Description

Title of the invention: COSMETIC AND / OR DERMATOLOGICAL COMPOSITION COMPRISING AT LEAST ONE EXTRACT OF CYLINDROTHECA FUSIFORMIS AND AT LEAST ONE EXTRACT OF DUNALIELLA SALINA, AND ITS COSMETIC USES Technical field

[0001] The present invention relates to a cosmetic and / or dermatological composition comprising at least one extract of Cylindrotheca fusiformis and at least one extract of Dunaliella Salina, as well as to a non-therapeutic cosmetic use of the cosmetic composition, and to a non-therapeutic cosmetic process implementing the cosmetic composition and a device comprising it.

[0002] The present invention finds application in the field of cosmetics and dermatology, more particularly in cutaneous cosmetics. State of the art

[0003] The skin is a vital organ in its own right, composed of three distinct tissues, each assuming different roles through different cell types and structures.

[0004] The outermost, and therefore most exposed, tissue is the epidermis. This stratified keratinized epithelium (Malpighian) is composed of different cells associated with numerous barrier and protective functions. The predominant cells are keratinocytes, which divide in the basal layer and begin their differentiation up to the stratum corneum (the outermost layer), then are shed by desquamation, on average in 21 to 28 days. The major role of the epidermis is to provide the skin, and therefore the human body, with a first line of protection against external aggressions, such as physical, chemical, water-related, and bacteriological aggressions. This protection is ensured in particular by the most differentiated layers and the stratum corneum, known for its hydrophobic properties and its compact and impermeable appearance. Good differentiation, and therefore good desquamation, is essential for a smooth, homogeneous, and regular surface. All of these mechanisms depend directly on the proliferative capacity of keratinocytes. However, external stresses and age alter keratinocytes and slow down this process, which can then become longer and more irregular. As desquamation becomes irregular and / or chaotic, defects in the smoothness and homogeneity of the skin surface develop.

[0005] In an intermediate position, the dermis is a connective tissue primarily composed of fibroblasts and matrix proteins, giving the skin its well-known compressibility and elasticity. Changes in its texture and composition are largely responsible for the skin alterations that occur during aging. Furthermore, the surface of the stratum corneum is directly determined by the quality and density of the underlying dermis. More so than the epidermis, which renews itself rapidly and constantly, the dermis undergoes significant age-related changes due to the aging of its cells and its structure. As we age and are exposed to environmental stressors, fibroblasts become less and less reactive and less and less proliferative. The synthesis of matrix macromolecules is no longer carried out by fibroblasts.Furthermore, these macromolecules that make up the dermis, such as collagen fibers, elastic fibers, proteoglycans, and glycosaminoglycans (including hyaluronic acid), tend to degenerate and fragment. These dermal tissue alterations cause a disruption in the organization of the epidermis on the surface and, more visibly and directly, a loss of firmness that can lead to sagging skin and / or the formation of wrinkles.

[0006] Within the connective tissue framework, other cells and structures are also interspersed, such as an important circulatory and nutritive network, consisting of blood vessels and lymphatic capillaries, as well as epidermal appendages: hair, body hair, nails, pilosebaceous glands, and sweat glands, which originate in the deep dermis. As previously observed with age and environmental stressors, cellular function slows down, and the endothelial cells constituting the vessels become less tightly joined and less functional, leading to impaired irrigation and nutrition of the integument and its appendages (glands or hair).

[0007] The hypodermis, located deep within and consisting mainly of fatty lobules (adipocytes), provides a primary support function, mechanical and thermal protection, and also plays a role in storing energy reserves that can be rapidly mobilized for all biological needs, such as cell renewal, defense of the body or muscle contraction.

[0008] The surface appearance of the skin is determined by all these tissues. In the epidermis, keratinocytes divide (via mitosis) in the basal layer and begin their differentiation up to the stratum corneum, then desquamation (i.e., their shedding), on average in 21 to 28 days. Good keratinocyte metabolism is essential for a smooth, homogeneous, and even surface. Deeper in the dermis lies the endothelial network, which irrigates the entire skin structure via the bloodstream, supplying water, nutrients, and oxygen. This mechanism depends directly on the body's proper physiological state and also gives the skin its natural color (rosy complexion). However, stress External factors, such as tobacco and UV radiation, slow down this process, which can then become more irregular and damaged: the entire network of endothelial cells suffers and deteriorates, leading to malnutrition, dehydration, and hypoxia. In addition, desquamation becomes irregular and / or chaotic. Uneven skin texture and surface uniformity, a more heterogeneous or yellowish complexion, and dehydration or dryness develop.

[0009] These mechanisms can occur as a consequence of exogenous or endogenous stresses, and can be completely disconnected from age-related mechanisms; on the other hand, the latter accentuate them.

[0010] From a general point of view, the aging process is a generalized process resulting from the advancement of age (intrinsic aging) or from the accumulation of effects due to repeated environmental aggressions (extrinsic aging).

[0011] For the cosmetics market, it is therefore necessary to biologically stimulate one or more mechanisms involved in the aesthetic appearance of the skin in direct or indirect relation to hydration, nutrition, radiance, and protection of the skin.

[0012] The cosmetics market is very large and consumer expectations are very high. In terms of anti-aging or not, they are increasingly turning to effective, natural and eco-designed products.

[0013] For example, vitamin C is very well known and has proven its effectiveness. Like other vitamins, it is commonly found in anti-aging products. Even when of natural origin, this compound is very unstable and therefore requires the addition of preservatives or the use of non-natural derivatives, which are not well-received by consumers.

[0014] Similarly, retinoic acid derivatives are known to stimulate keratinocyte renewal, but weaken the barrier function resulting from keratinocyte differentiation and do not affect cellular detoxification, or even cause inflammatory reactions.

[0015] Similarly, cosmetic responses to skin surface problems frequently focus on alpha hydroxy acids, such as glycolic acids, which have an immediate action due to their desquamating effect. However, they can cause damage to the epidermis and amplify UV-induced damage. Furthermore, their effects are often temporary and can lead to a rebound effect.

[0016] There therefore remains a real need to find new compositions and / or compounds of natural origin, enabling the improvement of the appearance and surface qualities of the skin, in particular for hydrating, anti-aging, soothing effects and enabling the prevention and fight against the effects of pollution. Description of the invention

[0017] The present invention is specifically designed to address these needs and drawbacks of the prior art.

[0018] The inventors are the very first to combine an extract of Cylindrotheca fusiformis and an extract of Dunaliella Salina, making it possible to respond effectively to the aforementioned needs.

[0019] The inventors of the present product have demonstrated properties of the combination that had never been described or suggested in the prior art. Indeed, surprisingly, the inventors have demonstrated that the combination of extracts of Cylindrotheca fusiformis and Dunaliella Salina has a cosmetic effect.

[0020] The inventors have in particular highlighted an effect of the combination of extracts of Cylindrotheca fusiformis and Dunaliella Salina on the improvement of the appearance and surface qualities of the skin, in particular for hydrating, anti-aging, soothing effects and to prevent and combat the effects of pollution.

[0021] Surprisingly, the inventors have also highlighted a synergy of Cylindrotheca fusiformis and Dunaliella Salina extracts for the cosmetic effects mentioned.

[0022] The inventors have also highlighted a synergy of extracts of Cylindrotheca fusiformis, Dunaliella Salina and Tetraselmis Suecica for the cosmetic effects mentioned.

[0023] Thus, a first object of the invention relates to a cosmetic or dermatological composition comprising at least one extract of Cylindrotheca fusiformis and at least one extract of Dunaliella Salina.

[0024] Advantageously, the composition of the invention may further comprise at least one extract of Tetraselmis Suecica.

[0025] Dunaliella salina is a halophilic green microalga, belonging to the class of Chlorophytes, which develops spontaneously and preferentially in very salty lagoon environments.

[0026] Cylindrotheca fusiformis is a marine diatom living in intertidal environments.

[0027] Tetraselmis suecica is a halophilic green microalga, belonging to the class of Chlorodendrophytes.

[0028] The microalgae Cylindrotheca fusiformis, Dunaliella Salina, and Tetraselmis Suecica can be obtained by any process known to those skilled in the art, for example, by culturing and extracting microalgae using a photobioreactor, in a completely sustainable process. The algae can, for example, be cultured in a suitable medium for the time necessary to reach a microalgae concentration of between 50,000 and 1,000,000 cells per ml. The culture is then, for example, centrifuged to harvest the microalgae. The paste of The resulting microalgae can be modified by bursting the microalgae cells, then removing the membrane debris by any suitable separation technique, such as centrifugation and / or filtration, and / or by the action of a solvent, in order to enrich the algal constituents with certain components while simultaneously removing unnecessary or undesirable components for the intended application. Preferably, according to the invention, the extracts are liquid microalgae extracts obtained preferably after culturing the microalgae, centrifuging, and grinding the pellet. These extracts can be water-soluble and / or fat-soluble.

[0029] In the present invention, the term "cosmetic composition" means any composition intended for cosmetic, i.e., aesthetic, purposes that may be brought into contact with the superficial parts of the human body, for example, the epidermis, hair and scalp, external organs, teeth, and external mucous membranes. Advantageously, a cosmetic composition allows, exclusively or primarily, for their protection, perfumement, maintenance in good condition, modification of their appearance, or correction of superficial defects. In particular, a cosmetic composition may include a cosmetically acceptable vehicle.

[0030] In the context of the present invention, the term "dermatological composition" means any composition intended for dermatological use, that is to say, a composition that can be brought into contact with the superficial parts of the human body for the treatment of the skin, mucous membranes, and skin appendages, for example, nails, hair, and / or body hair. In particular, a dermatological composition may include a dermatologically acceptable vehicle.

[0031] By "cosmetically and / or dermatologically acceptable vehicle" is meant a vehicle suitable for use in contact with human skin cells, in particular epidermal cells, without toxicity, irritation, undue allergic reaction and the like, and proportionate to a reasonable benefit / risk ratio. The cosmetically and / or dermatologically acceptable vehicle may be selected from water, allantoin, glycerin, methylpropanediol, this list not being exhaustive.

[0032] The composition of the invention may comprise a percentage by weight of at least one extract of Cylindrotheca fusiformis enabling the cosmetic effects mentioned above to be achieved. Preferably, the percentage by weight of the Cylindrotheca fusiformis extract may be between 0.0001 and 10% by weight relative to the total weight of the composition. For example, this could be 0.01 to 5%, or 0.01 to 3%, or 0.01 to 2%, or 0.01 to 1%, or 0.1 to 3%, or 0.1 to 1%, or 0.05 to 5%, by weight relative to the total weight of the composition.

[0033] The composition of the invention may include a weight percentage of at least one extract of Dunaliella Salina enabling the cosmetic effects mentioned above to be obtained. Preferably, the weight percentage of the Dunaliella extract Salina can be between 0.0001 and 10% by weight relative to the total weight of the composition. This could be, for example, 0.01 to 5%, or 0.01 to 3%, or 0.01 to 2%, or 0.01 to 1%, or 0.1 to 3%, or 0.1 to 1%, or 0.05 to 5%, by weight relative to the total weight of the composition.

[0034] Where appropriate, the composition of the invention may include a percentage by weight of at least one extract of Tetraselmis Suecica enabling the cosmetic effects mentioned above to be achieved. Preferably, the percentage by weight of the Tetraselmis Suecica extract may be between 0.0001 and 10% by weight relative to the total weight of the composition. For example, this could be 0.01 to 5%, or 0.01 to 3%, or 0.01 to 2%, or 0.01 to 1%, or 0.1 to 3%, or 0.1 to 1%, or 0.05 to 5%, by weight relative to the total weight of the composition.

[0035] The cosmetic composition of the invention may, for example, be a combination comprising the two microalgae Cylindrotheca fusiformis and Dunaliella Salina, in a ratio of between 0.1 and 5.0, for example between 0.2 and 4, or between 0.5 and 2. Advantageously, the extracts of Cylindrotheca fusiformis and Dunaliella Salina may be present in a ratio of 1.0 (i.e., one unit of Cylindrotheca fusiformis for one unit of Dunaliella Salina). For example, the composition of the invention may comprise approximately 0.01% by weight of each of these microalgae.

[0036] Alternatively, the cosmetic of the invention may, for example, be an association comprising the three microalgae that are Cylindrotheca fusiformis, Dunaliella Salina and Tetraselmis Suecica, in a ratio (between the extract of Cylindrotheca fusiformis or Dunaliella Salina and the extract of Tetraselmis Suecica) of between 0.1 and 5.0, for example between 0.2 and 4, or between 0.5 and 2. Advantageously, the extracts of Cylindrotheca fusiformis or Dunaliella Salina and Tetraselmis Suecica may be present with a ratio of 0.2 (i.e. one unit of Cylindrotheca fusiformis or Dunaliella Salina for five units of Tetraselmis Suecica). For example, the composition of the invention may comprise about 0.01% by weight of each of the microalgae Cylindrotheca fusiformis and Dunaliella Salina and about 0.05% of Tetraselmis Suecica.

[0037] The cosmetic or dermatological composition of the invention can be obtained by any suitable process known to those skilled in the art for manufacturing a cosmetic or dermatological composition. This may be, for example, a simple mixture. It may also be, for example, a process comprising a step of incorporating an internal phase into an external phase by means of an emulsifier, for example, a rotor-stator type turbine. It may also be, for example, a process using Temperature Inversion Phase (TIP), this process being classically used by those skilled in the art to obtain oil-in-water emulsions. whose dispersed droplets are particularly fine, for example with a diameter of 0.1 to 1 pm.

[0038] The cosmetic or dermatological composition of the present invention can be in any form suitable for cosmetic or dermatological application. Advantageously, the composition is a composition for topical use.

[0039] This may be, for example, a composition in a form chosen from the group comprising an oil-in-water emulsion, or water-in-oil emulsion, or a mixture of these emulsions.

[0040] According to the invention, the composition may, in particular, be in a form selected from a cream, ointment, oil, milk, ointment, solution, powder, gel, serum, balm, butter, lotion, suspension, shampoo, soap, or emulsion, which may be rinsed off or not. For example, the powder may be a loose powder or a compressed powder, for example, in the form of granules, pellets, possibly to be diluted, or in the form of a compact powder. The cosmetic or dermatological composition may, for example, be in a form selected from the group comprising an aqueous or hydroalcoholic gel, an aqueous or hydroalcoholic cream, and an aqueous or hydroalcoholic lotion. These formulations, which can be used to implement the present invention, are known to formulators in the prior art.In these composition examples, it is sufficient to add the combination of essential oils of the present invention to obtain a composition conforming to the present invention.

[0041] The combination of microalgae extracts of the present invention can be used in a cosmetic or dermatological composition, alone or in combination with one or more other substances or ingredients that are cosmetically or dermatologically active or inactive. Inactive substances or ingredients are those that do not have a cosmetic or dermatological effect. These are the elements of the composition that, in particular, accompany the extract, constitute a specific formulation, preserve the active extract over time, etc. They may also include any basic product found in conventional cosmetic or dermatological compositions. In contrast, active substances or ingredients are those that, in the intended cosmetic or dermatological application, have an aesthetic and / or medical effect. This could include, for example, at least one other extract chosen from the group comprising plant extracts, algae extracts and microalgae extracts other than those already mentioned previously.

[0042] Another object of the invention relates to a non-therapeutic cosmetic use of a cosmetic association or composition as defined above.

[0043] Advantageously, non-therapeutic cosmetic use may result in improved hydration of the skin, scalp and / or mucous membranes, and / or an anti-aging action on the skin, and / or an action to combat and / or prevent the effects of pollution on the skin, scalp and / or mucous membranes and / or a soothing action on sensitive and / or reactive skin and / or mucous membranes.

[0044] For the purposes of this invention, "mucous membrane" means, in particular, external mucous membranes such as the lips.

[0045] The cosmetic action aims in particular to improve the aesthetics of the skin. It is therefore a purely aesthetic action, excluding any therapeutic action, allowing to improve the surface appearance of the skin, and / or its appendages and / or mucous membranes, in particular to present, in the long term, a more aesthetic, regular and / or homogeneous appearance.

[0046] Advantageously, the hydrating action may be wholly or partly linked to an improvement in the protective barrier function of the skin, scalp, and / or mucous membranes. Without necessarily being linked by a specific mechanism of action, these actions / effects may be due, in particular, to an effect on the differentiation of epidermal keratinocytes and / or to an increase in filaggrin expression by keratinocytes.

[0047] Advantageously, the soothing action may involve a reduction of diffuse redness, and / or an improvement in skin tone and / or texture and / or radiance, and / or a decrease in sensations of tightness, tingling, diffuse heat, and irritability. Without being bound by a specific mechanism of action, these actions / effects may be due, in particular, to a reduction in the expression of pro-inflammatory mediators by epidermal keratinocytes.

[0048] Advantageously, the anti-aging action can be a preventive action against the visible effects of skin aging, including sagging and / or loss of firmness and / or loss of elasticity and / or the formation of wrinkles, preferably before the effects of aging become visible. Without being bound by a specific mechanism of action, these actions / effects may be due, in particular, to an inhibition of matrix metalloproteinase expression and / or an increase in hyaluronic acid synthesis.

[0049] Advantageously, the action of combating and / or preventing the effects of air pollution on the skin, scalp, and / or mucous membranes may be at least one action selected from among improving skin elasticity, skin radiance, and reducing skin desquamation. Without being bound by a specific mechanism of action, these actions / effects may be due, in particular, to an antioxidant action, a reduction in the expression of pro-inflammatory mediators, or an inhibition of matrix metalloproteinase expression.

[0050] As previously stated, the use of the cosmetic composition may be in a form chosen from a cream, ointment, oil, milk, pomade, solution, powder, gel, serum, balm, butter, lotion, suspension, shampoo, soap or emulsion which may be rinsed or not rinsed.

[0051] Another object of the invention relates to a non-therapeutic cosmetic treatment method for improving the appearance of the skin, comprising applying a cosmetic composition as described above to the skin, scalp, and / or mucous membranes. The non-therapeutic cosmetic method of the invention is therefore an aesthetic skin surface treatment method.

[0052] Advantageously, the non-therapeutic cosmetic process can improve the appearance of the skin, and / or improve the hydration of the skin, scalp and / or mucous membranes, and / or can have an anti-aging effect on the skin, and / or can help to combat and prevent the effects of pollution on the skin, scalp and / or mucous membranes.

[0053] In the context of the cosmetic processes according to the invention, use refers to non-therapeutic use, for example, for the treatment of normal skin. In the present context, "normal skin" means skin not exhibiting a pathological condition.

[0054] In general, any cosmetic use and any cosmetic process according to the invention are respectively non-therapeutic cosmetic uses and non-therapeutic cosmetic processes.

[0055] Another object of the invention relates to a device in a form chosen from a jar, a pump bottle, a wipe, a mask, a transdermal device, a patch, a spray, a capsule or a softgel, the device comprising a composition as defined above.

[0056] Other advantages may also become apparent to a person skilled in the art upon reading the examples below, given by way of illustration. EXAMPLES

[0057] Example 1: Preparation of extracts of Cylindrotheca fusiformis, Dunaliella Salina and Tetraselmis Suecica

[0058] The unicellular algae Tetraselmis Suecica (prasinophyceae / chlorodendrales / chlorodendraceae), Cylindrotheca fusiformis, Dunaliella Salina are cultured in a seawater medium.

[0059] When the algal concentration in the medium is sufficient, the culture is stopped and the algae are then centrifuged to harvest them. The resulting algal paste is then standardized by redispersion in the culture medium. This dispersion is modified by bursting the cells using a high-pressure APV homogenizer. A further centrifugation allows the removal of membrane debris from the burst algae. The cell homogenate, after APV, is resuspended in seawater and stabilized (sorbate / benzoate / hydrochloric acid or citric acid). [0060] Example 2: Effect of extracts from Cylindrotheca fusiformis, Dunaliella Salina and Tetraselmis Suecica, and their combinations, on filaggrin expression

[0061] Keratinocytes were seeded in 96-well plates and cultured in culture medium for 192 hours, with the culture medium being changed after 24 and 96 hours. The medium was then replaced with medium containing or not containing (control):

[0062] (1) either a composition of Cylindrotheca fusiformis at 0.01% by weight, or a composition of Dunaliella Salina at 0.01% by weight, or a composition of Tetraselmis Suecica at 0.05% by weight, or

[0063] (2) the combination Cylindrotheca fusiformis 0.01% + Dunaliella Salina at 0.01% or the association Cylindrotheca fusiformis 0.01% + Dunaliella Salina at 0.01% + Tetraselmis Suecica at 0.05%, or

[0064] (3) the reference (CaCl2 at 1.5 mM),

[0065] and the cells were incubated for 72 hours.

[0066] All experimental conditions were carried out in n = 3.

[0067] The cells were then rinsed with PBS solution, fixed, and permeabilized. The cells were labeled with a specific primary antibody (anti-filaggrin). The primary antibody was then detected using a suitable fluorescent secondary antibody (GAM-Alexa 488), and in parallel, the cell nuclei were stained with Hoechst 33258 solution (bisbenzimide). Image acquisition (5 images / well) was performed with an INCell Analyzer™ 2200 (GE Healthcare, 20x objective). Labeling was quantified by measuring fluorescence intensity and normalized to the total number of cells. (Integration of digital data with the Developer Toolbox 1.5, GE Healthcare software).

[0069] Extracts of Cylindrotheca fusiformis, Dunaliella Salina and Tetraselmis Suecica alone do not give significant results, i.e. no effect on filaggrin expression can be measured.

[0070] On the other hand, the results show a synergistic effect of the association Cylindrotheca fusiformis + Dunaliella Salina and of the association Cylindrotheca fusiformis + Dunaliella Salina + Tetraselmis Suecica on the expression of filaggrin.

[0071] Example 3: Effect of extracts of Cylindrotheca fusiformis, Dunaliella Salina and Tetraselmis Suecica, and their combination, on gene expression after exposure to urban pollution

[0072] Keratinocytes were seeded in 24-well plates and cultured in culture medium for 24 hours. The medium was then replaced with medium containing or not (toxicated control):

[0073] (1) either a composition of Cylindrotheca fusiformis at 0.01% by weight, or a composition of Dunaliella Salina at 0.01% by weight, or a composition of Tetraselmis Suecica at 0.05% by weight, or

[0074] (2) the combination Cylindrotheca fusiformis 0.01% + Dunaliella Salina at 0.01% + Tetraselmis Suecica at 0.05%,

[0075] and the cells were pre-incubated for 24 hours.

[0076] After pre-incubation, the treatments were repeated and the cells were poisoned with urban dust 1649b (Servilab), tested at 0.5 mg / ml. The cells were then incubated for 24 hours. In parallel, a non-poisoned control was performed. All experimental conditions were carried out at n=3. At the end of incubation, the cells were washed in phosphate-buffered saline (PBS) and immediately frozen at -80°C.

[0077] The expression of markers known to increase after exposure to pollution was analyzed by RT-qPCR on total RNA extracted from cell monolayers of each experimental condition (before RNA extraction, replicates of the same experimental condition were grouped).

[0078] Transcript analysis was performed in n=2 using a PCR chip ("mQPA-OX.STRESS-64") dedicated to research and adapted to the 'screening' format (Marker qPCR array or 'mQPA' designed by Bioalternatives) and targeting 64 genes selected for their importance in detoxification.
