The embodiment of the present application provides a pressure detection device and a manufacturing method and a pressure detection method thereof, comprising: a device body, an outer surface of the device body forms a handheld surface, the handheld surface is used for holding the device body by the handheld surface when pressure detection is performed, a device pressure interface is arranged on the device body, the device body is used for controlling a detection medium and outputting the detection medium with a detection pressure from the device pressure interface; a separator, a separation pressure inlet and a separation pressure outlet are arranged on the separator, the separator has a separation abutting surface, the separator is buckled to the device body, the separation abutting surface is abutted to the outer surface of the device body and forms an entirety with the device body, the separation pressure inlet is connected with the device pressure interface, the separation pressure outlet is used for being connected with a measured instrument, a separation structure is arranged in the separator, the separation structure is used for separating mixed impurities flowing into the separation pressure outlet and making the detection medium flow out from the separation pressure inlet.
