Abstract

Some aspects of the present disclosure provide strategies, systems, reagents, methods, and kits that are useful for targeted editing of nucleic acids, including editing a single site in the genome of a cell or subject, for example, in the human genome. [Solution] In some embodiments, fusion proteins between nucleic acid-programmable DNA-binding proteins (napDNAbp), such as Cpf1 or variants thereof, and a nucleic acid editing protein or protein domain, such as a deaminase domain, are provided. In some embodiments, methods for targeted nucleic acid editing are provided. In some embodiments, reagents and kits are provided for creating fusion proteins between targeted nucleic acid editing proteins, such as napDNAbp (e.g., CasX, CasY, Cpf1, C2c1, C2c2, C2c3, and Argonaute), and a nucleic acid editing protein or domain.
