Abstract

This application relates to a method for efficient propagation of plants using embryogenic cells from *Magnolia grandiflora*, belonging to the field of plant tissue culture technology. The method includes: (1) inducing and proliferating embryogenic cells from immature seeds of *Magnolia grandiflora*; (2) establishing an embryogenic cell suspension culture system through graded screening to achieve synchronized cell proliferation; (3) inoculating the embryogenic cells into a somatic embryo differentiation and maturation medium to obtain a large number of mature cotyledonary embryos; (4) obtaining rooted somatic embryo seedlings through two-step germination culture and transformation culture; and (5) domestication and transplanting. This method allows 0.5g of fresh-weight embryogenic cells to proliferate 15-fold in 4 weeks, and 1g of embryogenic cells can be transformed into approximately 60,000 somatic embryos, ultimately yielding approximately 2,000 seedlings. A cryopreservation technology pathway has also been established, allowing embryogenic cells to be re-established through regeneration culture after vitrification in liquid nitrogen, achieving both long-term germplasm preservation and efficient propagation. This method is not limited by season, providing stable and reliable technical support for the large-scale propagation and resource protection of *Magnolia grandiflora*.
