Abstract
Description
[Technical field]
[0001] [CROSS REFERENCE TO RELATED APPLICATIONS] This application claims the benefit of Chinese Patent Application No. 202110653751.3, filed on June 11, 2021, and No. 202210187387.0, filed on February 28, 2022, all of which are incorporated by reference in their entirety herein.
[0002] [Field of the Invention] The present invention belongs to the medical technology field and relates to compounds having TYK2 inhibitory activity. The present invention also relates to compositions containing the disclosed compounds, their preparation methods, and their applications in targeted therapy for the prevention and / or treatment of diseases related to TYK2, such as tumors, autoimmune diseases, neurodegenerative diseases, metabolic diseases, genetic diseases, etc. [Background technology]
[0003] Janus kinases or JAKs are non-receptor tyrosine kinases that bind to the intracellular portion of cell surface cytokine receptors. Currently, four members of the human JAK family are known: JAK1, JAK2, JAK3 and TYK2 (tyrosine kinase 2), each of which has a kinase domain and a pseudokinase domain (Trends Pharmacol.32 (2011), 25-34). JAK1, JAK2 and TYK2 are expressed in various human tissues, whereas JAK3 is mainly expressed in various hematopoietic cells. A common feature of cell surface cytokine receptors is that the receptors themselves have no kinase activity, but the intracellular segment of the receptors contains a binding site for the tyrosine kinase JAK. Thus, upon cytokine binding to the receptor, JAKs are activated, leading to phosphorylation of JAKs and associated receptors. Receptor phosphorylation then initiates the recruitment of STATs via the SH2 domain, which subsequently initiates the phosphorylation of signal transducer and activator of transcription (STAT) proteins. The phosphorylated STAT homodimers or heterodimers then translocate to the nucleus and bind to specific deoxyribonucleic acid (DNA) binding sites to regulate gene transcription, resulting in altered cellular functions (J. Med.Chem., 62 (2019), 8953-8972).
[0004] Different paired members of the JAK family are responsible for signaling between different cytokines and their respective receptors. For example, TYK2 regulates interleukin 12 (IL12) and IL23-mediated signaling when paired with JAK2, but regulates interferon alpha (IFN-α)-mediated signaling when paired with JAK1. The JAK / STAT pathway is involved in inflammatory responses and therefore may be a therapeutic target for diseases related to immune disorders (J. Med.Chem., 57 (2014), 5023-5038). As a possible therapeutic target for autoimmune diseases, TYK2 in particular has gained support in the research field. For example, mice lacking TYK2 survive and develop normally. However, mice lacking JAK1 (Cell, 93 (1998), 373-383) or JAK2 (Cell, 93 (1998), 397-409) are lethal. Moreover, JAK3-deficient mice show severe B-cell and T-cell exhaustion (Science, 270 (1995), 800-802). Moreover, TYK2 has been shown to be protective in several autoimmune deficiency disease models (multiple sclerosis, Crohn's disease, ulcerative colitis, ankylosing spondylitis, psoriasis, etc.) (Brain, 134 (2011), 693-703; Inflammation (London, UK) 7 (2010), 41; Nat.Rev. Rheumatol.12 (2016), 25-36). TYK2 has also been implicated in several cancers, such as T-lineage acute lymphoblastic leukemia (Cancer Disc.3 (2013), 564-567).
[0005] The value of inhibiting pathways involving TYK2 in the treatment of autoimmune diseases has been clinically demonstrated by various antibodies. Ustekinumab, an antibody targeting the p40 subunit of both IL-12 and IL-23, is currently marketed for the treatment of psoriasis, psoriatic arthritis, and Crohn's disease (Drugs, 71 (2011), 1733-1753; N. Engl. J. Med., 375 (2016), 1946-1960). This antibody has recently shown efficacy in patients with systemic lupus erythematosus (SLE) (Lancet, 392 (2018), 1330-1339). Guselkumab, an antibody that blocks IL-23 signaling by targeting the p19 subunit of IL-23 but not IL-12 signaling, has also been shown to be effective in psoriasis (J. Am. Acad. Dermatol., 76 (2017), 405-417). Several studies have shown that type 1 interferons have a pathogenic role in systemic lupus erythematosus (SLE), and as a result, sifalimumab and anifrolumab have been successfully developed in phase II clinical trials as treatments for SLE (Ann. Rheum. Dis., 75 (2016), 1909-1916; Arthritis Rheumatol., 69 (2017), 376-386).
[0006] Since TYK2 may be a therapeutic target, obtaining a highly selective TYK2 inhibitor may have a good therapeutic effect on the above diseases. Currently, the selective TYK2 inhibitor BMS-986165 (J. Med.Chem., 62 (2019), 8973-8995) and the JAK1 / TYK2 dual inhibitor PF-06700841 (J. Med.Chem., 61 (2018), 8597-8612) are in clinical trials. Therefore, obtaining a highly active and highly selective TYK2 inhibitor or TYK2 / JAK1 inhibitor with high druggability may lead to promising clinical applications. Summary of the Invention

[0063] The terms "isotopically labeled," "isotopically labeled," "isotopically labeled derivative," and "isotopically labeled" refer to an unnatural ratio of atomic isotopes at one or more of the atoms that constitute such a compound. For example, a compound may contain, for example, tritium ( 3 H), Iodine-125( 125 I), Carbon-14 (14 The compounds can also be radiolabeled with a radioisotope such as 2 H, 11 C. 13 C. 15 N, 17 O. 18 O. 18 F, 32 P, 35 S, 36 Certain isotopically labeled disclosed compounds (e.g., 3 H and 14 C) are useful in compound and / or substrate tissue distribution assays. 3 H) and carbon-14 isotopes (i.e., 14 C) can facilitate preparation and detection. In addition, substitution with heavy isotopes such as deuterium (i.e., H) can provide certain therapeutic advantages resulting from higher metabolic stability (e.g., increased half-life in vivo or reduced dosage requirements). Isotopically labeled disclosed compounds can generally be prepared by substituting an isotopically labeled reagent for a non-isotopically labeled reagent. In some embodiments, provided herein are compounds that may also contain unnatural proportions of atomic isotopes at one or more of the atoms that constitute such compounds. All isotopic variations of the disclosed compounds, whether radioactive or not, are encompassed within the scope of the disclosure.
[0064] The term "isomer" as used herein generally refers to different compounds having the same molecular formula, including all geometric isomers, tautomers, and stereoisomers. "Stereoisomers" are isomers that differ only in the arrangement of atoms in space. For example, "isomers" include geometric double bond cis- and trans-isomers, also referred to as E- and Z-isomers; R- and S-enantiomers; diastereomers, (d)- and (l)-isomers, racemic mixtures thereof; and other mixtures thereof, unless otherwise specified, as falling within the scope of this disclosure. [0065] The term "each independently" as used herein means that at least two groups (or ring systems) present in a structure having the same or similar range of values may have the same or different meanings under certain circumstances. For example, if the substituents X and Y are each independently hydrogen, halogen, hydroxyl, cyano, alkyl, or aryl, when the substituent X is hydrogen, the substituent Y can be hydrogen, halogen, hydroxyl, cyano, alkyl, or aryl. Similarly, when the substituent Y is hydrogen, the substituent X can be hydrogen, halogen, hydroxyl, cyano, alkyl, or aryl.
[0066] As used herein, the terms "optional" or "optionally" mean that the subsequently described event or circumstance may or may not occur, and that the description includes both the occurrence and non-occurrence of the subsequently described event or circumstance.
[0067] In some embodiments, the compound(s) of Formula (I), (II), (III), (IV), (V), (VI) or (VII) are used to treat a subject by administering the compound(s) as a pharmaceutical composition. To this end, the compound(s) are, in one embodiment, combined with one or more pharma- ceutically acceptable excipients, including carriers, diluents or adjuvants, to form a suitable composition, as described in more detail herein.
[0068] The term "excipient" as used herein generally refers to a pharma- ceutically acceptable additive, carrier, adjuvant, or other suitable ingredient, other than the active pharmaceutical ingredient (API), that is normally included for formulation and / or administration purposes.
[0069] The term "diluent" as used herein generally refers to an agent used as a filler to achieve the desired volume or weight of the composition. Diluent may be present in the pharmaceutical composition within the granule in the form of a single compound or in the form of a mixture of compounds. Non-limiting examples of diluents include lactose, starch, pregelatinized starch, microcrystalline cellulose, silicified microcrystalline cellulose, cellulose acetate, dextrose, mannitol, sodium phosphate, potassium phosphate, calcium phosphate, fructose, maltose, sorbitol, or sucrose.

However, when an adjuvant is used alone, no pharmacological effect is observed against the same target.
[0071] The terms "treat", "treating", "treatment" and "treatment" as used herein generally refer to treatments including, but not limited to, therapeutic, prophylactic and preventative treatments. Prophylactic treatments generally prevent the onset of a disorder altogether or delay the onset of a pre-clinically evident stage of a disorder in an individual. Treatment includes medical management of a patient with the intent of curing, ameliorating, stabilizing or preventing a disease, pathological condition or disorder. The term includes active treatments, i.e. treatments dedicated to ameliorating a disease, pathological condition or disorder, and also includes causal treatments, i.e. treatments directed at removing the cause of an associated disease, pathological condition or disorder. In addition, the term includes palliative treatments, i.e. treatments aimed at alleviating symptoms rather than curing a disease, pathological condition or disorder; preventive treatments, i.e. treatments aimed at minimizing or partially or completely suppressing the onset of an associated disease, pathological condition or disorder; and supportive treatments, i.e. treatments used to complement another specific treatment aimed at ameliorating an associated disease, pathological condition or disorder.
[0072] As used herein, the term "prevention" or "preventing" refers to preventing, avoiding, removing, forestalling, stopping, or hindering something from happening, especially by prior action. Where reduce, inhibit, or prevent are used herein, it is understood that the use of the other two words is also expressly disclosed unless specifically indicated otherwise.
[0073] The term "effective amount" or "therapeutically effective amount" as used herein refers to a sufficient amount of an administered drug or compound to alleviate to some extent one or more symptoms of the disease or condition being treated; achieve the goal of improving the severity and frequency of the disorder more than treatment with each drug alone; and result in a reduction and / or alleviation of the signs, symptoms, or causes of the disease, or any other desired change in a biological system; while avoiding side effects typically associated with alternative therapies. For example, an "effective amount" for therapeutic use is the amount of a composition as disclosed herein that is required to provide a clinically significant reduction in disease symptoms. The appropriate "effective" amount in any individual case can be determined using techniques such as dose escalation studies. An effective amount is, in one embodiment, administered in a single dosage form or in multiple dosage forms.
[0074] Regardless of the selected route of administration, the compounds of the present invention, which may be used in a suitable hydrated form, and / or the pharmaceutical compositions of the present invention, are formulated into a pharma- ceutically acceptable dosage form or by other conventional methods known to those skilled in the art.
[0075] The actual dosage of the active ingredient in the pharmaceutical compositions of the present invention can be varied, without toxicity to the patient, to obtain an effective amount of the active ingredient to achieve the desired therapeutic response for a particular patient, composition, and mode of administration.
[0076] The selected dosage level will depend upon a variety of factors, including factors well known in the medical art, such as the activity of the particular compound of the invention being employed, the route of administration, the time of administration, the rate of excretion of the particular compound being employed, the duration of treatment, other drugs, compounds and / or materials used in combination with the particular hedgehog inhibitor being employed, the age, sex, weight, condition, general health and prior medical history of the patient being treated.
[0077] A physician or veterinarian having ordinary skill in the art can readily determine and prescribe the effective amount of the pharmaceutical composition required. For example, the physician or veterinarian can start doses of the compounds of the invention employed in the pharmaceutical composition at levels lower than those required to achieve the desired therapeutic effect, and gradually increase the dosage until the desired effect is achieved.
[0078] In general, a suitable daily dose of the compound of the present invention is that amount of the compound that is the lowest effective dose to produce a therapeutic effect. Such an effective amount generally depends on the factors described above. In general, the intravenous, intracerebroventricular and subcutaneous dose of the compound of the present invention to a patient ranges from about 0.0001 to about 100 mg per kg of body weight per day. The mode of administration can have a large effect on the dosage. Higher doses can be used for localized routes of administration.
[0079] If desired, the daily effective amount of active compound can be administered as 2, 3, 4, 5, 6 or more subdoses at appropriate intervals throughout the day, and can be administered separately, optionally in unit dosage form. Those skilled in the art will readily understand that dosage levels can vary as a function of the specific compound, the severity of symptoms, and the subject's susceptibility to side effects. The dosage of a given compound disclosed herein can be readily determined by those skilled in the art by a variety of means.
[0080] [Pharmaceutical composition / formulation] One embodiment provides a pharmaceutical composition comprising a compound of Formula (I), (II), (III), (IV), (V), (VI) or (VII), or a stereoisomer, tautomer, hydrate, solvate or pharma- ceutically acceptable salt thereof, and at least one pharma- ceutically acceptable excipient.
[0081] In some embodiments, the compounds described herein are formulated into pharmaceutical compositions. Pharmaceutical compositions are formulated in a conventional manner with one or more pharma- ceutically acceptable inactive ingredients that facilitate the processing of active compounds into pharma- ceutically usable preparations. [0088] The compounds of the present invention can be prepared starting from commercially available materials using various synthetic routes, including those described below. The starting materials of the present invention are known, commercially available, or can be synthesized similarly or according to methods known in the art. Many starting materials can be prepared according to known processes, and in particular using the processes described in the examples. When synthesizing starting materials, functional groups are sometimes protected with suitable protecting groups as necessary. Functional groups can be removed according to procedures known in the art.
[0089] The protection of functional groups by protecting groups, the protecting groups themselves, and reactions for their removal (commonly referred to as "deprotection") are described in standard references, such as J. F. W. McOmie, Protective Groups in Organic Chemistry, Plenum Press, London and New York (1973), in T. W. Greene, Protective Groups in Organic Synthesis, Wiley, New York (1981), in The Peptides, Volume 3, E. Gross and J. Meienhofer editors, Academic Press, London and New York (1981).
[0090] All synthetic procedures described herein can be carried out under known reaction conditions, advantageously under the conditions described herein, either in the absence or in the presence (usually) of solvents or diluents.
[0091] The invention further encompasses "intermediate" compounds, including structures produced from the described synthetic procedures, whether isolated or not, prior to finally obtaining the desired compound. Structures resulting from carrying out steps from transient starting materials, structures resulting from branching out of the described methods at any stage, and structures forming starting materials under reaction conditions are all "intermediates" encompassed by the invention. Additionally, structures produced by using starting materials in the form of reactive derivatives or salts, or by compounds obtained by processes according to the invention, and structures obtained by treating compounds of the invention in situ are also within the scope of the invention.
[0092] Novel starting materials and / or intermediates, as well as processes for their preparation, are also subject of the present invention. In selected embodiments, such starting materials are used and reaction conditions are selected to obtain the desired compound(s).
[0093] The starting materials of the present invention are known, commercially available, or can be synthesized similarly or according to methods known in the art. Many starting materials can be prepared according to known processes, in particular using the processes described in the examples. When synthesizing starting materials, functional groups are sometimes protected with appropriate protecting groups as necessary. Protecting groups, their introduction and removal have been described above.
[0094] All reagents and solvents were used commercially unless otherwise noted. Commercially available reagents and solvents were used without purification unless otherwise noted. Some reagents and solvents were purified by standard techniques when necessary. For example, tetrahydrofuran can be purified by distillation from sodium. All thin-layer chromatography (TLC, GF254) analyses and column purifications (100-200 mesh) were performed on silica gel (Qingdao Haiyang Chemical Co., Ltd. or Yantai Chemical Co., Ltd.) using petroleum ether (bp 60-90 °C) / ethyl acetate (v / v) as the eluent; spots were revealed by UV visualization with I2 vapor or phosphomolybdic acid at 254 nm. All organic layers after extraction were dried over anhydrous Na2SO4 unless otherwise noted. All nuclear magnetic resonance spectra ( 1 H NMR was recorded on a Varian-400 spectrometer at 400 MHz with TMS as the internal standard. LC-MS was performed on an Agilent 1100 system equipped with an LC-MSD trap recorder, a diode array detector (DAD) with detection wavelengths of 214 nm and 254 nm, and an ESI source. The HPLC column was an Agela Durashell C18 3.5 μm 4.6 × 50 mm column. The gradient was run from 5 / 95 to 95 / 5 with 0.1 NH4HCO3 in water and acetonitrile for the indicated run time (e.g., 5 min) at a flow rate of 1.8 mL / min.
[0095] The size and scale of the synthesis varies depending on the amount of final product desired. Specific reactants and amounts are described in the examples, but it is understood that one skilled in the art would know of other alternative and equally viable sets of reactants that would yield the same compounds. Thus, where common oxidizing agents, reducing agents, and solvents of various natures (aprotic, non-polar, polar, etc.) are utilized, equivalents are known in the art and are contemplated herein for use in the present methods.
[0096] Many of the steps below show various work-ups after the reaction is complete. Work-ups generally involve quenching the reaction to terminate any remaining catalytic activity and starting reagents. This is typically followed by the addition of an organic solvent and separation of the aqueous and organic layers. The product is generally obtained from the organic layer, while unused reactants, spurious by-products, and unwanted chemicals are generally captured in the aqueous layer and discarded. Work-ups in standard organic synthesis procedures found throughout the literature are generally performed by drying the product by exposure to a drying agent such as anhydrous Na2SO4, removing excess water and aqueous by-products that remain partially dissolved in the organic layer, and concentrating the remaining organic layer. Concentration of the product dissolved in the solvent can be achieved by known means, such as evaporation under pressure, evaporation at elevated temperature and pressure, and the like. Such concentration can be achieved by using standard laboratory equipment such as rotary evaporator distillation, and the like. This is optionally followed by one or more purification steps, including, but not limited to, flash column chromatography, filtration through various media, and / or other preparative methods known in the art, and / or crystallization / recrystallization. (See, e.g., Addison Ault, “Techniques and Experiments for Organic Chemistry,” 6th Ed., University Science Books, Sausalito, Calif., 1998, Ann B. McGuire, Ed., pp. 45-59).

[0199] Example 15, Competitive Binding Assay
[0200] Testing Procedure:
[0201] For most assays, kinase-tagged T7 phage stocks were prepared using E. coli from the BL21 strain as the host. E. coli were grown to log phase, infected with T7 phage, and incubated at 32°C with shaking until lysis. The lysate was centrifuged and filtered to remove cellular debris. The remaining kinase was produced in HEK-293 cells and then tagged with DNA for qPCR detection. Streptavidin-coated magnetic beads were treated with biotinylated small molecule ligands for 30 min at room temperature to create affinity resins for kinase assays. Ligand beads were blocked with excess biotin and washed with blocking buffer (SeaBlock (Pierce), 1% BSA, 0.05% Tween20, 1 mM DTT) to remove unbound ligand and reduce nonspecific binding. Binding reactions were assembled by combining kinase, ligand affinity beads, and test compounds in 1x binding buffer (20% SeaBlock, 0.17x PBS, 0.05% Tween20, 6mM DTT). Test compounds were prepared as 111x stocks in 100% DMSO. Dissociation constants (Kds) were determined using an 11-point 3-fold dilution series, including three DMSO controls. All compounds for Kd measurements were dispensed in 100% DMSO by acoustic transfer (non-contact dispensing). Compounds were then diluted directly into the assay so that the final concentration of DMSO was 0.9%. All reactions were performed in polypropylene 384-well plates. The final volume of each was 0.02ml. The assay plates were incubated with shaking for 1 hour at room temperature, and the affinity beads were washed with wash buffer (1x PBS, 0.05% Tween20). The beads were then resuspended in elution buffer (1x PBS, 0.05% Tween 20, 0.5 μM non-biotinylated affinity ligand) and incubated at room temperature for 30 min with shaking. Kinase concentrations in the eluates were measured by qPCR.
[0202] Eleven-point 3-fold serial dilutions of each test compound were prepared in 100% DMSO to a final test concentration of 100× and then diluted to 1× in the assay (final DMSO concentration=1%). Most Kds were determined using the highest concentration of compound=30,000 nM. If the initial Kd determined was less than 0.5 nM (lowest concentration tested), measurements were repeated with serial dilutions starting from the lower highest concentration. A reported Kd value of 40,000 nM indicates that the Kd was determined to be >30,000 nM.
[0203] Binding constants (Kds) were calculated from standard dose-response curves using the Hill equation:
number
[0204] Hill slope was set to -1.
[0205] Curves were fitted using a nonlinear least-squares fit with the Levenberg-Marquardt algorithm.
[0206] [Table 2]
[0207] Conclusion According to Table 2 and Figure 2 (compound B2), the tested compounds B2-B5, B11, B19, B22, B24, B29, B32-B35, B37-B39 and B41 show strong affinity for the TYK2 pseudokinase domain, while at the same time showing little affinity for the JAK1 and JAK2 kinase domains.
[0208] Example 16. Luciferase assay
[0209] Testing Procedure: 1. L929 ISRE cells (5000 cells / well) were seeded into 96-well plates and cultured overnight at room temperature; 2. The sampling process is finished within 15 minutes. 5. Incubation: Seal the plate using plate film. Shake at 300 rpm and incubate at room temperature for 2 hours. 6. Discard the washing solution, add 300 μL of washing solution, and wash the plate six times. Complete removal of residual liquid is required to obtain the desired experimental performance. 7. Add enzyme: add 100 μL of diluted horseradish peroxidase-labeled streptavidin to each well (1:100 dilution). 8. Incubation: Seal the plate using plate film. Shake at 300 rpm and incubate at room temperature for 45 minutes. 9. Wash. Repeat step 8. 10. Add chromogenic substrate: Add 100 μL of chromogenic substrate TMB to each well and incubate at room temperature away from light for 5-30 minutes. 11. Add Stop Solution: Add 100 μL of Stop Solution to each well. The color should change from blue to yellow. If it appears green or the color change is uneven, gently tap the plate frame to mix well. 12. Assay readout: Within 30 minutes, measure the OD at the absorption maximum wavelength of 450 nm and the reference wavelength of 570 nm or 630 nm using a microplate reader with dual wavelength detection. The calibrated OD value is the measurement at 450 nm minus the measurement at 570 nm or 630 nm.
[0214] [Table 4]
[0215] The results are shown in Table 4 and Figure 3. Compounds B37, B38, B41 and BMS986165 exhibited inhibitory effects on IFNα-stimulated CXCL-10 production in PBMCs. Compounds B37 and B41 were both more active than BMS986165.
[0216] (Example 18, Selectivity Evaluation)
[0217] Aim 1: Verify the inhibitory activity of JAK2 / JAK2 pathway
[0218] Testing Procedure:
[0219] MEG-1 cells were digested with trypsin, resuspended in 1640 medium, and counted. Compounds to be measured were added to 6-well plates (1 million / well) for 2 h, and then TPO (final concentration: 100 ng / mL) was added to stimulate cells for 30 min. Cells were harvested and lysed with protein lysis solution. Protein was quantified with a BCA kit. Finally, p-STAT3 / STAT3 and the internal reference protein β-actin were detected by Western blot.
[0220] As shown in Figure 4, the JAK1 / JAK2 inhibitor ruxolitinib (RUX in the figure) could inhibit the JAK2 / JAK2 pathway. Compounds B37 and B41, which did not affect TPO-induced downstream STAT3 phosphorylation, were similar to BMS986165, indicating that they had no inhibitory activity against the JAK2 / JAK2 pathway. Therefore, compounds B37 and B41 had no inhibitory activity against JAK2.
[0221] Objective 2: Verify the inhibitory activity of the TYK2 / JAK2 pathway
[0222] Testing Procedure:
[0223] Spleens from mice were isolated and crushed into single cells with curved forceps. Splenic single cells were centrifuged (400g) for 5 min, red blood cells were lysed with ACK solution, neutralized with 1640 medium, then centrifuged (400g) for 5 min, and resuspended in 1640 medium. Splenic single cell suspensions were counted and incubated in 6-well plates (5 million / well), compounds to be tested were added for 2 h, and then IL-12 (50 ng / mL) was added to stimulate the cells for 1 h, and the cells were harvested and lysed with protein lysis solution. Protein was quantified with a BCA kit. Finally, p-STAT4 / STAT4 and the internal reference protein β-actin were detected by Western blot.
[0224] As shown in Figure 5, compounds B37 and B41 can effectively inhibit downstream STAT4 phosphorylation induced by IL-12, indicating that the compounds can inhibit the TYK2 / JAK2 pathway and their inhibitory activity is superior to BMS986165. Combined with the results in Figure 4 showing that compounds B37 and B41 did not inhibit JAK2, it is believed that their inhibitory activity against the TYK2 / JAK2 pathway comes from the inhibition of TYK2. Overall, the activity of B37 and B41 against TYK2 was superior to BMS986165.
[0225] Objective 3: Verify the inhibitory activity of the TYK2 / JAK1 pathway
[0226] Testing Procedure:
[0227] Commercially available human PBMCs (peripheral blood mononuclear cells) were counted by centrifugation (400g) for 10 min and seeded in 6-well plates (5 million / well). Then, the compounds to be tested were added for 2 h, and IFN-α (100 ng / mL) was added to stimulate the cells for 15 min. Protein lysis solution was used to lyse the cells, and the protein was quantified using a BCA kit. Finally, p-STAT1 / STAT1, P-TYK2 / TYK2 and the internal reference protein β-actin were detected by Western blot.
[0228] As shown in Figure 6, compounds B37, B38 and B41 can effectively inhibit downstream STAT1 phosphorylation induced by IFN-α, indicating that they can inhibit the TYK2 / JAK1 pathway. Among them, compounds B37 and B41 showed superior inhibitory activity to BMS986165.
[0229] Objective 4: Verify the inhibitory activity of the JAK1 / JAK2 pathway
[0230] Testing Procedure:
[0231] HT-29 cells were digested by trypsin, suspended in DMEM medium, and counted. Test compounds were added to 6-well plates (400,000 / well) for 2 h, and then IFN-γ (100 ng / mL) was added to stimulate cells for 30 min. Cells were harvested and lysed with protein lysis solution. Protein was quantified by BCA kit. Finally, p-STAT1 / STAT1 and the internal reference protein β-actin were detected by Western blot.
[0232] As shown in Figure 7, compound B37 was similar to BMS986165 and did not affect downstream STAT1 phosphorylation induced by IFN-γ, indicating that it had no inhibitory effect on the JAK1 / JAK2 pathway, whereas compound B41 inhibited the JAK1 / JAK2 pathway in a concentration-dependent manner. Combined with the results of Figure 4, compound B41 did not show inhibitory activity against JAK2, indicating that the inhibitory effect of B41 on the JAK1 / JAK2 pathway is derived from inhibition of JAK1. As a result, B41 showed higher biological activity than BMS986165. Thus, compound B41 or other compounds disclosed herein may be used to treat autoimmune diseases.
[0233] Objective 5: Verify the inhibitory activity of the JAK1 / JAK3 pathway
[0234] Testing Procedure:
[0235] Commercially available human PBMCs (peripheral blood mononuclear cells) were centrifuged (400g) for 10 min, counted and seeded in 6-well plates (5 million / well). After adding the compounds to be tested for 2 h, IL-2 (100 ng / mL) was added to stimulate the cells for 15 min. Finally, p-STAT5 / STAT5 and the internal reference protein β-actin were detected by Western blot.
[0236] As shown in Figure 8, compound B37, similar to BMS986165, did not affect IL-2-induced downstream STAT5 phosphorylation, indicating that it had no inhibitory activity against the JAK1 / JAK3 pathway, whereas compound B41 was able to inhibit STAT5 phosphorylation. The inhibitory activity of B41 was slightly lower than that of the JAK1 / JAK2 inhibitor ruxolitinib (denoted as RUX in Figure 8), indicating that B41 exhibited inhibitory activity against the JAK1 / JAK3 pathway. This experimental result again demonstrated that compound B41 had inhibitory activity against JAK1.

[0245] Example 21, Plasma Protein Binding Assay
[0246] Testing Procedure: 1. Test and control compounds were dissolved in dimethyl sulfoxide (DMSO) to prepare 10 mM stock solutions. DMSO working solutions were prepared at 400 μM. To prepare the loading matrix, compound working solution (5 μL) was added to blank matrix (995 μL) in a 1:200 ratio and mixed thoroughly. 2. To prepare time zero (T0) samples for use in measuring recovery, 50 μL aliquots of loading matrix were transferred in triplicate to a sample collection plate. Samples were immediately combined with blank buffer on the other side to prepare a final volume of 100 μL of 1:1 matrix / dialysis buffer (v / v) in each well. 500 μL of stop solution was added to these T0 samples. These samples were stored at 2–8 °C along with the other post-dialysis samples. 3. To load the dialysis devices, 150 μL of loading matrix was transferred in triplicate to the donor side of each dialysis well, and 150 μL of dialysis buffer was loaded into the receiver side of the wells. The dialysis plate was placed on a slowly rotating (approximately 100 rpm) shaking platform in a humidified incubator at 37°C with 5% CO2 for 4 hours. 4. At the end of dialysis, 50 μL samples were taken from both the buffer and matrix sides of the dialyzer. These samples were transferred to a new 96-well plate. Each sample was mixed with an equal volume of the opposite blank matrix (buffer or matrix) such that the final volume in each well was 100 μL of 1:1 matrix / dialysis buffer (v / v). All samples were further processed by adding 500 μL of stop solution containing internal standards. The mixtures were vortexed and centrifuged at 4000 rpm for approximately 20 min. Aliquots of 100 μL of the supernatant of all samples were removed for LC-MS / MS analysis. 5. Single blank samples were prepared by transferring 50 μL of blank matrix to a 96-well plate and adding 50 μL of blank PBS buffer to each well. The blank plasma must match the type of plasma used on the plasma side of the well. The matrix-matched samples were then further processed following the same sample processing method as the dialyzed samples, adding 500 μL of stop solution containing the internal standard.
[0247] Data Calculation:
[0248] The %Unbound and %Bound rates were calculated using the following formula: Non-coupling rate (%) = 100 × [F] / [T]; Coupling rate (%) = 100 - non-coupling rate (%)
[0249] where [F] is the analyte concentration or analyte / internal standard peak area ratio on the buffer (receiver) side of the membrane, [T] is the analyte concentration or analyte / internal standard peak area ratio on the matrix (donor) side of the membrane, and [T0] is the analyte concentration or analyte / internal standard peak area ratio in the loading matrix sample at time zero.
[0250] [Table 7]
[0251] According to Table 7, compounds B2, B22, and B41 exhibited moderate to high plasma protein binding.
[0252] (Example 22, hERG channel inhibitory activity test)
[0253] Testing Procedure:
[0254] A Chinese hamster ovary (CHO) cell line, CHO-hERG cells, was used in this study. CHO-hERG cells were obtained from Sophion Biosciences Inc. (Ballerup, Denmark), Suzhou, China. Cell reserves are frozen in liquid nitrogen tanks. Each batch of stored cell liquid is tested for mycoplasma contamination. The cells were discontinued after 30 generations.
[0255] hERG currents were recorded at room temperature using the whole-cell patch clamp technique. The patch clamp amplifier output was digital-to-analog converted and low-pass filtered at 2.9 kHz. Data were collected using Patchmaster Pro software.
[0256] The cells were planted in a cell recording chamber and placed on an inverted microscope stage. One cell was randomly selected from the recording chamber for the experiment. The perfusion system was fixed to the inverted microscope stage, and the cell was continuously perfused with ECS.
[0257] Microelectrodes for manual patch clamp recording were prepared using capillary glass tubes filled with intracellular solution. On the day of patch clamp testing, electrodes were prepared using borosilicate glass tubes (GC150TF-10, Harvard Apparatus Co.). The resistance of the electrodes after filling them with ICS was 2–5 MΩ.
[0258] The clamp voltage was -80 mV, and in the first step, the hERG channels were opened by depolarizing to +60 mV for 850 ms. The voltage was then set to -50 mV and maintained for 1275 ms, resulting in a rebound or tail current, the peak of which was measured and used for analysis. Finally, the voltage was returned to the clamp voltage (-80 mV). The command voltage program was repeated every 15 seconds during the test.
[0259] At the start of recording, the tail current peak can be monitored until the inhibition of the test substance / positive control working solution against the hERG current peak is stable, and then the test substance / positive control working solution can be perfused. In general, the peak value of the current curve is basically the same for the last three consecutive times as the criterion for the stable state. Once the stable state is reached, one concentration of the test sample is subsequently injected. One or more targets / positive controls, or multiple concentrations of the same drug, can be tested in one cell, and different targets / positive controls should be washed with the solvent control working solution until the hERG current returns to more than 80% of the value before drug administration. The standard deviation of the inhibition rate of all cells recorded at the same concentration was less than 15%.
[0260] The positive control cisapride was administered at a concentration of 0.1 μM in duplicate. Cisapride inhibited hERG currents by more than 50% at 0.1 μM.
[0261] [Table 8]
[0262] According to Table 8, compounds B2, B22, B37, and B41 did not inhibit the hERG channel.
