Abstract

The utility model discloses a method for using nano-enhanced loop-mediated isothermal amplification-recombinase polymerase amplification (LAMP-RPA) biosensor kit for rapid cage-side detection of Salmonella species in poultry farms. The method involves processing fecal sample collected from chickens, rehydrating the assay components with Silver nano particles (AgNPs) solution, activating the assay with Magnesium acetate (Mg(OAc)2). Interpreting results is achieved by a naked eye observation of change in color of the mixture. A positive result is indicated by the retention of the original dark gray to dark violet color of the mixture, otherwise, it is signaled by the appearance of a colorless to yellow mixture. This was done in a single-tube reaction, enabling a rapid, equipment-free, and userfriendly nucleic acid-based diagnostic platform ideal for on-site deployment. Diagnostic validation of specificity of the biosensor kit exhibited good results. Moreover, it can detect down to 64 picogram of DNA per reaction or a 10?³ dilution.
