Abstract
Description
[Technical Field]
[0001] CROSS-REFERENCE TO RELATED APPLICATIONS This application claims the benefit of priority to U.S. Patent Application No. 17 / 218,797, filed March 31, 2021, entitled "A NEW PRESERVATIVE SYSTEM," and French Patent Application No. 2107903, filed July 22, 2021, the disclosures of which are incorporated by reference as if fully rewritten herein.
[0002] The present disclosure relates to an antiseptic system for keratinous tissue, particularly skin, comprising salicylic acid, benzoic acid or a salt thereof, and a divalent transition metal salt. [Background technology]
[0003] Preservatives are commonly used in personal care products to protect such products from spoilage or spoilage, primarily caused by microorganisms. They typically have antimicrobial activity.
[0004] Because preservatives can cause adverse effects such as allergic reactions and dermatitis, it is desirable to use them in the lowest possible amounts in cosmetics or other personal care products. Therefore, a balance must be achieved by having an effective antimicrobial amount of the preservative or preservatives in the product, yet having as little amount as possible to avoid or reduce the chance of adverse effects. It is also desirable that the preservative or blend of preservatives be effective against the widest possible range of potentially harmful microorganisms that can cause spoilage or spoilage of the personal care product. Furthermore, such preservatives should be chemically and physically compatible with the other ingredients in the personal care product. [Prior art documents] [Patent documents]

However, one of ordinary skill in the art will appreciate that other ranges are within the scope of the present invention. [Example]
[0086] Practice of the present disclosure is provided by the following examples, which serve to clarify aspects of the technology without being limiting in nature.
[0087] Example 1 (Inventive Composition)
[0088] [Table 1]
[0089] Example 2 Stability and Microbial Challenge Evaluation The inventive and comparative compositions were evaluated for stability and microbial challenge testing. Efficacy was evaluated against a broad spectrum of microorganisms following a challenge testing protocol similar to the PCPC gas line.
[0090] result As shown by the results in Table 2, the use of salicylic acid and benzoic acid alone as preservatives was not strong enough to reduce E. faecalis (a gram-positive bacterium) in the formulation. Typical preservatives for the requisite log reduction of gram-positive bacteria include chlorphenesin and phenoxyethanol; however, these are petrochemical-based preservatives that do not meet the needs of highly natural formulations. Alcohol and capryloyl salicylate are also beneficial for preservation, but alcohol-free formulations are desirable, and capryloyl salicylate can cause instability in formulations even at low levels.
[0091] The addition of transition metal 2+ ions, such as copper and zinc, was tested to provide additional microbial protection against gram-positive bacteria. When copper gluconate was added to the composition, a blue-greenish tint was observed. Such a color was undesirable for the formulation. This was addressed by using an alternative transition metal 2+ ion, namely, zinc, which is colorless. Zinc salts are typically used for their anti-acne and anti-seborrheic properties. These properties motivated the inclusion of zinc gluconate and zinc PCA in formulations requiring additional strength against gram-positive bacteria, as bacteria are a common cause of acne. Gluconate salts are also beneficial for hydration as a skin conditioner, which led to the focus on zinc gluconate as a preservative additive for the present disclosure.
[0092] The goal of this disclosure was to provide preservation in non-protective packaging, such as jars, for highly natural formulations without the use of alcohol or petrochemical-based preservatives, and to provide stable formulations for highly sensitive compositions.
[0093] [Table 2]
[0094] [Table 3]
[0095] Stable compositions were demonstrated by Inventive Example 1, Inventive Example 2, Inventive Example 3, and Inventive Example 4. These compositions contained the preservative system described in this disclosure. These Inventive Examples passed microbiological testing as shown in Table 2.
[0096] Different preservative systems were also tested. For example, Comparative Examples 1, 2, 3, and 4 were prepared similarly to the inventive examples, except that one or two of the components of the preservative system were absent. It was observed that when only salicylic acid and benzoic acid were present, the microbiological tests failed (i.e., Comparative Examples 2, 3, and 4). In the case of Comparative Example 1, where the preservative system contained salicylic acid and copper gluconate, the microbiological tests were borderline and therefore unacceptable.
[0097] Other comparative examples were tested without divalent metal salts (i.e., Comparative Example 5, Comparative Example 6, Comparative Example 7, Comparative Example 8, and Comparative Example 9). Although some passed microbiological testing (i.e., Comparative Example 5, Comparative Example 6, Comparative Example 7, and Comparative Example 8), the compositions were not stable and / or exhibited undesirable textures. These results demonstrated the criticality of the preservative system. For example, in Comparative Example 5, the preservative system of the composition contained 0.1% salicylic acid and 0.15% benzoic acid along with 3.5% denatured alcohol. Despite successful microbiological testing, the composition was not stable and emulsion separation was observed.
[0098] In the case of Comparative Examples 10 and 11, the use of capryloyl salicylic acid resulted in an unstable formulation with emulsion separation.
[0099] In the case of Comparative Example 5, the use of denatured alcohol provided non-protective packaging that passed microbial testing, but resulted in undesirable aesthetics by having a glossy surface that appeared to lead to emulsion separation after 2 weeks at 45°C.
[0100] In the cases of Comparative Examples 6, 7, and 8, the use of chlorphenesin (a known non-natural preservative) provided non-protective packaging pass the microbial challenge test, but the use of this material was undesirable due to the high natural content of the formulation and loss of mousse-like texture and glossy surface appearance after two weeks of stability. Furthermore, chlorphenesin is also a petrochemical-derived preservative that was undesirable for the formulation.
[0101] In the case of Comparative Example 9, the use of the non-natural phenoxyethanol resulted in an unstable formulation due to emulsion separation.
[0102] As shown by the results in Table 2, the examples of the present invention demonstrated the microbial preservation of Gram-positive bacteria by an antiseptic system composed of salicylic acid, benzoic acid, and an association of transition metal 2+ ions such as copper and zinc.
[0103] While the present disclosure has been described in terms of the described embodiments, it will be understood by those skilled in the art that various changes may be made and equivalents may be substituted for the elements thereof without departing from the scope of the disclosure. In addition, many modifications may be made to adapt a particular situation or material to the teachings of the disclosure without departing from its essential scope. Therefore, the disclosure is not limited to the particular embodiment disclosed as the best mode contemplated for carrying out the disclosure, but the disclosure is intended to include all embodiments falling within the scope of the appended claims.
[0104] The articles "a" and "an," as used herein, mean one or more when applied to any feature in the embodiments of the present disclosure described herein and in the claims. The use of "a" and "an" does not limit the meaning to just one feature unless such a limitation is specifically stated. The article "the" preceding a singular or plural noun or noun phrase refers to the particular specified feature (singular) or the particular specified features (plural) and may have singular or plural connotations depending on the context in which it occurs.
[0105] "At least one," as used herein, means one or more and thus includes individual components as well as mixtures / combinations.
[0106] The transitional terms "comprising," "consisting essentially of," and "consisting of," when used in the appended claims, in their original and amended forms, delineate the scope of the claim in terms of which additional, unrecited claim elements or steps, if any, are excluded from the scope of the claim. The term "comprising" is intended to be inclusive or open-ended and does not exclude any additional, unrecited elements, methods, steps, or materials. The term "consisting of" excludes any elements, steps, or materials other than those specified in the claim and, in the latter case, impurities normally associated with the specified material. The term "consisting essentially of" limits the scope of the claim to those elements, steps, or materials specified and that do not materially affect the basic and novel characteristics of the claimed disclosure. All materials and methods described herein embodying the present disclosure may, in alternative embodiments, be more specifically defined by any of the transitional terms "comprising," "consisting essentially of," and "consisting of."
[0107] The terms "free" and "lacking" indicate that the excluded material is not present in the composition in a measurable amount, typically 0% by weight based on the total weight of the composition. The term "essentially free" indicates that the excluded material is preferably not present in the composition, but it is possible for the composition of the present invention to have very small amounts of the excluded material, provided that these amounts do not substantially affect the advantageous properties of the composition. In particular, "essentially free" means that the excluded material can be present in the composition in an amount of less than about 0.1% by weight based on the total weight of the composition.
[0108] Other than in the examples, or where specifically indicated, all numbers expressing amounts of ingredients and / or reaction conditions should be understood as being modified in all instances by the term "about," meaning within 10% of the indicated number (e.g., "about 10%" means 9% to 11%, "about 2%" means 1.8% to 2.2%).
[0109] All percentages and proportions are calculated by mass unless otherwise specified. All percentages are calculated based on the total composition unless otherwise specified.Generally, unless otherwise clearly stated, "mass" or "amount" as used herein in relation to the percentage amount of an ingredient refers to the amount of raw material containing the ingredient, where raw material may be described herein as containing less than 100% active of the ingredient. Therefore, the mass percentage of the active in a composition is expressed as the amount of raw material containing the active used, and may or may not reflect the final percentage of the active, where the final percentage of the active depends on the mass percentage of the active in the raw material.
[0110] The terms "weight percent" and "weight %" may be used interchangeably and refer to percent by weight, based on the total weight of a composition, article, or material, except as may be specified, for example, with respect to a component phase or system of the composition, article, or material. Ranges and amounts given herein are intended to include subranges and amounts using any disclosed endpoints. Thus, a range of "1% to 10%, such as 3% to 5%, such as 2% to 8%" is intended to encompass ranges such as "1% to 8%, 1% to 5%, 2% to 10%, etc. All numbers, amounts, ranges, etc. are intended to be modified by the term "about," whether or not expressly stated as such. Similarly, a given range of "about 1% to 10%" is intended to have the term "about" modify both 1% and 10%. Furthermore, when an amount of a component is given, it is intended to indicate the amount of active material, unless otherwise specified.
[0111] Notwithstanding that the numerical ranges and parameters setting forth the broad scope of the present disclosure are approximations, unless otherwise indicated, the numerical values set forth in the specific examples are reported as precisely as possible. However, any numerical value inherently contains certain errors necessarily resulting from the standard deviation found in their respective testing measurements. The following examples serve to illustrate embodiments of the present disclosure, but are not intended to be limiting in nature.
