[0119] The following examples are offered by way of illustration and not by way of limitation.

[0120] EXPERIMENTAL

[0121] Example 1 - Beneficial effects of A. muciniphila on overall food cravings

[0122] In this example, subjects experiencing food cravings were administered an oral composition comprising a microbial population consisting of Akkermansia muciniphila.

[0123] The oral composition comprises between 1 x106and 1x1015CFLIs (Colony Forming Units) or AFUs (Active Fluorescent Units) of the Akkermansia muciniphila, and one or more doses of the oral composition are administered per day. In some embodiments, the composition comprises one million AFUs of Akkermansia muciniphila per pill. The delivery form of the oral composition is an enteric-coated (e.g., pH sensitive polymer) capsule or tablet that does not fully degrade until after it exits from the stomach of the subject. In some embodiments, the oral composition can be administered as a capsule comprising a powdered Akkermansia muciniphila composition.

[0124] For purposes of this example, the participating subjects were administered one capsule per day, wherein the capsule comprised a microbial population consisting of one hundred million AFLIs of Akkermansia muciniphila. Administration of the oral composition comprising Akkermansia muciniphila is scheduled for 3 months. The food cravings in participating subjects are studied using a validated questionnaire called the Food-Craving Inventory II (FCI-I I) (Licensed from the Pennington Biomedical Research Center; White et al. (2002) Obesity Research 10(2):107-114, the disclosure of which is incorporated herein by reference in its entirety for all purposes), which featured a 28-item food list and asked participants to indicate how often they experienced a craving for that food over the past month. (See the full FCI-II in the appendix I). The FCI-I I defined food craving as an intense desire to consume a particular food (or food type) that is difficult to resist.

[0125] Participants indicated the frequency of cravings over the past month for individual food items using a 5-point Likert scale (Never, Rarely, Sometimes, Often, or Always). Foods were grouped into four scales for purposes of this study into: sweets (with foods such as cinnamon rolls and ice cream), high fats (with foods such as bacon and fried chicken), carbohydrates / starches (with foods such as baked potatoes and pasta), and fast-food fats (with foods such as pizza, French fries, and hamburgers).

[0126] The change in participants’ FCI-II scores from baseline (when the participants started taking the oral composition comprising Akkermansia muciniphila) to follow-up (when 3 months were completed) was recorded and analyzed at the end of 3 months of oral administration of the composition.

[0127] The results show, as demonstrated in Fig. 1 , that administration of the composition comprising a microbial population consisting of Akkermansia muciniphila resulted in significant decrease in Food Craving Inventory scores, independent of age and gender (p < 0.001 ). Overall, 91 % of participants experienced reduced overall food cravings.

[0128] Methods for Example 1

[0129] Strain, growth medium and conditions

[0130] A. muciniphila (ATCC Accession No. BAA-835) was grown in a medium having the ingredients and concentrations listed in the following table.

[0131] Supernatant obtention and lyophilization

[0132] A. muciniphila supernatant was collected approximately 16 days post growth under manufacturing conditions in Fed Batch fermentation settings. The OD6oo nm at the time of sample collection was ~ 8 (late exponential to early stationary phase). The culture samples were collected and dispensed into sterile 50mL Eppendorf conical tubes and stored at -80°C. Prior to mass spectrometry analysis, the Akkermansia muciniphila cultures were thawed and filtered-sterilized by a 0.2 pM filter.

[0133] The lyophilization process includes a first cycle at -40°C (5 min at 500mTorr), followed by a primary dry step at 20°C (10 min at 500mTorr), followed by a second cycle at -40°C (999 min at 3000 mTorr).

[0134] Example 2 - Reduction in food cravings is proportional to score at baseline

[0135] In this example, subjects experiencing from food craving are divided into three groups based on their baseline craving scores - (a) Mild category - subjects with baseline score of 1 -2 and experiencing food cravings rarely; (b) Moderate category - subjects with baseline score 2-2.5 and experiencing food cravings sometimes; (c) Higher category - subjects with baseline score 2.5 - 5 and experiencing very high food cravings quite often. None of these categories were limited to subjects experiencing food craving to a particular group of subcategories of foods but was scored for overall food cravings in these subjects. These subjects were administered an oral composition comprising a microbial population consisting of Akkermansia muciniphila.

[0136] The subjects were given an oral composition comprising a microbial population consisting of Akkermansia muciniphila for 3 months at a dosage of one pill daily. Each pill comprised 100 million AFLIs of the Akkermansia muciniphila.

[0137] In one embodiment, the composition of the capsule comprises of Akkermansia muciniphila, inulin, hypermellose and magnesium stearate.

[0138] Data as demonstrated in Fig. 2, groups the results of these three categories based on the scores as shown on the top of each rectangular box. The box on the extreme left is for the mild category (score 1 -2); the middle box is for the moderate category (2- 2.5) and the right most box is for the higher category (score 2.5 - 5).

[0139] The results show that subjects with higher baseline cravings score experienced a greater reduction in their scores at the end of the 3-month study period compared to the other two groups.

[0140] Example 3 - Reduction of specific categories of food cravings by administration of a composition comprising a microbial population consisting of A. muciniphila

[0141] Method: Subjects were provided with a validated questionnaire called the Food- Craving Inventory II and asked participants to indicate how often they experienced a craving for any of the foods listed in the inventory. The foods were categorized as four main groups of food - sweets (with foods such as cinnamon rolls and ice cream), high fats (with foods such as bacon and fried chicken), carbo hydrates / starches (with foods such as baked potatoes and pasta), and fast-food fats (with foods such as pizza, French fries, and hamburgers). See the full FCI-II in appendix I.

[0142] Based on their input, scores were given to the subjects. This score is termed as the baseline score.

[0143] Next, the subjects were administered a composition comprising a microbial population consisting of Akkermansia muciniphila. The subjects were given an oral composition comprising a microbial population consisting of Akkermansia muciniphila for 3 months at a dosage of one pill daily. Each pill comprised a microbial population consisting of 100 million AFLIs of Akkermansia muciniphila.

[0144] At the end of 3 months, the same survey questionnaire was again provided to the participating subjects and scored based on their responses. The scores this time are called follow-up scores.

[0145] Results: A significant reduction of cravings was observed for each of the four specific food categories in the FCI-II survey. That is, food cravings were reduced in all four of the following categories: • Sweets

[0146] • Fast-Food Fats

[0147] • Carbohydrates and starches

[0148] • High Fats

[0149] Scores in the subcategories Fast Food Fats and Sweets showed a slightly greater reduction than scores in the other two subcategories. The results for each food subcategory carbohydrates and starch, fast-food fats, high fats, and sweets, are shown in Figure 3A-3D, respectively.

[0150] Example 4 - Reduction of overall / total and specific food cravings by administration of a composition comprising A. muciniphila, Clostridium butyricum and Bifidobacterium infantis

[0151] In this example, subjects experiencing food cravings were administered an enterically coated oral composition (capsule) comprising a microbial population consisting of Akkermansia muciniphila Clostridium butyricum, and Bifidobacterium infantis. In particular, the microbial population consisted of 6.4 x 10A8 AFLIs of Akkermansia muciniphila, 1.9 x 10A9 AFLIs of Bifidobacterium infantis, and 4.3 x 10A8 AFLIs of Clostridium butyricum per dose. The composition was administered daily for six weeks.

[0152] 274 novel participants completed baseline & six week surveys. Shown in FIG. 4A is mean-change (+ / - 28SE) in food cravings from baseline to six weeks upon administration of the composition comprising a microbial population consisting of Akkermansia muciniphila, Clostridium butyricum, and Bifidobacterium infantis. Shown in FIG. 4B is reported pills per week over the past month.

[0153] The food cravings in participating subjects were studied using the validated questionnaire called the Food-Craving Inventory II (FCI-II) (Licensed from the Pennington Biomedical Research Center; White et al. (2002) Obesity Research 10(2):107-1 14), which featured a 28-item food list and asked participants to indicate how often they experienced a craving for that food over the past month, as used in Example 1 above. The FCI-II defined food craving as an intense desire to consume a particular food (or food type) that is difficult to resist.

[0154] The 274 participants indicated the frequency of food cravings over the past 1 .5 months using a 5-point Likert scale (Never (1 ), Rarely (2), Sometimes (3), Often (4), or Always(5)). As shown in FIG. 5A, total food cravings were significantly reduced (p < 0.001 ) after six weeks of daily administration of the composition. Of the 274 participants, 91% experienced reduced overall / total food cravings.

[0155] The 274 participants were divided into groups based on their baseline craving scores - (a) Mild category - subjects with baseline score of 1 -2 and experiencing food cravings rarely; (b) Moderate category - subjects with baseline score 2-2.5 and experiencing food cravings sometimes; (c) Higher category - subjects with baseline score 2.5 - 3; and (d) Highest category - subjects with baseline score 3-5 and experiencing very high food cravings quite often. None of these categories were limited to subjects experiencing food craving to a particular group of subcategories of foods but was scored for overall food cravings. As shown in FIG. 5B, participants with higher total food cravings at baseline experienced a greater reduction of cravings at the six week timepoint, i.e., the reduction in food cravings was proportional to the baseline score.

[0156] The 274 participants indicated the frequency of cravings over the past 1 .5 months for individual food items using the 5-point Likert scale. Foods were grouped into four types / categories for purposes of this study: sweets (with foods such as cinnamon rolls and ice cream), high fats (with foods such as bacon and fried chicken), carbohydrates / starches (with foods such as baked potatoes and pasta), and fast-food fats (with foods such as pizza, French fries, and hamburgers). As shown in FIG. 5C, participants experienced significant reductions (p < 0.001 ) in food cravings across all four categories tested: 88% reduction in cravings for sweets; 87% reduction in cravings for carbs / starches; 85% reduction in cravings for fast food fats; and 82% reduction in cravings for high fat food. Here as well, and as shown in FIG. 5D, higher total cravings at baseline experienced greater reduction of cravings at six weeks. Shown in FIG. 6 is binning by baseline total cravings value and % improvements.

[0157] As shown in FIG. 7, subdomain food cravings were reduced after six weeks of administration of the composition, where subjects experiencing higher subdomain cravings at baseline exhibited greater reduction of subdomain cravings at six weeks. Binning by subdomain baseline values, p values and % improvements is shown in FIG. 8.

[0158] Example 5 - Reduction of overall / total and specific food cravings by administration of a composition comprising A. muciniphila, Clostridium butyricum and Bifidobacterium infantis for 12 weeks

[0159] In this example, as in the previous Example 4, subjects experiencing food cravings were administered an enterically coated oral composition (capsule) comprising a microbial population consisting of Akkermansia muciniphila, Clostridium butyricum, and Bifidobacterium infantis. In particular, the microbial population consisted of 6.4 x 10A8 AFLIs of Akkermansia muciniphila, 1 .9 x 10A9 AFLIs of Bifidobacterium infantis, and 4.3 x 10A8 AFLIs of Clostridium butyricum per dose. The composition was administered daily for twelve weeks.

[0160] 310 novel participants completed baseline & twelve-week surveys. Shown in FIG. 9A is mean-change (+ / - 2*SE) in food cravings from baseline to twelve weeks upon administration of the composition comprising a microbial population consisting of Akkermansia muciniphila, Clostridium butyricum, and Bifidobacterium infantis. Shown in FIG. 9B is reported pills per week over the past month for all 310 participants.

[0161] The food cravings in participating subjects were studied using the validated questionnaire called the Food-Craving Inventory II (FCI-II) (Licensed from the Pennington Biomedical Research Center; White et al.

[0162] The 310 participants indicated the frequency of food cravings over the past 3 months using a 5-point Likert scale (Never (1 ), Rarely (2), Sometimes (3), Often (4), or Always (5)). As shown in FIG. 10A, total food cravings were significantly reduced (p < 0.001 ) after twelve weeks of daily administration of the composition. Of the 310 participants, 93% experienced reduced overall / total food cravings.

[0163] The 310 participants were divided into groups based on their baseline craving scores - (a) Mild category - subjects with baseline score of 1 -2 and experiencing food cravings rarely; (b) Moderate category - subjects with baseline score 2-2.5 and experiencing food cravings sometimes; (c) Higher category - subjects with baseline score 2.5 - 3; and (d) Highest category - subjects with baseline score 3-5 and experiencing very high food cravings quite often. None of these categories were limited to subjects experiencing food craving to a particular group of subcategories of foods but was scored for overall food cravings. As shown in FIG. 10B, participants with higher total food cravings at baseline experienced a greater reduction of cravings at the twelve-week timepoint, i.e., the reduction in food cravings was proportional to the baseline score.

[0164] The 310 participants indicated the frequency of cravings over the past 3 months for individual food items using the 5-point Likert scale. 10C, participants experienced significant reductions (p < 0.001 ) in food cravings across all four categories tested: 88% reduction in cravings for sweets; 88% reduction in cravings for carbs / starches; 87% reduction in cravings for fast food fats; and 80% reduction in cravings for high fat food. Here as well, and as shown in FIG. 10D, higher total cravings at baseline experienced greater reduction of cravings at six weeks. Example 6 - Reduction of overall / total and specific food cravings by administration of a composition comprising A. muciniphila, Clostridium butyricum and Bifidobacterium infantis for 6 weeks and 12 weeks

[0165] In this example, as in the previous Examples 4 and 5, subjects experiencing food cravings were administered an enterically coated oral composition (capsule) comprising a microbial population consisting of Akkermansia muciniphila, Clostridium butyricum, and Bifidobacterium infantis. In particular, the microbial population consisted of 6.4 x 10A8 AFUs of Akkermansia muciniphila, 1.9 x 10A9 AFUs of Bifidobacterium infantis, and 4.3 x 10A8 AFUs of Clostridium butyricum per dose. The composition was administered daily for six weeks and then continued upto twelve weeks.

[0166] 222 novel participants completed baseline, six-week & twelve-week surveys. Shown in FIG. 11 A is mean-change (+ / - 2*SE) in food cravings from baseline to six weeks and twelve weeks, respectively, upon administration of the composition comprising a microbial population consisting of Akkermansia muciniphila, Clostridium butyricum, and Bifidobacterium infantis. Shown in FIG. 1 1 B is reported pills per week over the past month for all 222 participants.

[0167] The food cravings in participating subjects were studied using the validated questionnaire called the Food-Craving Inventory II (FCI-II) (Licensed from the Pennington Biomedical Research Center; White et al.

[0168] The 222 participants indicated the frequency of food cravings over the past 1 .5 months and 3 months using a 5-point Likert scale (Never (1 ), Rarely (2), Sometimes (3), Often (4), or Always (5)). As shown in FIG. 12A, total food cravings were significantly reduced (p < 0.001 ) after twelve weeks of daily administration of the composition. Of the 222 participants, 90% experienced reduced overall / total food cravings at 6 weeks while 93% experienced reduced overall / total food cravings at 12 weeks.

[0169] The 222 participants were divided into groups based on their baseline craving scores - (a) Mild category - subjects with baseline score of 1 -2 and experiencing food cravings rarely; (b) Moderate category - subjects with baseline score 2-2.5 and experiencing food cravings sometimes; (c) Higher category - subjects with baseline score 2.5 - 3; and (d) Highest category - subjects with baseline score 3-5 and experiencing very high food cravings quite often. None of these categories were limited to subjects experiencing food craving to a particular group of subcategories of foods but was scored for overall food cravings. As shown in FIG. 12B, participants with higher total food cravings at baseline experienced a greater reduction of cravings at the twelve-week timepoint, i.e., the reduction in food cravings was proportional to the baseline score.

[0170] The 222 participants indicated the frequency of cravings over the past, 1 .5 months continued upto 3 months, for individual food items using the 5-point Likert scale. 12C, participants experienced significant reductions (p < 0.001 ) in food cravings across all four categories tested: all 222 participants showed sustained improvements by showing reduced food cravings over the 6 week and 12 weeks periods across all four food categories, as shown. Further, it was also observed as in the Examples 4 and 5 above, participants with higher total cravings at baseline experienced greater reduction of cravings at six weeks and consecutively at twelve weeks.

[0171] Methods for Examples 4, 5, and 6

[0172] Akkermansia muciniphila (ATCC Accession No. BAA-835) was prepared as described for Example 1 .

[0173] For preparation of Clostridium butyricum (CBUT), the fermentation process begins with the resuspension of CBUT powder seed stock in media, followed by streak plating onto agar plates. After 24 hours of incubation at 37°C, colonies are inoculated into tubes of media. Incubation at 37°C ensues, during which the optical density (OD) is monitored. When one of the tubes reaches an OD between 0.7-1.0, the culture is aseptically transferred to a bottle containing sterile media. The growth is monitored until OD is between 0.7 and 1.0. The entire culture is aseptically transferred to a bioreactor and further incubated at 37°C. Growth is monitored, and the harvest centrifuge process involves transferring the microbial culture from the fermentor to a centrifuge. Homogenous biomass accumulates within the centrifuge, cryopreservative is added, and is discharged into a collection vessel. Discharge is collected, weighed, and placed in liquid nitrogen, then stored until lyophilization.

[0174] Bifidobacterium infantis was obtained from Dupont Danisco as Bifidobacterium longum subsp. infantis Bi-26 (DGCC1 1473).
