The application is suitable for the field of biological medicine technology, and provides a preparation method and application of CAR-T cells loaded with biological calcified nanomaterials, which comprises the following steps: step one, mixing black phosphorus quantum dots, calcium peroxide and lipid components, and purifying by ultrasonic emulsification and ultrafiltration centrifugation to obtain BCLT with mitochondria-targeting molecule triphenylphosphine modified on the surface; step two, co-incubating CAR-T cells with membrane dye after staining, and adding a disulfide bond-containing reduction-responsive linker arm DSPE-S-S-NHS; and step three, co-incubating BCLT with CAR-T cells, so that the BCLT is connected to the surface of the CAR-T cells through a chemical bond to obtain CAR-T cells loaded with biological calcified nanomaterials. The application significantly improves the infiltration capacity and killing effect of the CAR-T cells on solid tumors, can relieve the problem of tumor heterogeneity, and has excellent biological safety and synergistic anti-tumor effect.
