Description

Composition for preventing hair loss or promoting hair growth containing elm extract

[0001] The present invention relates to a composition for preventing hair loss or promoting hair growth comprising an extract of the Korean elm (Ulmus parvifolia), and more specifically, to a pharmaceutical, cosmetic or functional food composition having a hair loss prevention and hair growth promotion effect comprising, as an active ingredient, an extract of the bark of the Korean elm, which has a proliferation effect on hair follicle dermal papilla cells.

[0002] The human scalp has about 80,000 to 90,000 hairs, but it is normal for the average person to lose 50 to 70 hairs a day. However, if you lose more than 100 hairs without any particular reason after washing your hair or going to bed, you may have alopecia. The exact cause of this hair loss has not been identified yet, but it is generally known to be due to genetic factors, hormonal imbalance, excessive stress, seborrheic dermatitis, poor blood circulation in the scalp (tension of scalp muscles), eating habits, nutritional and iron deficiencies, hair pulling (traction alopecia), atopic constitution, hair pollution (frequent perms, dyeing, use of hair dryers, etc.), air pollution, infectious and inflammatory diseases, chemical factors (alcohol, drugs, smoking), childbirth, and menopausal disorders.

[0003] In the human body, hair grows from hair follicles in the skin, and the hair follicle, from which hair grows, is a complex organ composed of the inner root epithelial sheath, the outer root epithelial sheath, the hair shaft, and the hair matrix, and is characterized by a cycle of growth, catagen, and telogen that continuously repeats throughout life. In particular, dermal papilla cells, among the cells that form the hair follicle, play an important role in hair growth, and during the anagen phase of the hair follicle, proliferation of cells constituting the hair follicle, including keratinocytes in the matrix surrounding the dermal papilla, occurs. During the catagen phase of the hair follicle, hair follicle growth stops, and apoptosis, condensation of the dermal papilla, and restructuring of the extracellular matrix occur. During the telogen phase, which is the stage in which the hair follicle prepares to enter the anagen phase again, only the permanently lasting part, including the raised area, remains.

[0004] Hair is composed of the cuticle, cortex, and medulla. The cortex accounts for over 80% of the hair, significantly affecting its characteristics. The cortex is divided into crystalline and amorphous regions, and is a fibrous layer composed of fibrous keratin proteins. Its physicochemical properties are determined by how the keratin molecules are bonded and the amount of sulfur (S) it contains.

[0005] Hair loss refers to an abnormal increase in hair loss due to a higher proportion of hair growing from hair follicles in the catagen or resting phase than from hair growing from hair follicles in the growth phase. It is generally known to be caused by unbalanced diet, excessive drinking and smoking, drug use, lack of sleep, excessive stress, childbirth, menopausal disorders, frequent perms and dyeing, and hormonal imbalances. Hair growing on a person's head maintains a certain density through the repeated cycle of growth and fall out, but when hair loss progresses due to the various causes mentioned above, the hair on the crown or forehead first begins to thin and lose strength, and the number of hair loss increases.

[0006] In other words, hair is rooted in the scalp and receives nutrients from the blood. However, if the capillaries shrink over a long period of time and the scalp becomes thinner, reaching a state where hair cannot grow normally, the hair in that area will gradually become thinner, shortening the lifespan of the hair in that specific area. Hair with a shortened lifespan will not only fall out easily, but hair will not grow back in the pores where hair has fallen out as described above, ultimately leading to symptoms of thinning hair.

[0007] Natural products that have been studied to date to be useful for hair growth include soybeans (Glycine linne, isoflavone), ginger (Zingiber officinale, 6-gingerol), red pepper (Capsicum annuum), aloe (Aloe arborescens), licorice (Glycyrrhiza glabra), tea tree (Camellia sinensis), ginkgo tree (Ginkgo biloba), avocado (Persea americana), and Polygonum multiflorum.

[0008] Additionally, research based on natural extracts has also been conducted. For example, there are results from extracts of Cnidium officinalis (Patent No. 2010-0119761), Phellodendron amurense (Patent No. 2012-0038426), Lonicera japonica (Patent No. 2012-0038426), Chamomile (Patent No. 2012-0015874, Patent No. 2010-0091571), Polygonum multiflorum (Patent No. 2013-0021464, Patent No. 2011-0097386), Atractylodes japonica (Patent No. 2012-0115980), Pomegranate (Patent No. 2012-7012465), and Ginkgo biloba leaf extract (Patent No. 2006-7008793). However, when these hair growth agents or hair tonics are actually used in clinical practice, their effects on the scalp are not that outstanding, and research is actively being conducted to find hair growth agents made of natural substances that are more effective and have fewer side effects than minoxidil or propecia, which are often avoided due to concerns about side effects.

[0009] However, despite efforts to utilize such natural products, commercialization of hair growth promoters or hair loss inhibitors is still minimal due to their low activity compared to pharmaceuticals.

[0010] Accordingly, the inventors of the present invention have conducted continuous research to develop a composition for preventing hair loss or promoting hair growth that can proliferate human dermal papilla cells (hDPCs), and as a result, they have discovered that an extract of the bark of the common elm tree has a proliferative effect on human dermal papilla cells, thereby completing the present invention.

[0011] The technical problem to be solved in the present invention is to provide a composition for preventing hair loss or promoting hair growth that has hair loss prevention and hair growth effects by proliferating hair follicle papilla cells.

[0012] In order to solve the above-mentioned technical problem, the present invention provides a pharmaceutical composition for preventing hair loss or promoting hair growth, characterized in that it contains an extract of elm tree as an active ingredient.

[0013] Preferably, the elm extract is characterized in that it is prepared by drying the elm and then extracting it at 20 to 60°C for 1 to 5 hours using water, an organic solvent, or a mixture thereof as an extraction solvent.

[0014] Preferably, the elm extract is characterized in that it is an ethanol extract of elm bark.

[0015] Preferably, the composition is characterized as being a composition for proliferation of hair follicle hair papilla cells.

[0016] The present invention provides a cosmetic composition for preventing hair loss or promoting hair growth, characterized in that it contains an extract of elm as an active ingredient.

[0017] Preferably, the cosmetic composition is characterized in that it is a hair care composition selected from the group consisting of hair conditioner, hair tonic, hair cream, hair lotion, hair face, hair gel, hair pack, hair massage, hair liquid, hair spray, hair mousse, treatment, non-aerosol mousse, non-aerosol spray, aerosol mousse, aerosol spray, perm, bleaching agent, dye, ampoule, shampoo, and rinse.

[0018] In addition, the present invention provides a functional food composition for preventing hair loss or promoting hair growth, characterized in that it contains an extract of elm as an active ingredient.

[0019] Thus, the elm extract of the present invention has hair loss prevention and hair growth effects by proliferating hair follicle papilla cells. The elm extract of the present invention is safe for the human body as a natural food raw material, and thus can be usefully utilized in pharmaceutical compositions, cosmetic compositions, and functional food compositions.

[0020] The following drawings attached to this specification illustrate preferred embodiments of the present invention and, together with the contents of the invention described above, serve to further understand the technical idea of the present invention, and therefore, the present invention should not be interpreted as being limited to matters described in such drawings.

[0021] Figure 1 shows the proliferation rate of mammary papilla cells according to the concentration of elm.

[0022] The present invention is described in more detail below.

[0023] The present invention provides a composition for preventing hair loss or promoting hair growth, characterized in that it contains an extract of the true elm (Ulmus parvifolia Jacq.) as an active ingredient.

[0024] The elm tree used in the present invention grows in Korea, Japan, China, and other countries. It is a large tree of the Ulmaceae family, growing 10 to 15 m tall. Among the elm species, the elm tree is the only one that blooms and bears fruit from late summer to early fall. The elm tree is known to have antioxidant and anti-inflammatory effects.

[0025] According to one embodiment of the present invention, the elm extract can be extracted using at least one of the leaves, stems, barks, fruits, and roots of the elm, and is preferably extracted using the bark of the elm.

[0026] The term "extract" of the present invention means a preparation obtained by squeezing raw materials with an appropriate extractant and evaporating the extractant to concentrate them, and is not limited thereto, but may be an extract obtained by extraction treatment, a diluted or concentrated extract, a dried product obtained by drying the extract, a controlled product, or a purified product thereof.

[0027] The extract used in the present invention can be extracted using each conventional extraction solvent known in the art, and the extracted liquid can be used in liquid form or after being concentrated and / or dried. At this time, the extraction solvent can be obtained using, for example, (a) water, (b) anhydrous or hydrous lower alcohol having 1 to 4 carbon atoms (methanol, ethanol, propanol, butanol, etc.), (c) a mixed solvent of the lower alcohol and water, (d) acetone, (e) ethyl acetate, (f) chloroform, or (g) 1,3-butylene glycol as an extraction solvent. Preferably, extraction is performed using methanol, ethanol, or butane. Since the degree of extraction and loss of the active ingredient of the extract may differ depending on the extraction solvent, an appropriate extraction solvent should be selected and used. The above extraction method is not particularly limited, and includes, for example, hot water extraction, cold immersion extraction, reflux cooling extraction, solvent extraction, steam distillation, ultrasonic extraction, dissolution, and pressing.

[0028] In addition, the extract can be obtained through a conventional purification process in addition to the extraction method using the extraction solvent. For example, the extract can be obtained through fractions obtained through various additional purification methods, such as separation using an ultrafiltration membrane having a certain molecular weight cut-off value, separation by various chromatographies (designed for separation according to size, charge, hydrophobicity, or affinity).

[0029] According to an embodiment of the present invention, a natural plant extract may be obtained by using an extraction method such as stirring extraction, hot water extraction, cold immersion extraction, reflux cooling extraction or ultrasonic extraction, using water having 1 to 5 carbon atoms including methanol, ethanol, propanol, isopropanol, butanol, or a mixed solvent of water and alcohol, in an amount of about 1 to 10 times, preferably about 2 to 5 times, the weight of the elm sample, and then performing hot water extraction at preferably 50 to 100°C for about 10 to 20 hours, and then filtering, concentrating under reduced pressure or drying the obtained extract.

[0030] Specifically, the above elm extract is characterized in that it is prepared by drying elm and then extracting it at 20 to 60°C for 1 to 5 hours using water, an organic solvent, or a mixture thereof as an extraction solvent.

[0031] According to one embodiment of the present invention, the elm extract is characterized in that it is included in an amount of 0.5 to 10 wt% based on the total weight of the composition for preventing hair loss or promoting hair growth. In this case, if the elm extract is included in an amount of less than 0.5 wt%, it is difficult to obtain the effect of preventing hair loss or promoting hair growth, and if the content of the elm extract exceeds 10.0 wt%, there is a concern that excessive nutrients will be supplied to the scalp, which may instead lead to the generation of scalp waste and dead skin cells.

[0075] <Comparative Example 3> Preparation of extract of the pine tree

[0076] Ulmus pumila extract was obtained by ultrasonically extracting the bark of Ulmus pumila using 70% ethanol at 50°C for 2 hours.

[0077] <Test Example 1> Proliferation efficacy of dermal papilla cells

[0078] Dermal papilla cells are mesodermal fibroblasts located at the base of the hair follicle, and are known to play an important role in hair growth through interaction with the hair matrix cells of the hair follicle (Jahoda et al., 1984; Oliver et al., 1986). The hair growth effects of extracts of the genus Ulmus, including the true elm, the elm, the king elm, and the burdock, were investigated using immortalized dermal papilla cells.

[0079] Immortalized dermal papilla cells isolated from rat whiskers (Rat vibrissa immortalized dermal papilla cells: Filsell et al., 1994) were cultured in DMEM (Hyclone Inc., USA) containing 100 units / mL penicillin-100 μg / mL streptomycin (Gibco Inc., NY, USA) and 10% heat-inactivated fetal bovine serum (FBS; Gibco Inc., NY, USA) at 37°C in a 5% CO2 incubator and subcultured every 3 days. The proliferation of dermal papilla cells was measured using the MTT assay. Dermal papilla cells (1.0 × 10 4 cells / mL) were placed in a 96-well plate in DMEM medium containing 1% FBS, and cultured for 24 hours. After treatment with concentrations of 0.01, 0.05, 0.1, and 1 μg / mL, the samples were treated. As a positive control, minoxidil (Sigma, MO, USA) was dissolved in 0.12 mM HCl and treated at a concentration of 10 μM, and after 72 hours of culture, 50 μL of MTT dye (Sigma, MO, USA) was added and reacted for 4 hours. The supernatant was removed, and 200 μL of DMSO was added to dissolve the precipitate, and the absorbance was measured at 540 nm using a VERSAmax microplate reader (Molecular Devices, CA, USA). The average absorbance value for each sample group was obtained, and the degree of proliferation was investigated by comparing it with the absorbance value of the control group.

[0080] All measurement results are expressed as mean ± standard deviation, and statistical significance was tested using student's t test, and significance was accepted if the p-value is less than 0.05. Statistical processing was performed using Sigma Stat software (Jandel Scientific Software, USA). The results show that the proliferation rate of hair papilla cells according to the concentration of the elm tree is shown in Table 1 below.

[0081] Figure 1 shows the proliferation rate of mammary papilla cells according to the concentration of elm.

[0082]

[0083] As shown in Table 1 and Figure 1, when the elm bark extract of the present invention was treated at concentrations of 0.01, 0.05, 0.1, and 1 μg / mL, the proliferation rates were 102.9±4.8%, 97.9±5.2%, 112.3±7.4% (p<0.01), and 97.7±8.3%, respectively. On the other hand, when the elm bark extract of Comparative Example 1 was treated at concentrations of 0.01, 0.05, 0.1, and 1 μg / mL, the proliferation rates were 94.0±10.1%, 92.7±10.7%, 92.5±11.0%, and 85.3±9.5%, respectively, showing a lower effect than the elm bark extract. The extract of the bark of the King Elm tree in Comparative Example 2 was low at 98.3±6.3%, 95.9±9.1%, 99.3±5.0, and 88.1±7.2% when treated at concentrations of 0.01, 0.05, 0.1, and 1μg / mL, and the extract of the bark of the Bisul tree in Comparative Example 3 was low at 94.9±11.1%, 100.5±8.4%, 101.9±9.7%, and 95.0±9.6% when treated at concentrations of 0.01, 0.05, 0.1, and 1μg / mL. In particular, the extract of the bark of the elm tree significantly increased the growth of hair papilla cells at a concentration of 0.1 μg / mL compared to the solvent-treated control group (100.0±7.0%), and showed a similar effect on hair papilla cell proliferation as the positive control group minoxidil (10 μM) (112.7±8.7% (p<0.001)%). While the specific parts of the present invention have been described in detail above, it is obvious to those skilled in the art that such specific descriptions are merely preferred embodiments and that the scope of the present invention is not limited thereto. Accordingly, the actual scope of the present invention will be defined by the appended claims and their equivalents.
