Description

Technical Field

[0001] The invention belongs to the field of traditional Chinese medicine preparations, and particularly relates to a method for preparing mulberry and ginger extract. Background Art

[0002] Usually, the preparation process of Chinese medicine extracts adopts the water extraction and alcohol precipitation method. However, in the long-term use process, many problems are found: first, it is impossible to extract all the effective ingredients of each medicine in the whole prescription during water extraction, resulting in a decrease in content, thereby affecting the clinical efficacy of Chinese medicine preparations. Second, the production cost is high, and the amount of alcohol consumed in the ethanol recovery process is large. Third, the finished product has many impurities, and there are still many impurities in the precipitation process, especially proteins, which will bring unexpected adverse reactions to the injection when used, causing pain and loss to patients.

[0003] Cold, also known as upper respiratory tract infection, is a general term for acute inflammation of the nasal cavity, pharynx or larynx. In a broad sense, upper respiratory tract infection is not a disease diagnosis, but a group of diseases, including common cold, viral pharyngitis, laryngitis, herpetic pharyngitis, pharyngoconjunctival fever, and bacterial pharyngitis-tonsillitis. In a narrow sense, upper respiratory tract infection, also known as common cold, is the most common acute respiratory infectious disease, which is mostly self-limiting, but the incidence is high. It occurs 2 to 4 times a year in adults, and the incidence in children is higher, 6 to 8 times a year. It can occur throughout the year, but more often in winter and spring. Mulberry and ginger cold injection is prepared from six traditional Chinese medicines: mulberry leaves, chrysanthemums, forsythia, bitter almonds, dried ginger, and perilla. It has the effects of dispersing wind and heat, removing phlegm and relieving cough. It is mainly used to treat colds, coughs, headaches, and sore throats. Summary of the invention

[0004] Ultra-high pressure extraction (UHPE), also known as ultra-cold isostatic extraction, refers to the use of 100-1000MPa of fluid static pressure on the mixture of extraction solvent and traditional Chinese medicine at room temperature, and maintaining it at a predetermined pressure for a period of time, so that the pressure inside and outside the plant cells reaches equilibrium and then the pressure is quickly released. Due to the sudden increase in osmotic pressure inside and outside the cells, the cell membrane structure changes, allowing the active ingredients in the cells to penetrate various cell membranes and transfer to the extract outside the cells, achieving the purpose of extracting the active ingredients of traditional Chinese medicine. Ultra-high pressure extraction technology has the advantages of high extraction efficiency, ability to retain the biological activity of the extract, low energy consumption, wide range of applications, simple and safe process operation, etc. in traditional Chinese medicine extraction.

[0005] Adsorption clarification is a high-tech method that adds an adsorption clarifier to the Chinese medicine extract or extract concentrate to remove coarse particles in the solution by adsorption, so as to achieve the purpose of refining and improving the clarity of the preparation. The adsorption clarifier used is a substance that flocculates and precipitates large particles and suspended particles with Stoke precipitation tendency in the system through adsorption, bridging, flocculation, and coagulation of inorganic salt electrolyte particles and surface charges, while retaining most effective high-molecular substances such as polysaccharides, and utilizing the protective effect of high-molecular natural hydrophilic colloids on hydrophobic colloids, thereby improving the stability and clarity of the preparation. Using a clarifier to adsorb and clarify the Chinese medicine extract has the advantages of less loss of effective ingredients, high clarification efficiency, simple operation, less time consumption, low cost, safety and non-toxicity, and no residue. Common natural clarifiers include chitosan, juice 101, and ZTC1+1Ⅱ. In order to overcome the problems of existing mulberry and ginger cold preparations, the inventors conducted a large number of experimental studies and found an efficient extraction method and an efficient purification method. At present, there is no report on the preparation of mulberry and ginger extract by combining ultra-high pressure extraction and adsorption clarification.

[0006] At present, there is no report on the preparation of Chinese medicine composition extract, especially mulberry ginger extract, by ultra-high pressure extraction or natural adsorption clarification.

[0013] (3) the wet medicinal material obtained in step (2) and its soaking solution are placed in special containers, respectively, and ultrahigh pressure extraction is performed on each container, and finally the two extracts are combined;

[0014] (4) adding a clarifier to the combined extract obtained in step (3), stirring evenly, maintaining the temperature at 35-65° C., standing for 4-5 hours, centrifuging at high speed, and taking the supernatant;

[0015] (5) performing microfiltration on the supernatant obtained in step (4) to obtain a mulberry ginger filtrate;

[0016] (6) concentrating the mulberry and ginger filtrate obtained in step (5) to obtain a mulberry and ginger extract;

[0017] The mulberry and ginger formula described in step (1) comprises 30 parts of mulberry leaves, 16 parts of forsythia, 12 parts of chrysanthemum, 10 parts of dried ginger, 16 parts of bitter almonds and 16 parts of perilla leaves.

[0018] The amount of water added in step (1) is preferably 3-5 times the total weight of the medicinal materials.

[0019] The ultra-high pressure extraction in step (3) has a pressure of 400-900 MPa, preferably 500 MPa.

[0020] The ultra-high pressure extraction in step (3) is performed 3-6 times, preferably 2-4 times.

[0021] The ultra-high pressure extraction in step (3) has an extraction time of 18-30 minutes.

[0022] The natural clarifier described in step (4) is one of chitosan, fruit juice 101 and / or ZTC1+1Ⅱ, preferably ZTC1+1Ⅱ.

[0023] The natural clarifier described in step (4) is ZTC1+1Ⅱ, wherein the order of adding components A and B is B first and then A.

[0024] The natural clarifier described in step (4) is ZTC1+1Ⅱ. Components A and B are prepared into 1% A and B solutions respectively with 1% acetic acid according to the instructions.

[0025] The natural clarifier described in step (4) is ZTC1+1Ⅱ, and the addition amounts of components A and B are 2-10% and 4-20% of the volume of the liquid respectively.

[0026] The standing time in step (4) is 3-8 hours, preferably 6-8 hours.

[0027] The pore size of the microporous filtration described in step (5) is 1.2 μm.

[0028] According to production needs, the prepared mulberry ginger extract is mixed with appropriate auxiliary materials and then subjected to corresponding process to obtain mulberry ginger tablets, mulberry ginger capsules, mulberry ginger granules, mulberry ginger oral liquid and other preparations.

[0029] The present invention adopts an ultra-high pressure extraction method to extract dried ginger, perilla, bitter almonds containing colds and colds and mulberry leaves, forsythia, and chrysanthemum containing colds and colds, respectively, and can efficiently extract effective ingredients for treating colds and colds from the two groups of medicinal materials. In the process of removing impurities, an adsorption clarification technology is used to add a natural clarifier to the extract, and finally the extract is further removed by microporous filtration technology, which ensures the efficient removal of impurities and the quality of the effective ingredients in the extract. The process of preparing mulberry and ginger extract by combining ultra-high pressure extraction and adsorption clarification technology simplifies the production process, shortens the production time, replaces the traditional ethanol impurity removal with a natural clarifier, saves costs, and avoids environmental pollution caused by ethanol. The mulberry and ginger preparation prepared by the mulberry and ginger extract prepared by the production process has a good market prospect. DETAILED DESCRIPTION
