AI Technical Summary
The existing drugs for the treatment of chronic obstructive pulmonary disease (COPD) are mainly single-component β2 receptor agonists or methyl receptor antagonists, which have poor efficacy, especially in complex diseases, with limited therapeutic effects.
A crystalline compound with dual activity that both agonizes the β2 receptor and antagonizes the M3 mesorreceptor, and a more effective treatment of COPD is achieved through this versatile drug.
Due to its dual activity, this drug can significantly improve the therapeutic effect on COPD, provide stronger bronchodilation, and is suitable as a pharmaceutical preparation due to its stability and ease of manufacture.

Abstract
Description
[Technical field]
[0001] (See related applications.) This application claims priority to the following:
[0002] This application claims priority to and the benefit of Chinese patent application No. 2022103077974, filed with the State Intellectual Property Office of China on March 25, 2022, the entire contents of which are incorporated herein by reference.
[0003] The present application relates to crystals of fused tricyclic derivatives or pharma- ceutically acceptable salts thereof and a method for preparing the same, as well as the use of said crystals in the manufacture of a medicament for treating diseases associated with the M3 receptor and β2 receptor. [Background technology]
[0004] Bronchodilators play an important role in the treatment of respiratory diseases (e.g., chronic obstructive pulmonary disease and asthma). β-adrenergic receptor agonists and muscarinic receptor antagonists are known bronchodilators that are widely used in clinical applications. Currently, β-adrenergic receptor agonists available via inhalation include short-acting drugs such as salbutamol and terbutaline, and long-acting drugs such as salmeterol, formoterol, vilanterol and indacaterol. These drugs produce bronchodilator effects by stimulating the adrenergic receptors of airway smooth muscle, and can reverse the bronchoconstrictor response to various mediators (e.g., acetylcholine). Currently available inhaled muscarinic antagonists include short-acting ipratropium bromide or oxitropium bromide, and long-acting tiotropium bromide. These drugs can produce bronchodilator effects by reducing vagal cholinergic activity of airway smooth muscle. These drugs can improve lung function, improve quality of life, and alleviate the exacerbation of the disease. Clinical practice has shown that the combination of β2 agonists and M3 antagonists is more effective in treating chronic obstructive pulmonary disease (COPD) than either component alone. At present, combination preparations of muscarinic receptor antagonists and β2 adrenergic receptor agonists have been prepared in clinical practice for the treatment of asthma and moderate to severe COPD. Such combination preparations mainly include Anoro Ellipta (umeclidinium bromide / vilanterol), Ultibro Breezhaler (glycopyrronium bromide / indacaterol) and Duaklir (aclidinium bromide + formoterol fumarate), etc. The combination preparation has a better therapeutic effect than the single preparation, but the preparation is more demanding.
[0005] A single molecule with dual activity at muscarinic M3 and β2-adrenergic receptors (MABA) is more likely to exhibit synergistic drug targeting effects due to its matching pharmacokinetic properties compared to two-component combinations, and shows related advantages in combination. It is also easier to obtain triple combination therapy by formulating it with other therapeutic agents (e.g., inhaled glucocorticoids). Therefore, novel drugs that simultaneously have activity as β2-receptor agonists and muscarinic receptor antagonists and are suitable for the treatment of respiratory diseases (e.g., asthma, COPD) are believed to be of great clinical value and significance. [0140] [Definition and Explanation] Unless otherwise specified, the following terms and expressions used herein have the following meanings: Certain expressions or terms are to be understood in their ordinary sense, not as open or unclear, unless specifically defined: When trade names are mentioned herein, they refer to the corresponding products or their active ingredients.
[0141] It is well known in the field of crystallography that for any given crystal morphology, the relative intensity of the diffraction peaks may change due to preferred orientation resulting from factors such as the crystal morphology. When the effect of preferred orientation is present, the peak intensity may change, but the diffraction peak position of the crystal will not change. It is also well known in the field of crystallography that for any given crystal, there may be a small error in the position of the peak. For example, the peak position may vary due to changes in temperature when analyzing the sample, movement of the sample, or calibration of the instrument, and the measurement error of the 2θ value may be about ±0.2°. Therefore, it is well known to those skilled in the art that such errors should be taken into account when identifying various crystal structures.
[0142] DSC measures the transition temperatures when a crystal undergoes a change in its crystal structure or absorbs or releases heat by melting. For the same type of crystal of the same type of compound, the error of thermal transition temperatures and melting points is typically within about 5°C or 3°C in consecutive analyses. When a compound is said to have a certain DSC peak or melting point, it means that the DSC peak or melting point is ±5°C or ±3°C. DSC provides an auxiliary method for distinguishing different crystals. Different crystal forms can be distinguished by their different transition temperature characteristics. For mixtures, the DSC peak or melting point may vary over a wider range. In addition, since decomposition occurs during the process of melting a substance, the melting temperature is related to the heating rate.
[0143] For the same crystal, the TGA weight loss temperature may vary depending on factors such as measurement equipment, measurement method / conditions, etc. For any particular crystal, there may be an error in the weight loss temperature, and the error may be about ±5°C, or about ±3°C.
[0144] In addition, when preparing crystals of a drug, in the process of contacting the drug molecules with the solvent molecules, due to external conditions or internal factors, the solvent molecules and the compound molecules will form a eutectic and remain in the solid substance, which inevitably produces solvates, specifically stoichiometric solvates and non-stoichiometric solvates. All of the above solvates are included within the scope of the present invention.
[0145] The above-mentioned "pharmaceutical acceptable adjuvant" refers to an inert substance that is administered together with an active ingredient and is advantageous to the administration of the active ingredient, and includes, but is not limited to, any glidant, sweetener, diluent, preservative, dye / colorant, flavor enhancer, surfactant, wetting agent, dispersant, disintegrant, suspending agent, stabilizer, isotonicity agent, solvent or emulsifier approved by the China Food and Drug Administration for use in humans or animals (e.g., livestock).
[0146] The term "crystalline composition" refers to a mixture of a crystalline compound of Formula (I) or Formula (II) or Formula (III) of the present application together with other crystalline or amorphous or other impurities of said compound. [0149] The term "treatment" refers to the administration of a compound or formulation described herein to improve or eliminate a disease or one or more symptoms associated with said disease; (i) inhibiting the disease or disease state, i.e., reducing its progression; (ii) Alleviating the disease or disease state, i.e., causing a reduction in the disease or disease state.
