Description

Nonviral dual plasmid vector capable of high-efficiency gene expression and stem cells and differentiated hematopoietic cells expressing the same

[0001] The present invention relates to a non-viral dual plasmid vector capable of highly efficient gene expression and stem cells and differentiated hematopoietic cells expressing the same.

[0002] This invention claims the benefit of priority from Korean Patent Application No. 10-2023-0103184, filed August 7, 2023, and Korean Patent Application No. 10-2024-0104051, filed August 5, 2024, the entire contents of which are incorporated herein by reference.

[0003]

[0004] CAR-T (Chimeric antigen receptor T cells; also referred to as “CAR-T cells”) cancer treatment is a cancer treatment that isolates T cells from the human body, manufactures them to directly recognize antigens on the surface of cancer cells and attack them, and administers the engineered cells to patients. CAR-T cell therapy has shown a groundbreaking effect in patients with blood cancers, including leukemia and lymphoma, who are no longer responding to existing anticancer drugs, and six products have been approved by 2023. Based on these effects, it is being developed as a treatment for various intractable solid cancers for which there are no alternatives.

[0005] Meanwhile, natural killer cells (NK cells), among the cells that make up the immune system, recognize abnormally altered cell membranes, such as virus-infected cells or cancer cells. NK cells play a crucial role in the early stages of viral infection or tumor formation, before activated cytotoxic T lymphocytes are produced in large numbers. Histologically, NK cells are large granular lymphocytes. Intracellular granules contain molecules with pre-formed, potent biological functions that are secreted when NK cells come into contact with target cells. Some of these molecules form pores in the target cell membrane, lysing the cell or invading the target cell, increasing nuclear DNA fragmentation and inducing apoptosis or programmed cell death.

[0006] Natural killer (NK) cells are known to have the ability to nonspecifically kill cancer cells. This killing ability is being utilized in the treatment of solid tumors using lymphokine-activated killer cells (LAK) and tumor-infiltrating lymphocytes (TILs), and in immunotherapy using donor lymphocyte infusions to prevent rejection during bone marrow or organ transplantation, a novel cell therapy approach.

[0007] In addition, defects in the differentiation and activity of NK cells have been reported to be associated with various cancers, including breast cancer, melanoma, and lung cancer, and NK cell therapy is emerging to treat these diseases.

[0008] Effective use of NK cells as anticancer immunotherapy requires securing a large number of NK cells. However, NK cells account for 10-15% of lymphocytes in the blood, and cancer patients often have reduced numbers, differentiation, and function, making securing sufficient numbers of NK cells challenging. Therefore, mass production of NK cells through proliferation or differentiation is required for their application as NK cell therapy.

[0009] Accordingly, the inventors of the present invention attempted to utilize the vector containing the CAR gene in stem cells such as induced pluripotent stem cells and differentiate them into hematopoietic cells such as T cells, hematopoietic stem cells, or NK cells that express the CAR, thereby preventing or treating cancer.

[0010]

[0011] The present inventors produced a dual plasmid vector that co-expresses a CAR gene and a CD24 or IL-15 gene, and introduced this into induced pluripotent stem cells to differentiate them into T cells, hematopoietic stem cells, or NK cells. They confirmed that cells with high expression of CAR, CD24, or IL-15 exhibited high cytotoxicity toward cancer cells, and completed the present invention based on this.

[0012] Accordingly, the purpose of the present invention is to provide a chimeric antigen receptor (CAR); and

[0013] A non-viral dual plasmid vector comprising one or more genes selected from the group consisting of CD24 and IL-15 is provided.

[0014] Another object of the present invention is to provide a cell transfected with the non-viral dual plasmid vector.

[0015] Another object of the present invention is to provide hematopoietic cells differentiated from stem cells transfected with a non-viral dual plasmid vector.

[0016] Another object of the present invention is to provide a composition for differentiating stem cells into hematopoietic cells, comprising a non-viral dual plasmid vector.

[0017] Another object of the present invention is to provide a kit for differentiating stem cells into hematopoietic cells, comprising a non-viral dual plasmid vector and a differentiation medium.

[0018] Another object of the present invention is to provide a method for producing a stem cell comprising the steps of: transfecting a non-viral dual plasmid vector into an isolated stem cell; and

[0087]

[0088] Hereinafter, the present invention will be described in detail.

[0089]

[0090] The present invention relates to a chimeric antigen receptor (CAR); and

[0091] A non-viral dual plasmid vector comprising one or more genes selected from the group consisting of CD24 and IL-15 is provided.

[0092] In the present invention, "chimeric antigen receptor (CAR)" means a synthetic receptor capable of targeting a specific antigen, and the CAR according to the present invention may target the human CD19 antigen.

[0093] In the present invention, the CAR may be #11, 13, 14, 15, or 16, and each of these may be encoded by a gene consisting of or including a base sequence having a sequence homology of 70% or more, 75% or more, 80% or more, 85% or more, 90% or more, 95% or more, 98% or more, 70 to 100%, 80 to 100%, 90 to 100%, 95 to 100%, 96 to 100%, 97 to 100%, 98 to 100%, 99 to 100%, or 100% with SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, or SEQ ID NO: 5, in that order, but is not limited thereto.

[0094] In the present invention, the CAR #11 is CD8sp-V L -(GGGGS)3-V H - It can have the structure of CD8h-CD8TM-41BB-CD3z (14aa: K), and CAR #13 is CD8sp-V L -(GGGGS)3-V H- It can have the structure of CD8h-CD8TM-41BB-CD3z (14aa: Q), and CAR #14 is CD8sp-V H -(GGGGS)3-V L - It can have the structure of CD8h-CD8TM-41BB-CD3z (14aa: K), and CAR #15 is CSF2RAsp-V L -whitlow linker-V H - It can have the structure of CD28h-CD28TM-CD28cyto-CD3z, and CAR #16 is GMCSFRsp-V L -whitlow linker-V H -It may have a structure of IgG4h-CD28TM-41BB-CD3z. In each CAR, each configuration may be encoded by a gene consisting of or including the base sequence shown in Table 2, but is not limited thereto.

[0095] In the present invention, the CAR may include, but is not limited to, one or more selected from the group consisting of a signal peptide, an antigen-binding domain, a hinge region, a transmembrane domain (TM), and an intracellular signaling domain (cytoplasmic domain or cytoplasmic signaling domain).

[0096] In the present invention, a "signal peptide" may be located in front of the antigen binding domain, and in one embodiment of the present invention, the signal peptide may be CD8sp, CSF2RAsp, or GMCSFRsp. In the present invention, the CD8sp may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 9, CSF2Rasp may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 17, and GMCSFRsp may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 23, but is not limited thereto.

[0097] In the present invention, "antigen-binding domain" refers to an antibody, protein, or peptide domain capable of specifically recognizing and binding to a target antigen. In the present invention, "antigen" refers to a polypeptide, compound, or other substance capable of specifically binding to a humoral immune mediator such as an antibody or a cellular immune mediator such as a T cell receptor. In the present invention, the target antigen may be the human CD19 antigen.

[0098] The antigen binding domain may be an antigen-specific antibody, an antigen-specific protein, an antigen-specific peptide, or a fragment of an antibody (preferably, an antigen-binding fragment).

[0099] In the present invention, "antibody" means an immunoglobulin molecule that immunologically reacts with a specific antigen (epitope), and includes polyclonal antibodies, monoclonal antibodies, and functional fragments thereof. In addition, the term may include forms produced by genetic engineering, such as chimeric antibodies (e.g., humanized murine antibodies) and heterologous antibodies (e.g., bispecific antibodies). The antibody comprises a variable region of a heavy chain and / or a light chain (V H , heavy chain variable region; V L , light chain variable region). The variable region includes a portion forming an antigen-binding site of an antibody molecule as a primary structure, and the antibody of the present invention may be composed of a fragment including the variable region. A linker may be positioned between the light chain and heavy chain variable regions of the antibody, protein, peptide, or antigen-binding fragment of the antibody. The "linker" refers to a polypeptide that connects the light chain variable region and the heavy chain variable region to each other without damaging their original antigen-binding properties.

[0100] In one embodiment of the present invention, the antigen binding domain is V L -(GGGGS)3-V H , V H -(GGGGS)3-V L , or V L -whitlow linker-V H It may include. In the present invention, the V L -(GGGGS)3-V H may be encoded by a gene consisting of or including the base sequence of sequence number 10, and V H -(GGGGS)3-V Lmay be encoded by a gene consisting of or including the base sequence of sequence number 16, and V L -whitlow linker-V H may be encoded by a gene consisting of or including the base sequence of sequence number 18 or 24, but is not limited thereto.

[0101] In the present invention, the "hinge region" refers to a region located between the antigen binding domain and the transmembrane domain and functions as a flexible linker. The hinge region allows the antigen binding domain to be appropriately positioned when bound to an antigen, thereby forming a stable binding with the antigen. For example, the hinge region has the purpose of extending the antigen binding domain from the cell membrane of a cell expressing a CAR. In one embodiment of the present invention, the hinge region may include a hinge region of CD8 (CD8h), CD28 (CD28h), or IgG4 (IgG4h). In the present invention, the CD8h may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 11, CD28h may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 19, and IgG4h may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 25, but is not limited thereto.

[0102] In the present invention, a "transmembrane domain (TM)" refers to any polypeptide or oligopeptide that functions to cross a cell membrane and connect extracellular and intracellular signaling domains. The transmembrane domain can penetrate the cell membrane so that the antigen binding domain of the chimeric antigen receptor can be located on the cell surface (outside the cell) and the intracellular signaling domain can be located within the cell. That is, the transmembrane domain serves as a support for the chimeric antigen receptor while simultaneously connecting the antigen binding domain (or hinge domain) and the intracellular signaling domain. In one embodiment of the present invention, the transmembrane domain may include a transmembrane domain of CD8 (CD8TM) or CD28 (CD28TM). In the present invention, the CD8TM may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 12, and CD28TM may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 20 or 26, but is not limited thereto.

[0103] In the present invention, the "intracellular signaling domain (cytoplasmic domain or cytoplasmic signaling domain)" refers to a polypeptide or oligopeptide located inside the cell membrane. The intracellular signaling domain receives a signal transmitted by the antigen binding domain and transmits the signal to the inside of a cell expressing the CAR. The signal is transmitted inside the cell and causes activation or inhibition of a biological process. The intracellular signaling domain is not particularly limited in type as long as it transmits a signal capable of inducing cell activation when the antigen binding domain binds to an antigen. In the present invention, the intracellular signaling domain may include one or more intracellular co-stimulatory domains. The co-stimulatory domain is located in the extracellular or intracellular portion of the CAR and serves to transmit a signal to a cell expressing the CAR. That is, the co-stimulatory domain contributes to inducing a sufficient response (activation) of a T cell following binding of a target antigen. The co-stimulatory domain may be selected from, but is not limited to, domains derived from CD27, CD28, 41BB, OX40, CD30, CD40, PD-1, ICOS, LFA-1 (lymphocyte function-associated antigen-1), CD2, CD7, LIGHT, NKG2C, and / or B7-H3. In one embodiment of the present invention, the intracellular signaling domain may comprise one or more intracellular signaling domains selected from the group consisting of 41BB, CD28cyto, and CD3ζ.In the present invention, the 41BB may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 13, CD28cyto may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 21, and CD3z may be encoded by a gene consisting of or including the base sequence of SEQ ID NO: 14, 15, 22, or 27, but is not limited thereto.

[0104] In the present invention, the genes encoding the components included in the CAR may be composed of or include a base sequence having a sequence homology of 70% or more, 75% or more, 80% or more, 85% or more, 90% or more, 95% or more, 98% or more, 70 to 100%, 80 to 100%, 90 to 100%, 95 to 100%, 96 to 100%, 97 to 100%, 98 to 100%, 99 to 100%, or 100% to the base sequence indicated by the corresponding sequence number, but are not limited thereto.

[0105] In the present invention, “dual plasmid vector” means a vector manufactured by inserting two or more genes into one plasmid.

[0106] In the present invention, a "vector" refers to any medium for cloning and / or transferring nucleotides into a host cell. A vector may be a replicating unit (replicon) capable of binding to another DNA fragment and causing replication of the bound fragment. A "replicating unit" refers to any genetic unit (e.g., a plasmid, phage, cosmid, chromosome, virus) that functions as an autonomous unit of DNA replication in vivo, i.e., is capable of replicating under its own control. The term "vector" encompasses viral and non-viral vectors for introducing nucleotides into a host cell in vitro, ex vivo, or in vivo.

[0107] In the present invention, the vector may include a #11 vector comprising the CAR #11 and CD24 genes and consisting of or including the base sequence of SEQ ID NO: 31, a #13 vector comprising the CAR #13 and CD24 genes and consisting of or including the base sequence of SEQ ID NO: 32, a #14 vector comprising the CAR #14 and CD24 genes and consisting of or including the base sequence of SEQ ID NO: 33, a #15 vector comprising the CAR #15 and CD24 genes and consisting of or including the base sequence of SEQ ID NO: 34, a #16 vector comprising the CAR #16 and CD24 genes and consisting of or including the base sequence of SEQ ID NO: 35, and a #22 vector comprising the CAR #13, CD24, and IL-15 genes and consisting of or including the base sequence of SEQ ID NO: 36.

[0108] In the present invention, "CD24" is a sialoglycoprotein expressed on the surface of most B lymphocytes and differentiating neuroblasts, and may be a "Don't eat me" signal gene. It has been reported that CD24 acts as an immune checkpoint molecule that avoids phagocytosis by interacting with sialic acid-binding Ig-like lectin 10 (Siglec-10), a receptor expressed on tumor-associated macrophages (TAMs). If the HLA gene is knocked out to eliminate immune rejection, the cell will be attacked by innate immune cells. Then, 1 mL of DPBS was added per well of a 6-well plate and washed. 1 ml of TrypLE reagent was added and incubated for 15 minutes at 37°C, 5% CO2 conditions. After confirming that the cells had detached, 1 ml of the reacted TrypLE reagent was added to a 15 ml tube containing 8 ml of complete medium. The medium was added once more to the existing 15 ml tube (8 ml of complete medium + TrypLE reagent), and the cells were collected again and placed into the 15 ml tube. Then, this was centrifuged at 300 g for 5 minutes, and the medium was removed after leaving only the cells. Afterwards, 3 ml of medium + Y-27632 was added, and cells were counted and seeded.

[0266]

[0267] Example 3. Introduction of dual plasmids into human induced pluripotent stem cells.

[0272]

[0273] 3-2. Harvesting human induced pluripotent stem cells

[0274] The proliferation complete medium prepared in Example 3-1 above was prepared by warming it at room temperature for more than 30 minutes.

[0275] To harvest human induced pluripotent stem cells, the culture medium of the plate containing the human induced pluripotent stem cells cultured in Example 2 was removed, and 1 mL of DPBS was added per well of a 6-well plate and washed. 1 ml of TrypLE reagent was added and incubated for 15 minutes at 37°C, 5% CO2 conditions. After confirming that the cells had detached, 1 ml of the reacted TrypLE reagent was added to a 15 ml tube containing 8 ml of complete medium, medium was added once more, and the cells were collected again and placed in the 15 ml tube. Then, this was centrifuged at 300 g for 5 minutes, and the medium was removed after leaving only the cells. Afterwards, 3 ml of medium + Y-27632 was added, and the cells were counted.

[0276]

[0277] 3-3. Electroporation

[0278] 4 ml of E buffer was added to the Neon electroporation kit tube and then mounted on the machine.

[0279] 5x10 human induced pluripotent stem cells harvested in Example 3-2 above 5 After placing the cells in an EP tube, centrifugation was performed at 300 g for 5 minutes, washing was performed with 1 ml of DPBS, and centrifugation was performed again at 300 g for 5 minutes. [0280] Then, 5 μg of the dual plasmid DNA prepared in Example 1 was added to the tube containing the cells, and resuspended in 100 μl of R buffer from the Neon electroporation kit.

[0281] After inserting a 100 μl tip into the pipette of the Neon electroporation kit, the cells and DNA in the R buffer were added, placed into a tube containing E buffer, and then transfected by electrical stimulation.

[0282] Afterwards, the seeds were seeded in the 100 mm dish coated in Example 3-1 and cultured for 3 days in the same manner as in Example 2-2-1.

[0283]

[0284] 3-4. Selection of human induced pluripotent stem cells with dual plasmids

[0285] Human induced pluripotent stem cells having dual plasmids transfected by the method of Example 3-3 above were selected through the following process, and this process is schematically shown in Figure 2a.

[0286] Day 0: Selective medium 1 was made by adding 100 μg / ml G418 (50 μg / ml Geneticin) to StemFit Basic04 medium. In Example 3-3, the growth medium (Stemfit Basic04) was suctioned out of the culture medium cultured in a 100 mm dish after transfection into the induced pluripotent stem cells, and 10 ml of selective medium 1 was added and cultured. After 24 hours, selective medium 1 was removed and replaced with selective medium 1, and this process was repeated until day 3.

[0287] Day 3: Selective medium 2 was prepared by adding 200 μg / ml G418 (50 μg / ml geneticin) to StemFit Basic04 medium. Selective medium 1 was removed and replaced with 10 ml of Selective medium 2. After 24 hours, the process of removing Selective medium 2 and replacing it with Selective medium 2 was repeated until day 6.

[0288] Day 6: Selective medium 3 was made by adding 400 μg / ml G418 (50 μg / ml geneticin) to StemFit Basic04 medium. Selective medium 2 was removed and replaced with 10 ml of selective medium 3. After 24 hours, selective medium 3 was removed and replaced with selective medium 3. This process was repeated until day 9-10.

[0289] Day 9~10: 4.8 μl of iMatrix-511 (175 μg) was added to each well of a 12-well plate and coated in a coating incubator for 1 hour. Then, the DPBS in the 12-well plate was removed, and 500 μl of StemFit Basic04 medium containing 200 μg / ml G418 and 10 mM Y-27632 was added.

[0290] The tip of a Pasteur pipette was heated with an alcohol lamp to create a ring shape, and then cultured repeatedly by replacing it with selective medium 3 until Day 9-10. One colony of human induced pluripotent stem cells selected from the culture (Day 6 to Day 9-10) was broken into clumps and transferred to a 12-month plate coated with the medium using a 20 μl pipette. Then, general culture was performed for 7 days using selective medium 3 in the same manner as in Example 2-2-1.

[0291] Day 16~17: 10 mM Y-27632 was dissolved at room temperature and added 1000x to the complete medium warmed at room temperature, and 400 μg / ml G418 was added.

[0292] Afterwards, the culture medium of the plate containing the cells to be subcultured (cells cultured normally from Day 9-10 to Day 16-17) was removed. Then, 1 mL of DPBS was added per well of a 6-well plate and washed. 1 ml of TrypLE reagent was added and cultured for 15 minutes at 37°C, 5% CO2 conditions. After confirming that the cells had detached, 1 ml of the reacted TrypLE reagent was added to a 15 ml tube containing 8 ml of complete medium, the medium was added once more, and the cells were collected again and placed in a 15 ml tube. Then, it was centrifuged at 300 g for 5 minutes, and the medium was removed after leaving only the cells. After that, 3 ml of selective medium 3 + Y-27632 was added and the cells were counted, and 3 x 10 4 Cells were seeded at 2.5 ml / well and cultured normally using selective medium 3 for 7 days.

[0293] Day 23~24: Selection was completed and cells were harvested for cell analysis.

[0294]

[0295] 3-5. Verification of human induced pluripotent stem cells with dual plasmids using flow cytometry (FACS)

[0296] After harvesting the cells selected through the process of Example 3-4 above, they were centrifuged at 300 g for 5 minutes. Then, the medium was removed, 10 ml of FACS buffer was added, and washed by centrifugation at 300 g for 5 minutes, and 1x10 5 Cells were added. Then, 1 μl of each antibody and isotype was added to the tube and incubated on ice for 1 hour. The antibodies and isotypes used were as follows: PE-SSEA-4 (Cat No. 560128), isotype: PE-IgG3k (Cat No. 556659), APC-Labeled Human CD19 (20-291) Protein (Cat No. CD9-HA2H5).

[0297] Afterwards, 1 ml of FACS buffer was added and centrifuged at 300 g for 5 minutes to wash, and the process of adding 1 ml of FACS buffer and centrifuging at 300 g for 5 minutes was repeated 3 times. Then, measurements were made using a FACS device.

[0298]

[0299] Four vectors (#11, 13, 15, 16) containing CARs #11, 13, 15, and 16 were introduced into iPSCs by electroporation, G418 selection was performed, and colonies were picked. As shown in Figures 2b to 2i, it was confirmed that the CAR expression level was higher than before G418 selection.

[0300] In Figure 2b, #11-3 refers to the 3rd iPSC colony picked after introducing the CAR #11 vector into iPSC, and the numerical value refers to the % value when CAR was detected by FACS.

[0301] In addition, as a result of introducing a vector (#22) containing CAR #13 and CD24-IL-15 genes into iPSCs, performing G418 selection, and then picking colonies, it was confirmed that iPSCs introducing #22 also showed a higher CAR expression level of 90% compared to before G418 selection (50%), as shown in Figures 2j and 2k.

[0302] The above results showed that the dual plasmid of the present invention was introduced into human induced pluripotent stem cells using electroporation.

[0303]

[0304] 3-6. Verification of human induced pluripotent stem cells with dual plasmids using ELISA

[0305] After harvesting the cells selected through the process of Examples 3-4 above, they were centrifuged at 300 g for 5 minutes. Then, the medium was removed, and ELISA was performed according to the manufacturer's instructions for the ELISA kit (Cat No. D1500), and IL-15 was quantified by measuring the absorbance.

[0306]

[0307] As a result, as shown in Fig. 2l, when a vector (#22) containing CAR #13 and CD24-IL-15 genes was introduced into iPSCs and the amount of IL-15 secreted was measured after G418 selection, it was confirmed that it was secreted at a high level.

[0378] Cells were seeded in 96-well plates as shown in Figure 4c and cultured for 3 hours and 15 minutes. The cultured plates were then removed, 10 μl of lysis buffer was added, and the plates were cultured for another 45 minutes, followed by centrifugation at 300 g for 5 minutes. Fifty μl of the supernatant was then transferred to new 96-well plates, and 50 μl of substrate solution was added. The plates were then incubated for 30 minutes in a darkened environment, 50 μl of stop solution was added, and the spectrum was measured at 490 nm using a microplate reader.
