Abstract
Description
[Technical field]
[0001] CROSS-REFERENCE TO RELATED APPLICATIONS This application is an international PCT application that claims priority to and the benefit of U.S. Provisional Application No. 62 / 961,035, filed January 14, 2020, U.S. Provisional Application No. 63 / 043,251, filed June 24, 2020, U.S. Provisional Application No. 63 / 092,407, filed October 15, 2020, and U.S. Provisional Application No. 63 / 125,842, filed December 15, 2020, the contents of each of which are incorporated herein by reference in their entirety.
[0002] The present invention relates to vicinal diol ether derivatives of certain polyether polymers, their preparation by reaction with substituted or unsubstituted epoxides, compositions containing them, and methods of their use. Polyether polymer precursors of said derivatives include, for example, polycitronellols, polyprenols, polyisocitronellols, and polyisoprenols. Epoxides include linear and branched alkane and cycloalkane epoxides, including terpene epoxides. [Background technology]
[0003] Liquid polymers have important utility in cosmetic and personal care applications, playing important roles in visual display, rheology, tribology and drug delivery. For example, they may be used as lubricants, emollients, or protective barriers for skin healing and UV protection. Ideally, these materials can be produced by convenient methods, easily derivatized to modify functionality, and preferably produced from safe and sustainable raw materials.
[0004] Citronellol, prenol and isoprenol are all naturally occurring molecules that are commercially available in large quantities. However, these molecules have an underutilized combination of isobutylene and alcohol functionalities that allow them to be polymerized and functionalized.
[0005] This type of chemistry has been largely overlooked in polymer chemistry. One reason for this may be that polymerization is an equilibrium reaction, and that isobutylene alcohol is not always readily available in large quantities. However, in recent years, the production of citronellol has increased rapidly, and one of the largest production routes also uses prenol and isoprenol as intermediates, which has greatly increased its availability.
[0006] WO 2019 / 028053 discloses novel polymers derived from the naturally occurring and commercially available monomers citronellol, prenol and isoprenol. These monomers can be efficiently polymerized in a controlled manner to produce a number of well-characterized polymeric ether alcohols. Furthermore, since the initially formed polymers have primary alcohol functionality, WO 2019 / 028053 further discloses the functionalization of the alcohols to derive ethers, esters and other derivative products. Summary of the Invention [Problem to be solved by the invention]
[0007] In a surprising advance in polymer science, the inventor's previous publications, U.S. Application No. 2017 / 0283553, U.S. Application No. 2017 / 0057940, and WO 2019 / 028053, the contents of each of which are incorporated herein by reference, teach general methods for the preparation of polyether polymers and their derivatives. These polyethers represent an advancement in liquid polymer technology and offer many desirable advantages for commercial applications. Polyether polymers and their simple ether and ester derivatives are utilized as lubricants, emollients, wetting agents, and surfactants.
[0008] The present invention extends the inventors' previous work by providing novel compounds, compositions and methods for the preparation of such polymer derivatives having a variety of physical and chemical properties. These structurally unique vicinal diol fragments can modify the properties of polyether polymers and provide a variety of novel applications.
[0009] The polymer derivatives disclosed herein can be used in cosmetic or specialty chemical formulations, and in some cases can be used as natural substitutes for silicone polymers. These polymers can be formulated for a variety of specialty chemical applications, including personal care compositions, to change and improve product function or application performance. However, the exact functionality of these polymers depends on their size and composition, and thus on their monomer distribution, polydispersity, and further functionalization.
[0010] Preferably, commercially available renewable terpene oxides (epoxides) such as limonene oxide, pinene oxide, styrene oxide, caryophyllene oxide, eugenol oxide, safrooxide, myrcene oxide, carene oxide, menthene oxide, etc. are used in the present invention. Alternatively, epoxides such as ethylene oxide, propylene oxide, styrene oxide and cyclohexane oxide may be used.

[0015] Detailed Description of the Invention Without being bound by theory, isobutylene groups may form ethers with alcohols via an acid-catalyzed mechanism, a chemistry that has been used in other instances to form ether bonds in organic synthesis.
[0016] The equilibrium nature of this reaction can make these ethers difficult to produce on a large scale. However, the inventors have discovered that by recycling monomers, choosing the right catalyst, and using high concentration reaction conditions, these molecules can reach a sufficient degree of polymerization for use in many different applications. Furthermore, these low molecular weight polymers can be further derivatized to reach higher molecular weights and realize new functionalities.

[0019] The compound of formula I has the formula I(a) [ka] [In the formula, n is 0 to 20 (e.g., 0, 1, or 2)] It is understood that the compound of formula (I) is comprised of a polymer backbone which may be formed by a controlled homopolymerization reaction between monomer units to form a compound of formula (I).
[0020] In some embodiments, the compound of formula I(a) is the following compound: [ka] [In the formula, n is 0 to 20 (e.g., 0, 1, or 2)]
[0021] In a subsequent reaction(s), the compound of formula I is reacted with the OH group of the compound of formula I(a) to form a compound of formula A: [ka] The epoxide may be reacted with
[0022] The resulting polymer derivative of formula I will necessarily substantially retain polymer structural features from the corresponding compound of formula I(a), such as the value of n and the identity and structural relationships between the various monomer units of the polymer backbone.
[0023] The polymerization reaction forming the precursor compound of formula I(a) necessarily results in a mixture of products with different values of the integer n, and as a result, the total molecular weight of the polymers also differs. This mixture of polymers can be difficult to purify completely, and it may be desirable to retain the mixture of polymers in order to take advantage of the unique physical and chemical properties that result from any mixture so formed. Thus, partial purification by means such as distillation or chromatography may be preferred to obtain a mixture of related polymers that, as a whole, has certain desirable physical or chemical properties (e.g., boiling point, melting point, viscosity, surface tension, solubility, density, reactivity).

[0025] It is understood that the 2nd and subsequent compositions described herein comprise a mixture of distinct polymers of formula I (and optionally further comprise some other components) that differ in the exact value of the integer n. Thus, the 2nd and subsequent compositions may be understood to be compounds of formula I having different integer n values, for example, a mixture of at least two compounds of formula I having different integer n values (e.g., a mixture of a compound of formula I where n is 0 and a compound of formula I where n is 1). Typically, in such compositions, substantially all of the polymers of formula I in the composition contain the same group R 1 , R 2 , R 3 , R 4 and R 5 i.e., the various polymers of formula I in the composition differ only in the value of the integer n. As used above (and as used herein) the term "substantially all polymers of formula I" refers to polymers having R 1 , R 2 , R 3 , R 4 and / or R 5 It is understood to recognize that there will be minor synthetic impurities present that differ from those of the majority of the composition (e.g., due to minor impurities in starting materials that may be present despite efforts at purification, minor by-products in the synthesis, or minor amounts of unreacted intermediates). [0037] Methods for producing polyethers of formula I(a) are described in U.S. Patent Application Publication No. 2017 / 0283553 and WO 2019 / 028053, the contents of which are incorporated herein by reference in their entirety. Such polymers can generally be produced with high degrees of polymerization in short periods of time under solvent-free, solvent-free conditions using a resin-bound acid catalyst, such as Amberlyst®. Amberlyst-type resins are known in the art and are macroreticular or porous resins covalently bonded to sulfonic or carboxylic acid groups, preferably sulfonic acid groups. Such polymerizations can be carried out at or below room temperature, preferably at slightly elevated temperatures, 30-110°C, or preferably 40-90°C (e.g., about 50°C). Such polymerizations may be carried out in a batch reactor, a semi-batch reactor, or more preferably in a continuous packed bed reactor of the type described in U.S. Provisional Application Nos. 62 / 384,939 and PCT / US2017 / 50808 and U.S. 2019-0210948, which are incorporated herein by reference.
[0038] The reversibility of the polymerization of the claimed compounds stems from the nature of these polymers having adjacent oxygen atoms and tertiary carbon atoms. As a result, under conditions that promote cleavage of the O-C bond, the resulting tertiary carbocation is extremely stable. This allows for the facile elimination of adjacent hydrogen atoms, regenerating the alcohol and alkene functionalities of the starting materials. Such depolymerization can be promoted by mildly acidic (e.g., with Lewis or Bronsted acids) or thermal or enzymatic (by enzymes found in naturally occurring bacteria) conditions.
[0039] This depolymerization property results in biodegradation. This property also allows for the formation of compositions containing compounds that can control the depolymerization of the polymer to allow for slow release of monomeric polymeric components (e.g., citronellol) or shortened polymeric components (e.g., release of citronellol dimers by depolymerization of larger polymers). The invention encompasses solid and / or liquid formulations (e.g., products) containing any of the compounds or compositions described herein that can be released from the composition (e.g., by vaporization at the surface of the composition) if the formulation provides for slow, controlled depolymerization of the polymer and diffusion of the resulting monomers or shortened oligomers. Such formulations can include components that accelerate such depolymerization (e.g., Lewis or Bronsted acids or enzymes) or such formulations can be combined with devices that include electric heating bodies to promote thermal depolymerization. The monomers and / or shortened oligomers (e.g., citronellol or citronellol dimers or trimers) purified in this way are themselves useful for many reasons, such as fragrances, insect repellents, antioxidants, antibacterial agents, or as active pharmaceutical ingredients (e.g., the composition is a pharmaceutical composition).
[0040] The compounds and compositions described herein are particularly suitable for replacing silicones, mineral oils and / or paraffins in cosmetic compositions, such as concealers, primers and / or moisturizers.

[0080] All percentages used herein are by volume unless otherwise specified.
[0081] All ratios used herein are molar ratios unless otherwise specified. EXAMPLES
[0082] Example 1: Continuous citronellol polymerization with cation exchange resin A 6 foot length of 0.25 inch smooth bore stainless steel tubing with 0.01 inch wall thickness was filled with Amberlyst resin, coiled and fitted at one end with PTFE tubing for continuous flow. The coil was heated to 50°C in an oil bath and 300 g of citronellol was pumped through the filled coil at a rate of 2 ml / min. The material emerging from the coil was 1 A high degree of polymerization is reached, as shown by H-NMR analysis. NMR shows a dramatic increase in the number of protons associated with the methylene groups adjacent to the ether oxygen atoms (approximately 3.3 ppm) compared to the protons associated with the methylene groups adjacent to the alcohol oxygen atoms (approximately 3.6 ppm). The integral ratio of the two different proton sets was found to be approximately 1:1.
[0083] Pressurized nitrogen gas was used to evacuate all material from the coil. The recovered material was further distilled under vacuum (0.7 mbar) and warmth (160° C.) to remove monomeric and dimeric species to give a clear, odorless liquid as shown below: [ka]
[0084] Various fractions can be collected, each of which has different values for number average molecular weight, weight average molecular weight, polydispersity and value of n. Such fractions have different physical properties, such as viscosity, refractive index, boiling point and surface tension.
[0085] A fraction is obtained in which the citronellol polymers have an average value of n between 0 and 4. Another fraction is obtained in which the citronellol polymers have an average value of n between 1 and 7.
[0086] Example 2: Polycitronellol-limonene oxide product [ka] A mixture of polycitronellol (141 g) and limonene oxide (34.4 g, 0.75 equiv.) is treated with Amberlyst resin (5%, 7 g) and stirred at room temperature for 2 h. During the treatment, the reaction was monitored by thin layer chromatography (TLC) and 1 The reaction is monitored by H NMR. No change is observed compared to the starting material. The reaction is then heated at 40° C. for another 2 hours. The product is observed in the mixture, but the reaction is not complete. The reaction is heated at 45° C. for another 4 hours. TLC and NMR show the consumption of the starting material (see FIG. 1). The reaction mixture is filtered through Celite to remove the resin. 70 g of reaction mixture is collected. The mixture is distilled under warming and reduced pressure (temperature 123.5° C.-132.3° C., pressure 2.43 mbar-2.51 mbar). The product thus obtained is characterized by NMR and is consistent with the desired product.
[0087] Example 3: Determination of the polydispersity of citronellol polymer derivatives Weigh out a sample of citronellol polymer derivative (e.g., 20-25 mg for a 10 mL volume flask) and dilute to flask volume with solvent (e.g., THF, volatiles). Stir the mixture well until homogenous. For analysis, use isocratic chromatographic conditions and the chromatographic parameters to separate the polymer peak are listed below: Column: Agilent Oligopore GPC, 6 μm, 3.5 mm x 700 mm Diluent: THF, no BHT inhibitor (unstable) Mobile phase: THF, no BHT inhibitor (unstable) Flow rate: 1.0mL / min Column temperature: 25±2℃ Detection: 220nm Injection volume: 30 μL Operating time: 25 minutes
[0088] Calculation: Since there is one double bond in each polymer molecule, which is the only functional group that absorbs light at 220 nm wavelength, the area % values obtained from the chromatograms are equivalent to the mole % values. Therefore, the peak values from the HPLC report data are used in the calculation, and only correction is required for the weight percent calculation. Mass = (theoretical molecular weight) x (area%) Mass%=(mass / total mass)×100 Mn (number average) = area of mAU / molecular weight Mw (weight average) = (area of mAU) x (molecular weight) PDI (Polydispersity Index)=Mw / Mn
[0089] From the HPLC chromatogram, each peak can be characterized by retention time, % area and molecular weight. Mass and mass % can be derived from these values. Number average molecular weight, weight average molecular weight and polydispersity are then calculated from these values.
[0090] A variety of citronellol polymers and citronellol polymer derivatives are tested for polydispersity according to this method. For one polymer, the data indicate that the major component of the citronellol polymer derivative composition is the dimer, present in an estimated amount of 50 weight percent. The remaining major components are the tetramer, pentamer, and hexamer, the total of which is estimated to be 50.36 weight percent. For another polymer, the data indicate that the major component of the citronellol polymer composition is the trimer, present in an estimated amount of about 45-57 weight percent. The remaining major components are the tetramer, pentamer, and hexamer, the total of which is estimated to be 42-46 weight percent, with the remainder being heptamers and octamers.
