Abstract
Description
[Technical field]
[0001] The present invention is in the field of immunotherapy. More specifically, the present invention relates to humanized antibodies targeting CD7 and their use in the prevention, treatment and diagnosis of diseases. [Background technology]
[0002] CD7 is a cell surface glycoprotein with a molecular weight of about 40 kDa, and is a member of the immunoglobulin superfamily. CD7 is expressed in most T cells, NK cells, myeloid cells, T cell acute lymphoblastic leukemia / lymphoma, acute myeloblastic leukemia, and chronic myelogenous leukemia. It is reported that CD7 molecule acts as a costimulatory signal during T cell activation by binding with its ligand K12 / SECTM1. It is also reported that disruption of CD7 molecule in mouse T precursor cells still results in normal T cell development and homeostasis, but CD7 does not significantly affect T cell development and function, which indicates that it is a very suitable therapeutic target for treating T cell acute lymphoblastic leukemia (T-ALL). In fact, CD7 has been widely studied as a target of cytotoxic molecules for treating leukemia and lymphoma.
[0003] The present invention aims to provide humanized antibodies targeting CD7 and their use in the prevention, treatment and diagnosis of disease. Summary of the Invention

[0019] Unless otherwise specified, all scientific and technical terms used herein have the same meaning as commonly understood by one of ordinary skill in the art. CD7 humanized antibody
[0020] As used herein, the term "antibody" has the broadest meaning as understood by those skilled in the art and includes monoclonal antibodies (including intact antibodies), polyclonal antibodies, multivalent antibodies, multispecific antibodies (e.g., bispecific antibodies), and antibody fragments or synthetic polypeptides having one or more CDR sequences capable of exhibiting the desired biological activity. Antibodies according to the present invention may be of any type (e.g., IgG, IgE, IgM, IgD, IgA, etc.) or subclass (e.g., IgG1, IgG2, IgG2a, IgG3, IgG4, IgA1, IgA2, etc.).
[0021] Typically, an intact antibody comprises two heavy chains and two light chains linked by disulfide bonds, with each light chain linked to its respective heavy chain by a disulfide bond, forming a "Y" shaped structure. Each heavy chain comprises a heavy chain variable region (VH) and a heavy chain constant region, where the heavy chain variable region comprises three complementarity determining regions (CDRs), such as CDR-H1, CDR-H2 and CDR-H3, and the heavy chain constant region comprises three constant domains, such as CH1, CH2 and CH3. Each light chain comprises a light chain variable region (VL) and a light chain constant region, where the light chain variable region comprises three CDRs, such as CDR-L1, CDR-L2 and CDR-L3, and the light chain constant region comprises one constant domain, CL. In the heavy / light chain variable regions, the CDRs are separated by more conserved framework regions (FR). The variable regions of the heavy / light chains are responsible for recognizing and binding to antigens, whereas the constant regions can mediate the binding of the antibody to host tissues or factors, including various cells of the immune system (e.g. effector cells) and the first component of the classical complement system.
[0022] The precise amino acid sequence boundaries of a given CDR or FR can be readily identified using a number of numbering schemes well known in the art, including, but not limited to, Kabat et al. (1991), "Sequences of Proteins of Immunological Interest," 5th ed. Public Health Service, National Institutes of Health, Bethesda, MD (the "Kabat" numbering scheme); Al-Lazikani et al., (1997) JMB 273, 927-948 (the "Chothia" numbering scheme); MacCallum et al., J. Mol. Biol. 262:732-745 (1996), "Antibody-antigen interactions: Contact analysis and binding sitetopography," J. Mol. Biol. 262, 732-745 (the "Contact" numbering scheme); and Lefranc MP et al., "IMGT unique numbering for immunoglobulin and T cell receptor variable domains and Ig superfamily V-like domains," Dev Comp Immunol, January 2003;27(1):55-77 ("IMGT" numbering scheme); Honegger A and Pluckthun A, "Yet another numbering scheme for immunoglobulin variable domains: an automatic modeling and analysis tool," JMol Biol, June 8, 2001;309(3):657-70 ("Aho" numbering scheme); and Martin et al., "Modeling antibody hypervariable loops: a combined algorithm," PNAS, 1989,86(23):9268-9272 ("AbM" numbering scheme).
[0023] The boundaries of a given CDR or FR may vary depending on the protocol used for identification. For example, the Kabat scheme is based on structural comparisons, while the Chothia scheme is based on structural information. The Kabat and Chothia schemes are based on the length of the sequences of the most common antibody regions, with insertions provided by inserting letters (e.g., "30a") and deletions occurring in some antibodies. The two schemes generate different numbers by placing certain insertions and deletions ("indels") in different positions. The Contact scheme is based on the analysis of complex crystal structures and is in many ways similar to the Chothia numbering scheme. The AbM scheme is a compromise between the Kabat and Chothia definitions and is the scheme used based on Oxford Molecular's AbM antibody modeling software.
[0024] Thus, unless otherwise specified, the "CDR" of a given antibody or a region thereof (e.g., its variable region) should be understood to cover the CDRs defined by any of the above or other known schemes. For example, when a particular CDR (e.g., CDR3) is specified to include a given amino acid sequence, it should be understood that such CDR may have the sequence of the corresponding CDR (e.g., CDR3) defined by any of the above or other known schemes. Similarly, unless otherwise specified, the FR of a given antibody or a region thereof (e.g., its variable region) should be understood to cover the FRs defined by any of the above or other known schemes.
[0025] As used herein, a "humanized" antibody refers to an antibody in which all or essentially all of the CDR amino acid residues are derived from a non-human CDR and all or essentially all of the FR amino acid residues are derived from a human FR. A "humanized form" of a non-human antibody refers to a variant of the non-human antibody that has been humanized to reduce immunogenicity to normal humans while retaining the specificity and affinity of the parent non-human antibody. In some embodiments, some FR residues in a humanized antibody are replaced by the corresponding residues from the non-human antibody (e.g., the antibody from which the CDR residues are derived), e.g., to restore or improve antibody specificity or affinity.
[0026] Humanized antibodies and methods for their preparation are well known to those skilled in the art, see, for example, Almagro and Fransson, Front. Biosci. 13:1619-1633 (2008). Human framework regions available for humanization include, but are not limited to, framework regions selected using the "best-fit" method, framework regions of consensus sequences of specific subgroups of human antibodies derived from light or heavy chain variable regions, human mature (somatically mutated) framework regions, human germline framework regions, and framework regions screened from FR libraries.
[0027] As used herein, the term "antibody fragment" or "antigen-binding portion" includes only a portion of an intact antibody, generally including the antigen-binding site of the intact antibody, thereby retaining the ability to bind to an antigen. Examples of antibody fragments in the present invention include, but are not limited to, Fab, Fab', F(ab')2, Fd fragment, Fd', Fv fragment, scFv, disulfide-bonded Fv (sdFv), the heavy chain variable region (VH) or light chain variable region (VL) of an antibody, linear antibodies, "double bodies" having two antigen-binding sites, single domain antibodies, nanobodies, natural ligands of said antigens or functional fragments thereof, etc. Thus, the "antibody" of the present invention covers the antibody fragments defined above.
[0028] In one embodiment, the CD7 humanized antibody of the present invention is an scFv. The terms "single chain antibody" and "scFv" can be used interchangeably herein and refer to an antibody in which the antibody heavy chain variable region (VH) and light chain variable region (VL) are linked by a linker. The optimal length and / or amino acid composition of the linker can be selected. The length of the linker obviously influences the folding and interaction conditions of the variable regions of the scFv. Indeed, the use of short linkers (e.g., between 5 and 10 amino acids) can prevent intrachain folding. For selection of linker size and composition, see, e.g., Hollinger et al., 1993 Proc Natl Acad. Sci. USA 90:6444-6448, U.S. Patent Application Publication Nos. 2005 / 0100543, 2005 / 0175606, 2007 / 0014794, and PCT Publication Nos. WO2006 / 020258 and WO2007 / 024715, which are incorporated herein by reference in their entireties. An scFv may comprise a VH and a VL linked in any order, e.g., VH-linker-VL or VL-linker-VH.
[0029] In one embodiment, the present invention relates to a CD7 humanized antibody comprising a light chain variable region and a heavy chain variable region, wherein the light chain variable region comprises CDR-L1 set forth in SEQ ID NO: 1, CDR-L2 set forth in SEQ ID NO: 2, and CDR-L3 set forth in SEQ ID NO: 3; the heavy chain variable region comprises CDR-H1 set forth in SEQ ID NO: 4, CDR-H2 set forth in SEQ ID NO: 5, and CDR-H3 set forth in SEQ ID NO: 6; the light chain variable region has at least 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99%, or 100% sequence identity to an amino acid sequence selected from SEQ ID NOs: 8, 11, 14, and 17; and the heavy chain variable region has at least 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99%, or 100% sequence identity to an amino acid sequence set forth in SEQ ID NOs: 9, 12, 15, and 18.
[0030] In one embodiment, the CD7 humanized antibody comprises a light chain variable region selected from SEQ ID NOs: 8, 11, 14 and 17, and a heavy chain variable region selected from SEQ ID NOs: 9, 12, 15 and 18.
[0031] In one embodiment, the amino acid sequences of the CD7 humanized antibodies are set forth in SEQ ID NOs: 10, 13, 16 and 19.
[0032] As used herein, the term "sequence identity" refers to the degree to which two (nucleotide or amino acid) sequences have the same residues at the same positions in a comparison, and is generally expressed as a percentage. Preferably, identity is determined over the entire length of the sequences being compared. Thus, two copies with the exact same sequence have 100% identity. Those skilled in the art recognize that sequence identity can be determined using several algorithms, such as Blast (Altschul et al. (1997) Nucleic Acids Res. 25:3389-3402), Blast2 (Altschul et al. (1990) J. Mol. Biol. 215:403-410), Smith-Waterman (Smith et al. (1981) J. Mol. Biol. 147:195-197) and ClustalW.
[0033] In one aspect, the present invention further provides a multispecific antibody (preferably a bispecific or trispecific antibody) comprising the CD7 humanized antibody described above and further comprising one or more second antibodies that specifically bind to other antigens.
[0034] As used herein, the term "multispecific" means that an antigen binding protein has polytopic specificity (i.e., is capable of specifically binding to two, three or more different epitopes on one biomolecule, or is capable of specifically binding to epitopes on two, three or more different biomolecules). As used herein, the term "bispecific" refers to an antigen binding protein having two different antigen binding specificities.
[0035] In one embodiment, the second antibody may have any antibody or antibody fragment format, such as, for example, a full length antibody, Fab, Fab', (Fab')2, Fv, scFv, scFv-scFv, microantibody, diabody or sdAb.
[0036] Thus, in one embodiment, the second antibody is selected from the group consisting of BCMA, CD4, CD5, CD8, CD14, CD15, CD19, CD20, CD21, CD22, CD23, CD25, CD33, CD37, CD38, CD40, CD40L, CD46, CD52, CD54, CD74, CD80, CD126, CD138, B7, MUC-1, Ia, HM1.24, HLA-DR, tenascin, angiogenic factors, VEGF, PIGF, ED-B fibronectin, IFN-glucose ... bronectin, oncogene, oncogene product, CD66a-d, necrosis antigen, Ii, IL-2, T101, TAC, IL-6, ROR1, TRAIL-R1 (DR4), TRAIL-R2 (DR5), tEGFR, Her2, L1-CAM, mesothelin, CEA, HBs antigen, antifolate receptor, CD24, CD30, CD44, EGFR, EGP-2, EGP-4, EPHa2, ErbB2, ErbB3, ErbB4, ErbB dimer, EGFR vIII, FBP, FCRL5, FCRH5, fetal acetylcholine receptor, GD2, GD3, G protein-coupled receptor type C family 5D (GPRC5D), HMW-MAA, IL-22R-α, IL-13R-α2, kdr, kappa light chain, LewisY, L1-cell adhesion molecule (L1-CAM), melanoma-associated antigen (MAGE)-A1, MAGE-A3, MAGE-A6, preferentially expressed antigen in melanoma (PRAME), survivin, EGP2, EGP40, TAG72, B7-H6, IL-13 receptor a2 (IL-13Ra2), CA9, CD171, G250 / CAIX, HLA-A1, HLA-A2, NY-ESO-1, PSCA, Folate receptor-a, CD44v6, CD44v7 / 8, avb6 integrin, 8H9, NCAM, VEGF receptor, 5T4, fetal AchR, NKG2D ligand, bi-antigen, common marker associated antigen, cancer-testis antigen, MUC1, MUC16, NY-ESO-1, MART-1, gp100, carcinoembryonic antigen, VEGF-R2, carcinoembryonic antigen (CEA), prostate-specific antigen, PSMA, The antibody targets an antigen selected from Her2 / neu, estrogen receptor, progesterone receptor, ephrin B2, CD123, c-Met, GD-2, O-acetylated GD2 (OGD2), CE7, Wilms' tumor 1 (WT-1), cytocyclin, cytocyclin A2, CCL-1, hTERT, MDM2, CYP1B, WT1, Bios, AFP, p53, cytocyclin (D1), CS-1, BAFF-R, TACI, CD56, TIM-3, CD123, L1-cell adhesion molecule, MAGE-A1, MAGEA3, cytocyclin (e.g., cytocyclin A1 (CCNA1)), and / or a pathogen specific antigen, a biotinylated molecule, a molecule expressed by HIV, HCV, HBV and / or other pathogens, and / or a neo-epitope or neo-antigen. Nucleic acids, vectors and host cells
[0037] In another aspect, the present invention relates to a nucleic acid molecule encoding the CD7 humanized antibody or multispecific antibody of the present invention. The nucleic acid of the present invention may be RNA, DNA or cDNA. According to one embodiment of the present invention, the nucleic acid of the present invention is an essentially separate nucleic acid.
[0038] In one embodiment, the nucleic acid molecule encoding the CD7 humanized antibody has at least 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99%, or 100% sequence identity to a nucleotide sequence selected from SEQ ID NOs: 20 to 23, and the encoded CD7 humanized antibody is capable of specifically binding to the CD7 antigen. Preferably, the nucleic acid molecule encoding the CD7 humanized antibody is set forth in SEQ ID NOs: 20 to 23.
[0039] The nucleic acid of the invention may be in the form of a vector, present in a vector and / or part of a vector, for example a plasmid, a sticky-end plasmid or a YAC. The vector is in particular an expression vector, i.e. a vector that allows the expression of the CD7 humanized antibody in vitro and / or in vivo (i.e. in a suitable host cell, host organism and / or expression system). This expression vector usually comprises at least one nucleic acid molecule of the invention, operably linked to one or more suitable expression control elements (e.g. promoters, enhancers, terminators, etc.). The selection of said control elements and their sequences for expression in a particular host is well within the skill of the art. [0044] However, the present invention may also be used with amphibian cells, insect cells, plant cells and any other cells known in the art for expressing heterologous proteins. Chimeric Antigen Receptor
[0045] In another aspect, the present invention further provides a recombinant receptor, such as a recombinant TCR receptor or a chimeric antigen receptor, comprising the above-mentioned CD7 humanized antibody. Preferably, the present invention further provides a chimeric antigen receptor comprising the above-mentioned CD7 humanized antibody.
[0046] As used herein, the term "chimeric antigen receptor" or "CAR" refers to an artificially constructed hybrid peptide that generally contains a ligand-binding domain (e.g., the antigen-binding portion of an antibody), a transmembrane domain, an optional costimulatory domain, and an intracellular signaling domain, with linkers between each domain. CARs can redirect the specificity and reactivity of T cells and other immune cells to selected targets using the antigen-binding properties of antibodies without MHC restriction.
[0047] In one embodiment, the present invention provides a chimeric antigen receptor comprising the above-described CD7 humanized antibody, or a multispecific antibody containing the CD7 humanized antibody, a transmembrane domain, and an intracellular signaling domain.
[0048] As used herein, the term "transmembrane domain" refers to a peptide structure that allows the chimeric antigen receptor to be expressed on the surface of immune cells (e.g., lymphocytes, NK cells, or NKT cells) and guides the cellular response of the immune cell to a target cell. The transmembrane domain may be natural or synthetic and may be derived from any membrane-associated or transmembrane protein. When the chimeric antigen receptor and the target antigen are bound, the transmembrane domain can transduce a signal. In particular, the transmembrane domain applied to the present invention may be derived from, for example, the TCR alpha chain, the TCR beta chain, the TCR gamma chain, the TCR delta chain, the CD3 zeta subunit, the CD3 epsilon subunit, the CD3 gamma subunit, the CD3 delta subunit, CD45, CD4, CD5, CD8 alpha, CD9, CD16, CD22, CD33, CD28, CD37, CD64, CD80, CD86, CD134, CD137, CD154, and functional fragments thereof. Alternatively, the transmembrane domain may be synthetic and may comprise predominantly hydrophobic residues, such as leucine and valine. Preferably, said transmembrane domain is derived from the CD8 alpha chain or CD28 and has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the amino acid sequence shown in SEQ ID NO: 24 or 26, or has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the nucleic acid molecule whose coding sequence is shown in SEQ ID NO: 25 or 27.
[0049] As used herein, the term "intracellular signaling domain" refers to a protein portion that transduces effector function signals and directs a cell to perform a predetermined function. In one embodiment, the intracellular signaling domain contained in the chimeric antigen receptor of the present invention may be any synthetic sequence having the same or similar function as the intracellular domain sequences of T cell receptors and co-receptors that function together to induce signal transduction after antigen receptor binding, and any derivative or variant of these sequences. The intracellular signaling domain may include a number of immunoreceptor tyrosine-based activation motifs (ITAMs). Non-limiting examples of intracellular signaling domains of the present invention include, but are not limited to, intracellular domains such as FcRγ, FcRβ, CD3γ, CD3δ, CD3ε, CD3ζ, CD22, CD79a, CD79b, and CD66d. In a preferred embodiment, the signaling domain of a CAR of the invention may comprise a CD3ζ intracellular domain, having at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the amino acid sequence set forth in SEQ ID NO: 32 or 34, or its coding sequence having at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to a nucleic acid molecule set forth in SEQ ID NO: 33 or 35.
[0050] In one embodiment, the chimeric antigen receptor of the present invention may comprise a hinge region located between the antibody and the transmembrane domain. As used herein, the term "hinge region" generally refers to any oligopeptide or polypeptide that functions to link the transmembrane domain to the antibody. Specifically, the hinge region is intended to provide greater flexibility and accessibility to the antibody. The hinge region may comprise up to 300 amino acids, preferably 10-100 amino acids, and most preferably 25-50 amino acids. The hinge region may be derived in whole or in part from a natural molecule, for example, derived in whole or in part from the extracellular region of CD8, CD4 or CD28, or derived in whole or in part from an antibody constant region. Alternatively, the hinge region may be a synthetic sequence that corresponds to a naturally occurring hinge sequence, or may be a completely synthetic hinge sequence. In preferred embodiments, the hinge region comprises a portion of the hinge region of CD8α, CD28, an FcγRIIIα receptor, IgG4 or IgG1, more preferably a CD8α, CD28 or IgG4 hinge, and has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the amino acid sequence set forth in SEQ ID NO: 40, 42 or 44, or the nucleotide sequence whose coding sequence has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the nucleotide sequence set forth in SEQ ID NO: 41, 43 or 45.
[0051] In one embodiment, the chimeric antigen receptor may include one or more costimulatory domains. The costimulatory domain may be an intracellular functional signaling domain from a costimulatory molecule, including the entire intracellular portion of said costimulatory molecule, or a functional fragment thereof. "Costimulatory molecule" refers to a cognate binding gametophyte that specifically binds to a costimulatory ligand in T cells, thereby mediating a costimulatory response (e.g., proliferation) of T cells. Costimulatory molecules include, but are not limited to, MHC Kreis 1 molecules, BTLA, and Toll ligand receptors. In one embodiment, the 4-1BB costimulatory domain has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the amino acid sequence set forth in SEQ ID NO: 30, or has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the nucleic acid molecule whose coding sequence is set forth in SEQ ID NO: 31.In one embodiment, the CD28 costimulatory domain has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the amino acid sequence set forth in SEQ ID NO:28, or its coding sequence has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the nucleic acid molecule set forth in SEQ ID NO:29.
[0052] In one embodiment, the CAR of the present invention may include a signal peptide so that when expressed in a cell, such as a T cell, the nascent protein is introduced to the endoplasmic reticulum and then to the cell surface. The core of the signal peptide may include a long hydrophobic amino acid segment and tends to form a single α-helix. At the end of the signal peptide, there is generally an amino acid segment that is recognized and cut by a signal peptidase. The signal peptidase is cut during translocation or after completion, generating a free signal peptide and a mature protein. The free signal peptide is then digested by a specific protease. Signal peptides applicable to the present invention are well known by those skilled in the art, such as signal peptides derived from B2M, CD8α, IgG1, GM-CSFRα, etc. In one embodiment, the signal peptide applicable to the present invention is derived from B2M or CD8α and has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the amino acid sequence set forth in SEQ ID NO: 36 or 38, or its coding sequence has at least 70%, preferably at least 80%, more preferably at least 90%, 95%, 97% or 99% or 100% sequence identity to the nucleic acid molecule set forth in SEQ ID NO: 37 or 39.
[0053] In one embodiment, the CAR comprises a CD7 humanized antibody provided herein, or a multispecific antibody comprising said CD7 humanized antibody, a CD8α transmembrane domain, a CD28 or 4-1BB costimulatory domain, and a CD3ζ intracellular signaling domain. In this embodiment, the CAR may further comprise a signal peptide derived from B2M, CD8α, IgG1, or GM-CSFRα.
[0054] The present invention further provides a nucleic acid molecule encoding a CD7-targeting chimeric antigen receptor as defined above, and a vector comprising said nucleic acid molecule.
[0055] As used herein, the term "vector" refers to a mediator nucleic acid molecule for the transfer of (exogenous) genetic material into a host cell, where said nucleic acid molecule may, for example, be copied and / or expressed. Vectors generally include targeting vectors and expression vectors. A "targeting vector" is a vehicle that delivers isolated nucleic acids to the interior of a cell, for example by homologous recombination or by using a hybrid recombinase of sequences at a specific target site. An "expression vector" is a vector that is used for the transcription of heterologous nucleic acid sequences (e.g., those sequences encoding the chimeric antigen receptor peptide of the present invention) in a compatible host cell and the translation of their mRNA. Suitable vectors applicable to the present invention are known in the art and can be obtained commercially in large quantities. In one embodiment, vectors of the invention include, but are not limited to, plasmids, viruses (e.g., retroviruses, lentiviruses, adenoviruses, vaccinia viruses, Rous sarcoma viruses (RSV, polyomaviruses, and adeno-associated viruses (AAV)), phages, phagemids, cosmids, and artificial chromosomes (including BACs and YACs). The vector itself is generally a nucleic acid molecule, generally a DNA sequence that contains an insert (gene transfer) and a larger sequence that is the "skeleton" of the vector. Modified vectors also generally contain an origin of autonomous replication in a host cell (if necessary for stable expression of the polynucleotide), a selection marker, and restriction enzyme cleavage sites (e.g., a multiple cloning site, MCS). Vectors may further include elements such as a promoter, a polyA tail (polyA), a 3'UTR, an enhancer, a terminator, an insulator, an operator, a selection marker, a reporter gene, a targeting sequence, and / or a protein purification tag. In a specific embodiment, the vector is an in vitro transcription vector. Engineered immune cells
[0056] In one aspect, the present invention further provides an engineered immune cell expressing a CAR according to the present invention.
[0057] As used herein, the term "immune cell" refers to any cell of the immune system that has one or more types of effector functions (e.g., killing activity of cytotoxic cells, secretion of cytokines, induction of ADCC and / or CDC). For example, the immune cell is a T cell, a macrophage, a dendritic cell, a monocyte, a NK cell and / or a NKT cell. In one embodiment, the immune cell is derived from a stem cell, such as an adult stem cell, an embryonic stem cell, an umbilical cord blood stem cell, a progenitor cell, a bone marrow stem cell, an induced pluripotent stem cell, a totipotent stem cell, or a hematopoietic stem cell. Preferably, the immune cell is a T cell. The T cell may be any T cell, such as an ex vivo cultured T cell, such as a primitive T cell, or from a T cell in vitro cultured T cell line, such as Jurkat, SupT1, etc., or a T cell obtained from a subject. Examples of subjects include humans, dogs, cats, mice, rats, and transgenic species thereof. T cells may be obtained from multiple sources, including peripheral blood mononuclear cells, bone marrow, lymph node tissue, umbilical cord blood, thymus tissue, tissue from a site of infection, ascites, pleural effusion, spleen tissue, and tumors. T cells may be enriched or purified. T cells may be at any stage of development, including, but not limited to, CD4+ / CD8+ T cells, CD4+ helper T cells (e.g., Th1 and Th2 cells), CD8+ T cells (e.g., cytotoxic T cells), tumor infiltrating cells, memory T cells, naive T cells, γδ-T cells, αβ-T cells, and the like. In a preferred embodiment, the immune cells are human T cells. T cells may be obtained from the blood of a subject using a variety of techniques known to those skilled in the art, such as Ficoll separation.
[0058] Common methods known in the art (e.g., transduction, transfection, transformation, etc.) are employed to introduce the nucleic acid sequence encoding the chimeric antigen receptor into immune cells. "Transfection" is the process of introducing a nucleic acid molecule or polynucleotide (including a vector) into a target cell. One example is RNA transfection, i.e., the process of introducing RNA (e.g., in vitro transcribed RNA, ivtRNA) into a host cell. This term is primarily used for non-viral approaches in eukaryotic cells. The term "transduction" is generally used to describe virus-mediated transfer of a nucleic acid molecule or polynucleotide. Transfection of animal cells generally involves a temporary opening of a pore or "hole" in the cell membrane to allow uptake of the material. Transfection can be achieved using calcium phosphate, electroporation, cell extrusion, or by mixing cationic lipids with materials to generate lipid bodies that fuse to the cell membrane and deposit these carriers inside. Exemplary techniques used for transfection of eukaryotic host cells include lipid vesicle-mediated uptake, heat shock-mediated uptake, calcium phosphate-mediated transfection (calcium phosphate / DNA co-precipitation), microinjection and electroporation. The term "transformation" is intended to describe the transfer of nucleic acid molecules or polynucleotides (including vectors) into bacteria, non-viral, non-animal eukaryotic cells (including plant cells). Thus, transformation is the genetic modification of bacteria or non-animal eukaryotic cells, which is produced by direct uptake from its surroundings by the cell membrane and then incorporating exogenous genetic material (nucleic acid molecules). Transformation may also be achieved by artificial means. For transformation to occur, the cell or bacteria must be in a competent state. For prokaryotic transformation, techniques include heat shock-mediated uptake, fusion of whole cells with bacterial protoplasts, microinjection and electroporation. Once the nucleic acid or vector has been introduced into immune cells, the skilled artisan can amplify and activate the obtained immune cells by common techniques.
[0059] In one embodiment, to reduce the risk of graft-versus-host disease, the engineered immune cells are engineered to express a specific gene that is specific for a particular gene, such as CD52, GR, dCK, TCR / CD3 genes (e.g., TRAC, TRBC, CD3γ, CD3δ, CD3ε, CD3ζ), MHC-related genes (HLA-A, HLA-B, HLA-C, B2M, HLA-DPA, HLA-DQ, HLA-DRA, TAP1, TAP2, LMP2, LMP7, RFX5, RFXAP, RFXANK, CIITA), and / or a specific gene that is specific for a particular gene, such as PD1, LAG3, TIM3, CTLA4, PPP2CA, PPP2CB, PTPN6, PTPN22, PDCD1, HAVCR2, BTLA, CD160, TIGIT, CD96, and immune checkpoint genes such as CRTAM, TNFRSF10B, TNFRSF10A, CASP8, CASP10, CASP3, CASP6, CASP7, FADD, FAS, TGFBRII, TGFRBRI, SMAD2, SMAD3, SMAD4, SMAD10, SKI, SKIL, TGIF1, IL10RA, IL10RB, HMOX2, IL6R, IL6ST, EIF2AK4, CSK, PAG1, SIT, FOXP3, PRDM1, BATF, GUCY1A2, GUCY1A3, GUCY1B2 and GUCY1B3, and the expression of at least one gene selected from these is suppressed or silenced. Preferably, the modified immune cells have suppressed or silenced expression of genes including at least one selected from TRAC, TRBC, HLA-A, HLA-B, HLA-C, B2M, RFX5, RFXAP, RFXANK, CIITA, PD1, LAG3, TIM3, and CTLA4, preferably TRAC, TRBC, HLA-A, HLA-B, HLA-C, B2M, RFX5, RFXAP, RFXANK, and CIITA.
[0060] Methods for suppressing gene expression or silencing genes are well known to those skilled in the art. For example, antisense RNA, RNA decoys, RNA aptamers, siRNA, shRNA / miRNA, transdominant negative proteins (TNPs), chimera / antibody complexes, chemokine ligands, anti-infected cell proteins, intracellular antibodies (sFv), nucleoside analogs (NRTIs), non-nucleoside analogs (NNRTIs), integrase inhibitors (oligonucleotides, dinucleotides and chemical agents) and protease inhibitors can be used to suppress gene expression. Genes can also be silenced by mediating DNA breaks, for example, by a wide range of nucleases, zinc finger nucleases, TALE nucleases or Cas enzymes in the CRISPR system.
[0061] In one embodiment, multiple types of immune cells are provided, with each type of immune cell modified to express one or more types of chimeric antigen receptors. For example, in some embodiments, one type of immune cell is modified to express a chimeric antigen receptor that binds to and / or targets CD7 (including, for example, a CAR of a CD7 humanized antibody described in the present invention), and another type of cell is modified to express a chimeric antigen receptor that binds to and / or targets another antigen. In one embodiment, the immune cells may express a multispecific chimeric antigen receptor that targets one or more types of antigens, including CD7. For example, such a multispecific chimeric antigen receptor may include a multispecific antibody that targets CD7, or may simultaneously include a CD7 humanized antibody described in the present invention and an antibody that targets another antigen. In such an embodiment, the multiple modified immune cells may be administered together or separately. In one embodiment, the multiple types of immune cells may be in the same composition or in different compositions. "Native Fc" refers to a molecule or sequence that is generated by digestion of an intact antibody and includes a non-antigen binding fragment that may be in the form of a monomer or polymer. The source of the immunoglobulin from which the native Fc is generated is preferably of human origin. A native Fc fragment may be composed of monomeric polypeptides that are linked as a dimer or polymer by covalent (e.g., disulfide bonds) and non-covalent bonds. Depending on the category (e.g., IgG, IgA, IgE, IgD, IgM) or subtype (e.g., IgG1, IgG2, IgG3, IgA1, IgGA2), there are 1-4 intermolecular disulfide bonds between monomeric subunits of the native Fc molecule. As an example of a native Fc, papain is used to digest disulfide-bonded dimers generated from IgG (see Ellison et al. (1982), Nucleic Acids Res. 10:4071-9). As used herein, "native Fc" generally refers to monomeric, dimeric and polymeric forms. "Variant Fc" refers to an amino acid sequence that differs from that of a "native" or "wild-type" Fc by at least one document-defined "amino acid modification", also referred to as an "Fc variant". Thus, "Fc" also includes single-chain Fc (scFc), i.e., a single-chain Fc consisting of two Fc monomers linked by a polypeptide linker, capable of naturally folding into a functional dimeric Fc domain. In one embodiment, the Fc is preferably a human immunoglobulin Fc, more preferably a human IgG1 Fc.

[0078] [Figure 1] 1 shows the scFv expression levels of CAR-T cells containing CD7 humanized antibodies. [Diagram 2] This shows the killing effect of CAR-T cells containing a CD7 humanized antibody on target cells. [Diagram 3] 1 shows cytokine release levels after co-culture of target cells with CAR-T cells containing a CD7 humanized antibody. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0079] Example 1. Preparation of CD7 humanized antibody A humanized antibody is prepared based on mouse CD7scFv (clone m189). Clone m189 contains CDR-L1 shown in SEQ ID NO: 1, CDR-L2 shown in SEQ ID NO: 2, CDR-L3 shown in SEQ ID NO: 3, CDR-H1 shown in SEQ ID NO: 4, CDR-H2 shown in SEQ ID NO: 5, and CDR-H3 shown in SEQ ID NO: 6, and its amino acid sequence is shown in SEQ ID NO: 7. A specific method for preparing a humanized antibody is as follows. First, a sequence similarity search is performed using Ig BLAST (http: / / www.ncbi.nlm.nih.gov / igblast / ) and IMGT (Immunity Gene Database IMGT: http: / / www.imgt.org), and an antibody having high sequence similarity in both light and heavy chains from the search results is used as a humanized antibody template. Then, the heavy and light chain CDRs in mouse CD7scFv are grafted into the framework of the humanized antibody template with high sequence similarity, and then back mutation is performed to ensure the affinity and specificity of the humanized antibody. The amino acid sequences of the finally obtained CD7 humanized single-chain antibody (scFv) are shown in Table 1 below.
[0080] Table 1. Sequences of humanized CD7scFv JPEG0007630190000001.jpg33170Example 2. Preparation of CAR-T cells containing CD7 humanized antibodies
[0081] Sequences encoding proteins such as CD8α signal peptide (SEQ ID NO: 38), CD7 humanized scFv (selected from SEQ ID NO: 10, 13, 16, 19), CD8α hinge region (SEQ ID NO: 40), CD8α transmembrane domain (SEQ ID NO: 24), 4-1BB intracellular domain (SEQ ID NO: 30), and CD3ζ intracellular signaling domain (SEQ ID NO: 32) are synthesized and cloned into a pLVX vector (Public Protein / Plasmid Library (PPL), Product No.: PPL00157-4a), and the correct insertion of the target sequence is confirmed by sequencing. Here, the amino acid sequence of the anti-CD7 scFv contained in the hCAR7-1 CAR is shown in SEQ ID NO: 10, the amino acid sequence of the anti-CD7 scFv contained in the hCAR7-2 CAR is shown in SEQ ID NO: 13, the amino acid sequence of the anti-CD7 scFv contained in the hCAR7-3 CAR is shown in SEQ ID NO: 16, and the amino acid sequence of the anti-CD7 scFv contained in the hCAR7-4 CAR is shown in SEQ ID NO: 19.
[0082] Add 3ml of Opti-MEM (Gibco, product number 31985-070) to a sterile tube to dilute the above plasmid, then add the packaging vector psPAX2 (Addgene, product number 12260) and envelope vector pMD2.G (Addgene, product number 12259) in a ratio of plasmid:viral packaging vector:viral envelope vector = 4:2:1. Then add 120ul of X-treme GENE HP DNA transfection reagent (Roche, product number 06366236001), mix evenly immediately, incubate at room temperature for 15 minutes, and then add the plasmid / vector / transfection reagent mixture drop by drop into the culture flask of 293T cells. After 24 hours and 48 hours, collect the virus, combine it, and then ultracentrifuge (25000g, 4℃, 2.5 hours) to obtain concentrated lentivirus.
[0083] Since CD7 is also expressed on T cells, to avoid mutual killing of CAR-T cells, the inventors knock out CD7 in T cells by the CRISP / Cas9 system. After culturing for 1 day, DynaBeads CD3 / CD28 CTS TM (Gibco, product no. 40203D) and subsequently cultured at 37°C and 5% CO2 for 1 day. Concentrated lentivirus is then added and after 3 days of continuous culture, CAR T cells expressing different CD7 humanized scFv are obtained. Unmodified wild-type T cells are used as negative control (NT).
[0084] Using Biotin-SP (long spacer) AffiniPure Goat Anti-Mouse IgG, F(ab`)2 Fragment Specific (min x Hu, Bov, Hrs Sr Prot) (Jackson Immunoresearch, product number 115-065-072) as the primary antibody and APC Streptavidin (BD Pharmingen, product number 554067) or PE Streptavidin (BD Pharmingen, product number 554061) as the secondary antibody, the expression levels of scFv in hCAR7-1 T cells, hCAR7-2 T cells, hCAR7-3 T cells, and hCAR7-4 were detected by flow cytometry, and the results are shown in Figure 1.
[0085] As can be seen, all of the CD7 humanized scFvs in the CAR T cells prepared in the present invention can be effectively expressed. Example 3: Target cell killing effect and cytokine release by CAR T cells 3.1 Killing effect of CAR-T cells on target cells
[0086] When there is killing of target cells by T cells, the number of target cells is reduced. After T cells are co-cultured with target cells having expressible luciferase, the number of target cells is reduced and the secreted luciferase is reduced accordingly. Luciferase can promote the conversion of fluorescein to oxidized fluorescein, during which bioluminescence occurs, and the intensity of such luminescence is determined by the level of luciferase expressed by target cells. Therefore, the detected fluorescence intensity can reflect the killing ability of target cells by T cells.
[0087] To detect the ability of CAR-T cells to kill target cells, we first 4 Jurkat target cells carrying the fluorescein gene in each well were placed in a 96-well plate, and then CAR T cells and NT cells were placed in the 96-well plate at effector:target ratios (i.e., the ratio of effector T cells to target cells) of 1:1, 0.5:1, and 0.25:1 and cultured together. After 16 to 18 hours, the fluorescence value was measured using a microplate reader. The killing efficiency was calculated and obtained based on the formula: (target cell fluorescence average value - sample fluorescence average value) / target cell fluorescence average value x 100%, and the results are shown in Figure 2.
[0088] As can be seen, compared to NT, the CAR T cells of the present invention both have specific killing of target cells. 3.2 Cytokine release from CAR-T cells
[0089] When T cells kill target cells, the target cells are reduced in number and also release cytokines. The release level of cytokine IFNγ when the CAR T cells of the present invention kill target cells is measured by sandwich method (ELISA) according to the following steps. (1) Collection of cell co-culture supernatant
[0090] 1x10 5Place the target cells into a 96-well plate at 1:1 / well, then co-culture CAR T and NT cells (negative control) with the target cells at a 1:1 ratio, and collect the cell co-culture supernatant after 18-24 h. (2) Detection of IFNγ secretion in the supernatant by ELISA
[0091] A 96-well plate was coated with a capture antibody Purified anti-human IFN-γ Antibody (Biolegend, product number 506502) and incubated at 4°C overnight, after which the antibody solution was removed and 250 μL of PBST (1X PBS containing 0.1% Tween) solution containing 2% BSA (sigma, product number V900933-1kg) was added and incubated at 37°C for 2 hours. The plate was then washed three times with 250 μL of PBST (1X PBS containing 0.1% Tween). 50 μL of cell co-culture supernatant or standard was added to each well and incubated at 37°C for 1 hour, after which the plate was washed three times with 250 μL of PBST (1X PBS containing 0.1% Tween). Then, add 50μL of detection antibody Anti-Interferon gamma antibody [MD-1] (Biotin) (abcam, product number ab25017) to each well, incubate at 37℃ for 1 hour, and wash the plate with 250μL of PBST (1XPBS containing 0.1% Tween) three times. Add HRP Streptavidin (Biolegend, product number 405210) and incubate at 37℃ for 30 minutes, discard the supernatant, add 250μL of PBST (1XPBS containing 0.1% Tween) and wash five times. Add 50μL of TMB substrate solution to each well. Allow the reaction to occur at room temperature in a dark place for 30 minutes, and then add 50μL of 1mol / L H2SO4 to each well to stop the reaction. During the 30 minutes that the reaction was stopped, the absorbance at 450 nm was detected using a microplate reader, and the cytokine content was calculated based on a standard curve (drawn based on the readings and concentrations of the standard). The results are shown in FIG. 3.
[0092] As can be seen, the cytokine release levels of the CAR T cells of the present invention are significantly higher than those of the control NT cells.
