Description

Technical Field

[0001] The invention belongs to the technical field of deep processing of plant protein, and specifically relates to a method for preparing a low-bitter sesame protein hydrolyzate with antioxidant properties and its application in food. The method uses high-temperature sesame cake as raw material, and obtains low-bitter sesame hydrolyzate through protein extraction, protein dispersion, multi-enzyme hydrolysis, resin adsorption, membrane separation, etc. Background Art

[0002] Sesame is one of the world's major oil crops. my country is a major producer and consumer of sesame. In 2023, sesame output and sesame consumption will be about 435,300 tons and 1.5 million tons respectively. At present, sesame in my country is mainly used for oil extraction. The production of sesame oil will produce high-temperature sesame cake, which is a by-product of oil extraction after high-temperature baking and fragrance production process (temperature above 150°C, heating time more than 30 minutes). About 600,000 tons of high-temperature sesame cake are produced each year. High-temperature sesame cake is rich in protein (protein content is about 40%), but due to high-temperature denaturation, the protein in high-temperature sesame cake has poor water solubility and is difficult to extract and utilize. It is mostly used as feed and fertilizer, and the protein in high-temperature sesame cake is not fully utilized.

[0003] Studies have shown that the enzymatic hydrolyzate of high-temperature sesame meal protein has a variety of biological activities, such as antioxidant and antihypertensive activities. Therefore, making high-temperature sesame meal protein into a bioactive hydrolyzate is an effective way to improve quality and efficiency. However, the high-temperature sesame meal protein hydrolyzate has a bitter taste, which damages the sensory quality of the product and affects product sales and promotion. In order to remove the bitterness of protein hydrolyzates, people have developed two types of methods: physicochemical debittering and biological debittering. Among them, physicochemical debittering includes embedding method, masking method, and separation method. Biological debittering mainly uses enzymes or microorganisms to modify or metabolize bitter substances to achieve a debittering effect. The physicochemical debittering method has the advantages of easy operation and low cost, but the addition of chemicals has safety risks and the debittering effect is unstable. Although the biological debittering method has the advantages of significant debittering effect and mild conditions, it takes a long time to process and requires strict control of the processing conditions. Therefore, the debittering of protein hydrolyzates requires a reasonable combination of physicochemical and biological debittering methods to obtain the best debittering effect. In addition, since the high-temperature sesame meal protein has poor dispersibility in water, it hinders enzymolysis, thereby reducing the polypeptide yield and increasing the production cost. It is necessary to use relevant technologies to improve the dispersion stability of high-temperature sesame meal protein in water to improve the enzymolysis effect. Therefore, in order to improve the sensory quality of sesame protein hydrolysate, improve enzymolysis efficiency, and control production costs, it is necessary to develop a preparation method for low-bitter sesame protein hydrolysate.

[0004] This application was developed based on this. Summary of the invention

[0005] The purpose of the present invention is to overcome the defects of the prior art and provide a method for preparing a low-bitter sesame protein hydrolyzate with antioxidant properties from sesame cake that can be industrially produced. The method has the advantages of high production efficiency, green environmental protection, low product bitterness, good antioxidant activity, etc.

[0006] To achieve the above object, the present invention adopts the following technical solutions:

[0007] A method for preparing a low-bitter sesame protein hydrolyzate with antioxidant properties, comprising defatting sesame cake, extracting protein, dispersing protein, multi-enzyme hydrolysis, resin adsorption, membrane separation, desalting and concentration, freeze drying, etc. The specific steps are as follows:

[0008] 1) Defatting sesame cake and extracting sesame protein:

[0009] The sesame cake was crushed to 40-100 mesh after removing stones, burnt materials, straws and other impurities, and defatted by Soxhlet extraction. Distilled water was added to the defatted sesame cake, and the pH was adjusted to 9-12 with NaOH solution, stirred at room temperature for 15-50 minutes, centrifuged, and the supernatant was collected. The pH was adjusted to 4-5 with HCl solution, allowed to stand, centrifuged, and the precipitate was collected and freeze-dried to obtain sesame protein;

[0010] 2) Protein dispersion treatment:

[0011] The sesame protein obtained in step 1) is crushed (crushed to 80-120 mesh), distilled water is added, and the pH is adjusted to 8-11 to obtain a sesame protein solution, which is subjected to high temperature treatment, cooled to room temperature, and then dispersed at high speed (10000-15000 r / min for 10-30 s, 3-6 times of treatment), and homogenized at high pressure. After homogenization, the solution is kept at a low temperature of 4-10° C. for standby use;

[0012] 3) Multi-enzyme combined hydrolysis:

[0013] The solution obtained in step 2) is heated to a set temperature of 40-60°C, the pH is adjusted to 7-10.5, and multiple enzymes are added for enzymolysis. After enzymolysis for 3-7 hours, the pH is adjusted to neutral, and centrifuged (8000-10000 r / min for 20-40 minutes), and the supernatant is taken;

[0014] 4) Resin adsorption:

[0015] Add the supernatant from step 3) to a resin adsorption column. After adsorption, wash the column with distilled water, elute with 50-85% ethanol solution, and collect the eluate.

[0016] Among them, the resin adsorption conditions are: the sample volume is 0.5-1.0BV (Bed Volume), the sample loading rate is 0.25-0.75BV / h, the column washing volume of distilled water is 2.5-4.0BV, and the elution volume of ethanol solution is 2-4BV;

[0017] 5) Membrane separation

[0018] The ethanol is removed from the eluate in step 4) by rotary evaporation, and then distilled water is added to the volume of the solution before rotary evaporation, ultrafiltration is performed using an ultrafiltration membrane, the permeate is collected, and the concentrated solution is concentrated by a nanofiltration membrane, and the concentrated solution is freeze-vacuum dried to obtain the product. [0021] Specifically, in step 2), 1 g: 10-20 mL of distilled water can be added to obtain a sesame protein solution; in the high temperature treatment, the heating temperature is 100-160° C. and the heating time is 10-50 min. The high temperature and high pressure treatment can be completed by a high temperature and high pressure reactor or a high temperature and pressure tube.

[0022] Furthermore, in step 2), the high-pressure homogenization pressure is 100 Mpa-200 Mpa, and the process can be performed 2-6 times.

[0023] Specifically, in step 3) multi-enzyme combined hydrolysis, the added amount of the multi-enzyme can be 0.5-2.5% of the mass of sesame protein; the multi-enzyme can be composed of three or more of alkaline protease, aminopeptidase, flavor protease, trypsin, and neutral protease, among which aminopeptidase and flavor protease are essential components, the sum of the two accounts for 25%-40% of the mass of the multi-enzyme, and the ratio of the two is 3-0.5:1.

[0024] Further, in step 4), the resin adsorption is composed of non-polar and weak polar macroporous resins, wherein the mass ratio of non-polar and weak polar macroporous resins is 1:4-8, and the concentration of the eluent ethanol solution is 50-85% (volume percentage). The non-polar macroporous resin can be D101, HP20, HPD100, HPD910, XAD-2, etc., and the weak polar macroporous resin can be CAD40, AB-8, HPD722, HZ-841, etc.

[0025] Furthermore, in step 5), the eluate is ultrafiltered using an ultrafiltration membrane with a molecular weight cutoff of 5000-10000Da, the permeate is collected, and concentrated using a nanofiltration membrane with a molecular weight cutoff of 200-500Da, and the nanofiltration concentration is to 10-20% of the volume of the feed liquid.

[0068] Table 1.

[0069]

[0070] The antioxidant application of low-bitter sesame polypeptides, the sesame protein hydrolysates obtained in Example 1 and Example 2 have high scavenging rates for DPPH and ABTS, which are about 89% and 95% respectively, and both show strong antioxidant activity (see Table 1). This shows that the sesame protein hydrolysate of the present invention can be used as an antioxidant for the preparation of antioxidant drugs, health products, foods, cosmetics, etc.

[0071] Table 1. Antioxidant activity of sesame protein hydrolysate products obtained in Examples 1 and 2

[0072] sample DPPH clearance rate (%) ABTS clearance rate (%) Example 1 90.56±0.38 97.37±0.61 Example 2 88.89±0.52 92.72±0.47

[0073] Note: When measuring the DPPH and ABTS clearance rates, the concentration of sesame protein hydrolysate solution was 2 mg / mL.

[0074] 3) The taste characteristics of sesame protein hydrolysate (bitterness, bitter aftertaste, astringency, astringent aftertaste, umami, umami aftertaste, sweetness, saltiness, and sourness) were determined using the SA402B electronic tongue. Test conditions: The sample was diluted to a sesame protein hydrolysate concentration of 5 mg / mL using 30 mmol / L KCl (containing 0.3 mmol / L tartaric acid). The data of each sample was collected 4 times, and the collection time was 120 s. The minimum error of 3 times was taken as the measurement data of each sample by the staggered method. The reference solution was a 30 mmol / L KCl solution (containing 0.3 mmol / L tartaric acid). When a certain taste data of the test sample was higher than the corresponding taste data of the reference solution, the sample presented that taste, and the greater the numerical difference, the stronger the taste, and vice versa, there was no such taste. When the difference between a certain taste data of the two samples was more than 1 scale, the difference was significant, and people could easily distinguish the difference in a certain taste of the above samples. The results are shown in Table 2.

[0075] Table 2. Taste characteristics of sesame protein hydrolysates obtained in Examples 1 and 2

[0077] It can be seen from Table 2 that the sesame protein hydrolyzate solutions obtained in Examples 1 and 2 have weak bitterness, and exhibit weak saltiness and umami, while other tastes are lost, which provides favorable conditions for use as auxiliary materials in food.

[0078] 4) The determination method of protein and polypeptide amino acids was carried out according to GB / T 18246-2000. The results are shown in Table 3.

[0079] As shown in Table 3, after debittering, the content of hydrophobic amino acid residues (valine, proline, phenylalanine, methionine, leucine, isoleucine, alanine) in sesame protein hydrolysate decreased, and the arginine content also decreased. Studies have shown that the presence of hydrophobic amino acids or arginine at the N-terminus in sesame protein hydrolysate will enhance bitterness.
