed that in all cases, the dissolution rate did not follow zero-order kinetics, which means that the dissolution rate was independent of the available amount of active ingredient. Compared to its pure form, the co-crystals had lower solubility, slower release rate, and therefore higher bioavailability (Figures 1 to 6).

[0164] experiment:

[0165] The following examples, including preferred embodiments, will serve to illustrate the practice of the invention, it being understood that the details shown are exemplary and for purposes of illustrative discussion of preferred embodiments of the invention.

[0166] General method for eutectic synthesis:

[0167] A general synthetic method for preparing novel co-crystals, the process includes adding the co-former (pterostilbene or resveratrol) to a suitable solvent and dissolving at 45-50°C to obtain a clear solution, followed by adding methylxanthines (theophylline, theobromine, methylcaproline, paraxanthine, caffeine and theophylline). Stirring is continued for 30 min at 45-50°C to obtain a clear solution. The reaction mass is gradually cooled to ambient temperature and maintained at the same temperature for 12-18 hours to crystallize the corresponding co-crystals. Suitable solvents that can be applied to synthesize co-crystals are selected from the group including: C1-5 alcohols, water, formic acid, acetic acid, propionic acid, butyric acid; ethers, esters, ketones, etc. alone or in mixtures thereof. The following additional techniques can also be used to prepare the methylxanthine co-crystals of the present invention; neat grinding, solvent drip grinding, reactive crystallization, anti-solvent addition, cooling crystallization and slow solvent evaporation.

[0196] Example 18: Debittering Study

[0197] The results were recorded by each panelist in their own form, indicating their preference (like) or dislike (dislike) for taking, as shown in the table below.

[0198]

[0199] Note: Theophylline (TC); Theobromine (TB); Methylcaproline (ML); Caffeine (CA), Paraxanthine (PX); Theophylline (TP); Pterostilbene (PB); Resveratrol (RS)

[0200] As is evident from the above, the panelists expressed a clear preference for cocrystals of methylxanthines compared to pure methylxanthines, which ultimately suggests that the coformers pterostilbene and resveratrol debitter the methylxanthines.

[0201] Example 19: Dissolution study of eutectic

[0202] The dissolution data of various methylxanthine cocrystals and pterostilbene / resveratrol systems are represented using 0th-order reaction rates, 1st-order reaction rates, and diffusion kinetics, as shown in Figures 1 to 5. Based on the data, the dissolution patterns of different cocrystal systems are summarized as follows:

[0203] Zero Order Kinetics: In all cases, the plots of % Dissolution versus time indicate that the dissolution rate does not follow zero order kinetics, meaning that the dissolution rate is independent of the available amount of active ingredient.

[0204] Solubility and release rate of active ingredients: From the following Figure (1.A ; 2.A; 3.A and 4.A) it is apparent that the methylxanthines, i.e. theophylline, methylcaproline, caffeine, paraxanthine and theophylline, in cocrystal form, have lower solubility, slower release rate and therefore higher bioavailability when compared to their pure forms. In the case of theobromine, the cocrystals provide improved solubility compared to pure theobromine, leading to better bioavailability. The dissolution patterns of the various cocrystal systems after a certain period of time are summarized in the figure as follows:
