Abstract
Description
Technical Field
[0001] The present invention relates to novel proteins specific for programmed (cell) death ligand 1 (PD-L1). The PD-L1-specific proteins of the present invention have high affinity binding to human and mouse PD-L1. The present invention further relates to PD-L1-specific proteins further comprising diagnostic or therapeutic active components. Further aspects of the present invention encompass the use of these PD-L1-specific binding proteins in medicine, such as in the diagnosis and treatment of PD-L1-related cancers. Background Art
[0002] The transmembrane protein programmed (cell) death ligand 1 (PD-L1; also known as B7-H, B7H1, CD274) belongs to the B7 family of immunomodulatory molecules and is involved in regulating cellular and humoral immune responses. The PD-L1 ligand interacts with PD-1 (programmed cell death 1). PD-L1 is expressed on a variety of normal cells and immune cells, and tumor cells express PD-L1 as a mechanism of immune evasion.
[0003] Both PD-1 and PD-L1 belong to the family of immune checkpoint proteins, which act as co-inhibitors. They can prevent or limit the development of T cell responses, such as inhibiting T cell activation and proliferation and inducing apoptosis of activated T cells. The PD-1 / PD-L1 interaction ensures that the immune system is activated only at the appropriate time to minimize the possibility of chronic autoimmune inflammation.
[0004] PD-L1 expressed on tumor cells binds to the PD-1 receptor on activated T cells, which results in the inhibition of cytotoxic T cells and, in turn, in reduced cytokine production and T cell proliferation. By upregulating PD-L1 expression, tumor cells evade detection and destruction by the immune system. In cancer, PD-L1 provides resistance to T cell-mediated cytotoxicity. By blocking the immune checkpoint receptor PD-1 or its ligand PD-L1, the immune system can overcome the ability of cancer to resist immune responses and stimulate the body's own mechanisms to maintain its effective defense against cancer.
[0005] A variety of immune checkpoint humanized IgG1 antibodies have been developed as PD-1 / PD-L1 inhibitors for cancer immunotherapy, such as atezolizumab, durvalumab, avelumab, BMS-936559, and MEDI4736.
[0006] Existing options often fail to adequately address the diagnosis and treatment of PD-L1-associated cancers, and thus many patients fail to fully benefit from current strategies.
[0007] Undoubtedly, new strategies are urgently needed for the diagnosis and treatment of PD-L1-overexpressing tumors as well as other PD-L1-associated diseases and conditions, including (viral) infections.
[0008] An object of the present invention is to provide proteins that specifically target PD-L1 to allow targeted diagnostic and therapeutic options, including detection of PD-L1 positive tumors (e.g., by radiodiagnostic methods). Targeting this tumor-associated protein can benefit patients who have not yet met the needs of new diagnostic and therapeutic approaches. Due to the low and limited distribution of PD-L1 in normal tissues, targeting PD-L1 indicates a potential non-toxic diagnostic and therapeutic method. Therefore, binding proteins with specificity for PD-L1 can provide effective medical options for cancer treatment and ultimately improve the quality of life of patients.
[0009] The present invention provides novel PD-L1 binding molecules, novel strategies and improved strategies for the diagnosis and treatment of PD-L1 positive tumors. In addition, the novel PD-L1 binding molecules of the present invention provide improved strategies for the diagnosis and treatment of infectious diseases (e.g., chronic viral infections).
[0010] The above objects and advantages are achieved by the subject matter of the appended claims. The present invention meets the above needs by providing new high affinity PD-L1 binding proteins. The above summary does not necessarily describe all problems solved by the present invention. Summary of the invention

The term "PD-L1" includes all polypeptides that show at least 70%, 75%, 80%, 85%, 90%, 95%, 96% or 97% or more, or 100% sequence identity with the PD-L1 (human) of Uniprot accession number Q9NZQ7. Human PD-L1 has 73% identity to mouse PD-L1 (accession number Q9EP73) and 74% identity to rat PD-L1. The term "PD-L1" includes the extracellular domain (residues 19-238) of PD-L1. Human PD-L1 has three domains: an extracellular domain (aa 19-238), a transmembrane domain (aa239-259), and a cytoplasmic domain (aa 260-290). The extracellular domain of human PD-L1 is shown in SEQ ID NO:11.
[0038] The term "PD-L1 binding protein" refers to a protein of the invention that binds to PD-L1 (programmed cell death ligand 1, or programmed death ligand 1) with high affinity, including its multimeric forms disclosed herein. Thus, the proteins of the present disclosure are PD-L1 binding proteins. As described elsewhere herein, the proteins of the present disclosure exhibit specific binding affinity to human PD-L1, which is a binding affinity in the nanomolar range.
[0039] The terms "protein" and "polypeptide" refer to any chain of two or more amino acids linked by peptide bonds, and do not refer to a specific length of the product. Thus, "peptide," "protein," "amino acid chain," or any other term used to refer to a chain of two or more amino acids is included in the definition of "polypeptide," and the term "polypeptide" may be used in place of, or interchangeably with, any of these terms. The term "polypeptide" is also intended to refer to the products of post-translational modifications of a polypeptide, as is well known in the art.
[0040] The term "modification" or "amino acid modification" refers to the substitution, deletion or insertion of a reference amino acid by another amino acid at a specific position in the parent polypeptide sequence. D The higher the value, the weaker the binding partners bind to each other (see Examples). Therefore, as described herein, the terms "less than 100 nM" and "at least 100 nM" can be used interchangeably herein, as they both refer to the high binding affinity of the protein of the present invention to PD-L1. More specifically, in the context of high binding affinity, the terms "less than 100 nM" and "at least 100 nM" each refer to an indication of a K DThe value range is ≤ 100 nM (= including 100 nM), because K D The lower the value, the greater the binding affinity. The same considerations regarding the interchangeable use of the terms "less than" and "at least" apply to all preferred binding affinities for PD-L1 disclosed throughout the specification, e.g., "less than 15 nM" and "at least 15 nM", etc. Again, the terms "less than 15 nM" and "at least 15 nM" each indicate an indication of K D The value range is ≤ 15 nM (= including 15 nM), because K D The lower the value, the greater the binding affinity.
[0072] In a preferred embodiment, the PD-L1 binding protein (including its multimeric form disclosed herein) binds to PD-L1 with a measurable binding affinity of less than 15 nM. Some of these methods are described in the examples below. Typically, the dissociation constant K is determined at 20° C., 25° C. or 30° C. D Unless otherwise specified, the K D The values were determined by SPR at 25°C. D The lower the value, the greater the binding affinity of the biomolecule for its binding partner. D The higher the value, the weaker the binding partners bind to each other (see Examples).

[0130] Example 1. Expression and purification of PD-L1 binding protein
[0131] The genes for proteins 224039, 224058, 224180, and 224121 (SEQ ID NOs: 5-8) were cloned into expression vectors using standard methods known to those skilled in the art, purified, and analyzed as described below. All proteins were expressed and highly purified by affinity chromatography and gel filtration. After affinity chromatography purification, the proteins were purified using System and Superdex TM Size exclusion chromatography (SEC) was performed on a 200 HiLoad 16 / 600 column (GE Healthcare). The column had a volume of 120 ml and was balanced using 2CV. The sample was applied at a flow rate of 1 ml / min using PBS as a running buffer. Fraction collection was started when the signal intensity reached 10 mAU. After SDS-PAGE analysis, positive fractions were pooled and their protein concentrations were measured.
[0132] Purity was confirmed by SDS-PAGE, SE-HPLC and RP-HPLC. Purity determined by RP-HPLC was at least 98%. Protein concentration was determined by absorbance measurement at 280 nm using the specific molar absorption coefficient. The protein was purified using an Ultimate 3000 HPLC system (Thermo Fisher Scientific) and PLRP-S (5 μm, ) column (Agilent) for RP chromatography (RP HPLC).
[0133] Example 2: Mammalian Expression of Human and Mouse PD-L1
[0134] Expi293-F- cells were cultured at 0.5-1 million cells / ml in Expi293-F expression medium (Fisher scientific, 13489756) at 135rpm, 37°C, 8% CO2 and 95% humidity in a shake flask. One day before transfection, cells were inoculated at a density of 2.0 million cells / ml. On the day of transfection, cells were inoculated at a density of 2.5 million cells / ml. 1 μg of the plasmid DNA of the extracellular domain of hPD-L1-Fc or mPD-L1-Fc was diluted in Opti-MEM I reduced serum medium (Life Technologies, 31985-062) per ml culture volume. According to the manufacturer's information, ExpiFectamine was diluted in Opti-MEM I reduced serum medium and incubated at room temperature for 5 min. Subsequently, the DNA-solution was added to the ExpiFectamine-mixture, incubated at room temperature for 20 min and mixed with the cells. After 16 h, the enhancer was added to the cells. After 96-120 h, the supernatant was collected, centrifuged and filtered through a 0.45 μm membrane.
[0135] Example 3. Stability at high temperature (above 60°C)
[0136] The thermal stability of proteins was determined by differential scanning fluorimetry (DSF). Each probe was transferred to a MicroAmp Optical 96-well plate at a concentration of 0.1 μg / μL, and SYPRO orange dye was added with an appropriate dilution. The temperature was programmed to 95°C from 25°C at a heating rate of 1°C / min. Fluorescence was continuously measured at 520nm for excitation and 623nm for emission (ViiA7 real-time PCR system (Thermo Scientific)). The midpoint of the transition (Tm, melting point) of thermal unfolding was determined, and as shown in Table 1.
[0137] Example 4. Biochemical binding analysis (surface plasmon resonance, SPR)
[0138] After NHS / EDC activation, recombinant protein A was immobilized on a high-capacity amine sensor chip (Bruker) using a Sierra SPR-32 system (Bruker) to generate approximately 1000RU. The chip was balanced with SPR running buffer (PBS 0.05%, Tween pH7.3). Ethanolamine was injected after ligand immobilization to block unreacted NHS groups. The Fc-tagged extracellular domains of hPD-L1 and mPD-L1 were injected at 30nM, followed by PD-L1 binding protein at a flow rate of 30μl / min. A flow cell without ligand was used as a reference. Once the ligand binds, the protein analyte accumulates on the surface, increasing the refractive index. This change in refractive index is measured in real time and plotted as a response or resonance unit (RU) versus time. The analyte was applied to the chip in serial dilutions. Association was performed for 120 seconds and dissociation was performed for 180 seconds. After each run, the chip surface was regenerated with 30 μl regeneration buffer (10 mM glycine pH 2.0) and equilibrated with running buffer.Binding studies were performed by using a Sierra SPR-32 system (Bruker) and data evaluation was performed by the Sierra Analyser software provided by the manufacturer.
[0139] The binding affinity to PD-L1 is shown in Table 1. Binding of 211828 to mouse PD-L1 was detectable, but an effective KD could not be determined using SPR.
