Abstract
Description
[Technical field]
[0001] The present invention relates to a cell culture device. [Background technology]
[0002] Medical technology is evolving every day. Unlike the past, when cancer and other diseases were treated with chemical drugs, with the advancement of regenerative medicine, gene therapy and cell therapy are now more commonly used instead of chemical drugs. For example, immune cell therapy in cell therapy uses the patient's own immune cells, which allows cancer cells to be attacked more effectively without destroying the patient's immune system, and causes fewer side effects.
[0003] To perform cell therapy, autologous or allogeneic cells must be cultured and expanded in vitro, and then reinjected into the patient's body. Cell culture is usually performed in a soft culture bag, and new culture medium is gradually injected according to the number of proliferating cells. The culture medium is then placed in a laboratory environment under specified temperature and humidity conditions.
[0004] Among them, in order to check the number of cells, it is necessary to manually extract a small amount of culture medium from the bag many times to calculate the number of cells, and then inject a corresponding amount of culture medium according to the number of cells. In the entire process of cell culture, such steps must be repeated many times, which increases the labor cost and also involves the risk of cell contamination and errors due to manual operation. This leads to a situation where the cost of cell culture is very high, cannot be borne by general patients, and the quality is unstable.
[0005] In conventional cell culture devices, such as WAVE Bioreactor (Cytiva), a culture bag of a certain size is used to culture cells. In addition, since the initial number of cells is small and the amount of culture solution in the culture bag is not large at the beginning of culture, the cells may be excessively dispersed when dispersed throughout the culture bag. It is known that interactions between cells promote cell growth, but if the number of cells per unit area is too small during the culture process, the interactions are reduced, which leads to problems such as a slower cell growth rate or uneven growth conditions, and there is room for improvement. In addition, in order to solve the problem of too few cells per unit area caused by a small initial number of cells, a method of replacing the culture vessel has been adopted in the past, for example, a vessel with a small volume is used at the beginning of culture so that the cells in the vessel are not excessively dispersed, and a vessel with a larger volume is changed as the number of cells increases. However, the above-mentioned method of replacing the culture vessel may increase the risk of cell contamination, and it is also difficult to achieve the purpose of automating the entire cell culture process, so there is room for improvement. [0019] As shown in Figures 1 to 7, the present invention provides an automated cell culture device that mainly includes a main body 10 and a bag clamping part 20, and may further include at least a pulling mechanism, a movement driving mechanism 40, and a push-up mechanism 50.
[0020] The main body 10 surrounds the receiving space and forms a platform 11 on the upper surface. Opposite sides of the platform 11 have slide rail parts 12, respectively. Each slide rail part 12 extends in a sliding direction, and two ends of the platform 11 opposite to each other along the sliding direction are defined as a first end 101 and a second end 102, respectively. The platform 11 is for horizontally placing a culture bag 70 on it. The culture bag 70 is for filling the target cells to be grown and the culture medium, and has at least one inlet 71 for injecting the culture medium. The inlet 71 is located at one end of the culture bag 70 close to the first end 101, and is for connecting to a culture medium supply source, such as a peristaltic motor. Preferably, a ventilation part 60 can be installed on the platform 11, and the surface of the ventilation part 60 has an uneven structure, and the ventilation part 60 is installed between the platform 11 and the culture bag 70 to allow air to pass through the bottom surface of the culture bag 70. For example, in this embodiment, the ventilation element 60 can be a metal net, a perforated board (the surface of the perforated board is a convex structure, and the holes of the perforated board are a concave structure), or other similar structures.
[0021] The two opposing ends of the bag clamping part 20 are slidably installed on the two slide rail parts 12, respectively. The bag clamping part 20 is for holding the culture bag 70 so that the liquid in the culture bag 70 does not flow to the side of the culture bag 70 closer to the second end 102 of the platform 11. For example, the bag clamping part may directly hold the culture bag in the direction of the platform, or may clamp and hold the culture bag in another way to achieve a blocking effect. The bag clamping part 20 can slide on the culture bag 70 along the sliding direction.
[0022] The at least one pulling mechanism is installed at least at the second end 102 of the platform 11 to clamp one end of the culture bag 70 close to the second end 102 of the platform 11. Preferably, the at least one pulling mechanism is further installed at the first end of the platform to clamp one end of the culture bag close to the first end of the platform. Specifically, the at least one pulling mechanism includes at least two clamping parts 30 to clamp opposite sides of one end of the culture bag 70 close to the second end 102 of the platform 11, and the two clamping parts 30 are respectively installed on opposite sides of the side of the main body 10 close to the two slide rail parts 12. When the at least one pulling mechanism is installed at the first end of the platform, it can be installed in the same arrangement. More specifically, in this embodiment, each of the clamping parts 30 includes two magnets 31, 32, the magnet 31 is fixedly installed on the platform 11, and the magnet 32 is magnetically attracted to the magnet 31 fixedly installed on the platform 11 in order to clamp one corner of the culture bag 70 between the two magnets 31, 32. However, each of the clamping parts can have other structures, such as a conventional clamping tool. It is particularly noted that in order to reliably prevent the culture medium from flowing to the second end due to the influence of gravity or capillary action, the height of the two magnets 31 is preferably not lower than the height of the bag clamping parts, and one end of the culture bag corresponding to the second end of the platform is higher than the other position of the culture bag.
[0023] In this embodiment, the bag clamping part 20 includes two horizontal bars 21, two connecting parts 22, and two sliding parts 23. The two horizontal bars 21 are horizontally installed so as to be perpendicular to the sliding direction. The two horizontal bars 21 are installed with a gap between them. The two horizontal bars 21 have different distances from the platform 11, that is, the heights of the two horizontal bars 21 relative to the platform 11 are different. The two opposing ends of each horizontal bar 21 are fixedly installed on the two connecting parts 22, respectively. The two connecting parts 22 are detachably installed on the two sliding parts 23, respectively. The two sliding parts 23 are slidably installed on the two slide rail parts 12, respectively. For example, each of the sliding parts 23 may have a groove for fitting the connecting part 22.
[0024] The moving drive mechanism 40 is installed in the receiving space of the main body 10, and in this embodiment, includes a belt set 41 and two magnet parts 42. The two magnet parts 42 are installed on the belt set 41, and can move along the sliding direction by driving the belt set 41. A magnet part 231 is installed on each of the slide parts 23. The magnet parts 231 of the two slide parts 23 are magnetically attracted to the two magnet parts 42, respectively, so that the belt set 41 drives the two slide parts 23 to move along the sliding direction. This eliminates the need to install a through-type slide rail on the platform, and the receiving space of the main body can be sealed, preventing the internal parts from rusting due to the influence of external temperature and moisture.
[0025] The push-up mechanism 50 is installed in the accommodation space of the main body 10 and is located at the first end 101 of the main body 10. The push-up mechanism 50 can move up and down so as to reciprocate and raise and lower one end of the culture bag 70 close to the first end 101. This allows the culture solution in the culture bag 70 to be gently shaken and ensured to be mixed uniformly. In this embodiment, the push-up mechanism 50 can include a rotating device 51, an eccentric wheel 52, and a push-up plate 53. The rotating device 51 can be a structure such as a rotating shaft that rotates by driving a motor. The eccentric wheel 52 is eccentrically installed on the rotating device 51 and rotates eccentrically together with the rotating device 51. The periphery of the eccentric wheel 52 is intended to abut against the push-up plate 53. The push-up plate 53 is intended to abut against the culture bag 70. When the eccentric wheel 52 rotates eccentrically, the height of the apex of the eccentric wheel 52 keeps changing, so that the push-up plate 53 is pushed by the eccentric wheel 52 and moves up and down continuously in a reciprocating manner. However, the push-up mechanism is not limited to the above mechanism, and other lifting mechanisms may be adopted.
[0026] In actual use, as shown in FIG. 4, one end of the culture bag 70 facing the inlet 71 passes between the two horizontal bars 21 toward the first end 101, then extends over the horizontal bars 21 toward the second end 102, and is fixed to the second end 102 by the two clamping parts 30. With the above structure, the middle part of the culture bag 70 is wrapped around the two horizontal bars 21, and is not only pressed and tightened by the two horizontal bars 21, but also the end of the culture bag 70 corresponding to the second end 102 is higher than the end of the culture bag 70 corresponding to the first end 101, preventing the culture medium from flowing toward the second end 102. It is particularly important to note that in this embodiment, the "automated cell culture device" according to the present invention refers to one in which the bag clamping parts move automatically, but the present invention does not exclude an embodiment in which the bag clamping parts are manually adjusted in position. The end of the culture bag 70 corresponding to the first end 101 may have a fixing part 72. The fixing part 72 is, for example, a sample collection tube of the culture bag 70. The push-up plate 53 can have a corresponding fixing part 531. The fixing part 72 can be arranged on the fixing part 531 so that both ends of the culture bag 70 are respectively arranged on the main body 10. As a result, as shown in FIG. 5, when the bag clamping part 20 moves along the sliding direction, the culture medium on the side close to the injection port 71 in the culture bag 70 is pushed toward the injection port 71, which can prevent the culture medium from dispersing throughout the culture bag 70. Even if the initial cell number is small, the number of cells per unit area in the culture bag is sufficient to promote cell-cell contact. In this embodiment, the fixing part 531 is a fastener that can elastically deform and fasten a tube, and is installed on the push-up plate 53 and used to arrange the fixing part 72 of the culture bag 70. However, if the fixing part 531 is changed to another type of clamping device, such as a spring clip, a magnetic clip, etc., and installed at a location close to the lifting plate 53 or the first end 101 of the main body 10, either can be placed at the other end farther from the culture bag pulling mechanism, and a similar usage effect can be achieved as in this embodiment.
[0027] In the cell culture process of gradually injecting new culture medium into the culture bag 70 and gradually increasing the number of culture medium and cells in the culture bag 70, the bag clamping part 20 can be moved gradually and continuously toward the second end 102, or can be moved in several steps, as shown in Fig. 6, to release the space in the culture bag. The time, speed and distance of the movement can be adjusted and controlled according to the desired number of cells per unit area and the amount of culture medium injected.
[0028] Furthermore, from Figures 5 to 7, it can be seen that in order to achieve a suitable mixing effect, promote uniform cell culture, and obtain excellent efficiency, the pushing-up mechanism 50 can gently rock the culture solution in the culture bag 70 by pushing one end of the culture bag 70 close to the first end 101 and raising and lowering it back and forth.
[0029] Regarding the actual culture effect of the cell culture device provided by the present invention, as shown in FIG. 8, natural killer cells isolated from peripheral blood samples of the same subject were cultured in vitro using three different devices and methods, namely (1) a method using the cell culture device of the present invention, (2) a method using a commercially available cell culture device WAVE Bioreactor, and (3) a method of manually culturing cells (i.e., without using a cell culture device). The peripheral blood sample was whole blood collected from the subject's arm according to a plan approved by the ethical committee. The initial cell number, the medium used, and the number of days of in vitro culture were all the same in the cell cultures performed by the three methods. It can be seen from FIG. 8 that the cell proliferation rate of the culture performed by the cell culture device of the present invention reaches 80% of that of the culture performed manually on the 15th day. This shows that the cell culture device of the present invention can replace the culture performed manually, and effectively reduce the labor cost. 8, the cell culture performed in the cell culture device WAVE Bioreactor, which is an existing commercially available product, achieved a cell proliferation rate of only 26% of the manually performed culture on the 15th day, which is obviously lower than the cell proliferation rate of the culture performed in the cell culture device according to the present invention. This shows that the cell culture device according to the present invention can certainly improve the effects of the conventional cell culture device.
[0030] With the above structure and operation, the present invention can further be combined with a monitoring and controlling mechanism for the culture medium supply source, other temperature, gas, pH value, and humidity. In order to perform the rocking and mixing of the culture medium, the injection frequency, speed, and injection amount of the culture medium or gas are obtained from a calculation model, and the moving time and moving speed of the bag clamping part are set based on this, and the pushing mechanism is driven at a suitable frequency at a suitable time. This allows the entire process to be automatically controlled, eliminating the need for manual injection and rocking, and reliably preventing the culture medium from dispersing, allowing cells to grow and proliferate in a constant state, thus improving the efficiency and yield of cell culture. Most importantly, it can greatly save on human costs and reduce errors caused by manual operation.
[0031] As a result, the cell culture device provided by the present invention can significantly reduce personnel costs under the premise of maintaining the efficiency and quality of cell culture, and can effectively mass-produce cells, thereby reducing the cost of cell culture, which is expensive and burdensome, and thus reducing the cost of cell therapy, thereby benefiting all patients and promoting medical welfare, which is what the medical industry and the general public desire. However, the above examples are merely intended to explain and interpret the technical content of the present invention, and the protection scope of the present invention is limited by the claims.
